This study was conducted to investigate the effect of cryoprotectants on the storage stability of Lactobacillus fermentum SK152, which was isolated as a probiotic candidate. Solutions of 10% glucose, trehalose, dextrin, and skim milk powder were used as cryoprotectants. The survival rates of L. fermentum SK152 after freeze-drying were 5.6% (dextrin), 2.2% (skim milk powder), 1.7% (glucose), and 1.5% (trehalose), suggesting that dextrin was most effective at minimizing the cell death of L. fermentum SK152 by lyophilization. The survival rates of L. fermentum SK152 stored at 4℃ ranged from 37% (dextrin)-90% (skim milk powder) after 8 weeks, while those at 20℃ ranged from 4% (dextrin)-12% (skim milk powder) after 7 weeks, indicating that skim milk powder was the best at minimizing the cell death of L. fermentum SK152 during storage, irrespective of storage temperature, among the cryoprotectants used.

Milk proteins have many potential sequences within their primary structure, each with a specific biological activity. In this study, we compared and investigated the bioactivities of hydrolysates of the domestic (A, B) and imported (C, D) skim milk powders generated using papain digestion. MALDI-TOF analysis revealed that all milk powder proteins were intact, indicating no autolysis. Electrophoretic analysis of hydrolysates showed papain treatment caused degradation of milk proteins into peptides of various size. The antioxidant activity of the hydrolysates, determined using 2,2-azino-bis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) and total phenolic contents (TPC) assays, increased with incubation times. In all skim milk powders, the antioxidant activities of hydrolysates were highest following 24 h papain treatment (TPC: A, 196.48 μM GE/L; B, 194.52 μM GE/L; C, 194.76 μM GE/L; D, 163.75 μM GE/L; ABTS: A, 75%; B, 72%; C, 72%; D, 57%). The number of peptide derived from skim milk powders, as determined by LC-MS/MS, was 308 for A, 283 for B, 208 for C, and 135 for D. Hydrolysate A had the highest antioxidant activity and the most potential antioxidant peptides amongst the four skim milk powder hydrolysates. A total of 4 β-lactoglobulin, 4 αs1-casein, and 56 β-casein peptide fragments were identified as potential antioxidant peptides in hydrolysate A by LC-MS/MS. These results suggest that domestic skim milk could have applications in various industries, i.e., in the development of functional foods.
This study was carried out to establish the optimal conditions for curd yogurt using saccharified rice solution. A central composite design was applied to investigate the effects of pectin (2.8~8.4 g), skim milk (14~70 g) and oligosaccharide (28~84 g) ratio on the physicochemical and sensory characteristics of curd yogurt using saccharified rice solution. pH, total acidity, and color value, were analyzed and sensory evaluation was performed. The pH of curd yogurt decreased with decreasing pectin and oligosaccharide ratio. The addition of pectin had a significant effect on the viscosity while skim milk had a significant effect on the color value (a and b value). The results of sensory evaluation showed that, oligosaccharide and skim milk had significant effects on sweetness and sour taste. Oligosaccharide and skim milk masked the sour taste of curd yogurt. The optimum range of ingredients for curd yogurt using saccharified rice solution was predicted to be 4.27~4.90 g of pectin, 30.80~41.30 g of skim milk, and 28.00~36.10 g of oligosaccharide. Based on the overlapped results of physicochemical and sensory evaluation, the optimal amounts of pectin, skim milk and oligosaccharide were 4.59 g, 36.50 g and 32.05 g, respectively.
Objective: This study was conducted to assess the level of pregnancy-associated glycoprotein (PAG) in whole and skim milk samples, and its suitability for early pregnancy diagnosis in goats. Methods: A two-step sandwich enzyme-linked immunosorbent assay (ELISA) system for estimation of milk PAG was developed and validated, which employed caprine-PAG specific polyclonal antisera. Whole and skim milk samples (n = 210 each) from fifteen multiparous goats were collected on alternate days from d 10 to d 30, and thereafter weekly till d 51 post-mating. PAG levels in milk samples were estimated by ELISA and the pregnancies were confirmed at d40 post-mating by transrectal ultrasonography (TRUS). Results: The level of PAG in whole and skim milk samples of both pregnant and nonpregnant goats remained below the threshold values until d 24 after mating. Thereafter, PAG concentration in whole and skim milk increased steadily in pregnant goats, whereas it continued below the threshold in non-pregnant does. The PAG profiles in whole and skim milk of pregnant goats were almost similar and exhibited strong positive relationship (r = 0.891; p<0.001). Day 26 post-mating was identified as the first time-point for significantly (p<0.05) higher milk PAG concentration in pregnant goats than to non-pregnant goats. When compared to TRUS examination for pregnancy diagnosis, the accuracy and specificity of PAG ELISA using whole and skim milk samples were 94.5% and 95.4%; and 95.3% and 100%, respectively. The high values of area-under-curve (0.904 [whole milk] and 0.922 [skim milk]), demonstrate outstanding discrimination ability of the milk assays. Among the sampling dates chosen, d 37 post-mating was identified as the best suitable time point for collection of milk samples to detect pregnancy in goats. Conclusion: The PAG concentration in whole and skim milk of goats collected between days 26 and 51 post-breeding can be used for the accurate prediction of pregnancy and may be useful for assisting management decisions in goat flocks.
