The objective of this study was to prove the effect of pig slurry application with charcoal on nitrogen use efficiency (NUE), feed value and ammonia (NH3) emission from maize forage. The four treatments were applied: 1) non-pig slurry (only water as a control), 2) only pig slurry application (PS), 3) pig slurry application with large particle charcoal (LC), 4) pig slurry application with small particle charcoal (SC). The pig slurry was applied at a rate of 150 kg N ha-1, and the charcoal was applied at a rate of 300 kg ha-1 regardless of the size. To determine the feed value of maize, crude protein, dry matter intake, digestible dry matter, total digestible nutrient, and relative feed value were investigated. All feed value was increased by charcoal treatment compared to water and PS treatment. Also, the NUE for plant N was significantly higher in charcoal treatments (LC and SC) compared to PS treatment. On the other hand, there is no significant difference for feed value and NUE between LC and SC. The NH3 emission was significantly reduced 15.2% and 27.9% by LC and SC, respectively, compared to PS. Especially, SC significantly decreased NH3 emission by 15% compared to LC. The present study clearly showed that charcoal application exhibited positive potential in nitrogen use efficiency, feed value and reducing N losses through NH3 emission.

Background: Congenital portosystemic shunt (cPSS) is one of the most common congenital disorders diagnosed in dogs. Hepatic encephalopathy (HE) is a frequent complication in dogs with a cPSS and is a major cause of morbidity and mortality. Despite HE been a major cause of morbidity in dogs with a cPSS, little is known about the cellular changes that occur in the central nervous system of dogs with a cPSS. Objectives: The objective of this study was to characterise the histological changes in the cerebral cortex and cerebellum of dogs with cPSS with particular emphasis on astrocyte morphology. Methods: Eight dogs with a confirmed cPSS were included in the study. Results: Six dogs had substantial numbers of Alzheimer type II astrocytes and all cases had increased immunoreactivity for glial fibrillary acidic protein in the cerebral cortex, even if there were minimal other morphological changes. Conclusions: This study demonstrates that dogs with a cPSS have marked cellular changes in the cerebral cortex and cerebellum. The cellular changes that occur in the cerebral cortex and cerebellum of dogs with spontaneously arising HE are similar to changes which occur in humans with HE, further validating dogs with a cPSS as a good model for human HE.
Tumor necrosis factor alpha (TNF-α) is a principal regulator of inflammation and immunity. The proinflammatory properties of TNF-α can be attributed to its ability to activate the enzyme cytosolic phospholipase A2 (cPLA2), which generates potent inflammatory lipid mediators, eicosanoids. L-glutamine (Gln) plays physiologically important roles in various metabolic processes. We have reported that Gln has a potent anti-inflammatory activity via rapid upregulation of mitogen-activated protein kinases (MAPKs) phosphatase (MKP)-1, which preferentially dephosphorylates the key proinflammatory enzymes, p38 MAPK and cytosolic phospholipase A2 (cPLA2). In this study, we have investigated whether Gln could inhibit TNF-α-induced cPLA2 activation. Gln inhibited TNF-α-induced increases in cPLA2 phosphorylation in the lungs and blood levels of the cPLA2 metabolites, leukotrine B4 (LTB4) (lipoxygenase metabolite) and prostaglandin E2 (PGE2) (cyclooxygenase metabolite). TNF-α increased p38 and cPLA2 phosphorylation and blood levels of LTB4 and PGE2, which were blocked by the p38 inhibitor SB202190. Gln inhibited TNF-α-induced p38 and cPLA2 phosphorylation and production of the cPLA2 metabolites. Such inhibitory activity of Gln was no longer observed in MKP-1 small interfering RNA-pretreated animals. Our data indicate that Gln inhibited TNF-α-induced cPLA2 phosphorylation through MKP-1 induction/p38 inhibition, and suggest that the utility of Gln in inflammatory diseases in which TNF-α plays a major role in their pathogenesis.

Yeast strains are capable of hydrolyzing non-digestible saccharides, such as melibiose, raffinose, and stachyose, found in soy meal components. This study revealed the biochemical properties of fermented soybean meal during 72 hours with kefir. Starchyose and raffinose, non-digestible components, were almost digested in kefir 150 mL + soybean meal 500 g + water 70 mL and galactose was produced. Proteolysis of the soybean meal produced most of the small molecule peptides in kefir 150 mL + soybean meal 500 g + water 70 mL. The production of the vitamin B group and C were the highest in kefir 250 mL + soybean meal 500 g. The yeast number of the fermented soybean meal was 7.0 × 106 CFU·mL-1 which was the highest in kefir 250 mL + soybean meal 500 g. The lactic acid bacteria of the fermented soybean meal was the highest at 3.5 × 109 CFU·mL-1 in kefir 70 mL + soybean meal 500 g. The antioxidant effect was the highest at 57% in kefir 250 mL + soybean meal 500 g. Expression of inflammation-related cytokine (interleukin [IL]-1β, tumor necrosis factor [TNF]-α, and interleukin [IL]-6) was significantly inhibited in fermented soybean meals with different treatments. These results suggest that fermented soybean meal by kefir has an antiinflammatory and anti-oxidation activity and could be utilized in feed manufacturing, and inhydrolyzing non-digestible soy meal components.
