  • The effects of some soil conditions on the degradation rate and decomposing pattern of parathion were investigated and the obtained results are summarized as follows: Parathion degraded more rapidly in flooded soils than in non-flooded, in wet soils than in dry soils under non-flooded soils. The degradation rates in paddy and upland soils increased at high temperature than low temperature, higher pesticide concentration than low concentration and higher soil pH level. Parathion in paddy and upland soils was more persistent under soil sterilization than under non-sterilization and degraded rapidly in glucose application. Parathion was more persistent in upland soils than paddy soils under several factors described above. The metabolites identified from the paddy and upland soils by TLC include para-oxon (Rf 0.5), aminoparathion(Rf 0.27), p-nitrophenol(Rf 0.2), p-aminophenol(Rf 0.15). Soil enzyme, acid phosphatase activities decreased more at flooded soils than non-flooded, higher pesticide concentration than low concentration and higher soil pH level and the activity in glucose application was increased. Soil enzymes, urease and dehydrogenase activity decreased more at higher pesticide concentration than low concentration. Comparing with soil enzyme activity in paddy and upland soil, the former was higher than the latter.

Comparison of Soil Physico-chemical and Microbial Characteristics in Soil of 'Niitaka' Pear Orchards between Organic and Conventional Cultivations (유기농법과 관행농법에 의해 재배한 '신고'배 과원 토양의 물리화학적 및 미생물학적 특성 비교)

  • Consumers' interest and government's support for the fruits rapidly increased organic fruit productions. This study was examined to compare the soil physicochemical and microbial properties of orchards soil in conventionally and organically management systems. Organic cultivation had lower soil bulk density, solid phase, and penetration resistance than the conventional cultivation. Soil pH and organic matter contents increased from March to August, and the values were greater in the organic cultivation than the conventional cultivation. Total nitrogen (N) and phosphorous concentrations decreased from March to August, and the organic soils had greater N but lower phosphorous concentrations than the conventional soils. Soil microbial carbon biomass increased 36% and 15% for organic and conventional cultivations, respectively, from March to August. Soil microbial N biomass was greater in June than March or August, and the organic cultivation had a greater biomass N compared to the conventional cultivation. Soil dehydrogenase and chitinase activities were greater in June than in March or August. ${\beta}$-glucosidase activity declined in both cultivations, while the phosphatase activity increased. Organic cultivation had greater enzyme activities in March, June, and August, except for the acid phosphatase activity in June.

Expression and Characterization of Fibrinolytic Enzyme Activity During Earthworm Tail Regeneration (지렁이 꼬리 재생시 발현되는 피브리노겐 분해효소의 활성과 특성에 관한 연구)

  • Fibrinolytic enzyme is thought to be involved in extracellular matrix remodeling during regeneration. We investigated the expression and characterization fibrinolytic enzyme activity during earthworm tail regeneration. Electrophoretic analysis of fibrinolytic enzymes induced during regeneration revealed that at least seven types of fibrinolytic enzymes were expressed, which had molecular weight of 12, 19, 23, 27, 32, 45 and 58 kDa, respectively. These fibrinolytic enzyme activities were dramatically increased within 1 day after amputation. These activities were maintained by 7 days postamputation, followed by decrease to control level from 14 days after amputation. Alltypes of fibrinolytic enzyme activities were inhibited by treatment of PMSF and aprotinin, and were insensitive to EDTA and exogenous Ca$^{2+}$. These results indicate that the fibrinolytic enzymes are serineproteinase. Other characteristics including specificities for extracellular matrix proteins are under investigation. Based on these results, we are trying to find out the relationship among expression of proteinases, extracellular matrix remodeling, and dedifferentiation, which are believed to be essential processes during regeneration.

  • This study was conducted to find out the effect of herbicides on soil micro-organism and soil enzyme in loam soil, and on pathogenic microorganism in continuous pepper cropping soil. The result was summarized as follows: When herbicides were treated, the number of soil microorganism generally decreased at the early stage of incubation, and gradually increased at 30 days incubation. The number of fungi was significantly decreased seven times comparison with control plot. The number of actinomycetes was increased at 20 days incubation and that of pytium was increased for all days incubation in Linuron treatment plot. Although the activity of soil enzyme tended to decrease when herbicides were treated, the activities of urease and phosphatase were higher than that of control plot at the early stage of incubation in MO, Linuron and Simetryne treatment plot. In Simetryne treatment plot, the activity of protease was increased at the early stage, and suddenly decreased after 30 days incubation. There was no significant effect of herbicides on the activities of ${\beta}$-glucosidase and polygalacturonase. The activity of cellulase was inhibited at the early stage of incubation, but that of cellulase was higher than that of control plot after 20 days incubation. High significance was showed the correlation coefficient between soil microorganisms except fungi and soil respiration as herbicides were treated, and the total microorganism and soil respiration in Linuron plot. when Dicamba and Simetryne were treated, the correlation coefficient between the total microorganism and the activities of protease and urease were considerably significant.

