Effects of Annealing Conditions of Corn Starch Slurry on the Formation of Phosphorylated Cross-linked Resistant Starch (옥수수 전분유의 Annealing 조건이 인산가교 저항 전분의 형성에 미치는 영향)

  • The optimum annealing conditions of corn starch slurry were studied for RS4 type resistant starch production by phosphorylated cross-linking. When a corn starch slurry was cross-linked by using phosphate salts (STMP/STPP mixture) in the presence of 0.9%, 1.2% and 1.5% NaOH/st.ds, a high concentration of NaOH resulted in a rapid increase of the RS contents at the early reaction stage. However, similar RS contents were obtained after 12 h of cross-linking regardless of NaOH concentrations. The annealing treatment was conducted under various conditions such as pH between 2-10, temperature $40-60^{\circ}C$, time 0-14 h followed by phosphorylated cross-linking. The lower slurry pH was for the annealing treatment, the higher RS contents were obtained after cross-linking. When the slurry annealed for various period of time and temperature, a maximal amount of RS was formed after 2 h of annealing at $50^{\circ}C$ of annealing temperature of the starch slurry (pH 2.0). Therefore, an optimal annealing conditions at pH 2.0 and $50^{\circ}C$ for 2 h were proposed under the cross-linking conditions of sodium sulfate 10%/st.ds, NaOH 1.2%/st.ds and 12 h of the reaction time. The RS contents were linearly increased with the increase of phosphate salt addition. The RS4 prepared under the optimal conditions contained RS 72.3% and its phosphorus content was 0.36%/st.ds, which was below the limit (0.4%/st.ds) of modified starch by Korea Food Additives Code.

Processing of Powdered Smoked-Dried Anchovy Soup and Its Taste Compounds (훈건멸치 분말수프의 가공 및 정미성분)

  • The study was carried out to develop the powdered smoked-dried anchovy products as a natural flavoring substance. The processing conditions, chemical and taste compounds of products were as follows: The raw anchovy were washed, and then boiled 5 minutes in $5{\sim}6%$ NaCl and 1.0% sodium erythorbate solution. Boiled anchovy were smoked in smoking house at $40^{\circ}C$ for 4 hours as the first stage, and then increased temperature up to $80^{\circ}C$ as the second stage, and finally smoked 8 hrs at $80^{\circ}C$ to maintain the moisture content between 9 and 10 percent. The smoked-dried anchovy were pulverized and screened to be 50 mesh of particle size, and finally packed in PET/Al/CPP film bag. The moisture, crude lipid content and salinity of powdered smoke-dried anchovy were 9.4%, 9.6% and 6.9%, respectively. Fatty acid composition of product was mainly consisted of polyenes (43.4%) such as 22 : 6 and 20 : 5, followed by saturates (36.9%), monoenes (19.7%). The principal taste compounds of product were IMP, 466.5 mg/100g; free amino acids such as His, Tau, Pro, Lys, Ala and Glu, 1179.2 mg/100g; non-volatile organic acids such as lactic acid and succinic acid, 617.9 mg/100g; total creatinine, 595.9 mg/100g; small amount of betaine and TMAO. To make a instant soup, it was desirable for taste of products that powdered smoked-dried anchovy were mixed with 20% salt, 4.0% sugar, 3.0% MSG, 1.0% onion powder, 1.0% garlic powder and 1.0% black pepper.

Risk assessment for water quality of a river using QUAL2E model (QUAL2E 모형을 이용한 하천수질의 위해성평가)

  • In this study, we consider ability of self-purification for a rational water quality management. And we assess the risk of Alkyl Benzene Sulfonic acid sodium salt(ABS) of harmful ingredients in Anseong Cheon watershed using QUAL2E model. The observations and simulated results were fitted well for BOD and ABS, but even though the trend of DO concentration change was well represented, the error between observation and simulation values was existed. We assessed the Risk assessment by calculating Risk quotient(RQ) by Predicted Exposure Concentration(PEC) and Predicted No-Effect Concentration(PNEC). Results of the impact of ABS on the self-purification of the river were Anseongcheon[0.0003(Bressan), 0.06(Criteria of Ministry of environment)], Jinwicheon[0.0002(Bressan), 0.04(Criteria of Ministry of environment). And result of the impact of ABS on the Aquatic ecosystem of the river were Anseongcheon[0.0667(Bressan), 0.005(Criteria of Ministry of environment)], Jinwicheon[0.1(Bressan), 0.0075(Criteria of Ministry of environment). All of these results were smaller than the 1.0 which is the reference value suggested by Norification No.30 of the National Institute of Environment Research. So, ABS did not affect a self-purification and aquatic ecosystem of the river. The method suggested in the study is a simple one and can provide more information for harmful ingredients than criteria of Ministry of environment.

