Processing Conditions and Quality Stability of Frozen Seasoned Sardine Meat during Frozen Storage (냉동 정어리 조미육의 가공 및 저장중의 품질안정성)

  • Seasoned sardine meat was prepared to extend the use of sardine for human consumption, and processing conditions and storage stability of frozen seasoned sardine meat were studied during storage at $-20^{\circ}C$. The fish was beheaded, gutted and cleaned in a washing tank. The washed fish was then put through a belt-drum type meat separator which separates the flesh iron the bone and skin. Mechanically deboned fish meat was mixed with $20.6\%$ emulsion curd, $0.5\%$ table salt, $2.0\%$ sugar, $0.4\%$ sodium bicarbonate, $0.2\%$ polyphosphate, $0.1\%$ monosodium glutamate, $0.3\%$ onion powder, $0.1\%$ garlic powder, $0.1\%$ ginger powder, $3.0\%$ soybean protein and $0.1\%$. In sodium erythorbate. This seasoned sardine meat was frozen with contact freezer, packed in a carton box and then stored at $-20^{\circ}C$. The pH, volatile basic nitrogen, viable cell counts, peroxide value, carbonyl value, thiobarbituric acid value, taste compounds, fatty acid composition, salt extractable nitrogen, drip, texture, and color values of the products were determined during frozen storage. The results showed that lipid content in products could be controlled by using emulsion curd, and flavor and texture could be improved by adding spices and soybean protein, and lipid oxidation could be retarded by $0.1\%$ sodium erythorbate. Judging from the results of chemical experiments and sensory evaluation, the products can be preserved in a good quality for 120 days during frozen storage.

Effects of Sodium Bicarbonate and Vitamin Supplementation on Milk Production and Composition in Lactating Holstein Cows Under Heat Stress Condition (고온기에 있어서 중조와 Vitamin 급여가 착유우의 산유량 및 유조성분 변화에 미치는 효과)

  • Sixteen multiparous lactating Holstein cows were used to compare effects of supplementing 1)no additive(Control), 2)1.2% sodium bicarbonate(NaHCO3); 3)niacin(80g/d), 4)vitamin A+E (140,000IU+1000IU) on feed intake, milk production, milk composition and somatic cell counts during the summer months. Insofar as possible, treatment groups were balanced for lactation number and days in milk. Cows were fed a diet of 9.1kg DM of concentrate and 10.2kg DM of corn silage. Daily maximum air temperature in free stall barn was 35℃ for 3 days of the pretreatment periods and decreased gradually up to 27℃ during the treatment periods of 15days. Dry matter intake of corn silage was higher(p<0.05) for cows consuming NaHCO3 than those not consuming NaHCO3. Daily milk production for niacin and vitamin A+E supplementations resulted in significant(p<0.001) increase in milk production from 3 day of trials than control and NaHCO3. Milk fat percentage tended(p=0.09) to increase and milk lactose percentage was increased significantly(p<0.001) for cows supplemented with NaHCO3, niacin and vitamin A+E. Milk protein percentages was higher significantly(p<0.05) with supplemental niacin and somatic cell counts was higher significantly(p<0.001) with supplemental vitamin A+E. These data strongly suggest that supplementation of NaHCO3, niacin or vitamin A+E should be increased for improving milk production and mammary gland health of dairy cows under heat stress.

  • Purpose : Nowadays, drinks, foods and snacks have frequently been intensified with calcium and the insights into the importance of calcium-intake in general has developed in Korea. In this decade, we found the numbers of children who was visited to our hospital for evaluation of hematuria defined with hypercalciuria were increased. So we tried to compare the mean levels of serum calcium, alkaline phosphate, sodium, potassium, chloride, BUN, creatinine, bicarbonate and urinary pH who visited our hospital in 1991, 1992 with in 2000, 2001. Materials and methods : Between January 1991 to December 1992, and between January 2000 to December 2001, each 366 children and 488 children, aged 1 month to 15 years, who presented in our hospital for tonsilectomy and adenoidectomy or for inguinal herniorrhaphy were enrolled in the study, The children in the study were checked the level of serum calcium, alkaline phosphate, sodium, potassium, chloride, BUN, creatinine, bicarbonate and urinary pH with the machine which was corrected the similar levels of practical chemical levels in serum. We compared each mean levels in 1991s' group with in 2001s' group totally and separately through the age and sex. We used t-test to analysis data. Results : The levels of serum calcium, alkaline phosphate, creatinine, sodium, potassium, and urinary pH of 2001s' group were significantly higher than the levels of 1991s' group(P<0.05). The each level was $9.91{\pm}0.50\;mg/dL,\;248.58{\pm}94.98\;U/L,\;0.61{\pm}0.14\;mg/dL,\;138.64{\pm}2.22\;mM/L,\;4.35{\pm}0.40\;mM/L,\;6.18{\pm}0.86$ in 2001s' and $9.13{\pm}0.68;mg/dL,\;198.26{\pm}79.34\;U/L,\;0.433{\pm}0.18\;mg/dL,\;137.86{\pm}2.67\;mM/L,\;4.22{\pm}0.36\;mM/L,\;5.83{\pm}0.95$ in 1991s'. And the levels of serum bicarbonate, $23.64{\pm}2.57\;mM/L$ in 2001s' was significantly lower than the 1991s', $24.60{\pm}2.23\;mM/L$(P<0.05). The similar results were detected each age and sex group. Conclusion : The levels of serum calcium increase in this decade. The results will be used as a basic data for the national health plan in the years to come.