  • Effects of different amounts of fertilizer and manure application on changes of chemical and biological properties of soil were studied in plastic film house. Application amount of fertilizer was determined on the basis of soil test, standard application rate, and conventional rate of farmers. Lettuce in the first and second seasons and spinach in the third season were cultivated. Crop yields were highest in the plot fertilized on the basis of soil test during the three crop seasons. In the third crop season, spinach yield was lower in conventional plot applied with larger amount of pig manure compost than any other treatment. Organic matter, available phosphorus, and exchangeable potassium were accumulated in soil of the conventional plot with successive cropping. There was no difference in the abundance of soil invertebrates among the treatments, but soil enzyme activity was highest in the conventional plot.

Production of Mannooligosaccharides by the Penicillium purpurogenum Mannanase (Penicillium purpurogenum 유래의 Mannanase에 의한 Mannooligosaccharide의 생산)

  • Penicillium purpurogenum , which produces a copra galactomannan degrading enzyme extracellularyl, was isolated from soil , and its properties and formation condition of mannooligosaccharides were investigated. The optimum ph and temperature for the activity of the mannanase were 5.5 and 55$^{\circ}C$, respectively. The mannanase was stable in between pH 3.5 and 7.0 after 2 hr incubation at 3$0^{\circ}C$ lost 90% of the original activity after incubation at 55$\AA$ and pH 5.5 for 2 hr. With two different substrate concentration, hydrolysis of white coprameal proceeded rapidly at the early stage of the reaction, but gradually solwed thereafter especially at a higher concentration of copra meal (20 %). The enzyme hydrolyzed white copra meal to monosaccharides, mannobiose and mannotriose at the final stage of the reaction.

Isolation and Identification of Dextranase Production Strains and Enzyme Production (Dextranase 생산균주의 분리, 동정 및 효소생산)

  • In order to screen dextranase with high dextranolytic activity from microbial origins, dextranase producing fungal isolates were isolated from soil of the Taeion area. 197 strains with dextranolytic activities were isolated, out of which 3 strains with high dextranolytic activities were selected in the first screening. A strain (GR-98) with a best dextranolytic activity was selected in the second screening. The strain was identified to be similiar Aspergillus ustus by the morphological and cultural characteristics. The optimum culture temperature and initial pH for the dextranase production of the strain was 30$\circ$C and 7.0, respectively. The optimum culture medium was composed of 2% dextran, 0.3% KNO$_{3}$, 0.05% K$_{2}$HPO$_{4}$, 0.02% MgSO$_{4}$-7H$_{2}$O, 0.05% KC1, and 2.5 $\mu$g/ml pyridoxamine, and the enzyme production was maximum when the strain was subcultured at 30$\circ$C for 7 days.

Studies on acid protease produced from Aspergillus tubingensis I (Aspergillus tubingensis의 acid protease에 관한 연구 I)

  • The strain of Aspergillus, 6368A, producing acid protease showing high activity was isolated from soil, as a result of wide research about mold group. This strain was identified as a species of Aspergillus tubingensis by the investigation of morphological characteristics. The change of the enzyme production under the various media and culture condition was also studied. The optimum pH and stability of crude acid protease are 2.5, 2.0~4.5 and the optimum temeprature and thermal inactivation waas shown $50^{\circ}C,\;55^{\circ}C$, respectively. From the result of the study on the effects of metal ions, it was found that $MnCl_2,\;CoCl_2,\;CuCl_2,\;SrCl_2,\;and\;NiCl_2$ slightly increased the enzyme activity, on the other hand $ZnCl_2,\;CaCl_2,\;MgCl_2,\;SLS,\;and\;KMnO_4$ decreased it.

  • The inulinase producing microorganism was isolated from soil and tentatively identified as Arthrobacter protophormiae/ramosus. Inulinase was pruified by ethanol precipitation, DEAE-Sephadex ion exchange chromatography and Sephadex gel filtration chromatography. The molecular weight of the purified enzyme was 34 kDa. The specific activity, yield and purity were 31.5 Unit/mg, 19.5% and 18.5 fold, respectively. Optimal pH and temperature for reaction of the purified inulinase were 8.5 and $55^{\circ}C$, respectively. The enzyme was stable at pH 7.5, below$ 55^{\circ}C$, and the activity was stimulated Mg2+.

  • Purification and characterization of pectate lyase from Bacillus sp HSA-925. Bacillus sp. HSA-925 isolated from soil produced constitutively an extracellular pectate lyase when cultivated in LB broth. The pectate lyase(EC 4.2.2.2) was purified from the cuylture broth by preciptation with ammonium sulfate, followed by column chromatography on CM-cellulose C-50 and repeated gel filtration on Sephadex G-75G. The enzyme had a molecular weight of 32-33 kDa. The activity was mazimum at pH 9.5 AND 45$\CIRC $C. The enzume activity was stable at 55$\circ $C for 15 min and between pH7-12. The activation energy, Km and V$_{max}$ for the pectate lyase were 5.8779 kcal/mol, 6.33$\times $10$^{-2}$ mol/ml and 2.09$\times $10$^{2}$ $\mu $mol/min respectively. The enzyme was activated by Ca$^{2+}$, Cu$^{2+}$ and inhibited by Li$^{+}$, Hg$^{2+}$, EDTA.
