Effect of dietary supplementation of grape skin and seeds on liver fibrosis induced by dimethylnitrosamine in rats

  • Grape is one of the most popular and widely cultivated fruits in the world. Although grape skin and seeds are waste product of the winery and grape juice industry, these wastes contain large amounts of phytochemicals such as flavonoids, phenolic acids, and anthocyanidins, which play an important role as chemopreventive and anticancer agents. We evaluated efficacies of grape skin and seeds on hepatic injury induced by dimethylnitrosamine (DMN) in rats. Treatment with DMN significantly increased levels of serum alanine transaminase, aspartate transaminase, alkaline phosphatase, and bilirubin. Diet supplementation with grape skin or seeds (10% daily for 4 weeks) prevented these elevations. The grape skin and seeds also restored serum albumin and total protein levels, and reduced the hepatic level of hydroxyproline and malondialdehyde. Furthermore, grape skin and seeds reduced DMN-induced collagen accumulation, as estimated by histological analysis of liver tissue stained with Sirius red. Grape skin and seeds also reduced hepatic stellate cell activation, as assessed by ${\alpha}$-smooth muscle actin staining. In conclusion, grape skin and seeds exhibited in vivo hepatoprotective and antifibrogenic effects against DMN-induced liver injury, suggesting that grape skin and seeds may be useful in preventing the development of hepatic fibrosis.

The Effectiveness of Infrared Thermography in Patients with Whiplash Injury

  • Objective : This study aims to visualize the subjective symptoms before and after the treatment of whiplash injury using infrared (IR) thermography. Methods : IR thermography was performed for 42 patients who were diagnosed with whiplash injury. There were 19 male and 23 female patients. The mean age was 43.12 years. Thermal differences (${\Delta}T$) in the neck and shoulder and changes in the thermal differences (${\Delta}dT$) before and after treatment were analyzed. Pain after injury was evaluated using visual analogue scale (VAS) before and after treatment (${\Delta}VAS$). The correlations between ${\Delta}dT$ and ${\Delta}VAS$ results before and after the treatment were examined. We used Digital Infrared Thermal Imaging equipment of Dorex company for IR thermography. Results : The skin temperature of the neck and shoulder immediately after injury showed $1-2^{\circ}C$ hyperthermia than normal. After two weeks, the skin temperature was normal range. ${\Delta}T$ after immediately injuy was higher than normal value, but it was gradually near the normal value after two weeks. ${\Delta}dT$ before and after treatment were statistically significant (p<0.05). VAS of the neck and shoulder significantly reduced after 2 week (p=0.001). Also, there was significant correlation between ${\Delta}dT$ and reduced ${\Delta}VAS$ (the neck; r=0.412, p<0.007) (the shoulder; r=0.648, p<0.000). Conclusion : The skin temperature of sites with whiplash injury is immediately hyperthermia and gradually decreased after two weeks, finally it got close to normal temperature. These were highly correlated with reduced VAS. IR thermography can be a reliable tool to visualize the symptoms of whiplash injury and the effectiveness of treatment in clinical settings.

Healing Effect of 635 nm Laser Diode in Skin Injury (635nm 레이저다이오드의 피부 결손 치유 효과)

  • Low level laser therapy has various therapy effects. This paper performed the basic study for fabricating the low level laser therapy apparatus, and one of the goals of this paper was to make this apparatus used handily. The apparatus has been fabricated using the laser diode and microprocessor unit. The apparatus used a 635 nm laser diode for laser medical therapy and was designed for a pulse width modulation type to increase stimulation effects. In this study, the designed device was used to find out how 635 nm laser diode affected the skin injury of SD-Rat(Sprague-Dawley Rat). We divided the participants into two groups; irradiation group which was irradiated 10 minutes a day for 9 consecutive days, and none irradiation group. The results showed that the study group had lower incidence of inflammation and faster recovery, compared with the control group.

  • Physilogical disorders of P. ginseng occurred in farmer's field were reviewed in relation to symptom. In root, red skin, rough skin, rust, root rot complex, round root, fine root stripe, freezing injury, cracking, sleeping and uneven emergence were frequently appeared. In leaf and stem, yellowing, early defoliation, leaf bum, Papery leaf, white freezing injury, wind injury, stem cracking were the main troubles. Red skin of root and leaf yellowing gave the greatest negative impact on ginseng production. Some cases of damage by pesticides, excess boron and industrial pollutants were reported. Physiological disorders related to quality factors, such as inside cavity, inside white sponge-like ect. after processing were discussed.