Skim milk progesterone($P_4$) profiles in 74 dairy cows were determind to monitor postpartum ovarian activity by radioimmunoassay. Milk samples were collected from each cow every 5 days from 10 to 90 days postpartum. Signs of estrus were observed twice daily, and status of the ovaries and uterus were examined every 10 days by rectal palpation. Results are summarized as follows: 1. Cows were categorized into five types by the change of skim milk $P_4$ profiles; Type I(normal) : Cyclic changes of skim milk $P_4$ profiles appeared within 20 days postpartum(12 cows, 16.2%), Type II(cycle delayed) : Cyclic changes of skim milk $P_4$ profiles appeared from 21 to 60 days postpartum(39 cows, 52.7%), Type III(cycle ceased with low $P_4$) : Onset of the estrous cycle within 20 days postpartum but ceased later with low levels of $P_4$ (7cows, 9.5%), Type IV(cycle ceased with high $P_4$) : Onset of the estrous cycle within 20 days postpartum but ceased later with high levels(>3.0 ng/ml) of skim milk $P_4$ (4 cows, 5.4%), Type V(acyclicity) : Skim milk $P_4$ concentration remained low(<1.0 ng/ml) until 80 days postpartum(12 cows, 16.2%). 2. Out of the 17 cows classified as the Type III and Type V by skim milk $P_4$ profiles, 13 cows had inactive ovaries and remaining 6 cows had single or multiple follicular cysts in their ovaries by rectal palpation. All 4 cows of Type IV had a persistent corpus luteum in their ovaries. 3. Approximately eighty percent of the cows had begun ovarian activity by 60 days postpartum and 90.6% by 90 days by skim milk $P_4$ profiles, but only 39.2% by 60 days and 71.7% by 90 days had shown visible estrus signs. The mean days from parturition to the first, second and third ovulations determined by skim milk $P_4$ profiles was $28.0{\pm}11.0$, $46.4{\pm}13.3$ and $66.4{\pm}11.5$ days and the visible estrus signs were 9.3%, 38.1%, and 48.6%, respectively. The mean days from parturition to the first visible estrus was $57.2{\pm}15.9$ days. These results indicated that milk $P_4$ profile of each Types by radioimmunoassay can be utilized for monitoring postpartum ovarian and would be useful for the early detection of ovarian dysfunction in dairy cow.

The object in this study is to investigate the effects of high pressure and freezing processes on the curdling of skim milk depending on the presence of transglutaminase (TGase) and glucono-δ-lactone (GdL). Skim milk was treated with atmospheric freezing (AF), high pressure (HP), pressure-shift freezing (PSF) and high pressure sub-zero temperature (HPST) processing conditions. After freezing and pressure processing, these processed milk samples were treated with curdling agents, such as TGase and GdL. Pressurized samples (HP, PSF and HPST) had lower lightness than that of the control. In particular, PSF had the lowest lightness (p<0.05). Likewise, the PSF proteins were the most insoluble regardless of whether they were activated by TGase and GdL, indicating the highest rate of protein aggregation (p<0.05). Furthermore, the TGase/GdL reaction resulted in thick bands corresponding to masses larger than 69 kDa, indicating curdling. Casein bands were the weakest in PSF-treated milk, revealing that casein was prone to protein aggregation. PSF also had the highest G' value among all treatments after activation by TGase, implying that PSF formed the hardest curd. However, adding GdL decreased the G' values of the samples except HPST-treated samples. Synthetically, the PSF process was advantageous for curdling of skim milk.
The optimum conditions for lactic acid fermentation by Lactobacillus acidophilus in the media containing malt syrup, skim milk or skim milk and malt syrup were studied to develope marketable fermented beverages from barley. The optimum sugar concentration of malt syrup was $10^{\circ}$ Bx. Addition of skim milk to malt syrup enhanced bacterial growth and acid formation. The best flavor of the product was obtained from the media containing 5% skim milk and sterilized at $90^{\circ}C$ for 30 minutes. Optimum temperature and pH for the formentation were $40^{\circ}C$ and 6.0, and bacterial cells grew logarithmically upto 16 hours.