Effect of Sodium Bicarbonate Supplementation on Carcass Characteristics of Lambs Fed Concentrate Diets at Different Ambient Temperature Levels

  • The objective of this study was to investigate the influence of ambient temperatures on carcass characteristics of lambs fed concentrate diets with or without $NaHCO_3$ supplementation. A slaughter study was carried on 12 male Black Belly Barbados lambs randomly drawn from a growth trial (35 weeks). The lambs were divided into four equal groups and allotted in a $2{\times}2$ factorial design. The lambs were allotted at random to two dietary treatments of a basal diet (35:65 roughage:concentrate) or basal diet supplemented with 4% $NaHCO_3$ at different ambient temperatures ($20^{\circ}C$ and $30^{\circ}C$) in an environment controlled chamber for 10 days. Lambs were slaughtered for carcass evaluation at about 262 days of age (245 days of growth trial, 7 days adaptation and 10 days of experimental period). Ambient temperature had significant (p<0.05, p<0.05, p<0.01, and p<0.001) effects on meat color from the ribeye area (REA), fat, leg and longissimus dorsi muscles with higher values recorded for lambs in the lower temperature group than those from the higher ambient temperature group. Significant differences (p<0.05) in shear force value ($kg/cm^2$) recorded on the leg muscles showed higher values (5.32 vs 4.16) in lambs under the lower ambient temperature group compared to the other group. Dietary treatments had significant (p<0.01, p<0.01, and p<0.05) effects on meat color from the REA, fat, and REA fat depth ($cm^2$) with higher values recorded for lambs in the $NaHCO_3$ supplementation group than the non supplemented group. Similarly, dietary treatments had significant differences (p<0.05) in shear force value ($kg/cm^2$) of the leg muscles with the $NaHCO_3$ groups recording higher (5.30 vs 4.60) values than those from the other group. Neither ambient temperature nor dietary treatments had any significant (p>0.05) effects on pH, and water holding capacity on both muscles. These results indicated that $NaHCO_3$ supplementation at low ambient temperatures had caused an increase in carcass characteristics leading to significant effect on meat quality.

Skin Transcriptome Profiling of the Blass Bloched Rockfish (Sebastes pachycephalus) with Different Body Color Patterns (체색 패턴이 다른 개볼락(Sebastes pachycephalus) 피부 전사체 프로파일링)

  • The body color pattern in fish is a distinctive feature for species identification. The blass bloched rockfish Sebastes pachycephalus is a commercially important marine fish species, distributed in the central and southern parts of Korea and south Hokkaido of Japan. It has a morphological feature divided into four subspecies according to with or lacking distinct spots on the body surface, and to the location of markings on the body surface. However, the genetic basis of body color pattern of S. pachycephalus is still unknown. Thus we analyzed the transcriptome of S. pachycephalus skin samples using RNA-seq analysis to investigate functional genes related to body color patterns. The experimental skin samples were prepared by classified into 'Wild type' (lacking distinct spots and markings) and 'Color type' (with distinct spots and marking). Two skin sample transcriptomes were compared pairwise and the results revealed that were 164 differentially expressed unigenes in the skin samples of 'Wild type' and 'Color type'. Gene Ontology analysis of 164 differentially expressed unigenes showed that these genes were included in the functional group of molecular function (2 genes), biological process (46 genes), and cellular component (6 genes). There were several genes that body color type skin specific expression and the genes were CTL (Galactose-specific lectin nattectin), CUL1 (Cullin-1), CMAS (N-acylneuraminate cytidylyltransferase), NMRK2 (Nicotinamide riboside kinase 2), ALOXE3 (Hydroperoxide isomerase ALOXE3), SLC4A7 (sodium bicarbonate cotransporter 3). Our study is the first attempt to search for functional genes involved in the formation of body color patterns in S. pachycephalus. The differentially expressed unigenes obtained in this study can be used as candidate genes for functional gene study related to body coloration of fish.

