The Effect of Salt and Organic Solvents on the Interaction of Thionin-Sodium Dodecyl Sulfate System (Thionin-Sodium Dodecyl Sulfate계의 상호작용에 있어서 염 및 유기용매의 첨가효과에 관한 연구)

  • The interaction between the cationic dye, thionin(Th) and the anionic surfactant, sodium dodecyl sulfate (SDS) has been investigated by absorption spectra. As the temperature of surfactant solution was increased in premicellar range(S/D=10, 80, and 160) which was much lower than the critical micelle concentration(CMC), the increment or decrement of the molar extinction coeffecient ratio appeared. It was found that the most stable temperature range of the oligomer aggregate in Th-SDS system at S/D=160 was below $60^{\circ}C$. With increasing the concentration of inorganic salt and organic solvents in Th-SDS system, ${\alpha}$-band was increased, but ${\gamma}$=band or J-band was decreased. The orders of ${\alpha}$-band increasing power were $Cl^-$>$ClO{_4}{^-}$>$SO{_4}{^{2-}}$>$NO{_3}{^-}$ and 2-propanol>ethanol>methanol>ethylene glycol.

Effects of Euphorbia Pekinensis RUPR. on Rats with Acute Renal Failure induced by Gentamicin Sulfate (대극전탕액(大戟煎湯液)이 Gentamicin sulfate로 유발(誘發)된 백서(白鼠)의 급성(急性) 신부전(腎不全)에 미치는 영향(影響))

  • This article is to investigate the effects of Euphorbia Pekinensis RUPR. on rats with acute renal failure induced by gentamicin sulfate. So this experimental research was focused on measuring (1)the levels of creatinine, blood urea nitrogen, glutamic oxaloacetic transaminase, glutamic pyruvic transaminase, total protein, sodium, potassium, chloride in the serum and (2)the levels of protein, blood, sodium, potassium, chloride, pH, specific gravity and volume in the urine and (3)intake water. The results were summarized as follows. 1. While the levels of creatinine and glutamic oxaloacetic transaminase, glutamic pyruvic transaminase in the serum were significantly increased, the level of BUN in the serum were partially increased and the levels of sodium and potassium in the serum were partially decreased on the Euphorbia Pekinensis $R_{UPR}$. group. 2. While the urine volume was significantly increased, the levels of protein, blood, sodium and chloride in the urine were partially increased and the specific gravity, pH in the urine were partially decreased. According to the above results, it is shown that Euphorbia Pekinensis $R_{UPR}$. without refinning had a diuretic effect on rats with acute renal failure induced by gentamicin sulfate, but aggravated the glomerular filtration rate and tubular reabsorption. So, it is supposed that Euphorbia Pekinensis $R_{UPR}$. without refinning needs to be used carefully on renal disease and futhermore research about the kinds of extracts form is necessary.

Determination of Sodium Chondroitin Sulfate by Enzymatic Digestion and HPLC (효소분해와 HPLC를 이용한 황산콘드로이틴나트륨의 정량)

  • In order to determine sodium chondroitin sulfate in the mixture, chondroitinase ABC was used for enzymatic reaction. The procedure was rapid, simple, quantitative and the HPLC analysis of ${\Delta}Di-6S$(2-actamido-2-deoxy-3-0-(${\beta}$-D-gluco-4-enepyranosyluronic acid)-6-0-sulfo-D-galactose) in the sodium chondroitin sulfate was obtained. The absorbance was measured at 230nm and detection limit was $1{\mu}g/ml$. When we applied this method to the drugs(capsule, opthalmic solution), it gave the mean contents of $100.01{\pm}1.58%$ and $99.89{\pm}1.80%$ respectively.

The Anti-oxidative and Anti-inflammatory Effect of Lonicera Japonica on Ulcerative Colitis Induced by Dextran Sulfate Sodium in Mice (금은화 추출물의 항산화, 항염증 효과가 Dextran Sulfate Sodium으로 유도된 생쥐의 궤양성 대장염에 미치는 영향)

  • Objectives : This study was to investigate the anti-oxidative and anti-inflammatory effect of Lonicera japonica water extracts (LE) on Ulcerative Colitis Induced by DSS (Dextran Sulfate Sodium) in Mice. Methods : Colitis was induced by DSS in Balb/c mice. The sample group was divided into three. The mice in control group were not inflammation-induced. The pathological group was composed of untreated colitis elicited mice. The experimental group was administered Lonicera japonica water extracts (LE) after colitis elicitation. The effects on ulcerative colitis were evaluated the anti-oxidant effect, inhibition of COX-2 mRNA expression, the morphological change of colonic mucosa, decrease effect of HSP 70 and COX-2 in mucosa. Results : The SOD ability of LE was dose-dependently increased and the LPS-induced COX-2 mRNA expression of LE was dose-dependently decreased. LE showed the protective effects on DSS-induced experimental colitis. LE inhibited shortening of colon length, the hemorrhagic erosion in colonic mucosa. LE also showed the decrease effect for HSP70 and COX-2 in mucosa. Conclusions : The current results demonstrate the clinical utility of LE in traditional medicine and indicate the possible treatments for ulcerative colitis from natural products. Further investigations for exact mechanisms will be needed.

