The Nutritive Value of Thin Stillage and Wet Distillers' Grains for Ruminants - Review -

  • Thin stillage and distillers' grains are byproducts remaining after alcohol distillation from a fermented cereal grain mash. Both byproducts are used as energy and protein sources for ruminants. Due to its liquid nature, more than 50% of thin stillage bypasses the rumen. Thin stillage can be fed alone or in combination with distillers' grains. However, a better utilization by beef cattle is anticipated when thin stillage replaces water as a fluid source. Ruminal undegraded protein content of distillers' grains is greatly affected by type of cereal grain and by drying. Corn distillers' grains have a higher ruminal undegraded protein content than wheat distillers' grains while dried distillers' grains have a higher ruminal undegraded protein content than the wet distillers' grains. Wet and dried distillers' grains can replace up to 50% of corn grain in beef cattle diets without affecting animal performance. The estimated NEg of corn distillers' grains for beef cattle ranges from 100 to 169% of that of corn. In general, wet corn distillers' grains have a higher NEg value than dried corn distillers' grains and the addition of thin stillage improves the NEg of distillers' grains. Improved performance of ruminats fed distillers' byproducts can be attributed to high digestible fiber content, improved rumen environment and a shift in organic matter digestion from the rumen to the small intestine.

Cost-effective isotope labeling technique developed for 15N/13C-labeled proteins

  • A newly developed cost-effective approach to prepare $^{15}N/^{13}C$-labeled protein for NMR studies is presented. This method has been successfully applied to isotopically labeling of PTK6 SH2 domain and MTH 1880 protein. The production method generates cell density using a growing media containing $^{15}NH_4Cl$, $^{12}C_6$-D-glucose. Following a doubling time period for unlabeled metabolite exhaustion and then addition $^{13}C_6$-D-glucose into a M9 growing media, the cells are induced. Our results demonstrate that in order to get full incorporation of $^{13}C$, the isotopes are not totally required during the initial growth phase before induction. The addition of small amounts of $^{13}C_6$-D-glucose to the induction phase is sufficient to obtain more than 95% incorporation of isotopes into the protein. Our optimized protocol is two-thirds less costly than the classical method using $^{13}C$ isotope during the entire growth phase.

  • Extract of Japanese red pine needles has been used in Asia pacific regions since long periods believing its valuable properties as tonic and ability of curing diseases of unidentified symptoms. Some selective compounds present in the extract and their effects were analyzed. Carbohydrates and vitamin c were identified using HPLC; terpenoid compounds by GC-MS; anti-bacterial analysis by paper discs, plates count and gastrointestinal motility by whole cell patch clamp. The extract is a mixture of compounds therefore its diverse effect was expected. Self-fermentation in extract proceeds after spontaneous appearance of yeast strains without inoculation. Effects and composition of the extract vary with varying period of self-fermentation. Extract inhibits the growth of bacteria dose dependently exhibiting its antibacterial properties however effectiveness increases with increase in fermentation period. The extract also can modulate gastrointestinal motility in murine small intestine by modulating pace maker currents in ICC mediated through ATP sensitive potassium channel.

Effects of Curcumin, the Active Ingredient of Turmeric(Curcuma longa), on Regulation of Glutamate-induced Toxicity and Activation of the Mitogen-activated Protein Kinase Phosphatase-1 (MKP-1) in HT22 Neuronal Cell