Quantification of Ceruloplasmin in Wale Rats Exposed to ${\gamma}$-radiation by Enzyme Linked Immunosorbent Assay (ELISA방법에 의한 방사선 피폭 후 흰쥐 혈액 내에서의 ceruloplasmin 정량)

  • Adult male rats were exposed to a whole body with a single dose of 1.0, 2.0, 3.0, 5.0, and 7.0 Gy. The animals were sacrificed 48, 72, 96 and 216 hours following exposure. A competitive enzyme linked immunosorbent assay(ELISA) with antigen immobilized on the solid phase has been developed to measure ceruloplasmin in rat serum and complete dose response curves. Ceruloplasmin was purified from the plasma of turpentine treated male rats. Coating of ceruloplasmin had more effectiveness in 10 mM Tris-HCI, 150 mM sodium chloride, pH 7.4 than in 50 mM carbonate/bicarbonate buffer, pH 9.6. The coating range for ceruloplasmin was $70{\sim}140ng$/well. Levels of ceruloplasmin increased to maximum on the $72{\sim}96$ hours after irradiation. Slope of between response and dose was greatest value 96 hours following irradiation. Normal ceruloplasmin levels were not recorded 216 hours following exposure. In 0.1 Gy irradiated group, levels of ceruloplasmin also increased to maximum on the $72{\sim}96$ hours following irradiation. The concentration of this protein remained significantly different from control value, 196 hours after exposure. Ceruloplasmin was identified as one of the major acute phase protein following irradiation and further studies about gene expression and regulation would be necessary for radiation protection.

Rapid and Simple Analytical Method for Removing Patulin from Apple Juice Using Solid Phase Extraction (고체상 추출법(SPE: Solid Phase Extraction)을 이용한 국내 사과주스 중 Patulin 간편.신속 분석방법)

  • Patulin, a secondary metabolite of mold, is commonly found in rotten apples. Many countries regulate patulin at levels ranging from 30 to $50\;{\mu}g/L$. Most analytical methods for removing patulin from apple juice include liquid-liquid extraction (LLE), which is time and labor intensive. To replace the LLE method, a solid-phase extraction (SPE) method has been developed for apple juice and unfiltered apple juice. A portion of the test sample was applied to a macroporous copolymer cartridge and washed with 5 mL of 1% sodium bicarbonate, followed by 5 mL of 1% acetic acid. Patulin was eluted with 5 mL of 2% acetonitrile in anhydrous ethyl ether. The mobile phase was tetrahydrofuran in water (0.8:99.2) and was detected with a UV detector at 276 nm. Recoveries ranged from 95 to 101% in test samples, and the minimum detectable level was 30 ppb. Because this SPE method is fast, easy, reliable, and inexpensive, it could be applicable for companies or analytical agencies to analyze patulin concentrations in apple juice.

  • Further elucidation of the mechanism of halothane's negative inotropic action has resulted from a study of the effect of various substrates on halothane-depressed rat atria. Approximately 6 mg% halothane was required to maintain a 50% depression of the contractility of rat atria suspended in a modified Krebs-Ringer bicarbonate glucose medium, pH 7.4, $30^{\circ}C$ for 2hr. Both lactate and acetate were found to restore partially the contractility of halothane-depressed atria. The maximally effective concentration of lactate was 5 mM; for acetate it was 2.5mM. Neither 5 nor 20 mM of additional glucose was effective in restoring the force of contraction of halothane-depressed atria. The results are consistent with the hypothesis that halothane exerts at least a part of its negative inotropic effect on rat atria by inhibiting either the uptake or utilization of glucose by the myocardium. The site of blockade must be prior to the conversion of pyruvate to acetyl CoA. In our previous report dealing with the mechanism of cardiac depressant action of inhalation anesthetic halothane, it has been demonstrated that: 1) approximately 6 mg/100 ml halothane is required to maintain 50% depression of the force of contraction of isolated rat atria in Krebs-Ringer bicarbonate glucose medium; 2) pyruvate partially restores the contractility of halothane-depressed atria, but has no effect on normal atria; the partial recovery of depressed atria by the addition of sodium pyruvate is due to the effect of the pyruvate ion itself, not to the sodium ion; 4) addition of pyruvate, to atria depressed with hypertonic medium, produced only further depression. From these findings we concluded that the cardiac depressant action of halothane on rat atria is a manifestation of inhibition of glucose uptake or utilization. The present studies were undertaken to observe the effect of other substrates on halothane-depressed atria in order to substantiate our conclusion. As with the case of pyruvate, lactate and acetate also partially restored the force of contraction of halothane-depressed atria. These data are consistent with the hypothesis that halothane inhibits glucose uptake or utilization in the glycolytic cycle of the myocardium.