Effect of Molecular Weight of Chitosan on Flocculation of Suspended Solids in Rhodamine 6G-Sodium Dodecyl Sulfate System (키토산 분자량에 따른 Rhodamine 6G-Sodium Dodecyl Sulfate계의 응집효과에 관한 연구)

  • Chitin was extracted from crab shell of Portuns triberculatus and deacethylated to yield chitosan with various molecular weights. The absorption and the fluorescence spectra of Rhodamine 6G(Rh 6G)-sodium dodecyl sulfate(SDS) and Rh 6G-chitosan systems were obtained. From the spectra, we observed that the absorption and the fluorescence intensity of Rh 6G-SDS system decreased when S/D(the concentration of SDS to that of Rh 6G ratio) was below or at 32, while they increased when S/D was above 32. From the suspended solid(SS) removal rate and the transmittance of Rh 6G-SDS-chitosan system, we found that when S/D ratio was 32 its flocculating behaviour was much stronger than Rh 6G-SDS system. As the concentration and the molecular weight of chitosan increased, we also found that S/D range was extended from 32 to 100. With increasing the molecular weight of chitosan, the SS removal rate increased around pH 2~9 but decreased remarkably at pH>12.

Effects of Auklandia Lappa on Dextran Sulfate Sodium-Induced Murine Colitis (당목향(唐木香)이 DSS(Dextran sulfate sodium)로 유발된 염증성 장질환 동물모델에 미치는 영향)

  • Objectives : Auklandia Lappa (ALE) is one of the herbs used frequently to treat abdominal pain and diarrhea and reported anti-inflammatory activity by suppressing proinflammatory cytokines. This study was designed to investigate whether ALE could show protective activities on experimental colitis induced by dextran sulfate sodium (DSS) models. Methods : Colitis was induced by DSS in Balb/c mice. ALE 10, 30, 100 and 300 mg/kg were orally administered twice a day for 7 days in DSS model. Mice weight was measured daily. Scoring of clinical findings was measured every other day. Colon length, edema, fecal blood and histological damages were assessed at day 7 in DSS model. In histological analysis, we checked cryptal glands, surface epithelium, submucosa, transmural, stroma and scored degree of inflammatory cell damage by modified histological scoring. We also calculated cytokines concentrations including IFN-${\gamma}$, TNF-${\alpha}$, IL-6, IL-$1{\beta}$, IL-17, IL-23, IL-10 and TGF-${\beta}1$ by Biometric Multiplex Cytokine Profiling method. Results : ALE showed the protective effects on DSS-induced experimental colitis. ALE inhibited shortening of colon length and histological damages of colon does-dependently, but it did not inhibit weight loss. ALE also inhibited IFN-${\gamma}$ and IL-6 expression, and upregulated cytokines (IL-10, TGF-${\beta}1$) related to regulatory T cell differentiation and proliferation. Conclusions : The current results demonstrate the clinical utility of ALE in traditional medicine and indicate the possibility of potent drug development of inflammatory bowel diseases from natural products. Further investigations for exact mechanisms will be needed.

The Effects of Pulsatilla Koreana NAKAI Pharmacopuncture Therapy on the Chunchu (ST25) for Dextran Sulfate Sodium-Induced Colitis in Mice

  • Background: This research was performed to investigate the effects of Pulsatilla Koreana NAKAI pharmacopuncture (PPA) therapy on intestinal disease in rats with dextran sulfate sodium (DSS)-induced colitis. Methods: The subjects were divided into five groups : A control group, saline group, pharmacopuncture group PPA1 ($0.2mg/1kg/40{\mu}{\ell}$), pharmacopuncture group PPA2 ($0.5mg/1kg/40{\mu}{\ell}$), and pharmacopuncture group PPA 3($1mg/1kg/40{\mu}{\ell}$). The experimental model of colitis was induced by infection of dextran sulfate sodium (DSS) for eighteen days. After colitis was induced, PPA therapy was practiced on the Chunchu (ST25) once every two days for a total six times. Thereafter Disease Activity Index (DAI), colon length, damage to the colonic mucosa, body weight, IL-6, IL-10, $IL-1{\beta}$, $IFN-{\gamma}$, $TNF-{\alpha}$, $TGF-{\beta}1$, IL-23 and IL-17 were measured. Results: The results were as follows. 1. DAI was significantly decreased in the PPA groups. 2. Colon length was significantly increased in the PPA groups. 3. Damage of colonic mucosa was observed less in the PPA groups. 4. Body weight was significantly increased in the saline group and the PPA groups. 5. The PPA2 group showed a significant decrease in the intensity of IL-6, $IL-1{\beta}$, $IFN-{\gamma}$ and $TNF-{\alpha}$ levels and the mean of IL-23. 6. The PPA3 group showed a significant increase in the intensity of IL-10 and $TGF-{\beta}1$ levels. 7. No significant differences were shown in the mean of IL-17. Conclusion: These results suggest that PPA therapy on Chunchu (ST25) can be used as an effective treatment for inflammatory bowel disease.

