Study on the adsorption of berberine hydrochloride (Berberine hydrochloride의 흡착에 관한 연구)

  • When a large quantity of insoluble drugs was added into a solution containing a small amount of soluble drugs, the latter was adsorbed to the former. Their adsorption was expected to alter the drug activity. Berberine hydrochloride (BH) was selected as a soluble drug and tested with frequently combined insoluble drugs and antacids for their adsorption phenomena in prufied water, gastric and intetinal fluid test solutions, respectively. The adsorption isotherms of kaolin and natural anuminum silicate with BH in the three media fitted the Langmuir (LM) equation, and that of talc in purified water and gastric fluid fitted it, but in intestinal fluid it fitted the Freudlich quation. The adsorption isotherm of aluminum hydroxide fitted the LM equation only in intestinal fluid. The degree of adsorption of BH in purified water and gastric fluid is in the following order: magnesium trisilicate, kaolin, natural aluminum silicate and talc; in intestinal fluid: magnesium trisilicate, kaolin, talc, natural aluminum silicate and aluminum hydroxide. Magnesium stearate did not adsorb BH.

Status of intestinal helminthic infections in some residents of North Korea (북한 함경북도 일부 주민의 기생충 감염 실태 조사)

  • A small scale survey on intestinal parasitic infections was investigated by subjecting residents of North Korea to recommend a plausible control strategy. The Kato-Katz method was applied to 235 residents in a borderline city and 46 in a refugee camp in borderline China. Only eggs of Ascaris and Trichuris were detected in 41.1% and 37.6% respectively but most of them were light infected. Females were infected in 61.2% while 53.1% males were egg positive. By age, residents in their 30s showed highest egg positive rates. The survey confirmed high prevalence of soil-transmitted intestinal nematodes in rural areas in North Korea It is recommended to implement mass chemotherapy for al residents with a part sample examination in rural areas, North Korea.

Effects of Carthami flos on pacemaker potentials of small intestinal and colonic interstitial Cells of Cajal (홍화의 생쥐 소장 및 대장 카할 간질세포의 향도잡이 전위 조절에 미치는 효능에 관한 연구)

  • Objectives : The purpose of this study was to investigate the effects of Carthami flos on pacemaker potentials of small intestinal and colonic Interstitial Cells of Cajal (ICC). Methods : To dissociate the ICC, we used enzymatic digestions from the small intestine and colon in mice. In the ICC, the electrophysiological whole-cell patch-clamp configuration was used to record pacemaker potentials in the cultured ICC. Results : 1. The ICC generated pacemaker potentials in the murine small intestine and colon. 2. Pretreatment with a Ca2+ free solution and thapsigargin, a Ca2+-ATPase inhibitor in the endoplasmic reticulum, stopped the pacemaker potentials. In the case of Ca2+-free solutions, Carthami flos did not induce membrane depolarizations in the murine small intestine and colon. However, when thapsigargin in a bath solution was applied, Carthami flos induced membrane depolarizations only in the murine colon. 3. Pretreatment with 2-APB (transient receptor potential melastatin (TRPM) channel inhibitor) abolished the pacemaker potentials and suppressed Carthami flos-induced effects in the murine small intestine and colon. 4. However, pretreatment with T16Ainh-AO1 (Ca2+ activated Cl- channel; anoctamin 1 (ANO1) inhibitor) did not affect the pacemaker potentials and induced Carthami flos-induced effects only in the murine small intestine. Conclusions : These results suggest that Carthami flos can modulate the pacemaker activity of ICC and the mechanisms underlying pacemaking in ICC might be different in the small intestine and the colon.

Activation of Platelet Rich Plasma by Soluble Canine Small Intestinal Submucosa Gel and Bovine Thrombin (개 소장점막하 겔과 소 트롬빈을 이용한 혈소판풍부혈장의 활성화 연구)

  • This study evaluated the efficacy of soluble canine small intestinal submucosa gel in comparison to bovine thrombin in activating rabbit platelet rich plasma (PRP) by detecting growth factors. PRP from rabbits was activated by using soluble canine SIS gel, bovine thrombin, or both. The surface morphology of each group of samples was examined by scanning electron microscopy. The release of transforming growth factor (TGF)-${\beta}1$ from each set of samples was measured over 7 days using enzyme-linked immunosorbent assay. The PRP-canine SIS gel group exhibited the highest total amount of released TGF-${\beta}1$. However, there were no significant differences between any groups. The use of soluble type of canine SIS gel could be an effective alternative to bovine thrombin.