Objective: One hundred hybrid male ducks (Mojosari×Alabio) were used to examine the efficacy of chicory supplementation as nutritional feed manipulation on production performance, and blood lipid profile of hybrid ducks. Methods: The ducks were tagged, weighed, and then allotted randomly to one of the four treatment diets using a completely randomized design. The experimental diets were: i) P0 (100% basal diets+0% chicory as control), ii) P1 (95% basal diets+5% chicory), iii) P2 (90% basal diets+10% chicory) and iv) P3 (85% basal diets+15% chicory). For each treatment group, there were 5 replicates of 5 birds each. All experimental diets were isonitrogenous and isocaloric using locally available ingredients. Results: Hybrid ducks with fed diets supplemented fresh chicory (5%, 10%, and 15%) showed increased feed intake and body weight gain, as well as feed conversion ratio to be smaller than those ducks fed diets without chicory supplementation (control). The ducks fed 10% chicory supplementation contained significantly (p<0.05) lower ash and higher organic matter contents of meat than those ducks fed other diets. The ducks fed 15% chicory supplementation showed the lowest crude protein and cholesterol content of meat among the treatment diets. Ducks fed chicory supplementation showed lower (p<0.05) blood cholesterol and triglyceride levels than those ducks fed without chicory supplementation, while dietary interventions had no major (p>0.05) influence on low-density lipoprotein and high-density lipoprotein levels in duck blood. Conclusion: In this study, 10% chicory supplementation showed the best results characterized by an increase in growth performance, carcass quality, small intestinal histomorphology, and lower cholesterol levels of meat.

Using antibiotics as growth promoter has been banned in poultry feed industry, thus various researchers try to seek an alternative to replace the growth-promoting antibiotics. In this study, we aimed to evaluate the growth performance via intestinal nutrient utilization and cecal microbial composition of broiler after dietary supplementation with most commonly using antibiotics, zinc bacitracin, and sophorolipid. A total of 180 1-day-old chicks were randomly assigned, and dietary treatment was as follow: CON, basal diet; ZB, 100 ppm of zinc bacitracin supplemented diet; and SPL, 250 ppm of sophorolipid supplemented diet. Their growth performance was evaluated and the samples of blood, small intestine, and ileal and cecal digesta were collected for biochemical, histological, and genomic analyses. The body weight and average daily gain of 7-day-old chicks were higher in ZB and those in overall experimental period were improved by ZB and SPL supplementation (p < 0.05). Their intestinal characteristics were not affected by dietary treatments in duodenum and ileum. Nonetheless, villus height was increased by SPL supplementation in jejunum (p < 0.05). Moreover, dietary SPL supplementation could down-regulate the expression level of pro-inflammatory cytokine, IL-1β (p < 0.05). mRNA levels of lipid and protein transporters did not differ among the treatments, however, relative expression levels of carbohydrate transporters, GLUT2 and SGLT1 were increased in broiler chicken's jejumum fed zinc bacitracin and sophorolipid supplemented diets (p < 0.05). Dietary zinc bacitracin supplementation could increase the population of Firmicutes in phylum level, and the portion of Turiciacter in genus level. On the other hands, the portion of Faecalibacterium was increased by dietary SPL supplementation compared to the other treatments. Our findings suggest that SPL supplementation improves growth performance through enhanced carbohydrate utilization capacity via improvement of gut morphological status and modulation of the cecal microbial population of broilers.
Objectives: To confirm the antibacterial effects of each mouth rinse on multiple oral biofilms in vitro. Methods: The antibacterial effects of different mouth rinses were examined by ATP and counted colony forming units (CFU). Preformed oral biofilms on saliva coated hydroxyapatite (sHA) disks were treated with essential oil and saline; then, the multiple oral biofilms were observed by Scanning electron microscope (SEM). RNA sequencing analysis was performed on total RNA isolated from old biofilms of P. intermedia ATCC 49046. Results: In the CFU measured result compared to controls, preformed multiple oral biofilms were reduced from a low of 39.0% to 95.7% (p<0.05). The size of bacterial cells changed after treatment with the essential oil, and some of the cells ruptured into small pieces of cell debris. Gene expression in P. intermedia ATCC 49046 significantly altered in RNA transcribed and protein translated genes after exposure to essential oil. Conclusions: Mouth rinse solutions with different ingredients had different antibacterial effects and may alter surface structure and gene expression as determined by RNA sequencing.

