The Effect of Moutan Cortex on Pro-inflammatory Cytokines through NF-κB & MAPKs pathway in HMC-l (목단피의 NF-κB와 MAPKs 억제를 통한 Pro-inflammatory Cytokines 활성에 미치는 효과)

  • Objective : Moutan Cortex (the root bark of Paeonia suffruticosa Andr.) is widely used in oriental medicine as a remedy for inflammation. However, as yet there is no clear explanation of how MC(Moutan Cortex) affects the production of inflammatory cytokine. This study was to determine the effects of Essence extracted MC on the mast cell-mediated inflammatory responses. Method : We observed the effect of MC on compound 48/80-induced histamine release of rat peritoneal mast cells and the effect of administering MC on PCA in rat. We measured the amount of inflammatory cytokine production induced by the phorbol myristate acetate (PMA) plus calcium ionophore(A23187) in the human mast cell line (HMC-1) incubated with various concentrations of MC. The TNF-$\alpha$ protein levels were analysised by Western blot. The TNF-$\alpha$, IL-6 and IL-8 secreted protein levels were measured by the ELISA assay. The TNF-$\alpha$, IL-6 and IL-8 mRNA levels were measured by the RT-PCR analysis. NF-$\kappa$B, phospho-I$\kappa$B and MAPKs were exmined by Western blot analysis. The NF-$\kappa$B promoter activity was examined by luciferase assay. Result : 1. Enzyme immunoassay indicated that MC suppressed histamine secretion of rat peritoneal mast cells. 2. In PCA dependent on IgE, MC had anti-allergic effect of the internal surface of rat skin. 3. Western blot indicated that MC decreased TNF-$\alpha$ protein levels. 4. ELISA indicated that MC decreased TNF-$\alpha$, IL-6 but MC had no significant effect on IL-8 in HMC-1 cells. 5. RT-PCR indicated that MC decreased TNF-$\alpha$, IL-8 but MC had no significant effect on IL-6 in HMC-l cells. 6. Western blot indicated that MC suppressed the induction of MAPKs, NF-$\kappa$B & phospho-I$\kappa$B activity in HMC-1 cells. 7. Luciferase assay indicated that MC suppressed the PMA plus A23187-induced NF-$\kappa$B promoting activityin HMC-1 cells. Conclusion : In this study, we have found that MC is an inhibitor of NF-$\kappa$B, MAPKs & cytokines on the mast cell-mediated inflammatory responses.

The influences of Phaseolus radiatus L.'s Ethanol Extracts and Fractions on Skin Whitening and Anti-inflammatory Effects (녹두(綠豆) 에탄올 추출물 및 분획물이 피부의 미백, 항염증 효과에 미치는 영향)

  • Objectives : This study is designed to clarify whitening, anti-inflammatory effect of fractions extracted from the mixture of Phaseolus radiatus L. and ethanol. Methods : In this experiment, we were intended to reveal whitening, anti-inflammatory effect of fractions extracted from the mixture of Phaseolus radiatus L. and ethanol. The whitening activity was confirmed by UV blocking activity, tyrosinase inhibiting activity, and melanin formation inhibiting activity. Anti-inflammatory activity is confirmed by measurement of cytotoxicity level by MTT assay and measurement of Cytokine expression, which is the main mediator of inflammation reaction. Results : As a results, overall activity was high in the ethyl acetate fraction. Tyrosinase inhibitory activity was less than 20% at all concentrations, but the activity to inhibit melanin self-production was higher than that of ethyl acetate fraction at $32.19{\pm}2.79%$ at $100{\mu}g/m{\ell}$. And ethyl acetate fraction had a relatively high UV blocking activity. In the anti-inflammatory test, the concentration-dependent activity was shown, and the chloroform and ethyl acetate fractions showed significant NO production inhibitory activity. Cytokine expression was superior to that of the final stage of B cell differentiation, and cell viability was over 80% except for the chloroform fraction at the concentration of $200{\mu}g/m{\ell}$. Conclusions : The results of this experiment confirmed the whitening effect and anti-inflammatory effect of Phaseolus radiatus L.'s extracts and fractions and report the possibility of application as external medicine.

