Melanin synthesis and skin wrinkle inhibitory effects of the medicinal mushroom Ganoderma applanatum

  • Anti-melanogenesis and skin anti-wrinkle effects of methanol (ME) and hot water (HE) extracts from the fruiting bodies of Ganoderma applanatum were investigated in this study. The total phenolic contents of the ME and HE of the mushroom were 11.68 and 3.15 ㎍ GAEs/mg, respectively, whereas the total flavonoid contents of the ME and HE were 21.82 and 2.69 ㎍ QEs/mg, respectively. The survival rate of B16-F10 murine melanoma cells treated with 750 ㎍ ME and HE were 83.46% and 85.54%, respectively, thereby suggesting that mushroom extracts were slightly cytotoxic at the tested concentration. The in vitro tyrosinase inhibition by ME (83.15%) and HE (83.44%) was significantly lower than that of kojic acid (99.61%), the positive control, at 2.0 mg/mL. Although the inhibition of cellular melanin synthesis in B16-F10 melanoma cells by 2.0 mg/mL of ME (50.24%) and HE (51.24%) was lower than that of arbutin (64.84%), the inhibition by both ME and HE was higher than 50%. Collagenase inhibition by HE was comparable to 2.0 mg/mL epigallocatechin (EGCG), the positive control; however, elastase inhibition by ME and HE was lower than that of EGCG at the concentration tested. The results showed that the fruiting bodies of G. applanatum had good anti-tyrosinase, good anti-collagenase, and moderate anti-elastase activities, which might be useful for developing novel skin-whitening and anti-wrinkle agents.

  • Cosmetics have mild activity on the human body, and intended to be used for cleaning, beautifying the human body, and also keeping healthy skin or hair, promoting attractiveness or altering the appearance. Functional cosmetics, in other words cosmeceuticals, are restricted for following functions: 1) Whiten the skin tone by preventing deposition of melanin pigmentation or lightening of the color of melanin of skin, 2) alleviate or improve wrinkles of the skin, and 3) protect the skin from the ultra violet rays from the sun. According to the functions of the functional cosmetics, skin whitening products, anti-wrinkle products, and suntan & sunscreen products are manufactured. In order to manufacture and import the functional cosmetics in Korea, the approval process in KFDA is necessary. The review process in KFDA is performed based on The Korea Food and Drug Administration Notification 2007-44, "The Regulation of Reviewing the Functional Cosmetics" (June 29, 2007). Only after the approval of KFDA, functional cosmetics are allowed to advertise to the consumers for their functionality.

Anti-wrinkle Activity of Acanthopanax senticosus Extract in Ultraviolet B (UVB)-induced Photoaging (가시오가피 추출물의 광노화에 의한 주름형성 억제 효과)

  • This study was performed to investigate anti-wrinkle effects of Acanthopanax senticosus (AS) on ultraviolet B (UVB)-induced photoaging with wrinkle formation. AS extract showed higher DPPH radical scavenging activity (3 ${\mu}g/mL$ as $IC_{50}$) and collagenase inhibition (1.52 mg/mL as $IC_{50}$) than those of ascorbic acid (50 ${\mu}g/mL$ and 2.17 mg/mL, respectively). Cell proliferation and type I pN collagen synthesis were increased by 11.4% and 96.4%, respectively, compared with non treatment control. In vivo, SKH-1 hairless mice were administrated AS 400 mg/kg for 10 weeks with UVB irradiation three times a week. After 10 weeks, a visual assessment and replica assay were performed on each mouse. According to visual assessment of close-up photos and skin replica, oral administration of A. senticosus affected on inhibition of wrinkle formation caused by UVB irradiation on the skin of mice as compared to the vehicle treated control mice. These results indicated that A. senticosus could protect skin wrinkle formation caused by collagen synthesis of fibroblast cells and photo-irradiation of UVB in hairless mice.