Aggregation of Partially Quarternized Poly(4-vinylpyridine) with Anionic Surfactant Sodium Dodecvl Sulfate (부분적으로 4차아민화된 폴리(4-비닐피리딘)과 음이온 계면활성제인 도데실 황산 소듐과의 응집체 형성)

  • Modified poly(4-vinylpyridine) was obtained by partial quarternization of nitrogen atoms in pyridine rings of poly(4-vinylpyridine) with methyl group. By means of laser light scattering and fluorescence, it was found that this modified polymer chains were aggregated in the aqueous solution and its structure was core-shell type. The hydrophobic parts of the chains were densely condensed in core part and the hydrophilic part of quarternized amino with positive charge formed the shell part. In the mixed system of modified poly(4-vinylpyridine) and anionic surfactant, sodium dodecyl sulfate, it was observed that a critical aggregation concentration existed and that this critical concentration was suddenly decreased above 0.1 M NaCl. The size change of aggregates was also investigated by dynamic light scattering while sodium dodecyl sulfate was added into polymer solution upto the critical aggregation concentration.

Effect of Hwangyeonhaedok-tang on Experimental Mouse Colitis Induced by Dextran Sulfate Sodium (황련해독탕(黃連解毒湯)이 Dextran Sulfate Sodium 유도 마우스 대장염에 미치는 영향)

  • Objectives : Hwangyeonhaedok-tang(HHDT) has been traditionally used for various clinical symptoms associated with gastrointestinal disorder, cardiovascular diseases, and inflammation in the Oriental medicine. However, little is known for antioxidant and anti-inflammatory effects of HHDT on dextran-sulfate sodium(DSS)-induced colitis in mice. Methods : In this study, we investigated an antioxidant and anti-inflammatory effects of HHDT on DSS-induced colitis in mice. An experimental colitis was induced by daily treatment with 5% DSS. HHDT was orally administrated the various concentrations(25-100 mg/kg, body weight/day) for 7 days with one time per day. Results : HHDT reduced significantly clinical sign of DSS-induced colitis, including body weight loss, shorten colon length, disease activity index(DAI), and histological colon injury. HHDT also inhibited significantly serum NO and prostaglandine $E_2(PGE_2)$ productions in DSS-induced colitis mice. Furthermore, HDDT increased significantly an superoxide anion(SOD), catalase, and glutathione peroxidase(GPx) activity of the colon tissue in DSS-induced colitis mice. Conclusions : These results suggest that HHDT administration could reduce significantly the clinical signs and inflammatory mediators, and increase antioxidant activity in DSS-induced colitis model mice and is a good candidate for further evaluation as an effective anti-ulcerative agent.

Synergic Effect of Methanol extracts of Schizandrae Fructus and Mume Fructus on Experimental Mouse Colitis Induced by Dextran Sulfate Sodium (Dextran Sulfate Sodium 유도 마우스 대장염에 미치는 오미자와 매실의 상승효과)

  • The fruits of Schisandra chinensis and Prunus mume have been traditionally used in the Oriental countries as an astringent against diarrhea and abdominal pain, a protectant for liver disease, an antimicrobial, and a blood tonic. However, little is known about the extract of Schizandrae Fructus and Mume Fructus (SMF-Ex) on dextran-sulfate sodium (DSS)-induced colitis in mice. In this study, we investigated the protective effects of SMF-Ex on DSS-induced colitis in mice. An experimental colitis was induced by daily treatment with 5% DSS. SMF-Ex was orally administrated the single dose (80 mg/kg, body weight/day) for 7 days with one time per day. SMF-Ex reduced significantly clinical sign of DSS-induced colitis, including body weight loss, shorten colon length, increased disease activity index (DAI), and histological colon injury. SMF-Ex also inhibited significantly nitric oxide (NO) and prostaglandine $E_2$ ($PGE_2$) productions in DSS-induced colitis mice. Furthermore, SMF-Ex increased significantly an superoxide anion (SOD), catalase, and glutathione peroxidase (Gpx) activity of the colon tissue in DSS-induced colitis mice. These results suggest that SMF-Ex administration could reduce significantly the clinical signs and inflammatory mediators, and increase antioxidant activity in DSS-induced colitis model mice and is a good candidate for further evaluation as an effective anti-ulcerative agent.