Stability of Protein Colloids in the Mixture of Cheese Whey and Soy Milk (유청(乳淸) 및 두유(豆乳) 혼합액에서의 단백질 콜로이드 안정성)

  • To investigate effectors on the colloidal stability of whey and soybean proteins, characteristics of tofu-gel formation, effects of heat treatment and salt composition on the colloidal stability, and effects of heat treatment on storage stability were analyzed. When experimental tofus were made from the mixture of whey and soybean, the calcium in the whey precipitated the soy proteins, and disrupted the gel formation, which resulted in the curd of poor texture. In the heat treatment at $60{\sim}100^{\circ}C$, whey and the whey proteins dialyzed against distilled water were readily preciptated at over $70^{\circ}C$, but the mixture of whey and soy extract as well as soy extract were stable at the range of temperature. The proteins of soy extract, whey dialyzed against sodium phosphate buffer, and the mixture were stable at the same heat treatment, and this suggested that phosphates in the soy extract stabilize specialty the whey proteins. Soy proteins were easily destabilized by adding $CaCl_2(0.05{\sim}0.07M)$ at $80{\circ}C$ and $70{\sim}85%$ of the proteins in soy extract and the mixture were preciptated, while only $30{\sim}55%$ of the proteins in whey dialyzed against distilled water were destabilized at the same conditions. Storage stability at $4^{\circ}C$ of the mixture was increased when the mixture was treated at $63^{\circ}C$ and lower temperature.

Rapid Analysis of Boric Acid in Nickel Plating Solutions (니켈도금액중의 붕산 신속정량법)

  • Only mannitol or glycerine is generally used for the determination of boric acid in a nickel plating solution in order to make its acidic property so strong that it can be titrated with NaOH. However, these solutions give very amgiguous color change of indicator due to the precipitation of nickel salts . Therefore, only experienced dchemistsorwell trained workimen can accurately confirm the actual end point of the titratiion. For eliminating such interference of nickel salts and easily confirming the end point by any persons , the author attempted to find out a solution which produces no precipitates during the titration in these experiments, and also he tried to funish the reason for ambiguousness in titration. The following results were obtained after many experiments. (1) In any titrations which produce nickel salts such nI(oh)$_2$, the salt is formed umption very approximate to the end point, which shows some error by the consumption of titrant(NaOH) . Then, the pink color of phenolphthalein is absorbed by Ni(OH)$_2$ and the pH jumping at the end pint is also diminished to as little as less than 15% of the total phenophthalein ph range. (2) Known methods by complex salts of citrate,w hich do not produce precipitates of Ni(OH)$_2$, are also not very satisfactory, because, the pH jumping at the end point is only about 35% and the color change of phenolphthalein is form blue-green to purple-blue. (3) New method by complex salts of oxalate were attempted in these experiments. They also did not produce precipitates of Ni(OH)$_2$ and were very satisfactory in color change at the end of point was about 65% and the color change was from blue-green to purpled. In this methods, analytica cost was minimized by the use of less amounts of cheaper chemicals than the conventional citrates complex methods. The mixture of chemicals used was composed methods. The mixture of chemical used was composed of 37g/ι of sodium oxalate(Na$_2$C$_2$O$_4$$.$5H$_2$O), 2g/ι of phenolphthalein, and 400ml /ι of glycerin. The accuracy of analysis was within the error of 0.5%. (4) The procedure of analysis was as follows. One ml of nickel plating solution was taken out and to it were added 20ml of water and 20 ml of the above mixture for the indicator. The solution was titrated with 0.1N NaOH. The quantity of boric acid was calculated by the following equation. Boric acid (g/ι) = 6.184${\times}$F${\times}$ml .

Studies on the Development of Food Resources from Waste Seeds V. Chemical Composition of Water-melon Seed (폐기종실(廢棄種實)의 식량자원화(食糧資源化)에 관(關)하여 제(第)5보(報) : 수박씨의 화학적(化學的) 조성(組成))

  • An attempt was made to find out the possibility of utilizing water-melon seed as resources of food fats and protein. The water-melon seed contained 40.40% of crude fat and 28.36% of crude protein. The lipid fraction obtained by silicic acid column chromatography was composed of about 97.35% neutral lipid, and the main components of neutral lipid by thin layer chromatography were triglyceride(50.40%), diglyceride(21.84%) and sterol(11.48%). The predominant fatty acids of total and major lipid classes were linoleic acid(55.30-67.85%), palmitic acid(12.07-28.12%) and oleic acid(9.06-16.40%), whereas stearic acid and linolenic acid were detected as small amounts. The salt soluble protein of watermelon seed was highly dispersible in 0.02M sodium phosphate buffer containing about 0.7M $MgSO_4$, and the extractability of seed protein was about 27%. Glutamic acid and arginine were major amino acids, and the essential amino acids such as lysine, threonine, valine, methionine, isoleucine, leucine and phenylalanine were also detected. The electrophoretic analysis showed 6 bands in water-melon seed protein, and the collection rate of the main protein fraction purified by sephadex G-100 and G-200 was 52.4%. The amino acids of the main fraction protein were also mainly composed of glutamic acid and arginine. The molecular weight for the main protein of the water-melon seed was estimated to be 120,000.

