백합뿌리 추출물의 멜라닌 생성 억제효과 (Hypopigmenting Effects of Extracts from Bulbs of Lilium Oriental Hybrid 'Siberia' in Murine B16/F10 Melanoma Cells)

  • 피부미백 소재를 개발하기 위해 멜라닌 함량, 세포내 tyrosinase 활성의 측정 및 Western blotting 실험이 수행되었다. 백합(Lilium Oriental Hybrid 'Siberia') 뿌리의 80% 에탄올 추출물로부터 얻은 에틸아세테이트 분획물(R-EA)은 ${\alpha}$-melanocyte stimulating hormone(${\alpha}$-MSH)에 의해 멜라닌 생성이 유도된 B16/F10 흑색종 세포에서 농도 의존적으로 멜라닌 생성을 저해하였다. 정확하게 세포내 tyrosinase 활성과 멜라닌 함량은 에틸아세데이트 분획물 $100{\mu}g/mL$ 처리 시 ${\alpha}$-MSH 단독 처리군에 비해 각각 45%와 74%의 저해율을 보였다. ${\alpha}$-MSH에 의해 멜라닌 생성이 유도된 B16/F10 흑색종 세포에서 단백질 발현양상을 살펴본 결과 TRP-1이 가장 많이 억제된 양상을 확인할 수 있었고 이 결과는 세포내 tyrosinase 활성저해보다 멜라닌 생성저해가 더 많이 일어난 것과 일맥상통하는 것이다. 이를 종합해 볼 때 p-coumaric acid와 resveratrol 함량이 백합뿌리의 에탄올 추출물에 비해 많이 함유된 에틸아세테이트 분획물은 멜라닌 생성 유도물질에 의해 촉진된 ERK의 활성화를 억제하는 피부미백 소재로서 그 가치가 입증된다고 사료된다.

Royal jelly enhances migration of human dermal fibroblasts and alters the levels of cholesterol and sphinganine in an in vitro wound healing model

  • Oral administration of royal jelly (RJ) promotes wound healing in diabetic mice. Concerns have arisen regarding the efficacy of RJ on the wound healing process of normal skin cells. In this study, a wound was created by scratching normal human dermal fibroblasts, one of the major cells involved in the wound healing process. The area was promptly treated with RJ at varying concentrations of 0.1, 1.0, or 5 mg/ml for up to 48 hrs and migration was analyzed by evaluating closure of the wound margins. Furthermore, altered levels of lipids, which were recently reported to participate in the wound healing process, were analyzed by HPTLC and HPLC. Migration of fibroblasts peaked at 24 hrs after wounding. RJ treatment significantly accelerated the migration of fibroblasts in a dose-dependent manner at 8 hrs. Although RJ also accelerated the migration of fibroblasts at both 20 hrs and 24 hrs after wounding, the efficacy was less potent than at 8 hrs. Among various lipid classes within fibroblasts, the level of cholesterol was significantly decreased at 8 hrs following administration of both 0.1 ug/ml and 5 mg/ml RJ. Despite a dose-dependent increase in sphinganines, the levels of sphingosines, ceramides, and glucosylceramides were not altered with any concentration of RJ. We demonstrated that RJ enhances the migration of fibroblasts and alters the levels of various lipids involved in the wound healing process.

무, 양파의 시료제조 방법에 따른 흰쥐의 지방대사와 항산화능에 관한 연구 (Effect of Dry Powders, Ethanol Extracts and Juices of Radish and Onion on Lipid Metabolism and Antioxidative Capacity in Rats)

