Effect of Pre-Sowing Treatments on Germination and Initial Seedling Growth of Castanopsis Indica- An Endangered Tree Species in Bangladesh

  • Castanopsis indica (Sil Batna) is an ecologically valuable multipurpose indigenous tree species of Bangladesh. Considering its high value but poor natural regeneration due to seed dormancy, the authors conducted an experiment at the Institute of Forestry and Environmental Sciences Chittagong University (IFESCU) during 2012-2013 to find out effective pre-sowing treatments. Ten pre-sowing seed treatments were applied on mature, even-sized seeds namely-control; sandpaper rubbing; nail clipping; immersion in normal water (at room temperature: 24℃) for 24 hrs., 48 hrs. and 72 hrs.; immersion in hot water (80℃) for 1 minute; soaking in 10% dilute H2SO4; soaking in 10% HCl for 5 minutes; and sowing in propagator house. Seeds sown after sandpaper rubbing at the distal end revealed best performances by providing highest germination percent (66.7%), germination energy (30%), germination index (0.17), germination rate (0.0145), germination value (30%) and plant percent (66.7%) within shortest period (38 days). The treatment also produced most vigor seedlings with 20.9 cm shoot height, 15 node number, and largest leaves (11.1 cm×2.9 cm). Hence, it is recommended to adopt sandpaper rubbing method for maximum germination and quality seedlings.

Effect of Different Herbicides on Initial Rice and Weedy Rice Seedling Growth under Iron-coated Seeds in Flooded Direct Seeding (담수직파 파종 전 제초제 처리에 따른 철분코팅종자와 잡초성 벼의 출아, 입모 및 초기생육 영향)

  • This research was conducted to determine a pre-germinated herbicides with iron-coated seeds in water and wet hill seeded rice. Days of rice seedling emergence was the faster germination at the application of benzobicyclon > oxadiazon > thiobencarb > butachlor and the sealed iron-coated seeds with pre-germinated seeds> pre-germinated seeds> iron-coated seeds with pre-soaking. The seedling establishment was relatively high in untreated control and benzobicyclon among seed treatments by 63.7% and 75.7%. There was 100% seeds killed of pre-germinated seeds in terms of herbicide phytotoxicity and sealed iron-coated seeds with pre-germinated seeds in butachlor but benzobicyclon was of safe with 2% rice seeds killed. Infant rice seedling height was of 9.2-12.9cm in benzobicyclon and 11.9-16.3cm in untreated control and thus there was relatively normal development and growth at the initial rice seedling.

  • This study was investigated seed germination of Rheum undulatum L. depends on storage temperature (-20, $5^{\circ}C$ and room temperature), germination temperature (5, 15, 25 and $40^{\circ}C$) and a soaking treatment (no soaking, 2 days and 4 days). There was not much difference from the germination rate of R. undulatum L. in storage temperature conditions for 8 weeks. The highest of germination rate occurred over 80 percentage at $25^{\circ}C$. When R. undulatum L. was soaked with tap water for four days, the average values of germination velocity and germination day were represented of 1.7 ea/day and 1.68 days. These results may help our understanding of variations in germination characteristics for R. undulatum L. seeds treated under various germination conditions.

Priming Effect of Rice Seeds on Seedling Establishment under Adverse Soil Conditions

  • An experiment was carried out to find out the priming effects of rice seeds, Oryza sativa L. (cv. Ilpumbyeo) on. the seedling establishment and early emergence under excess soil moisture conditions. Seeds were primed by soaking in -0.6 MPa polyethylene glycol (PEG) solution at $25^{\circ}C$ for 4 days. The primed seeds were sown in soils with various soil moistures (60, 80, 100, 120, and 140% field capacity) at 17 and $25^{\circ}C$, respectively. Germination and emergence rates, plumule height, and radicle length of primed seeds were higher than those of untreated seeds at any soil moisture and temperature examined. The time from planting to 50% germination ($T_{50}$) of primed seeds was less than that of untreated seeds by 0.9~3.7 days. Germination rate, emergence rate, plumule height, and radicle length were highest at the soil moisture of 80% field capacity among the soil moistures. Priming effects of rice seeds on germination and emergence rates were more prominent under the unfavorable soil moistures (60, 100, 120, and 140% field capacity) than those under the optimum soil moisture condition (80% field capacity). However, priming effects on seedling growth were greater at near optimum soil moisture compared with too lower or higher soil moistures. Therefore, these findings suggest that priming of rice seeds may be a useful way for better seedling establishment under the adverse soil conditions.