A Study on Efficiency of Essential Oil for Dandruff Symptoms Relief - Focused on Tea tree oil - (비듬 증상 완화를 위한 에션셜 오일의 유효성 연구 - 티트리 오일을 중심으로 -)

  • The reason that aromatherapy began to draw attention again as the alternative therapy is because the essential oils that are used for aromatherapy not only allow the human body to maintain homeostasis but also have an effect on curing a mental feeling of fatigue and because these efficacies and effects are proceeding with being proved scientifically. Also, with coming to be high in a voice of concern about severally side effects that the modern medicine and the synthetic chemistry have, the aromatherapy came to be paid attention as the replacement therapy that can substitute for and supplement this. In particular, at the time of requiring a study on a substance to substitute for this after having been announced in Japan for 1998 a risk of Zincpyrithione that is being used as a component of dandruff treatment, it is thought that the aroma oil that was known for having an anti-dandruff effect has a merit of being easy in a user's access, thus it will be effective in curing the symptom of dandruff. Accordingly, this study conducted the following experiment in a bid to examine whether the aroma oil has the anti-microbial effect on the actual malassezia. First of all, it measured the minimum concentration of aroma oil in which the growth of malassezia is inhibited, by carrying out MIC(Minimum Inhibition Concentration) test in terms of anti-microbial activity against Malassezia furfur KCTC 7744 while using oils of Patchouli, Rosemary, Lavender, and Tea tree among aroma oils that are being used for dandruff treatment. As a result of that, Tea tree oil has the most excellent anti-microbial activity, and the measurement result of anti-microbial activity was shown in order of Lavender oil>Rosemary oil>Patchouli oil. Next, it carried out the is actually eased by applying Tea tree oil directly to the scalp of dandruff. In consequence of the experiment, the massage of Tea tree oil indicated the result of relieving the symptoms of dandruff such as scale, itch, pain, inflammation, and sebum, and among these symptoms of dandruff, it was shown to be most effective in easing itch. Especially, this clinical experiment was indicated that the massage of Tea tree oil has the most effect on relieving itch of targets with a dry-skin type who had no experience of dandruff treatment.

Group A Streptococcal Bacteremia in Pediatric Patients: A Single-Center Study (소아환자에서의 A군 사슬알균 혈류감염: 단일기관 연구)

  • Purpose: Group A streptococcus (GAS) is a common pathogen in pediatric patients and often causes acute pharyngotonsillitis and skin and soft tissue infections. In addition, bacteremia with significant morbidity and mortality can also occur. This study was conducted to describe the clinical manifestations and treatment outcomes of pediatric GAS bacteremia patients in Korea. Methods: This was a single-center, retrospective study. From January 2000 to December 2016, pediatric patients aged ≤18 years with GAS bacteremia were studied. Clinical manifestations, underlying diseases, intensive care unit stay, and antibiotic susceptibility were evaluated. Results: During the study period, 19 patients had GAS bacteremia. Ten (53%) were male, and the median age was 7.4 years (range, 0.3-17.4 years). Fourteen (74%) had chronic underlying diseases. Five (26%) were immunocompromised (leukemia and chronic kidney disease). Eight (42%) had lymphatic or vascular malformations, of which seven had lesions with signs of inflammation. Three (16%) developed pneumonia, and two of them received ventilator care. The 30-day mortality rate was 6% (1/19), and the cause of death was bacteremic pneumonia. All GAS isolates were sensitive to penicillin. Fifteen (79%) were sensitive to both erythromycin and clindamycin. Conclusions: This study identified various clinical manifestations of GAS bacteremia. GAS should be considered as a potential pathogen that can cause bacteremia and result in a serious clinical course.