Effects of Moxibustion at Combined Acupoints of ST36, BL21 and CV12 on Small Intestinal Motility for Sex and Ages of Rats (흰쥐의 성별과 주령에 따른 족삼리(足三里) 위유(胃兪) 중완(中脘) 배혈(配穴) 시구(施灸)가 소장(小腸) 수송능에 미치는 효과)

  • Objectives : The aim of this study was to observe the effect of moxibustion at combined acupoints of ST36, BL21 and CV12 in rats with sex and age. Methods : This study measured small intestinal motility in rats. First, cauterize with moxa was applied 5 times to the ST36, BL21 and CV12 in rats classified by sex and age under enflurane anesthesia. And then same treatment is done to mached acupoints of ST36, BL21 and CV12. Results : In single acupoint groups, cauterizing with moxa on ST36 increased with 5, 7 weeks in female and all weeks in male. The BL21 group increased with 5, 6, 7 weeks in female, and only 5 weeks in male. The CV12 group showed increasing with 5, 6 weeks in female and only 7 weeks in male. In combined acupoints groups, the ST36+BL21 with 8 weeks in male, the ST36+CV12 with 5, 8 weeks in female and 5, 6, 8 weeks in male, the CV12+BL21 only with 5 weeks in female, the ST36+BL21+CV12 only with 5 weeks in female showed increasing, respectively. Conclusions : Although these differents according to the sex and age in rats do not have a established tendency, the results suggested that the effects of moxibustion are experimentally should be considered the sex and age of individual.

Rat Peripheral Nerve Regeneration Using Nerve Guidance Channel by Porcine Small Intestinal Submucosa

  • Objective : In order to develop a novel nerve guidance channel using porcine small intestinal submucosa (SIS) for nerve regeneration, we investigated the possibility of SIS, a tissue consisting of acellular collagen material without cellular immunogenicity, and containing many kinds of growth factors, as a natural material with a new bioactive functionality. Methods : Left sciatic nerves were cut 5 mm in length, in 14 Sprague-Dawley rats. Grafts between the cut nerve ends were performed with a silicone tube (Silicon group, n=7) and rolled porcine SIS (SIS group, n=7). All rats underwent a motor function test and an electromyography (EMG) study on 4 and 10 weeks after grafting. After last EMG studies, the grafts, including proximal and distal nerve segments, were retrieved for histological analysis. Results : Foot ulcers, due to hypesthesia, were fewer in SIS group than in Silicon group. The run time tests for motor function study were 2.67 seconds in Silicon group and 5.92 seconds in SIS group. Rats in SIS group showed a better EMG response for distal motor latency and amplitude than in Silicon group. Histologically, all grafts contained some axons and myelination. However, the number of axons and the degree of myelination were significantly higher in SIS group than Silicon group. Conclusion : These results show that the porcine SIS was an excellent option as a natural biomaterial for peripheral nerve regeneration since this material contains many kinds of nerve growth factors. Furthermore, it could be used as a biocompatible barrier covering neural tissue.

Effects of Rice Bran and Wheat Bran on Intestinal Physiology and Small-bowel Morphology in Rats

  • The present study was aimed at investigating the nutritional and physiological significance of rice bran as a source of dietary fiber as compared to pectin and wheat bran. The parameters for comparison included hypertrophy and morphology of intestines, stool weights and villus marker enzyme activity. For 6 weeks, 10 Sprague Dawley male rats were given one of six experimental diets: 1% cellulose control (CC), 5% pectin (P5), 5% rice bran(RB5), 10% rice bran(RB10), 5% wheat bran (WB5) or 10% wheat bran (WB10) based on the level of dietary fiber. Among experimental groups, food efficiency ratio and body weight gain was comparable. RB10 increased cecal and colonic tissue weights and content weights of cecum and colon as much as P5 did. Stool weight was positiviely correlated with colonic tissue weight (r=0.727, P<0.001), with colonic content weight(r=0.647, P<0.001). Small intestine length increased most in the P5 group, followed by the RB10 group. The scanning electron micrograph of jejunal villi from rice bran groups showed a leaf-shaped, smooth and regular pattern, whereas that of CC group produced a rather long shape. The wheat bran groups showed an irregular leafshaped pattern, and the pectin group typically produced leaf-shaped villi with surface damage. The activities of villus marker enzymes (maltase and sucrase) were higher in the bran-fed rats than in the control or pectin-fed rats. The results indicate than not only dietary fiber amounts but also fiber sources are closely related to the physiology and morphology of the large and small intestines in rats. Rice bran exerted effects on fecal output and trophic effects on the intestines similar to those of pectin.

Capsule enteroscopy versus small-bowel ultrasonography for the detection and differential diagnosis of intestinal diseases

  • Background/Aims: Capsule enteroscopy (CE) and intestinal ultrasonography (IUS) are techniques that are currently used for investigating small-bowel (SB) diseases. The aim of this study was to compare the main imaging findings and the lesion detection rate (LDR) of CE and IUS in different clinical scenarios involving the SB. Methods: We retrospectively enrolled patients who underwent CE and IUS for obscure gastrointestinal bleeding (OGIB), complicated celiac disease (CeD), and suspected or known inflammatory bowel disease (IBD). We evaluated the LDR of both techniques. The accuracy of IUS was determined using CE as the reference standard. Results: A total of 159 patients (113 female; mean age, 49±19 years) were enrolled. The LDR was 55% and 33% for CE and IUS (p<0.05), respectively. Subgroup analysis showed that the LDR of CE was significantly higher than that of IUS in patients with OGIB (62% vs. 14%, p<0.05) and CeD (55% vs. 35%, p<0.05). IUS showed a similar LDR to CE in patients with suspected or known IBD (51% vs. 46%, p=0.83). Conclusions: CE should be preferred in cases of OGIB and CeD, whereas IUS should be considered an early step in the diagnosis and follow-up of IBD even in patients with a proximal SB localization of the disease.