The effects of manual acupuncture, invasive laser acupuncture and laser skin irradiation at Liver Seunggyeok(肝勝格) on the repair of D-galN-induced Liver Injury in Rats (간승격(肝勝格) 혈위(穴位)에 시술된 침자(鍼刺)와 침습(侵襲) 및 비침습(非侵襲) 레이저침요법(鍼療法)이 간손상(肝損傷) 회복에 미치는 영향)

  • Objective: This study was performed to investigate the effect of manual acupuncture, invasive laser acupuncture and laser skin irradiation at LU8, LR4, HT8 and LR2(Liver Seunggyeok) on D-galN-induced liver injury in rats. Method: Liver injury was induced with D-galN. The experimental rats were divided four groups(Control group, EXP-1, EXP-2, EXP-3). In the Control group, liver injury-induced and not treated. EXP-1 group was liver injury-induced and carried out manual acupuncture with Young-Su(against the meridian course and following the course of the meridian) & Won-Bang(by twisting and rotating the needle) acupuncture method at Liver Seunggyeok. EXP-2 group was liver injury-induced and carried out invasive laser acupuncture at Liver Seunggyeok. EXP-3 group was liver injury-induced and carried out laser skin irradiation at Liver Seunggyeok. Result: In the change of body weight(in 1 week), EXP-1, EXP-2 and EXP-3 groups were significantly increased as compared with control group. In the change AST & ALT, EXP-1 and EXP-2 groups were significantly decreased as compared with control group. In the change of SOD, EXP-1, EXP-2 and EXP-3 groups were significantly increased as compared with control group. In the change of WBC, EXP-2 group was significantly increased as compared with control group. Conclusion: Manual acupuncture, invasive laser acupuncture and laser skin irradiation at Liver Seunggyeok had hepatotherapeutic effect on the treatment of hepatocytotoxity. Invasive laser acupuncture was as effective as manual acupuncture on the treatment of hepatocytotocity.

A literature study on dermatological efficacy and drug induced liver injury of Dictamnus dasycarpus Turcz (백선피(白鮮皮)의 피부과적 효능과 약인성 간손상에 대한 문헌 연구)

  • Objectives : The root bark of Dictamnus dasycarpus has been frequently used to treat patients with skin diseases in Korea. Recently, wild root of D. dasycarpus are reported to induce liver injury. Methods : Traditional uses of D. dasycarpus for skin diseases were analysed bibliographically. In addition, reported cases were collected and analysed using pubmed and national digital library. Results : In taiwan, D. dasycarpus revealed to be one of major herbs for skin diseases and many researchers in worldwide had reported its dermatological efficacies. Reported cases related in liver injury described that hepatocellular or cholestatic liver injury were seen in patients eating wild root of D. dasycarpus. In addition, 6 cases in worldwide and 1 case in Korea showed that patients with drug induced liver injury (DILI) ingested not root bark of D. dasycarpus but prescriptions containing root bark of D. dasycarpus. These mean that wild root of D. dasycarpus (Bongsam or Bongwhangsam) was estimated to be closely related in DILI. Whereas, it was difficult to confirm direct correlation between root bark of D. dasycarpus used as herbal medicine by doctor of Korean medicine and DILI. Conclusions : these results imply that wild root of D. dasycarpus is closely related in DILI and strong recommendation not to take it without consultation by experts is needed. In addition, although there are no evidences of direct correlation between root bark of D. dasycarpus and DILI, doctor of Korean medicines should pay attention to use root bark of D. dasycarpus in their clinics.

Effects of lipopolysaccharide and CpG-DNA on burn-induced skin injury

  • Destruction of the skin barrier by thermal injury induces microbial invasion, which can lead to the development of systemic infection and septic shock. Microbial pathogens possess pathogen-associated molecular patterns (PAMPs), which are recognized by conserved receptors. To understand the role of PAMPs in thermal injury-induced mice, LPS or CpG-DNA were topically applied to dorsal skin after thermal injury. We observed an increase in the number of inflammatory cell infiltrates as well as thickening in the dermis upon treatment with LPS or CpG-DNA. We also found that expression of IL-$1{\beta}$, MIP-2, and RANTES induced by thermal injury was enhanced by LPS or CpG-DNA. In addition, the proportions of $CD4^+$ and $CD^8+$ T cells in the spleen and lymph nodes were altered by LPS or CpG-DNA. These results provide important information concerning PAMPs-induced inflammation upon thermal injury and provide a basis for studying the role of PAMPs in thermal injury-induced complications.