  • Glutamate causes neurotoxicity through formation of reactive oxygen species and activation of mitogen-activated protein kinase (MAPK) pathways. MAPK phosphatase-1 (MKP-1) is one of the phosphatases responsible for dephosphorylation/deactivation of three MAPK families: the extracellular signal-regulated kinase-1/2 (ERK-1/2), the c-Jun N-terminal kinase-1/2 (JNK-1/2), and the p38 MAPK. In this report, the potential involvement of MKP-1 in neuroprotective effects of curcumin, the active ingredient of turmeric (Curcuma longa), was examined using HT22 cells. Glutamate caused cell death and activation of ERK-1/2 but not p38 MAPK or JNK-1/2. Blockage of ERK-1/2 by its inhibitor protected HT22 cells against glutamate-induced toxicity. Curcumin attenuated glutamate-induced cell death and ERK-1/2 activation. Interestingly, curcumin induced MKP-1 activation. In HT22 cells transiently transfected with small interfering RNA against MKP-1, curcumin failed to inhibit glutamate-induced ERK-1/2 activation and to protect HT22 cells from glutamate-induced toxicity. These results suggest that curcumin can attenuate glutamate-induced neurotoxicity by activating MKP-1 which acts as the negative regulator of ERK-1/2. This novel pathway may contribute to and explain at least one of the neuroprotective actions of curcumin.

Isolation of whey protein and hydrolysis pattern of whey protein by proteolytic enzyme (유청단백질의 분리 및 단백질 분해 효소에 의한 유청단백질의 가수분해 양상)

  • The aim of this study was to introduce a simple method for isolation of ${\alpha}$-lactalbumin, ${\beta}$-lactoglobulin and bovine serum albumin from cow's milk, and peptides produced by enzymatic hydrolysis of ${\alpha}$-lactalbumin, ${\beta}$-lactoglobulin and bovine serum albumin with alcalase. Whey protein were precipitated from whey by ammonium sulfate and, ${\alpha}$-lactalbumin and ${\beta}$-lactoglobulin were isolated using Hi Prep 26/60 Sephacryl S-100 column gel filtration chromatography. Bovine serum albumin and ${\beta}$-lactoglobulin were isolated by Mono-Q 5/50 GL column anion exchange chromatography of the 50% Ammonium Sulfate-supernatant. Isolated whey proteins were hydrolyzed by proteolytic alcalase. Tricine SDS-PAGE and reverse-phase HPLC analyses revealed that almost hydrolyzed all the ${\alpha}$-lactalbumin, ${\beta}$-lactoglobulin and bovine serum albumin with alcalase. Molecular weight of various peptides derived from alcalase hydrolysate were small molecular weight than 3.5 kDa.

Sequence Homologies of GTP-binding Domains of Rab and Rho between Plants and Yeast/Animals Suggest Structural and Functional Similarities

  • Small GTP-binding proteins are divided into three major group: Ras, Rho and Ypt/Rab. They have the conserved regions designed G1 to G5 that are critical in GDP/GTP exchange, GTP-induced conformational change and GTP hydrolysis. We isolated and characterized genomic DNA or cDNAfragments encoding G1 to G3 domains of small GTP-binding protein Rab and Rho from several plant species using two different PCR-based cloning strategies. Seven rab DNA fragments were isolated from 4 different plants, mung-bean, tobacco, rice and pepper using two degenerate primers corresponding to the GTP-binding domain G1 and G3 in small GTP-binding proteins. The amino acid sequences among these rab DNA fragments and other known small GTP-binding proteins shows that they belong to the Ypt/Rab family. Six rho DNA fragments were isolated from 5 different plants, mung-bean, rice, Arabidopsis, Allium and Gonyaulax using the nested PCR method that involves four degenerate primers corresponding to the GTP-binding domain G1, G3 and G4. The rho DNA fragments cloned show more than 90% homology to each other. Sequence comparison between plant and other known Rho family genes suggests that they are closely related (67 to 82% amino acid identity). Sequence analysis and southern blot analysis of rab and rho in mung-bean suggest than thses genes are encoded by multigene family in mung-bean.