The Anti-wrinkle Effects of Cultivated Wild Ginseng Pharmacopuncture (산삼 약침의 피부 주름 개선 효과에 대한 연구)

  • Objectives : This study was aimed to report the anti-wrinkle effects of cultivated wild ginseng pharmacopuncture. Methods : Data were collected from 23 patients who had been treated with cultivated wild ginseng pharmacopuncture for skin wrinkles. Patients were treated twice a week, for a total of 5 times. To observe the effect of the treatment, we measured the width and depth of skin wrinkles with ECOSKIN(portable fluorescent dermal diagnostic equipment). We also conducted a survey with patients to measure total satisfaction and the anti-wrinkle effect using a semantic differential scale. The evaluation was done 2 times, once before the first treatment and once 1 week after the last treatment. The relationship was measured by Wilcoxon signed rank test. Results : Skin wrinkle mean width decreased significantly from $9.45{\pm}6.60$ to $5.54{\pm}4.78$. Depth decreased significantly from $0.68{\pm}0.34$ to $0.39{\pm}0.24$ total satisfaction increased significantly from $2.46{\pm}0.743$ to $4.13{\pm}0.51$. Anti-wrinkle effect increased significantly from $2.46{\pm}0.63$ to $3.93{\pm}0.593$. Conclusions : These results suggest that cultivated wild ginseng pharmacopuncture may be effective for decreasing wrinkles.

Safflower Seed Oil and Its Active Compound Acacetin Inhibit UVB-Induced Skin Photoaging

  • Ultraviolet (UV) is one of the major factors harmful to skin health. Irradiation with ultraviolet accelerates the decline of skin function, causing the skin to have deep wrinkles, dryness, decreased procollagen production, and degradation of collagen. Novel materials are needed to prevent the aging of the skin by blocking the effects of UV. Safflower seed oil (Charthamus tinctorius L., SSO) contains significantly high levels of unsaturated fatty acids and phytochemicals. SSO has been traditionally used in China, Japan, and Korea to improve skin and hair. Our objective in this study was to determine the effect of SSO and its active compound acacetin on UVB-induced skin photoaging in HaCaT cells and human dermal fibroblasts (HDF). SSO inhibited UVB-induced matrix metalloproteinase-1 (MMP-1) at both protein and mRNA levels in HaCaT cells and HDF. MMP-1 is known to play important roles in collagen degradation and wrinkle formation. Acacetin, a type of flavonoid, is present in SSO. Similar to SSO, acacetin also inhibited UVB-induced MMP-1 protein and mRNA levels in HaCaT cells and HDF. MMP-1 mRNA is primarily regulated by the mitogen-activated kinase (MAPK) signaling pathway. Acacetin regulated the phosphorylation of JNK1/2 and c-jun, but did not inhibit the phosphorylation of ERK1/2, p38 and AKT. Taken together, these results indicate that SSO and its active compound acacetin can prevent UVB-induced MMP-1 expression, which leads to skin photoaging, and may therefore have therapeutic potential as an anti-wrinkle agent to improve skin health.

Effects of Dendrobii herba and Punica granatum Extract on the Anti-oxidant, Anti-inflammatory, Anti-wrinkle and Whitening (석곡(石斛), 석류(石榴)의 항산화, 항염증, 주름, 미백에 미치는 영향)

  • Objective : The aim of this study is to determine the effects of Dendrobii herba extract and Punica granatum extract on skin disease and skin beauty. Methods : To investigate in vitro anti-oxidant activity assay, ethanol extracts of medicinal plants tested by DPPH radical, xanthine oxidase activity. In the next experiment, to investigate anti-inflammatory activity assay, examined by relations in NO synthesis, IL-$1{\beta}$, IL-6, TNF-${\alpha}$, NF-${\kappa}B$, COX-2, MAP kinase. To study Skin wrinkle formation effect, we were examined by tyrosinase activities, melanin synthesis in MNT-1 cell. Results : 1. In an anti-oxidant test, Dendrobii and Punica granatum extract showed high radical scavenging activity. 2. In an anti-inflammatory test, Dendrobii herba and Punica granatum extract weakly inhibited the lipopolysaccharide(LPS)-induced nitric oxide(NO) release from RAW 246.7 macrophage cells. Dendrobii herba and Punica granatum extract also inhibited LPS-induced IL-$1{\beta}$ and COX-2 expressions. The inhibitory effect of Dendrobii herba and Punica granatum extract on macrophage activation were via the inhibition of NF-${\kappa}B$, evidenced by transient transfection assay. however, Dendrobii herba and Punica granatum extract did not have any effects about activation of Jun-N-terminal kinase(JNK) and inhibition of p38 MAP kinase in RAW 264.7 cells. 3. In the skin wrinkle formation assay, Dendrobii herba and Punica granatum extract weakly inhibited collagenase and elastase, however it was not statistically significant. 4. In the skin whitening assay, Dendrobii herba and Punica granatum extract weakly inhibited tyrosinase activity, however, it was not statistically significant. They did not have any effect on melanin synthesis, indicating that they could not be applicable for skin whitening. Conclusion : Dendrobii herba extract and Punica granatum extract may play a significant role in skin disease and skin beauty.