  • This study was performed to investigate the effects of dry powders, ethanol extracts and juices of radish and onion on lipid metabolism, lipid peroxidation and antioxidative enzyme activity in rats. Forty-nine male Sprague-Dawley rats weighing 157$\pm$6g were blocked into seven groups according to body weight and raised for four weeks with diets containing 5%(w/w) dry powders of two different vegetables consumed frequently by Korean-radish(Raphanus sativus L.) and onion(Allium cepa L.), ethanol extracts and juices from equal amount of each dry powder. All the powders, ethanol extracts and juices of radish and onion decreased total lipids, triglycerides and total cholesterol concentrations in plasma and liver. Above all, onion ethanol extract decreased them most remarkably. It was thought that organosulfur compounds and flavonoids extracted from onion by ethanol inhibited biosynthesis and absorption of lipid and promoted degradation of lipid. Radish powder also decreased them by increasing fecal excretions of total lipids, triglycerides and total cholesterol most effectively. Catalase and glutathine peroxidase(GSH-px) activities in red blood cell(RBC) were most remarkably increased by radish powder and onion powder respectively. Superoxide dismutase(SOD), catalase and GSH-px activities in liver were most remarkably increased by onion ethanol extract, radish powder and onion ethanol extract respectively. Xanthine oxidase(XOD) activities in liver were most effectively decreased by ethanol extracts of radish and onion. Thiobarbituric acid reactive substance (TBARS) levels in plasma and liver of experimental groups were significantly lower than those of controls. Above all, onion powder decreased them most effectively. It was thought that vitamin E and high flavonoids in onion powder inhibited lipid peroxidation, promoting liver and RBC SOD, catalase and GSH-px activities and inhibiting XOD activities effectively. Flavonoids in onion ethanol extract inhibited lipid peroxidation, promoting three antioxident enzyme activities and inhibiting XOD activities most remarkably. Also flavonoids and high vitamin C in radish powder inhibited lipid peroxidation, promoting liver and RBC catalase most remarkably and inhibiting XOD activities. In conclusion, radish and onion were effective in lowering lipid levels and inhibiting of lipid peroxidation in animal tissue. From these data, radish and onion can be recommended in the treatment and prevention of diseases such as cardiovascular disease and cancer and in delaying aging. As ethanol from onion were most effective in lowering lipid level and promoting three antioxident enzymes, and inhibited lipid peroxidation as did we should try to utilize onion skin which is discarded though reported to have abundant flavonoids. 인체 두피 모낭의 장기간 배양을 위한 기관 배양 배지의 개발 (Development of Organ Culture Medium for Long Term Culture of Human Hair Follicle)

  • 본 연구에서는 인체 두피조직에서 미세수술법을 이용하여 모낭을 성공적으로 분리하였으며 다양한 조건으로 액침기관배양을 수행하였다. 우태아 혈청첨가시 모낭의 길이 성장이 저해되는 것으로 확인되어 무혈청 배지 조성을 시도하였다. 무혈청 배지로는 모낭 기관배양에 널리 이용되는 Williams'E medium을 기본으로 하는 Philpott medium과 자체 개발한 고농도 아미노산과 비타민(B군) 조성의 DHGM(Dongguk hair growth medium)을 이용하였다. 그리고 IMDM은 DHGM의 비교 대조군으로 이용하였다. 연구 결과 Philpott medium과 IMDM으로 배양한 모낭은 구조상으로는 길이 성장이 각각 9일과 12일 정도에 멈추며, 낮은 알카라인 포스파테이즈 발현, CK19 발현이 거의 없는 것으로 보아 세포사멸에 의한 퇴화(regression)가 빠르게 일어나는 것으로 판단되었다. 반면, DHGM으로 배양한 모낭은 상대적으로 긴 기간 동안 성장기의 구조를 보이며 25일까지 지속적인 길이 증가 및 3배 높은 알카라인 포스파테이즈 발현, 전반적인 CK19 발현을 나타내었다. 따라서 고농도의 아미노산 및 비타민 배지 조성이 생체 외에서 모낭을 장기간 배양하는데 중요한 역할을 하는 것으로 판단된다. 이러한 배양 방법은 장기간 검사를 필요로 하는 모낭에 대한 기초 생물학적 연구뿐만 아니라 새로운 탈모치료제의 효능 평가 분야에 이용될 수 있을 것이다.

NF-κB 억제를 통한 대회향(大茴香)의 항염증효능 (Anti-inflammatory Effects of Illicium verum Hook. f. via Suppression of NFκB Pathway)

  • Objectives : Illicium verum Hook. f. has been known to possess antimicrobial, antioxidant, antifungal, anti-inflammatory, insecticidal, analgesic, sedative, convulsive activities, it has been rarely conducted to evaluate the immuno-biological activity. The present study was examined to evaluate the anti-inflammatory effects of the Illicium verum Hook. f. water extracts (IVE) in vivo and in vitro. Methods : Cell viability was measured by MTT assay. The relative levels of NO were measured with Griess reagent. iNOS, COX-2, $NF-{\kappa}B$ and target proteins were detected by immunoblot analysis, and levels of cytokines were analyzed by ELISA kit. Anti-edema effect was determined in the carrageenan (CA)-induced paw edema model in rats. Results : All dosages of IVE used in MTT assay had no significant cytotoxicity. The increases of NO production and iNOS expression were detected in LPS-treated cells compared with control. However, these increases were attenuated by treatment with IVE. Also, IVE reduced the elevated production of $TNF-{\alpha}$, $IL-1{\beta}$ and IL-6 by LPS. IVE inhibited the $p-I{\kappa}B$ and translocation of $NF-{\kappa}B$ to nuclear. Furthermore, IVE significantly inhibited the increases of hind paw swelling, skin thicknesses and inflammatory cell infiltrations induced by CA injection. Therefore, IVE will be favorably inhibited the acute edematous inflammations. Conclusion : These results provide evidences that anti-inflammatory effect of IVE is partly due to the reduction of some inflammatory mediators by suppression of $NF-{\kappa}B$ pathway.