Free radical scavenging, anti-inflammatory and melannin synthesis inhibitory activities of Gloeostereum incarnatum (느릅나무버섯 자실체의 메탄올과 열수추출물의 항산화, 항염증 및 멜라닌 생성저해 활성)

  • Gloeostereum incarnatum is an edible and medicinal mushroom belongs to Family Cyphellaceae of Polyporales, Basidiomycota. The purpose of this study was to investigate the free radical scavenging, anti-inflammatory, and melanin synthesis inhibitory activities of fruiting bodies of Gloeostereum incarnatum. In the free radical scavenging activities, the mushroom extracts showed good 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging and chelating activity on the ferrous ions compared with the positive control, BHT. The mushroom extract suppressed nitric oxide (NO) production in LPS-induced RAW 264.7 macrophage cells in dose dependant manners. Significant reduction of paw edema of rats were observed at 2~6 h after administration with 50 mg/kg of the methanol and hot-water extracts, which were comparable with treatment of 5 mg/kg of indomethacin, the positive control. The melanin synthesis of Melanoma B16/F10 cells treated with $100{\mu}g/mL$ of the methanol and hot water extracts decreased melanin concentration to 50% and 45% compared with the control, arbutin. Therefore, the experimental results showed that methanol and hot-water extracts of Gloeostereum incarnatum fruiting bodies might be used for good sources of anti-inflammatory, free radical scavenging, and skin whitening agents for human health.

Biological Evaluation of the Methanolic Extract of Eriobotrya japonica and Its Irradiation Effect (비파 메탄올 추출물의 생리활성 및 방사선 조사 효과)

  • Eriobotrya japonica has been used as a folk medicine for treatment of skin diseases, inflammation, coughing, phlegm, and ulcers in Korea and other Asian countries. In a search for possible bioactive agents from natural sources, we found that the methanolic extracts from various parts of E. japonica showed moderate antioxidative and antimicrobial activities in several in vitro bioassay systems. Additionally, the respective parts of E. japonica were irradiated at 20 kGy to investigate the effects of irradiation. Gamma irradiation of E. japonica extracts removed the deep greenish color without affecting its natural biological activities such as its antioxidative and antimicrobial properties. Based on these findings, the methanolic extracts of this plant source may be not affected by gamma irradiation as its bioactive constituents may be insensitive to this irradiation. Moreover, the methanolic extract of E. japonica may serve as a good natural resource for beneficial functions in food and other related industries.

  • Components of extracellular matrix and the matrix-degrading enzymes are some of the key regulators of tumor metastasis and angiogenesis. Hyaluronic acid (HA), a matrix glycosaminoglycan, is known to promote tumor adhesion and migration, and its small fragments are angiogenic. Until now, we have compared levels of hyaluronidase, an enzyme that degrade HA, in normal adult prostate, benign prostate hyperplasia and prostate cancer tissues and in conditioned media from epithelial explant cultures, using a substrate (HA)-gel assay and ELISA-like assay (Kim et al., unpublished results). The present review described an overall characterization of hyaluronidases and its application to human diseases. The hyaluronidases are a family of enzymes that have, until recently, deed thorough explication. The substrate for these enzymes, hyaluronan, is becoming increasingly important, recognized now as a major participant in basic processes such as cell motility, wound healing, embryogenesis, and implicated in cancer progression. And in those lower life forms that torment human beings, hyaluronidase is associated with mechanisms of entry and spread, e.g. as a virulence factor for bacteria, for tissue dissection in gas gangrene, as a means of treponema spread in syphilis, and for penetration of skin and gut by nematode parasites. Hyaluronidase also comprises a component of the venom of a wide variety of organisms, including bees, wasps, hornets, spiders, scorpions, sh, snakes and lizards. Of particular interest is the homology between some of these venom hyaluronidases and the enzyme found in the plasma membrane of mammalian spermatozoa, attesting to the ancient nature of the conserved sequence, a 36% identity in a 300 amino acid stretch of the enzyme protein. Clearly, hyaluronidase is of biological interest, being involved in the pathophysiology of so many important' human disorders. Greater effort should be made in studying this family of enzymes that have, until recently, been overlooked. Also, oriental medical application of the hyaluronidase will be discussed with respect to inhibition and suppression of inflammation and malignacy.