Under normal conditions, women produce a single dominant follicle that participates in a single ovuation each menstrual cycle. But Polycystic ovary syndrome(PCOS) conditions, folliculogenesis does not proceed normally. This condition leads to the accumlation of large numbers of small graffian follicles in which the theca interstitial cells (TIC) produce abnormally large amounts of androgen. PCOS is probably the most common endocrine disorder, affecting women of reprodutive age with 5-10% prevalence estimate. Chronic anovulation, hyperandrogenism, hirsutism, obesity, infertility and polycystic ovaries are clinical hallmarks of women with PCOS. Its etiology remains unknown. To investigate the gene expression pattern of ovary in PCO-induced rat, we used cDNA expression analysis. Total RNA was extracted from the ovary of PCO-induced rat and reverse-transcribed in the presence of[$\alpha$$^{32}$P]-dATP Which were hybridized to Atlas$^{TM}$ Rat Toxicology 1.2 array (Clontech) representing approximately 1176 rat genes. We compared gene expression between ovary of pco-induced immature female rats and control. Differential gene expression profiles were revealed (LIFR-alpha, ADRA1A, Heat shock 90-kDa protein A, PDGFRA). Reverse transcription-polymerase chain reaction(RT-PCR) was used to validate the relative expression pattern obtained by the cDNA array. The purpose of this research is to improve sustainable clothes care by comparing household washer's standard course and quick course. Detergency at each course was classified by laundry weight, detergent concentration, and soils. Also, fabric damage from each course was compared. Washing experiments were carried out using two types of washing machines and three types of detergents. Using the standard soiled fabrics of EMPA 108 set, detergency was compared by laundry weight, soil, and detergent concentration. Additionally, fabric damage was evaluated using the mechanical action of MA-40. The results of the research were as follows. First, a standard course, having more working time exhibited better detergency than a quick course. However, the detergency deviation under 6kg laundry weight was as low as 9.0%. Second, detergency by the type of soil was more effective in standard course than in a quick course, but hydrophilic protein soils had a small detergency deviation at 7.6%. Moreover, hydrophobic oil, complex, and particulate soils had a higher deviation at 19.7% Third, fabric damage was in proportion to operating time. Fourth, a quick course showed approximately 80% detergency regardless of the type of detergent. in the case of using 50% of the recommended allowance by the detergent manufacturer. In conclusion, reducing the operating washing time and detergent concentration is in accordance with increasing sustainability, in the case of washing with lightly soiled fabrics under 6kg of laundry weight.
Kidney ischemia/reperfusion (I/R) injury, a common cause of acute kidney injury (AKI), is associated with the migration of inflammatory cells into the kidney. Ras-related C3 botulinum toxin substrate 1 (Rac1), a member of the Rho family of small GTPase, plays an important role in inflammatory cell migration by cytoskeleton rearrangement. Here, we investigated the role of Rac1 on kidney I/R injury and macrophage migration. Male mice were subjected to either 25 min of bilateral ischemia followed by reperfusion (I/R) or a sham operation. Some mice were administrated with either NSC23766, an inhibitor of Rac1, or 0.9% NaCl (vehicle). Kidney damage and Rac1 activity and expression were measured. The migration and lamellipodia formation of RAW264.7 cells, mouse monocyte/macrophage, induced by monocyte chemoattractant protein-1 (MCP-1, a chemokine) were determined using transwell migration assay and phalloidin staining, respectively. In sham-operated kidneys, Rac1 was expressed in tubular cells and interstitial cells. In I/R-injured kidneys, Rac1 expression was decreased in tubule cells in correlation with the damage of tubular cells, whereas Rac1 expression increased in the interstitium in correlation with an increased population of F4/80 cells, monocytes/macrophages. I/R increased Rac1 activity without changing total Rac1 expression in the whole kidney lysates. NSC23766 administration blocked Rac1 activation and protected the kidney against I/R-induced kidney damage and interstitial F4/80 cell increase. NSC23766 suppressed monocyte MCP-1-induced lamellipodia and filopodia formation and migration of RAW 264.7 cells. These results indicate Rac1 inhibition protects the kidney against I/R via inhibition of monocytes/macrophages migration into the kidney.
