A shell layer entrapping aerobic ammonia-oxidizing bacteria for autotrophic single-stage nitrogen removal

  • In this study, a poly(vinyl) alcohol/sodium alginate (PVA/SA) mixture was used to fabricate core-shell structured gel beads for autotrophic single-stage nitrogen removal (ASNR) using aerobic and anaerobic ammonia-oxidizing bacteria (AAOB and AnAOB, respectively). For stable ASNR process, the mechanical strength and oxygen penetration depth of the shell layer entrapping the AAOB are critical properties. The shell layer was constructed by an interfacial gelling reaction yielding thickness in the range of 2.01-3.63 mm, and a high PVA concentration of 12.5% resulted in the best mechanical strength of the shell layer. It was found that oxygen penetrated the shell layer at different depths depending on the PVA concentration, oxygen concentration in the bulk phase, and free ammonia concentration. The oxygen penetration depth was around $1,000{\mu}m$ when 8.0 mg/L dissolved oxygen was supplied from the bulk phase. This study reveals that the shell layer effectively protects the AnAOB from oxygen inhibition under the aerobic conditions because of the respiratory activity of the AAOB.

Biosynthesis of polyhydroxyalkanoate by mixed microbial cultures from hydrolysate of waste activated sludge (혼합미생물배양체를 이용한 폐활성슬러지 가용화 산물로부터 polyhydroxyalkanoate 생합성)

  • A new approach to the solubilization of waste activated sludge (WAS) using alginate-quaternary ammonium complex beads was investigated under controlled mild alkaline conditions. The complex beads were prepared by the reaction of sodium alginate (SA) with 3-(trimethoxysilyl)propyl-octadecyldimethylammonium chloride (TSA) in acid solution, followed by crosslinking with $CaCl_2$. Treatment of WAS with SA-TSA complex beads was effective for enhancing the efficacy of WAS solubilization. The highest value of soluble chemical oxygen demand (SCOD) concentration (3,900 mg/L) was achieved after 10 days of treatment with 30% (v/v) SA-TSA complex beads. The WAS solubilization efficacy of the complex beads was also evaluated by estimating the concentrations of volatile fatty acids (VFAs). The maximum value of VFAs was 2,961 mg/L, and the overall proportions of VFAs were more than 75% of SCOD. The main components of VFAs were acetic, propionic, iso-butyric, and butyric acids. These results suggest that SA-TSA complex beads might be useful for enhancing the solubilization of WAS. The potential use of VFAs as the external carbon substrate for the production of polyhydroxyalkanoate (PHA) by a mixed microbial culture (MMC) was also examined. The enrichment of PHA-accumulating MMC could be achieved by periodic feeding of VFAs generated from WAS in a sequencing batch reactor. The composition of PHA synthesized from VFAs mainly consisted of 3-hydroxybutyrate. The maximum PHA content accounted for 25.9% of dry cell weight. PHA production by this process is considered to be promising since it has a doubly beneficial effect on the environment by reducing the amount of WAS and concomitantly producing an eco-friendly biopolymer.

Dried Noodle Making of Composite Flours Utilizing Buckwheat and Wheat Flour (메밀가루와 밀가루 복합분의 건면 제조시험)

  • In this study, buckwheat four was classified by the order of extraction from the inmost layer to the outmost layer, and was designated as $B_1,\;B_2,\;B_3,$ and the mixed flour of $B_1,\;B_2,\;B_3$ (1:1:1) was named $B_0$. To improve the dried noodle-making properties, various levels of strong wheat flour and additives such as sodium alginate (SA) and xanthan gum(XG) were added to buckwheat. 1) The buckwheat $B_2$ and $B_3$ flours contained much higher amounts of crude ash, protein and fat than the $B_1$ flour. 2) Buckwheat $B_3$ flour demonstrated lower maxium viscosity value by amylograph than the $B_1$ flour, but it demonstrated 2 times higher maximum viscosity value than wheat flour. Buckwheat $B_1$ flour demonstrated 4-5 times higher maximum viscosity value than wheat flour. Addition of SA and XG increased the viscosity of the flour mixtures. 3) It was possible to make dried noodles from the composite flour of (buckwheat 40 %+wheat 60 %). The lesser the quantity of buckwheat flour was, the better the kneading property of mixed flour was. 4) SA and XG, which were added at the level of 1.0-1.5%, significantly improved dough making properties and the cooking quality of the noodles.

  • Nanofiltration (NF) composite membranes based of poly (vinyl alcohol) (PVA) and sodium alginate (SA) were prepared by coating PVA/SA (95/5 in wt %) mixture solutions on the microporous polysulfone (PS) supports, followed by the crosslinking with glutaraldehyed. The composite membranes prepared were characterized with a scanning electron microscopy (SEM), a fourier transform infrared spectroscopy(FTIR), an elecrtokinetic analyzer (EKA) and permeation tests.