Seed Gravity, Germination, and Optimum Seeding Rate for Machine Transplanting in Colored Rice Varieties (유색벼의 품종별 종자 비중, 발아 특성 및 기계이앙 육묘시 적정 파종량)

  • Seeds of colored rice generally have high anthocyanin and flavonoid content but some cultivars have low seed germination rate. This defect poses unstable seedling establishment in nursery bed. The seed gravity variations of 10 colored rice varieties and its effect on germination, seed soaking duration and their optimum seeding rates for seedling raising in machine transplanting were investigated in the experiment. Based on seed gravity distribution of the 7 black- and 3 red-colored rice varieties at three levels of seed gravity (1.0<, 1.0~1.06, 1.06>), the black colored seeds such as Josaengheugchal and Sinmyungheugchal had high amount of specific gravity of below 1.0 ranging from 86~ 96%, while they had only 3~13% of seeds in specific gravity above 1.06. Sintoheugmi, Heugjinju, Heugnam, Heugkwang had various mass of seeds, showing 29~44% for specific gravity of below 1.0, 24~39% for 1.0~1.06 and 25~45% for above 1.06. On the other hand, the red colored rice such as Hongjinju, Jeogjinju and Geonkanghongmi had high percentage of specific gravity of above 1.06 with 84~86% while they had only 9~12% of specific gravity below 1.0 similar to Ilmibyeo of noncolored rice. The black colored seed generally showed low germination percentage, slow germination speed and long mean germination time, and low water absorption rate as compared with seed of the red colored rice which was similar to those of Ilmibyeo used as a control cultivar. The black colored seeds took 2~4 days longer seed soaking duration than the red colored rice before germination. This was related to high seed amount of specific gravity below 1.0 in black colored rices. The high amount of seed gravity above 1.06 in the colored rice seeds was positively correlated with percentage of germination, germination speed and mean germination time, ripened grain ratio and water absorption. Seed gravity distribution effect were less pronounced between back- and red- colored seeds in seedling emergence. The black colored rice has slightly lower normal seedling emergence rate than the red colored rice and Ilmibyeo in the seedbed soil due to high percentage of ungerminated seed. Normal seedling emergence rate of the black colored rice in the seedbed was 75.2~82.2% for 10-day old seedling and 85.3~ 90.9% for 30-day old seedling which was lower by 4.5~ 8.0% and 0~3.3%, respectively, than the red colored rice. Based on the normal seedling number per tray of Ilmibyeo for seeding rate of 10-day and 30-day old seedlings, the recommended seeding rate of black colored rice is 200~ 220g seeds for the 10-day old seedling and 110~130g for the 30-day old seedling in transplanting rice while the seeding rate of the red colored rice was 220g seeds for the 10-day old seedling and 130g for the 30-day old seedling.

Morphological Characteristics and Germination Conditions of Seeds in Arabis pendula L. (늘어진장대(Arabis pendula L.) 종자의 형태적 특성 및 발아조건)

  • This study aimed to investigate the morphological characteristics and optimal germination conditions in seeds of Arabis pendula L., a traditionally edible and medicinal plant. The external seed shape was circular-obovate with narrow wings and dark brown. The seed length and width were 1.25 mm and 1.47 mm, respectively. The seeds were exalbuminous and the embryo was a bent type. Seed germination was the highest (49.7%) at $20^{\circ}C$ under dark conditions among the various temperature and light conditions applied. However, under the dark condition, the seedling was weak, overgrown, and the cotyledons were small and folded. To improve the germination and growth of seedlings, the seeds were pre-treated with $GA_3$ solutions of varying concentrations (0, 200, 500, and 1,000 mg/L). The seed germination and seedling growth were effectively improved by $GA_3$ pre-treatment. The germination rate was the highest (97.3%), mean germination time was the shortest (8.1 days), and a vigorous growth of seedlings was observed upon pre-soaking the seeds in 500 mg/L $GA_3$ solution. In conclusion, the best method for germination was pre-soaking in 500 mg/L $GA_3$ ($4^{\circ}C$, dark, 24 h) and incubating the seeds at $20^{\circ}C$ for 15 days.