Effects of Copper / Zinc-Containing Superoxide Dismutase (Cu, Zn-SOD) and Catalase on Paraquat-Induced Injury in Primary Cultured Rat Skin Fibroblast (일차 배양한 백서 피부섬유아세포에서 Paraquat 독성에 미치는 SOD 와 Catalase 의 영향)

  • The participation of superoxide in initiating tissue damage derived from xenobiotics is best illustrated by paraquat intoxication. In the present study, the roles of superoxide dismutase and catalase on paraquat-induced cell injury were investigated using primary cultured rat skin fibroblast. The degree of cell injury was assassed by the conversion of reduced MTT to a blue formazan. Paraquat produced concentration-and time-related cell injury in cultured rat skin fibroblast. Paraquat induced-cell injury was aggravated by pretreatment of aminotriazol (AT: 10 mM), an catalase inhibitor, and attenuated by addition of catalase (100∼500 unit/ml). However, the effects of diethyldithiocarbamate (DDC : 10 mM), copper- and zinc-containing superoxide dismutase (Cu, Zn-SOD) inhibitor, and Cu, Zn-SOD on paraquat-induced injury were not significant. These results suggest that hydrogen peroxide might be more responsible factor than superoxide in the pathogenesis of paraquat-induced cell injury.

The Effects of Anti-Histamine and Mast Cell Stabilizer against Ischemia-Reperfusion Injury to TRAM Flap in Rat (백서 복직근피판의 허혈-재관류 손상에 대한 히스타민 차단제의 효과)

  • Purpose: The purpose of this study was to evaluate the role of mast cell and histamine as typical product of mast cell in ischemia-reperfusion injury of muscle flap using H2 receptor blocker and mast cell stabilizer. Methods: Thirty-five Sprague-Dawley rats weighing 250-300 gm were divided into four groups; Group I: Control group without ischemia, Group II: Normal saline injection group with ischemia, Group III: Cimetidine injection group with ischemia, Group IV: Sodium cromoglycate injection group with ischemia. Well established single pedicled transverse rectus abdominis musculocutaneous(TRAM) flap was designed in all rats and were rendered ischemia by clamping the artery for 150 minutes. All injections were applied intramuscular around gluteal area 30 minutes before reperfusion. The flap survival was evaluated at 7 days after operation. Neutrophil counts and mast cell counts were evaluated 24 hours after reperfusion. Results: The difference of skin flap survival between control group and cimetidine injection group was not significant. In the normal saline injection group flap survival was markedly decreased compared to that of control group. The muscle flap survival was similar to the results of skin flap survival. The neutrophil counts were significantly decreased in control group and sodium cromoglycate injection group than normal saline injection group. The mast cell counts were significantly decreased in cimetidine injection group and control group than both normal saline injection and sodium cromoglycate injection groups. The protective effect of sodium cromoglycate was not seen in the skin flap, but the muscle flaps showed protective effects of sodium cromoglycate compared to normal saline injection group. Conclusions: It is suggests that commonly used antihistamine(H2 receptor blocker) has protective effect against ischemia-reperfusion injury to skin and muscle flaps by reducing neutrophil and mast cell. The mast cell stabilizer was not effective for skin flap but, possibly, for muscle flap.

The effect of 470nm LED Irradiation on skin injury of RAT (470nm LED 광이 RAT 피부결손에 미치는 효과)

  • This paper performed the basic study for developing the Photodynamic Therapy Equipment for medical treatment. We developed the 4channel light medical therapy apparatus for external injury cure using a LED light source. This equipment was fabricated by using microcontroller, and designed to enable us to control light irradiation timer, intensity and reservation. In this paper, the designed device was used to find out how 470nm LEDs light source affects the skin wound of RAT. In the experiment, $1cm_2$ wounds on the External injury of RAT were made. Light irradiation RAT and none light irradiation RAT divided, each RAT was irradiated one hour a day for 15 days. In result, compared with none light irradiation RAT, the lower incidence of inflammation and faster recovery was shown in light irradiation RAT.