  • Forward osmosis (FO) is an emerging membrane technology with potential applications in desalination and wastewater reclamation. The osmotic pressure gradient across the FO membrane is used to generate water flux. In this study, flux performance and foulant deposition on the FO membrane were systematically investigated with a co-current cross-flow membrane system. Sodium alginate (SA), bovine serum albumin (BSA) and tannic acid (TA) were used as model foulants. Organics adsorbed on the membrane were peeled off via oscillation and characterized by Fourier transform infrared (FTIR) spectroscopy and scanning electron microscopy (SEM). When an initial flux of $8.42L/m^2h$ was applied, both flux reduction and foulant deposition were slight for the feed solution containing BSA and TA. In comparison, flux reduction and foulant deposition were much more severe for the feed solution containing SA, as a distinct SA cake-layer was formed on the membrane surface and played a crucial role in membrane fouling. In addition, as the initial SA concentration increased in FS, the thickness of the cake-layer increased remarkably, and the membrane fouling became more severe.

Development of Seasoned Semi-Dried Oyster (조미 반건조 굴 가공품의 개발)

  • This study was carried out to examine the optimum processing conditions of seasoned semi-dried oyster, and to investigate its food component characteristics. Three types of semi-dried oyster were prepared: semi-dried oyster prepared without seasoning and coating (C), seasoned semi-dried oyster without coating (S) and seasoned semi-dried oyster coated with alginate (SA). SA was high in moisture (48.6%), while low in lipid (2.8%), and crude protein (25.9%) compared to those of C and S. Hardness and sensory scores of SA were $209.8g/cm^2$ and $3.9\sim4.5$ points, respectively. Total amino acid content (24,299 mg/100 g) of SA was lower than that (27,181 mg/100 g) of C, and the major amino acids were aspartic acid, glutamic acid, leucine and Lysine. The major fatty acids of SA were 16:0 (25.5%) as saturates and EPA (23.5%) and DHA (9.3%) as polyenes. Calcium and phosphorus contents of SA were 42.6 mg/100 g and 245.5 mg/100 g, respectively.

Release of 5-Fluorouracil from Ethylene-Vinyl Acetate Matrices Containing Hydrophilic Additives (수용성 첨가제를 함유하는 에틸렌초산비닐 매트릭스로부터 5-플루오로우라실의 방출)

  • In our previous work, we have studied the effect of lactose and sodium alginate (SA) on the rate of release of 5-fluorouracil (5-FU) from ethylene-vinyl acetate (EVA) matrix. These hydrophilic additives promoted the rate of 5-FU release and the increase in rate was larger when SA was used. Both additives showed better ability to increase the rate than 5-FU itself. In this paper, we extended our study to another hydrophilic additive, Carbopol 940 (CP). Compared to SA or lactose, CP increased the rate of 5-FU release markedly. Release rate increased as the loading amount and the pH of the release medium increased. After release experiment, matrix volume increased up to 15 times of that before release experiment, depending on the amount of CP dispersed in the matrix and the pH of the release medium. On the other hand, the volume of the matrix containing lactose or SA decreased. The weight changes of the dry matrix before and after release experiment imply that CP is not released out of the matrix, to the contrary of lactose and SA. Scanning electron microscope study clearly showed that large cavities and pores are generated on the surface and the inside of the matrix. These results indicate that the mechanism by which CP increases the release rate is quite different from that of monomeric additives such as lactose or SA.

Mechanical behaviours of biopolymers reinforced natural soil

  • The mechanical behaviours of biopolymer-treated soil depend on the formation of soil-biopolymer matrices. In this study, various biopolymers(e.g., xanthan gum (XG), locust bean gum (LBG), sodium alginate (SA), agar gum (AG), gellan gum (GE) and carrageenan kappa gum (KG) are selected to treat three types of natural soil at different concentrations (e.g., 1%, 2% and 3%) and curing time (e.g., 4-365 days), and reveal the reinforcement effect on natural soil by using unconfined compression tests. The results show that biopolymer-treated soil obtains the maximum unconfined compressive strength (UCS) at curing 14-28 days. Although the UCS of biopolymer-treated soil has a 20-30% reduction after curing 1-year compared to the maximum value, it is still significantly larger than untreated soil. In addition, the UCS increment ratio of biopolymer-treated soil decreases with the increase of biopolymer concentration, and there exists the optimum concentration of 1%, 2-3%, 2%, 1% and 2% for XG, SA, LBG, KG and AG, respectively. Meanwhile, the optimum initial moisture content can form uniformly biopolymer-soil matrices to obtain better reinforcement efficiency. Furthermore, the best performance in increasing soil strength is XG following SAand LBG, which are significantly better than AG, KG and GE.

  • Poly(phenylene oxide)s were used to prepare flat, integrally skinned self-supporthed asymmetric membranes by dry-wet phase separption. The intrinsic ideal gas selectivity of poly- (2,6-dimethy-1,4-phenylene oxide) (PMPO) was retained in the membranes, and improved by a coating with silicone rubber. Polymers of the same class were coated of UF supports with a silicon rubber gutter layer, yielding composite membranes with high flux but lower selectivity. The effect of th glutaraldehyde cross-linking of sodium alginate (SA) membranes on the mobility of water and ethanol has been studied with pfg nmr. Crosslinking reduces water self-diffusion, and does not seem to be stable on the timescale of weeks.