한국원자력연구소 방사선방어기술 개발 및 연구 현황

  • 1959년 한국원자력연구소가 창립됨과 동시에 &Health Physics&, 즉 보건물리라고 하는 명칭과 조직이 탄생되어, 방사선안전관리의 실무와 보건물리의 연구가 시작되었다. 최초 10년간은 선진제국의 보건물리분야의 연구와 기술을 추적하여 우리나라의 방사선안전관리 기술의 기초를 다지는 시기로서 개인방사선모니터링기술, 환경방사선(능) 모니터링기술 및 방사선방어용계측기기의 교정기술 개발에 중점을 두고 연구개발이 추진되었으며, TRIGA Mark-II 연구용원자로의 가동에 따라 원자로 생체차폐체의 건전성 검증에 관한 유익한 방사선량 측정자료도 얻게 되었다. 즉 이 기간은 방사선안전관리의 체제정비 및 기초기술 확립에 노력한 기간이었다. 1970년대는 원자력 연구개발에 대한 기본방향과 정책의 변경등으로 보건물리 연구조직은 방사선안전관리, 환경연구 그리고 방사화학분야로 분산되었으며, 그로인하여 연구개발활동은 거의 정체되어 겨우 방사선안전관리 실무만이 그 명맥을 유지하였다. 그 결과 우리나라 방사선안전관리 및 그와 관련된 연구개발의 기반이 흔들리게 되었으나, 그러한 환경하에서도 방사선량측정평가기술, 방사선차폐설계기술 및 원자로사고시 피폭선량평가기술의 선진화에 필요한 지식을 얻었으며, 방사선 안전관리에 유익한 실무경험도 축적하게 되었다. 1980년대는 통합된 원자력 연구개발체제의 구축으로 방사선작업종사자 및 일반공중의 피폭저감화 기술개발에 필요한 각종 최신기술을 도입하였고, 관리업무에 있어서도 측정의 정확도와 신뢰성향상 및 새로운 관리기술의 개발에 많은 노력을 한 결과, 유익한성과를 얻게된 기간이다. 특히, 이 기간은 방사선안전관리기술의 선진화를 위한 지식이 축적되어 90년대의 방사선안전관리기술자립화를 위한 전환기로서, 이와같이 축적된 기술은 원자력의 평화적 이용에 크게 기여할 것으로 기대된다.서 dithiothreitol를 투여한 군에서는 우라늄단독투여군에 비해 cretinine의 배설이 상당히 증가하였다(P<0.05). 6. 우라늄오염에 의한 신장의 소견에 있어 우라늄단독투여군은 근위곡세뇨관상피의 공포화 및 종창, microvilli와 brush border의 손실, 세뇨관 상피의 괴사가 관찰되었으며, 간장의 충혈, 중심성 괴사 및 모세관 확장증도 관찰되었다. 그리고 sodium bicarbonate와 생리적 식염수를 병행투여한 군과 우라늄을 투여하고 30분이 지나서 dithiothreitol를 투여한 군에서는 우라늄 단독투여군에 비해 높은 방호효과가 관찰되었으나 다른 실험군에서는 큰 효과가 없는 것으로 나타났다. 결론적으로 우라늄의 체내오염시에는 sodium bicarbonate와 생리적 식염수를 가능한 빨리 병행투여하거나 dithiothreitol을 체내오염후 30분이 지나서 투여하는 방법이 우라늄오염에 대한 제염에 매우 유효할 것으로 생각되며, 특히 우라늄에 의한 인체장해를 유의하게 경감시켜줄 것으로 사료되었다.내의 어떤 부위와도 관계가 되는 것으로 간주되는데 이것이 $(^3H)$ QNB가 $(^3H)$ NMS보다 높은 최대 결합능력 $(B_{max})$을 나타낼 이유이다.   • The processing conditions and food components of meaty textured sardine burgers were studied to develope a new form of burger, The separated sardine meat was chopped, mixed with 14.1% emulsion curd, 1.5% table salt, 2.0% sugar, 0.4% sodium bicarbonate, 0.2% polyphosphate, 0.1% monosodium glutamate, 8.0% bread powder, 0.4% onion powder, 0.1% garlic powder, 0.1% ginger powder and 3.0% soybean protein by remodeled stone mortar. This seasoned sardine meat was fried in soybean oil $(165{\pm}2^{\circ}C,\;3min)$. The main fatty acids of sardine burger were palmitic acid, oletic, acid, linoleic acid, eicosapentaenoic acid and docosahexaenoic acid. Amino acid composition of sardine burger were mainly consisted of histidine, glutamic acid, leucine and lysine. The major taste compounds in the product were revealed nucleotides and their related compounds $(11.19{\sim}11.96\;{\mu}mole/g)$ such as IMP and free amino acids (1824.8 mg/100g) such as histidine, glutamic acid, leucine and lysine. Total creatinine, betaine and trimethylamine oxide were seemed to act an auxiliary role in taste of product.
