효과적인 효소 소화율을 위한 볏짚 전처리 (Pretreatment of Rice Straw for Efficient Enzyme Digestibility)

  • Rice straw was pretreated with aqueous ammonia in order to enhance enzyme digestibility. Soaking in ammonia aqueous (SAA) was conducted with 15% ammonia, at $60^{\circ}C$. for 24 h. Optimization of both saccharification conditions and enzyme loading of SAA rice straw was carried out. Especially enzyme loading test was performed using statistical method. Moreover proton beam irradiation (PBI) was also performed to overcome the problem which inhibit the enzyme digestibility at 1-25 kGy doses with 45 MeV of beam energy. Optimal condition for enzymatic saccharification was follows; pH 4.8, $50^{\circ}C$, 60 FPU of enzyme activity, 1:4 ratio of celluase and ${\beta}$-glucosidase. Also, optimal doses of PBI on rice straw and SAA-treated rice straw for efficient sugar recovery were found to be 3 kGy, respectively. When saccharification was performed with optimal condition, glucose conversion yield was 89% of theocratical maximum in 48 h, and 3 kGy of PBI was applied to SAA-treated rice straw, approximately 90% of the theoretical glucose yield was obtained in 12 h. The results of X-ray diffractometry (XRD) support the effect of both SAA and PBI on sugar recovery, and scanning electron microscopy (SEM) images unveiled the physical change of the rice straw surface since rugged rice straw surface was observed.

Effect of Pre-Sowing Treatments on Germination and Initial Seedling Growth of Castanopsis Indica- An Endangered Tree Species in Bangladesh

  • Castanopsis indica (Sil Batna) is an ecologically valuable multipurpose indigenous tree species of Bangladesh. Considering its high value but poor natural regeneration due to seed dormancy, the authors conducted an experiment at the Institute of Forestry and Environmental Sciences Chittagong University (IFESCU) during 2012-2013 to find out effective pre-sowing treatments. Ten pre-sowing seed treatments were applied on mature, even-sized seeds namely-control; sandpaper rubbing; nail clipping; immersion in normal water (at room temperature: 24℃) for 24 hrs., 48 hrs. and 72 hrs.; immersion in hot water (80℃) for 1 minute; soaking in 10% dilute H2SO4; soaking in 10% HCl for 5 minutes; and sowing in propagator house. Seeds sown after sandpaper rubbing at the distal end revealed best performances by providing highest germination percent (66.7%), germination energy (30%), germination index (0.17), germination rate (0.0145), germination value (30%) and plant percent (66.7%) within shortest period (38 days). The treatment also produced most vigor seedlings with 20.9 cm shoot height, 15 node number, and largest leaves (11.1 cm×2.9 cm). Hence, it is recommended to adopt sandpaper rubbing method for maximum germination and quality seedlings.

Effects of Chitosan on Production and Rot Control of Soybean Sprouts

  • The practicality of utilizing chitosan as a natural antimicrobial compound to reduce soybean sprout rot was tested. Domestic and imported soybean seeds were soaked for 6 hours in solutions containing different levels of chitosan and acetic acid (glacial), and cultured at $25^{\circ}C$ for 6 days. In case of domestic seeds, soaking with 1,000ppm chitosan increased germination percentage, hypocotyl thickness, total length, and fresh weight of sprouts by 4%, 5%, 2%, and 1%, respectively. The total sprout yield was increased by chitosan in a concentration-dependent manner in that 1,000ppm chitosan resulted in 8% increment of total yield (7.47kg sprouts/kg seed). Chitosan significantly reduced sprout rot percentage to 7.0% compared to control (13.8%), and consequently enhanced marketable sprout yield by 39%. Compared to domestic seeds, the imported soybean seeds exhibited very low germination percentage regardless of chitosan treatments. Chitosan, nevertheless, consistently induced yield increment and rot decrement in imported soybean sprouts. Although 100ppm acetic acid was effective in reducing sprout rot percentage down to 11.8%, its yield-increasing effects were not as prominent as chitosan. In conclusion, soaking soybean seeds with chitosan seems to be a practical method to enhance the efficiency of soybean sprout production.

