Steatosis in a Slaughtered Korean Native Cattle

  • Muscular lesion was detected in a 23-month-old castrated bull encountered at Kyungsan slaughter house. The lesion appeared as fat intervening muscle fibers. The affected animal had no clinical signs. On microscopic examination, there was replacement of many muscle fibers by normal fat cells. Numerous fat cells were located between muscle fibers. Remaining skeletal muscle cells were in degenerative process, and thus abnormal skeletal muscle cells had loose fibers while normal had intact ones. The advent of inflammatory cells is not at the lesion, which is unique view in steatosis.

이온화칼슘이 도계과정 중 도체표면의 미생물 교차오염에 미치는 영향 (Effects of ionized calcium on microbial cross-contamination in surface of carcass via slaughter process of chickens)

  • The purpose of this study was to investigate the effects of ionized calcium treatment on total bacterial cross-contamination of chicken carcass surface in the slaughtering process. The growth of Escherichia coli was strongly inhibited in a medium prepared by using a 0.5% ionized calcium solution. The total bacterial cross-contamination of chicken carcass surface and the scalding water was significantly increased as the number of scalding was increased (p<0.05). The total bacterial cross-contamination of chicken carcass surface reached a plateau without a further increase as scalding was performed consecutively for 10 or more times. The total bacterial cross-contamination of the scalding water was significantly increased as the number of scalding was increased (p<0.05). The total bacterial cross-contamination of chicken carcass surface of the chickens raised on a floor type farm was significantly higher than that of the chickens raised in a battery cages (p<0.05). The total bacterial cross-contamination of chicken carcass surface of the chickens raised on a floor type farm was significantly lower in the 0.5% ionized calcium solution treatment group than in the control group (p<0.05).

식육의 처리 단계별 미생물 오염실태와 병원성 미생물의 분포 (Hygienic Quality of Beef and Distribution of Pathogens during Cut-Meat Processing)

  • 위생적인 신선육 처리 방안을 모색하기 위하여 도체처리 과정과 식육처리 과정 중 고기의 표면 및 작업도구의 위생실태를 계절별로 검토하였다. 하절기에 총균수, 저온성 균수, 대장균군수 모두 높게 나타났으며, 특히 도체 세척전보다 세척 후에 미생물 오염도가 높았고, 계절에 관계없이 수송 후의 오염도가 가장 높았다. 칼, 도마와 장갑에서 하절기에 $10^{5}$ $ extrm{cm}^2$을 나타내어 높은 미생물 오염도를 나타내었다. 도체 처리 과정 중 도체 표면에서 Escherichia. coli O157, Pseudomonas aeruginosa, Klebsiella, ornithinolytica, Staphylococcus aureus, E. Tatumella, ptyseos, Serratia odorifera, Aeromonas, Aeromonas sobria, Enterobacter colacae, Flavimonas oryzihabitans oryzihabitans를 분리하였으며, 도마에서는 S. aureus, Listeria grayi, L. monocytogenes, 장갑에서 L. grayi, Erwinia spp., Salmonella spp. S aureus, 골발육에서는 Citrobacter freundii, L. momocytogenes, S. aureus가 분리.동정하였다.

  • 2002년 5월 도축중 발견된 거세수소의 근육 병변에 대한 검사 의뢰가 들어왔다. 이 병변의 육안적 소견은 광범위한 근육의 지방침윤이었고, 병리조직학적 검사에서 근섬유가 지방조직에 의해 대치된 것이 관찰되었으며, 기타 염증 소견은 관찰되지 않았다. 본 증례는 골격근의 광범위한 부분이 지방조직으로 대치된 지방증(steatosis)으로 진단되어 이에 보고하는 바이다.

생물고분자응집제 Biopol32의 산업폐수에 대한 응집활성 및 탈수효과 (Flocculating Activity and Dehydration Efficiency of Biopolymer Flocculant Biopol32 in Industrial Wastewater Treatment)

  • Pseudomonas sp. GP32가 생산하는 생물고분자 응집제 Biopol32의 실제 응용, 개발을 위하여 식품폐수, 도축폐수, 염색폐수에 대하여 응집효과를 검토하였다. 식품폐수의 경우 pH 6.0에서 70%의 화학적산소요구량(COD) 감소율과 49%의 부유고형물(SS) 제거율을 나타내었다. 도축폐수에서는 pH 4.0에서 61%의 화학적산소요구량 감소율과 91%의 부유고형물 제거율을 보였으며, 염색페수의 경우 pH 5.0에서 72%의 화학적산소요구량 감소율과 92%의 부유고형물 제거율을 보였다. 응집과정에서 형성되는 floc의 크기는 Biopol32 용액을 최종농도 20 ppm으로 첨가하였을 때 10 mm였고 이때 탈수효율은 62%였다. 생물응집제 Biopol32 첨가구와 합성응집제 PAA 첨가구에서 탈수효율이 전반적으로 가장 좋은 응집제 첨가 농도는 20 ppm으로 나타났으며, 또한 이때 자연한계함수율(78.1%)에 이르는 최단의 여과시간을 얻을 수가 있었다.