Nondestructive Vigor Test of Seed (비파괴적 방법에 의한 종자의 활력 검정)

  • Experiments were conducted to obtain information on separation of nonviable seeds from seed lots of rape, Chinese cabbage and radish, by nondestrutive ways. Seeds were artificially aged at 90% relative humidity(RH) and 45$^{\circ}C$ to get different seed qualities. Large amounts of amino acids, proteins and sinapine were leaked from the dead seeds into water in the course of 4~8 hours soaking, while high quality seeds did not leak in all crops. Percentage of normal seedlings from nonfluorescent cellulose coated seeds of rape, Chinese cabbage and radish under ultraviolet light was 96, 96 and 74%, while that of fluorescent seeds was 8, 9 and 1%, respectively. Cellulose coating and storage of Chinese cabbage and rape seeds at 60 and 75% RH for 3 months and 90% RH for 2 months did not deteriorate seed quality. But percentage of normal seedlings from nonfluorescent radish seeds stored at 60, 75 and 90% RHs for 3 months was 63, 64 and 2%, respectively.

Effects of Seed Soaking Treatment of Diniconazol on the Inhibition of Stretching of Tomato and Cucumber Seedlings (Diniconazol의 종자침지 처리가 토마토와 오이 플러그묘의 도장억제에 미치는 영향)

  • This study was conducted to find out the effect of diniconazole treatment by seed soaking on the stretching of tomato and cucumber seedlings. The emergence rate of tomato and cucumber seed was decreased as diniconazole concentration was higher and soaking period was longer. The emergence rate of tomato seedlings was lower as the soaking period was longer in $1mg{\cdot}L^{-1}$ concentration treatments, although there was no significant difference. The other concentration treatment (10, 50, and $100mg{\cdot}L^{-1}$) reduced emergence rate of tomato seedlings outstandingly in higher concentration and longer treated period, but 90% of treated seeds emerged at 10 days after sowing. As the concentration of diniconazol was higher and soaking period was longer, plant height and growth rate of seedlings were retarded. There covery of internodes growth was faster in lower concentration and shorter soaking period. The top and root fresh weight and dry weight of tomato and cucumber seedlings was not influenced by $1mg{\cdot}L^{-1}$ concentration, but these growth characteristics were reduced in higher concentration and longer soaking period. The dwarfed cucumber seedlings treated by diniconazol showed normal growth rate after planting and their male and female flower was set normally. These results suggest that the proper concentration and soaking period of diniconazol may be $10mg{\cdot}L^{-1}$ concentration for 12 hours in tomato seed, and $1mg{\cdot}L^{-1}$ concentration for 24 hours in cucumber seed.

Effect of NaCl on Germination of Artiplex gmelini and Pharagmites communis (갈대 및 가는갯능쟁이의 발아에 미치는 NaCl의 효과)

  • The results of the conducted experiment obtained basic data on seed germination for Phragmites communis and Atriplex gmelini were; Seed germination was not influenced till 0.5% NaCl, but at over 1.0% NaCl it dropped remarkably. The germination limit fro degree of NaCl was 2.0% in Phragmites communis. Atriplex gemelini was 2.5% and in accordance with the increase of the degree of NaCl. Germination speed showed a negative correlation being highly significant and the germination period lengthened. Compared with growing top plants, growth of roots was largely influenced by a high degree of NaCl. In accordance with the rise of temperature, the germination rate, and speed of both plants remarkably increased and the germination period was shortened. Ubride of Atriplex gmelini was germinated at the early days of picking but was not as the passing of the period. The seeds also did not germinated likewise Ubride. By a seedcoat breaking germination became 81%. During 20 min soaking treatment in conc H2SO4 seed germination possibility of 63% was known to be hard. Adequate soaking time in conc H2SO4 was 17.5 min.

Effects of Seed Pretreatment and Environment Controls on Germination of Atractylodes japonica Seeds (종자 전처리 및 환경조절이 삽주 종자 발아에 미치는 영향)

  • This study was carried out in order to examine the germination characteristics of Atractylodes japonica seeds, and it was to develop the more efficient pre-treatment and production system of the seeds. Experiment was performed by two ways-temperature control (10, 15, 20, $25^{\circ}C$) and shading treatment (full sunlight, 35, 50, 75, 95% of full sunlight). Seed pre-treatment before the each experiment was carried out by temperature (with low temperature and wetting treatment (LTW) for 0, 15, 30, 45 and 60 days) and shading treatment (with drying at room temperature (DRT), drying at low temperature (DLT) and water soaking (WS) for 48 hours). Seeds of A. japonica were germinated well under temperature control (overall 85.3~100%), especially high temperature. And, the seeds pre-treated with LTW for 45 days germinated with a 100% success rate. Under the surveying shading treatment, the highest germination rate was 95.8% with DRT and overall 63.2~95.8%. Germination rate under seeds with WS was 63.2~7.8%. As a result of surveying the whole experiment, A. japonica seeds don't need to pre-treat cause of high germination, but if it is performed with LTW for a certain period of time, it would be more productive.