Competitive Extraction of Chlorinated Solvents by Headspace SPME GC/FID (Headspace SPME GC/FID를 이용한 Chlorinated Solvents의 경쟁적 추출효과에 관한 연구)

  • In this study, Solid-phase microextraction (SPME) with GC/FID was studied as a possible alternative to liquid-liquid extraction for the analysis of chlorinated solvents (PCE and TCE) and these by-products (cis-DCE, VC, and Ethylene). Experimental parameters affecting the SPME process (such as kind of fibers, adsorption time, desorption time, volume ratio of sample to headspace, salt addition, and magnetic stirring) were optimized. Experimental parameters such as CAR/PDMS, adsorption time of 20 min, desorption time of 5 min at $250^{\circ}C$, headspace volume of 50mL, sodium chloride (NaCl) concentration of 25% combined with magnetic stirring were selected in optimal experimental conditions for analysis of chlorinated solvents and these by-products. The general affinity of analytes to CAR/PDMS fiber was high in the order PCE>TCE>cis-DCE>VC>Ethylene. The linearity of $R^2$ for chlorinated solvents and these by-products was from 0.912 to 0.999 when analyte concentrations range from $10{\mu}g/L$ to $500{\mu}g/L$, respectively. The relative standard deviation (% RSD) were from 2.1% to 3.6% for concentration of $500{\mu}g/L$ (n=5), respectively. Finally, the limited of detection (LOD) observed in our study for chlorinated solvents and these by-products were from $0.5{\mu}g/L$ to $10{\mu}g/L$, respectively.

A Study on the Solubilization of $\alpha$-Chymotrypsin Using AOT Reverse Micelles; Effects of pH and salts (AOT 역미셀을 이용한 $\alpha$-chymotrypsin의 가용화에 대한 연구;pH와 염의 영향)

  • Micellar aggregates are known to be useful for the selective isolation of biologically active materials such as amino acids, proteins, and enzymes from crude mixtures sparsely dispersed in water. In this study, the effects of pH, salt type and its concentration on the solubilization of $\alpha$-chymotrypsin into the organic micellar phase, which consisted of AOT (sodium 야(2-ethylhexy)sulfosuccinate) and iso-octane, were comprehensively examined. It was found that maximum extraction efficiency was attained at a pH below the isoelectric point of $\alpha$-chymotrypsin; at pH=5.0 for NaCl and KCl, and at pH=7.0 for $CaCl_2$and $MgCl_2$. In order to avoid complications stemming from the precipitationof protein at low pH interfaces, the protein concentrations in the organic and aqueous phases were directly measured. The size of the micelle water pool was estimated by measuring the molar ratio of the surfactant to the water, W(sub)o. The resulting values of W(sub)o were nearly constant at 30 and 19 for NaCl and KCl, respectively, and were independent of pH. The addition of 1:2 salts like $MgCl_2$and $CaCl_2$ led to much lower, but a constant value of, W(sub)o than the 1:1 salts.

  • Poly (vinyl alcohol) (PVA) and carboxymethyl cellulose (CMC) have received increasing attention in biomedical and biochemical applications because of their properties such as being water-soluble and biocompatible. In this study, a PVA/CMC hydrogel applicable to artificial cartilage was prepared by a freezing-thawing technique and a gamma-ray irradiation. The solid concentration of PVA was 7 wt% and the concentration of CMC was 4 wt%. The freezing/thawing process was repeated twice and the dose of gamma-ray irradiated was 30 kGy. Results of gelation before and after gamma-ray irradiation were similar, but the swelling degree decreased and compressive strength increased. The cytotoxicity was investigated with CCK-8 assay.

Processing Conditions and Quality Stability of Filefish Steak during Frozen Storage (말쥐치 스테이크가공조건(加工條件) 및 동결저장중(凍結貯藏中)의 품질안정성(品質安定性))

  • Processing conditions of filefish steaks and effect of soybean protein on quality during frozen storage were investigated. Additives which is added to the filefish meat were 1% of table salt, 0.2% polyphosphate, 0.5% of sodium bicarbonate, 0.2% of monosodium glutamate, 0.2% of red pepper powder, 0.4% of white pepper powder, 0.2% of garlic powder and 0.2% of nutmeg. The mixture was minced in the stone mortar and then stored at -3 to $-5^{\circ}C$ for two days prior to frozen storage. The benefical effects of adding 5% of soybean protein to the filefish steaks were the control of free drip, oxidative rancidity and in texture that exhibited the improvement of quality. The quality of frozen filefish steaks, by sensory evaluvation, was not inferior to that of hamburger on the market.