  • The ulcerative colitis is a chronic inflammatory bowel disease with an unknown etiology. The major symptoms of ulcerative colitis are diarrhea, abdominal pain and hematochezia. However, arthritis, skin disorders, hepatobiliary inflammation and uveitis are occasionally recognized as systemic complications. Although there are few reports of coexistent pulmonary and inflammatory bowel disease, the lung is not generally considered to be a target organ in ulcerative colitis. We report a patient with ulcerative colitis-related bronchilolitis obliterans organizing pneumonia confirmed by video-assisted thoracoscopic surgery, who responded to corticosteroid therapy.

Pathological changes of the heart of olive flounder Paralichthys olivaceus in experimental Streptococcus parauberis infection (Streptococcus parauberis 인위 감염에 의한 넙치, Paralichthys olivaceus 심장의 병리학적 변화)

  • We conducted bacteriological and histopathological analysis for olive flounder Paralichthys olivaceus after experimental infection with Streptococcus parauberis (FP2284) isolated from diseased olive flounder under different stress conditions. Experimental challenge was performed in healthy flounder (40.4 g in average body weight) by intraperitoneal (i.p.) injection with $2{\times}10^{8}$ CFU/fish under normal (no stress) or netting (for 2 min, twice/day) stress condition. The cumulative mortalities of no-stress and netting stress group were 70% and 95%, respectively. The most prevalent changes observed in experimentally infected flounder were darkness of skin and inflammation of the heart. Severe pericarditis, myocarditis and fibrosis were observed in the heart of the affected flounder. The results of viable counts showed the number of bacteria of the heart tissue was maintained over the $10^{4}$ CFU $g^{-1}$ heart for 3 weeks after inoculation. Histological lesions of the heart was more extensive and gradual decrease in bacterial numbers of heart tissue was delayed under stress condition.

  • A variety of experiments concerning the development of ideal prosthetic grafts for correcting circumferential tracheal defects have been performed. The requirements for an ideal tracheal prosthesis are impermeability to air, consistency to prevent collapse, and acceptance by the host tissue causing a minimum inflammatory reaction, allowing fibroblastic infiltration and epithelialization. The synthetic material, polyurethane(PU), is known as a biocompatible polymer with an inert component. In this study, the tracheal prosthesis was made from microporous PU(30 micrometer in diameter) coated with gelatin and reinforced with isoplastic rings. This procedure provides the prosthesis with a compression strength. The out side diame er of the prosthesis was 20 mm with a length of 30 mm. The gelatin used in the study was obtained from pig skin and immobilized and cross-linked by irradiation(60 Co gamma ray) to promote host tissue incorporation and render the prosthesis epithelization after implantation. Animal experiments using 10 mongrel dogs were performed to compare three kinds of prosthesis; gelatin coated polyurethane graft, uncoated polyurethane graft, and prosthesisf pericadium complex graft. After 6 weeks of implantation, the epithelialization of implants was seen on the gelatin-coated and prosthesisfpericadium complex grafts. Implanted prosthesis were complicated by airway obstruction due to anastomosis granuloma. Early tracheal stenosis was found in the uncoated graft group. Two kind of anastomosis techniques were tested on the gelatin-coated prosthesis. Everted anastomosis resulted severe granuloma than the inverted anastomosis. In the prosthesislpericadium complex graft, bacteria and inflammation at a anastomotic site was found. Based on these results, gelatin coated porous polyurethane trachea prosthesis is biocompatible and may be useful in clinical application with further investigation.