  • A serious problem of the 21st century is the supply of energy resources. Reserves of fossil fuels are facing depletion: renewable energy resources must be developed in this era. Dye sensitizedsolar cells(DSC) have been very economical and easy method to convert solar energy to electricity. DSC can reach low costs in future outdoor power applications. However, to commercialize the DSC, there are still many shortages to overcome. When the DSC is commercialized in the near future, the productivity is an important factor. In the process of soaking in a dye, it usually takes $12\sim24$ hours. In this study, we varied the dye coloring temperature from $0^{\circ}C$ to $60^{\circ}C$. At the temperature of $40^{\circ}C$, DSC cell showed the best performance. We also expect the reduction of the time soaking in a dye. Counter electrode surface of DSC is deposited by RF magnetron sputtering under the conditions of Ar $2.8{\times}10^{-3}$ torr, RF power of 120W and substrate temperature of $100^{\circ}C$.

  • This report described the corrosion structure of excavated iron artifacts in terms of simple model based on the knowledge of the corrosion process. (Fig.1)(Table 1,2)In storing the objects, there are basically three ways in which they either break in wedges, flakes and dish-shaped flakes. Completely mineralized objects or those with only a small iron core tend to break into wedges and more solid objects either split small dish-shaped flakes or large flat ones.(Fig. 2,3,4)There are two ways, therefore, to prevent this from happening. One is to keep the artifacts rigorously dried in Silica-gel, never allowing the relative humidity to rise. This is feasible which the artifacts are in store but causes great difficulty if they are wanted for museum display. Because they still contain $ FeCl _2$ they are always at risk ; they contain the seeds of their own destruction. The other alternative is to use of washing process to dissolve out the $ FeCl _2$. In this connection, many different methods to stabilize the artifact have been employed; boiling iron in frequent changes of water, soaking in Na-sesquicarbonate solution, soaking in alkaline Na-sulphite solution. In this report, introduced the alkaline sulphite method by the N.A. 치수안정화를 위한 목질재료의 고분자 하이브리드화에 관한 연구 (Study on the Hybridization of Polymer-macromers for the Dimensional Stabilization of Woody Materials)

  • This study was carried out to experiment the dimensional stabilization of woods of red pine (Pinus densiflora S. et Z.) and sugi(Cryptomeria japonica D. Don) by vacuum impregnation of polyehtylene glycols(PEG) with mo. wt. 200, 400, 600, 1000; polypropylene glycols (PPG) with mo. wt. 425,725 ; PEG-acryloylates, and PPG-acryloylates synthesized, and then by water soaking. The results obtained are as follows: 1. The density of sapwood and heartwood was different from each other in both species. 2. The PEG and PEG-macromers with lower molecular weight by impregnation has increased the density of wood specimens more higher, thereby caused their higher volume expansion, and those with higher molecular weight than 600 has tended to down their density increment. 3. Before and after water soaking, the density decrease of specimen impregnated was high in woods impregnated with simple PEG and PPG, while lower in specimens impregnated with PEG-macromers and PPG-macromers. 4. So PEG-macromer was expected to hold the original dimension of decayed wood for antiques, but it was necessary to develop another penetration method as well as aqueous solvent.

조선전기 고문헌에 수록된 식품저장법에 대한 문헌고찰 (Review of Literature on Food Preservation of the Early Joseon Dynasty)

  • This study aims to investigate the food preservation methods adopted by the Joseon Dynasty, which existed before the 17th century. A total of 232 food preservation methods were discovered in 25 books, and could be classified by their targeted food types: vegetables (84), sea foods (60), meats (41), fruits (37), and others (10). Depending on the preservation method applied, they are classified as food drying, soaking, mud cellar preservation, and other preservation. Food drying is further classified into 8 sub-types: drying, sun-drying, shadow-drying, wind-drying, dry heat, combined drying, smoking, and others. Soaking could be sub-divided into using salt, ash, dry sand, bran, fermented paste, wet distillers grains, oil, and others. Mud cellar preservation is sub-classified into installing shelf inside the mud cellar, making the mud cellar for food preservation, and making hole or underground tunnel for food preservation. Other food preservation methods include minimizing moisture loss by applying beeswax on a section of the vegetable stem, and cutting the vegetables or fruits with their branches and leaves for food preservation.

Efficiency of transformation mediated by Agrobacterium tumefaciens using vacuum infiltration in rice (Oryza sativa L.)