  • The hazard analysis critical control point (HACCP) system was introduced to the Korean livestock slaughter and product processing business in 1997. Ice cream manufacturers focus on the sterilization process in order to reduce microorganism, so the HACCP-accredited ice cream manufacturers particularly set the sterilization process as critical control point (CCP). Therefore, the microbiological test results on the finished products can be considered as one of the indicators which show the level of microbiological control in the ice cream manufacturers. Accordingly, this study was carried out to examine if there is a meaningful difference in terms of the microbiological control level between the HACCP-accredited and non-accredited ice cream manufacturer groups by comparing aerobic plate count (APC) of ice creams. The mean of APC was 1.68 Log CFU/g in the HACCP-accredited ice cream manufacturer group and 2.59 Log CFU/g in the non-accredited ones respectively and there was a statistically significant difference (P<0.05). The test results suggest that the level of microbiological control in the HACCP-accredited ice cream manufactures is much higher than that of non-accredited ones.

도축되는 한우 혈액에서 회수한 혈청의 생물학적 안전성 분석 (Assessing Biological Safety of the Hanwoo Serum Obtained During Slaughtering Process)

  • 본 연구에서는 성체 한우의 도축과정 중에 채혈된 혈액으로부터 분리 정제한 혈청의 생물학적 안전성을 검증하기 위하여 세균, 마이코플라즈마 및 바이러스의 검출과 동정을 실시하였다. 검사를 실시한 대부분의 혈청에서 전염성 병원체가 검출되지 않았고 두 개의 혈청에서 토양 유래의 것으로 추정되는 Bacillus thuringiensis가 검출 되었다. 본 연구의 결과는 국내에서 도축되는 한우는 생물학적 안전성을 담보할 수 있는 최소한의 기본적인 안전장치를 통해 도축된다는 사실을 보여주며 한우의 도축과정에서 부산물로 얻을 수 있는 혈액을 안전한 생물학적 제제로 개발할 수 있다는 가능성을 보여주는 것으로 생각된다.

Effects of Whey Powder Supplementation on Dry-Aged Meat Quality

  • The objective of this study was to determine the effect of dietary supplementation with whey powder (WP, 1g/kg feed) from weaning to slaughter (150 d) on dry-aged loin quality of pigs. Fifty-eight pigs were randomly divided into two dietary treatment groups (seven replications of four pigs per treatments). Basal diet with 0.1% whey powder was supplied to the WP group. Basal diet was used for the control group (CON). Diet whey protein did not appear to influence the moisture or protein contents. However, ash and fat contents were significantly (p<0.05) decreased in the WP group compared to the control group. Drip loss was significantly (p<0.05) lower in the WP group than that of the control group. Increasing redness with decreasing lightness was found in the inner loin of the WP group. Calcium and iron contents in the WP group were significantly higher than those in the control group. Protein degradation was higher in the WP group than that in the control group (p<0.05), whereas shear force was lower in the WP group than that in the control group (p<0.05). In conclusion, the basal diet supplemented with 0.1% whey powder influence negatively the lipid oxidation of meat whereas the texture property and mineral composition of meat from whey powder fed pigs are developed.

A Whole Genome Association Study on Meat Palatability in Hanwoo

  • A whole genome association (WGA) study was carried out to find quantitative trait loci (QTL) for sensory evaluation traits in Hanwoo. Carcass samples of 250 Hanwoo steers were collected from National Agricultural Cooperative Livestock Research Institute, Ansung, Gyeonggi province, Korea, between 2011 and 2012 and genotyped with the Affymetrix Bovine Axiom Array 640K single nucleotide polymorphism (SNP) chip. Among the SNPs in the chip, a total of 322,160 SNPs were chosen after quality control tests. After adjusting for the effects of age, slaughter-year-season, and polygenic effects using genome relationship matrix, the corrected phenotypes for the sensory evaluation measurements were regressed on each SNP using a simple linear regression additive based model. A total of 1,631 SNPs were detected for color, aroma, tenderness, juiciness and palatability at 0.1% comparison-wise level. Among the significant SNPs, the best set of 52 SNP markers were chosen using a forward regression procedure at 0.05 level, among which the sets of 8, 14, 11, 10, and 9 SNPs were determined for the respectively sensory evaluation traits. The sets of significant SNPs explained 18% to 31% of phenotypic variance. Three SNPs were pleiotropic, i.e. AX-26703353 and AX-26742891 that were located at 101 and 110 Mb of BTA6, respectively, influencing tenderness, juiciness and palatability, while AX-18624743 at 3 Mb of BTA10 affected tenderness and palatability. Our results suggest that some QTL for sensory measures are segregating in a Hanwoo steer population. Additional WGA studies on fatty acid and nutritional components as well as the sensory panels are in process to characterize genetic architecture of meat quality and palatability in Hanwoo.

한국 재래돼지 브랜드 돈육 원산지 검증을 위한 유전자 원산지 감식 기법 활용 연구 (Application of DNA Test for Individual Traceability in the Brand Marketing of Korean Native Pig.)

  • Identification of animals has been used with an e ar tag with dummy code and blood typing has been used for paternity and individual identification in live animals. Various genetic markers are different for breeds of pig and hence, it is necessary to identity the discrete genetic marker in korean native pig. A total of 240 pigs were used to find korean native pig population specific markers that expressed in population of korean native pigs. To identify the individual traceability, 20 animals were randomly chosen and tested for a whole process from being live to slaughter stages. The candidate genetic marker used in the study were 18 DNA microsatellites which were identified in pig genome. The number of alleles of those DNA microsatellites ranged form a minimum of 3 to maximum of 6. The heterozygote frequency rang6d from 0.44 to 0.69. Effective number of alleles for each DNA microsatellotes were 2 to 4. By choosing 6 candidate genetic markers among all, the traceability of individual identification was estimated as accurate as 99.99%(p>0.0014), nearly.