A Multicenter Noncomparative Clinical Study on Midface Rejuvenation Using a Nonabsorbable Polypropylene Mesh: Evaluation of Efficacy and Safety

  • Background Facial rejuvenation can be achieved using a variety of techniques. Since minimally invasive procedures for face lifting have become popular because of their convenience and short operating time, numerous minimally invasive surgical procedures have been developed. In this study, a nonabsorbable polypropylene mesh is introduced as a new face lifting instrument, with the nasolabial fold as the main target area. In this paper, we report the efficacy and safety of a polypropylene mesh in midface rejuvenation. Methods Thirty-three subjects with moderate-to-severe nasolabial folds were enrolled from two medical institutions for a noncomparative single-sample study. A mesh was inserted above the superficial muscular aponeurotic system layer, reaching the nasolabial folds through a temporal scalp incision. After 3 weeks, the temporal end of the mesh was pulled to provide a lifting effect. Then, the mesh was fixed to the deep temporal fascia using nonabsorbable sutures. To evaluate efficacy, we compared the scores on the Wrinkle Severity Rating Scale and a visual analog scale for patient satisfaction between the baseline and 7 weeks postoperatively. In addition, we evaluated safety based on the incidence of adverse events. Results The treatment was deemed effective at improving wrinkles in 23 of 28 cases, and patient satisfaction improved significantly during the study period. There were seven cases of skin or subcutaneous tissue complications, including edema and erythema, but there were no suspected serious adverse events. Conclusions Face lifting using a nonabsorbable mesh can improve nasolabial folds without serious adverse effects. Thus, this technique is safe and effective for midface rejuvenation.

맥동전자장에너지의 미세전류가 가토의 상처치유에 미치는 영향 (The Effects of Pulsed Electromagnetic Energy and Microcurrent on Wound Healing in Rabbits.)

  • The purpose of this study w8s to evaluate the effects of pulsed electromagnetic energy(Diapulse) and microcurrent on the wound healing in rabbits. 15 domestic rabbits were randomly assigned to the PRME(n=5). MC(n=5) and CON(n=91 group. Each rabbits were anesthetized with lidocaine HCL $2\%$. Skin wounds were created laterally on the back of IS domestic rabbits(33cm). From 24 hours after being injured, the rabbits of the PEME group were irradiated with an intensity of 3 at a 300 pulses per second, which were applied for 15 minutes every day during the 12 days. The MC group were stimulated with an intensity of $50{\mu}A$ at frequency of40 pulses per second, which were applied for 15 minutes every day during the 12 days. The CON group were not stimulated. The rabbits were sacrificed and the incised wound pans were processed appropriately for the light microscopic examination on the 3rd day, 6th day and 12th day before the beginning of wound treatment. The areas of wound were measured with metric graph paper. The results were as tallows. 1 The PRME and MC group compared with control group showed that wound closure rate increased on 6th, 12th day. 2. It was found that the CON group did not show a complete maturation and had a chronic inflammatory response. Judging from the irregularity of intercellular space and the loose alignment of connective tissue. these findings showed that wound healing was delayed. 3. It showed that inflammatory cells. fibroblasts and epithelial cells activity rapidly processed in the PEME group compared with the CON group. It was found that the PEMI; group showed a advanced remodeling of epithelial layer and a positive repair of connective tissue. 4. It showed that fibroblasts, epithelial cells and inflammatory cells activity rapidly processed in the MC group compared with the CON group. It was found that the MC group showed a improved remodeling of epithelial layer and a dense connective tissue.