  • Agrobacterium-mediated gene transfer has recently been developed to improve rice transformation. In this study, 3 different transformation methods were tested including soaking, co-cultivation, and vacuum infiltration. Agrobacterium tumefaciens GV3101 harboring the binary vector pGreen:: LeGSNOR was used in this experiment. This study aimed to identify the most appropriate method for transferring LeGSNOR into rice. Vacuum infiltration of the embryonic calli for 5 min in Ilpum resulted in high transformation efficiency based on confirmation by PCR, RT-PCR, and qRT-PCR analyses. In conclusion, we described the development of an efficient transformation protocol for the stable integration of foreign genes into rice; furthermore, the study results confirmed that PCR is suitable for efficient detection of the integrated gene. The vacuum infiltration system is a potentially useful tool for future studies focusing on transferring important genes into rice seed calli, and may help reduce time and effort.

감자의 전처리 방법에 따른 환원당 함량과 potato chip의 색상 (Reducing sugar contents of potato tubers and potato chip color by pretreated methods)

  • 우리나라에서 널리 보급되어 재배되고 있는 감자 품종인 수미(秀美, Superior) 및 남작(男爵, Irish Cobbler) 두 품종을 원료로 potato chip을 제조할 때 품질에 직접적으로 영향을 미치는 Maillard 반응의 중요한 요인으로 작용하는 환원당의 함량을 감소시키기 위하여 화학적 저해제로 알려진 $CaCl_2$와 NaCl을 사용하여 감자를 전처리한 후 환원당 함량을 분석하고 potato chip을 제조하여 색상을 비교한 결과 NaCl로 침지 처리한 경우에는 환원당의 감소가 $15{\sim}30%$에 불과하고 chip의 색상도 좋지 못하였다. 그러나 NaCl 침지 가열처리의 경우에는 환원당의 감소가 평균 40%였으며, 특히 두 품종 모두 0.08% NaCl 용액으로 5분 및 7분간 처리한 것이 좋은 효과를 나타내었다. $CaCl_2$ 용액에 침지처리한 것은 NaCl 용액으로 처리한 것보다 효과는 있었으나 색상이 고르지 못하고 부분적으로 갈색화되었다. $CaCl_2$용액에 침지 가열처리한 것이 가장 좋은 결과를 나타내어 환원당의 감소가 약 50% 정도였으며 그 중에서도 0.05% 및 0.07% $CaCl_2$ 용액에 5분 및 7분간 처리한 것이 환원당의 감소는 물론 chip의 색상도 가장 좋았다. 침지시 상온에서 처리하는 것보다는 $72{\sim}82^{\circ}$ 에서 NaCl 용액 보다는 $CaCl_2$ 용액으로 blanching하는 것이 가장 효과적이었다.

상어육(肉)중의 수분유지제(水分維持劑) 침투(浸透) (Penetration of Some Humectants into Shark Muscle)

  • 상어육을 시료로 하여 sucrose, sorbitol, glycerol, propylence glycol등 4 종류의 수분유지제(水分維持劑) 용액 중에서 침투량을 측정하였다. 또한 침투에 미치는 수분유지제(水分維持劑) 농도 및 온도의 영향을 검토하고 침투속도식을 유도하였다. 상어육을 10% 수분유지제(水分維持劑) 용액에 침지할 때는 10시간 전후, 30% 용액에서는 7 시간 정도에서 거의 평형에 도달하였다. 수분유지제(水分維持劑) 종류에 따른 침투 속도는 분자량이 작을수록, 온도가 높을수록 빨랐다. 그리고한종류의 수분유지제(水分維持劑)를 10%로 하여 침지할 경우와 여러종류의 수분유지제(水分維持劑)를 각각 10% 되도록 혼합한 용액에 침지할 경우 침투속도에는 차이가 없었다. 상어육을 10% 및 30% 용액에 침지할 경우 수분유지제(水分維持劑) 침투량은 다음의 회귀직선식으로 예측가능 하다는 결론을 얻었다. $$M\;=\;a\;{\log}(c{\cdot}t)\;+\;b$$ (M : 침투량, t : 침지시간, c : 수분유지제(水分維持劑)농도, a, b : 상수)