Malignant gliomas can be converted to non-proliferating glial cells by treatment with a combination of small molecules

  • Gliomas, the most highly malignant central nervous system tumors, are associated with an extremely poor patient survival rate. Given that gliomas are derived from mutations in glial precursor cells, a considerable number of them strongly react with glial precursor cell-specific markers. Thus, we investigated whether malignant gliomas can be converted to glial cells through the regulation of endogenous gene expression implicated in glial precursor cells. In the present study, we used three small-molecule compounds, [cyclic adenosine monophosphate (cAMP) enhancer, a mammalian target of rapamycin (mTOR) inhibitor, and a bromodomain and extra-terminal motif (BET) inhibitor] for glial reprogramming. Small-molecule-induced gliomas (SMiGs) were not only transformed into exhibiting a glial-specific morphology, but also showed positive reactions with glial-specific markers such as glial fibrillary acidic protein (GFAP), 2',3'-cyclic nucleotide 3'-phosphohydrolase (CNP) and anti-oligodendrocyte (RIP). A microarray analysis indicated that SMiGs exhibited a marked increase in specific gene levels, whereas that of a malignant cancer-specific gene was greatly decreased. Moreover, proliferation of the cells was markedly suppressed after the conversion of malignant glioma cells into glial cells. Our findings confirmed that malignant gliomas can be reprogrammed to non-proliferating glial cells, using a combination of small molecules, and their proliferation can be regulated by their differentiation. We suggest that our small-molecule combination (with forskolin, rapamycin and I-BET151) may be the next generation of anticancer agents that act by reprogramming malignant gliomas to differentiate into glial cells.

Gastrointestinal Stromal Tumor with Extensive Lymphatic Metastasis: A Case Report

  • Gastrointestinal stromal tumor is a rare tumor which arises from the whole gastrointestinal tracts and most of it is detected in the stomach. It is uncommon with small intestine originated gastrointestinal stromal tumor and more uncommon with lymphatic metastasis. We experienced an unusual case of the small bowel gastrointestinal stromal tumor during experimental autopsy. Two primary tumors with central necrosis were detected in the ileum. The sizes of each tumor were $6.1{\times}3.4{\times}4.0$ cm and $3.7{\times}4.2{\times}3.2$ cm. There was extensive lymphatic metastasis on the greater omentum and mesenteric, iliac lymph nodes were also involved. With histologic findings, the eosinophilic spindle cells were densely distributed. Immunohistochemical findings were CD117 (-), CD34 (+), desmin (-), and S-100 protein (-). Therefore, we diagnosed the tumors as small bowel gastrointestinal stromal tumors with broad lymph node mestasis.

FEED RESOURCE AVAILABILITY AND UTILIZATION IN SMALLHOLDER PIG FARMS IN SRI LANKA

  • Data on available feed resources, feeding practices and nutrient adequacy of rations under small farm conditions in Sri Lanka were obtained in a baseline survey involving 104 pig farms. The results showed that a wide range of non-conventional feedstuffs are used for pig feeding under typical small farm conditions and that dietary protein quality is a major factor limiting productivity. Following the survey, two on-farm trials were conducted to evaluate cheaper, alternative feeding strategies. In trial 1, a test diet was formulated using several non-conventional feedstuffs and compared with a commercial feed that is normally fed in the farms. In trial 2, the possibility of improving growth rates by amino acid supplementation was evaluated. The results demonstrated that feed costs can be considerably lowered through these packages. Some problems inherent to on-farm livestock trials are highlighted.

Microwave-enhanced gasification of sewage sludge waste

  • To convert sewage sludge to energy, drying-gasification characteristics during microwave heating were studied. During the gasification of carbon dioxide, the main products were gas, followed by char, and tar in terms of the amount. The main components of the producer gas were carbon monoxide and hydrogen including a small amount of methane and light hydrocarbons. They showed a sufficient heating value as a fuel. The generated tar is gravimetric tar, which is total tar. As light tars, benzene (light aromatic tar) was a major light tar. Naphthalene, anthracene, and pyrene (light polycyclic aromatic hydrocarbon tars) were also generated, but in relatively small amounts. Ammonia and hydrogen cyanide (precursor for NOx) were generated from thermal decomposition of tar containing protein and nitrogen in sewage sludge. In the case of sludge char, its average pore diameter was small, but specific area, pore volume, and adsorption amounts were relatively large, resulting in superior adsorption characteristics.