유제품(乳製品)의 품질(品質)과 밀접(密接)하게 관련(關聯)되어 있는 skim milk protein의 가열(加熱)에 의(依)한 상대변화(狀態變化)를 단백질(蛋白質) 화학적(化學的) 측면(側面)에서 연구(硏究)하였다. 1. Skim milk 중의 calcium농도(濃度)가 높아질수록 가열시(加熱時) 점도(粘度)는 급증(急增)하였으며 gel화(化)가 일어나는 온도(溫度)는 점점 저하(低下)되었다. 이러한 사실(事實)로 보아 calcium ion은 유단백질(乳蛋白質)의 열안정성(熱安定性)에 결정적(結定的) 영향(影響)을 미치며 calcium존재(存在) 하(下)에서 가열(加熱)시킬 때에는 우유단백질(牛乳蛋白質)들을 균일분산계(均一分散系)로부터 불균일회합상태(不均一會合狀態)로 전이(轉移)시키는 것으로 결론(結論)되었다. 2. 일정(一定) 온도(溫度) skim milk의 안정성(安定性)에 대(對)한 가열시간(加熱時間)의 효과(效果)는 전혀 나타나지 않았다. 3. 등전침전(等電沈電)시킨 casein을 재용해(再溶解)시킨 후(後), skim milk 내(內)에서와 동일(同一)한 calcium농도(濃度) 환경(環境)으로 조절(調節)하고 가열(加熱)하였을 때 skim milk가 불안정성(不安定性)을 보이는 온도(溫度)보다도 저온(低溫)에서 침전(沈澱)으로 분리(分離)되었다. 따라서 가열(加熱)에 불안정(不安定)한 구분(區分)은 whey protein일 것으로 추정(推定)하였으며, 이는 skim milk 내(內)에서 casein에 의(依)하여 보호(保護)되는 것으로 해석(解析)되었다. 5. pH를 변경(變更)시킨 skim milk의 점도(粘度) 증가(增加)에는 calcium ion에 의(依)한 기여(寄與)가 컸으며, calcium은 유단백질(乳蛋白質)의 응집(凝集)을 촉진(促進)할 때 Ca-bridge로 작용(作用)하는 것이라고 결론(結論)되었다. 6. 비가열(非加熱)의 skim milk는 6개의 fraction으로 분획(分劃)되었음에 비(比)하여, $95^{\circ}C$에서 10분(分) 가열(加熱)했을 때는 fraction A 및 B가 소실(消失)되었고 새로운 fraction F@를 출현(出現)시켰다. 7. Electrophoretogram에서 fraction C 및 C#는 모두 3개의 band를, fraction F 및 F#는 모두 4개의 band를 나타냈으며 가열(加熱) 전후(前後)의 이동도(移動度)에도 차이(差異)가 없었다. Fraction D는 6개의 band로 D#에는 그 외(外)에도 3개의 band가 더 나타났다. fraction E 및 E#는 모두 4개의 band를 보였으나, 그 성분(成分)들의 이동도(移動度)는 모두 대응(對應)되지 않았다. Fraction A 및 B, 그리고 fraction D 및 E에는 가열(加熱)로 인(因)하여 변화(變化)를 받게되는 유단백성분(乳蛋白成分)이 포함(包含)되어 있을 것으로 추정(推定)되었다.
Behavior of Listeria monocytogenes in Skim milk during fermentation by Lactobacillus bulgaricus YI-2 and Streptococcus lactis FYI-1 were determined. Autoclaved skim milk was inoculated with ca. 10$^{3}$ L. monocytogenes (Strain LM91-1 or LM 96-2) cells/ml, and with 5.0, 1.0, 0.5 or 0.1% of a milk culture of either L. bulgaricus TI-2 or S. lactis FYI-1. Skim milk containing ca. 10$^{3}$ L. monocytogenes was incubated at 37 or 42$\circ $C for 15 h with L. bulgaricus YI-2, and at 21 or 30$\circ $C for 15 h with S. lactis FYI-1. Cultured skim milks were stored at 4$\circ $C in the refrigerater. Samples were plated on Oxford Agar with oxford antimicrobic supplement to enumerate L. monocytogenes and on either modified MRS agar to enumerate lactic acid bacteria. L. monocytogenes survived the 15-h fermentation with S. lactis FYI-1 in all combinations of level of inoculum and temperature of incubation, but inhibition of growth ranged from 94 to 100%. When incubated with over the 1.0% of L. bulgaricus, L. monocytogenes inhibited or disappeared in fermented skim milk from 9 h after incubation. Especially, incubation at 42$\circ $C with 5.0% L. bulgaricus YI-2 as inoculum appeared to be the most effective inhibitory combination for strain LM 91-1, causing 100% inhibition in growth based on maximum papulation attained. In most instances of incubated with L. bulgaricus YI-2, growth of the pathogene appeared to be completely inhibited when the pH dropped below 4.38.
This study was undertaken to assess the effect of the Korean red ginseng extracts on the growth of lactic acid bacteria, such as Lac. acidophilus and Str. thermophilus. The results are summarized as follows: 1. The growth of all the lactic acid bacteria tested in this study was not influenced when the skim milk was added with the red ginseng extracts in concentration of less than 4.0%. But the growth was remarkably inhibited in the presence of 8.0% red ginseng extracts in skim milk. 2. Titratable acidity was more or less increased by addition of red ginseng extracts as compared with control except when the amount of 8.0% red ginseng extracts were added into skim milk. 3. No particular effect was observed in the growth of lactic acid bacteria in case of addition of saponins. But the remarkable inhibitory effect was observed in the skim milk with the amount of 1.2mg/ml the fractions extracted by ethyl ether from red ginseng extracts.