Effects of Mugil cephalus Extract on Wrinkle Improvement (숭어 추출물을 이용한 주름개선 효과)

  • This study was to suggest the method for wrinkle improvement using Mugil cephalus extract. In order to evaluate the cosmetic product made of M. cephalus extract, the experiments were conducted with subjects and skin states for 4 weeks. Effect of cosmetics containing M. cephalus extract on the water holding capacity and evaporation of the women skin were investigated. The water holding capacity was increased after 1 h of application. But after 2 h, it was decreased. In the case of the water evaporation, it was increased with the increase of time. On the clinical trial, it was also found that water, oil, and rough level on the women skin were increased and wrinkles were improved. Side effects were also not detected during the application and treatment of cosmetics. Based on this result, the application of M. cephalus extract had positive effects on the improvement of wrinkles and skin in women. Therefore, it will be used to develop the wrinkle improvement therapy for women.

The Efficacy of Korean Herbal Cosmetics for Facial Skin Elasticity (한방화장품이 안면부의 탄력 개선에 미치는 영향)

  • Objective : The purpose of this study was to evaluate the effect of korean herbal cosmetics for facial skin elasticity. Methods : We recruited patients and divided them to cosmetics group and control group randomly. We provided korean herbal cosmetics to cosmetics group and subjects in cosmetics group applied the provided cosmetics for 4 weeks. Control group waited and applied cosmetics that had previously used for 4 weeks. We assessed all subjects in 0 week, 2 weeks and 4 weeks. Primary assessment instrument was moire topography photo scale. Second assessment instrument was skin moisture, oiliness and elasticity which assessed with skintouch system v1.0, detox/regeneration questionnaire score and self-evaluating facial wrinkle/looseness score. Result : Moire topography photo scale improved in cosmetics group than control group significantly. Skin moisture and elasticity assessed with skintouch system improved in cosmetics group than control group significantly while skin oiliness assessed with skintouch system have not improved significantly. Detox/regeneration questionnaire score and self-evaluating facial wrinkle/looseness score improved in cosmetics group than control group significantly. Conclusion : Korean herbal cosmetics application might be effective method for facial skin elasticity.

  • The purpose of this study was to investigate the effects of Cuscuta chinensis Lamark ethanol extract (CL) on wrinkle improvement. Cuscuta chinensis Lamark is known to contain dried saccharide, alkaloids, flavonoids, lignans and rein glycoside as major components of dried mature seeds of Cuscuta japonica Choisy. In this study, we evaluated the anti-wrinkle effects of CL and investigated bio-markers (e.g ; MMP-1, MMP-3, MMP-9, TIMP-1, type I procollagen) associated with skin wrinkle improvement. We tested the anti-wrinkle effect of CL using human fibroblast called CCD-986Sk cell. We observed an increase in MMPs, TIMP-1, and type 1 pro-collagen CL in CCD-986Sk cells irradiated with UVB at an intensity of $2mJ/cm^2$ for 60 seconds. As a result, CL decreased UVB-induced MMPs levels and mRNA expressions in CCD-986Sk cell. The levels and mRNA expressions of type I procollagen and TIMP-1 were increased by CL. These results suggest that CL has activities on improvement of skin wrinkle, which is induced by UVB radiation. Taken together, this study proposed the possibility of developing herbal medicine and functional herbal cosmetic materials with wrinkle-improving effects of Cuscuta chinensis Lamark.

Design of 850 nm Near Infrared and Galvanic Current Based Eyeglass-Type Device for Periorbital Wrinkle Treatment and Verification of Treatment Performance through Image Analysis (850 nm 파장대 근적외선과 갈바닉 전류기반의 눈가 주름 치료기 개발 및 영상 분석을 통한 치료성능 검증)

  • In this paper, we proposed eyeglass type periorbital wrinkle treatment device for reducing and improving periorbital wrinkles using near infrared LED of 850nm wavelength and galvanic current. The proposed periorbital wrinkle treatment device is equipped with a control system based on F-PCB. It consists of eight near-infrared LEDs and four indicator LEDs for treatment of right and left periorbital wrinkles. The eyeglass frame is coated with conductive material, so galvanic current can flow to the skin of periorbital wrinkle contacted to it. One male adult in the mid-40s was allowed to use the device for 10 minutes every day for 4 weeks. After 4 weeks, image analysis using optical equipment for measuring wrinkles indicated that wrinkle indexes were reduced.