피부화상으로 유도된 심근손상에서 Protein Kinase C의 역할 (The Role of Protein Kinase C in the Cardiac Injury Induced by Skin Burn)

  • 본 연구는 피부화상으로 유도된 심근손상에서 protein kinase C (PKC)의 역할을 알아보고자 하였다. 수컷 흰 쥐(SD계)에 15%의 피부전층화상을 유도한 뒤, PKC activator인 phorbol 12-myristate 13-acetate (PMA)와 PKC inhibitor인 bisindolylmaleimide (BIS)를 투여하여 5시간, 24시간 후에 심장을 적출하여 생화학적 미세구조적 입체해석학적 방법을 실시하였다. 혈청 AST와 creatinine 은 화상 후 5시간군과 화상 후 5시간+BIS 투여군에서 높게 나타났고, KC와 MPO 활성은 PMA 투여군이 BIS 투여군보다 낮게 나타났다. 미세구조적 관찰 결과 PMA 투여군에서는, 화상으로 인한 핵의 분열, 과수축대 형성, 사이원반의 분리 현상이 다소 완화된 형태로 관찰되었고, BIS 투여군에서는 화상 단독군에서 나타나는 형태적 변화 뿐만 아니라 비정상적인 형태의 사립체도 일부 관찰되었다. 전체적으로 5시간에서 24시간군으로 가면서 손상이 완화된 양상을 나타내었다. 입체해석학적 결과에서는 화상으로 인한 근원섬유의 체적밀도 감소가 PMA와 BIS 투여로 인해 증가되었고, 사립체의 체적밀도와 수밀도의 증가는 BIS군에서 가장 높게 나타났다. 결론적으로 PKC의 활성화는 화상으로 인해 손상된 심근에서 염증반응을 감소시켜 심근 손상을 보호한다고 사료된다.

애완토끼에서 발생한 Cheyletiellosis 피부염에 대한 selamectin 치료 1례 (A Case of Naturally Acquired Cheyletiellosis in a Rabbit: Therapeutic Trial of Selamectin)

  • 5개월령의 암컷 토끼가 소양증, 탈모 및 점액화농성 안루로 내원하였다. 전신에서 경미에서 중등도 인설이 관찰되었다. 안면부, 이개내측, 사지 중수골과 중족골의 내측, 좌측슬관절 앞쪽의 탈모, 발적, 구진, 농포와 가피등의 다양한 피부병변이 나타났다. 채취된 인설의 현미경 검사에서 다수의 Cheyletiella spp가 관찰되었다. 다수의 이염색성 백혈구, 호산구 및 림프구가 세포학적 검사에서 나타났으며 곰팡이 배양검사는 음성이었다. 세균배양결과 Pasteurella spp가 검출되었다. 검사결과에 따라 Cheyletiella 피부염으로 진단하였다. 치료는 selamectin 6mg/kg을 2주 간격으로 2회 국소 적용하였으며 치료 1개월 후에 임상증상이 현저하게 경감되었고 피부병변은 대부분 정상으로 회복하였다. 따라서 selamectin의 국소적용은 토끼의 Cheyletiellosis의 치료에 효과적인 것으로 판단되었다.

개에서 발생한 피부 상피친화성 T-세포 림프종: Lomustine 및 Gemcitabine에 대한 임상적 반응 (Cutaneous Epitheliotropic T-Cell Lymphoma in a Dog: Clinical Responses to Lomustine and Gemcitabine)

  • 5년령의 중성화된 암컷 말티즈가 1개월 동안 지속된 전신적 발적, 탈모 및 소양감으로 내원하였다. 피부조직구증으로 진단되어 면역억제치료를 1개월 간 지속하였지만 다발성의 발적된 반 및 판 병변들이 새롭게 발생하였다. 피부병변에 대한 압박도말검사에서 림프종을 지시하는 원형세포들이 관찰되었다. 조직학적 검사에서는 종양세포들의 외피 및 부속구조물에 대한 친화성이 나타났다. 면역염색결과 종양세포들은 CD3에 대하여 양성, CD79a에 대해서는 음성반응을 나타내어 피부 상피친화성 T-세포 림프종으로 확진하였다. $70mg/m^2$의 lomustine을 2-3주 간격으로 총 2회 투여하였으며 부분적인 치료반응이 관찰되었다. 피부병변의 악화와 lomustine의 이용제한으로 gemcitabine ($500mg/m^2$, 1주일당 1회 30분간 혈관주입)을 이용한 화학치료를 시작하였다. 총 3회 치료가 실시되었으나 질병의 진행을 억제하지 못하였으며, 최초 lomustine 치료 69일 후 안락사 되었다.
