The myostatin (MSTN) gene, as a negative regulator of skeletal muscle growth, has been proposed to be associated with production traits in farm animals. In the present study, a T/C variant at -125 bp (relative to ATG start codon) of 5'regulatory region of rabbit MSTN was identified by direct sequencing. Two hundred and twenty two rabbits, which were randomly sampled from 3 breeds (Ira rabbits, Champagne rabbits and Tianfu black rabbits), were genotyped by high-resolution melting (HRM). Comparing the genotyping results of 47 samples with direct sequencing, the HRM showed high sensitivity (0.96) and high specificity (0.98). In the three rabbit breeds, the allele C was the predominant allele. The polymorphic site showed high heterozygosity (He = 0.48) and high effective number of alleles (Ne = 1.91). Zooplankton community dynamics were studied after establishment of an artificial vegetation island (AVI) in Lake Paldang, from April 2005 to November 2006. There were distinct seasonal and inter-annual changes of total zooplankton abundance at the survey site. Total zooplankton abundance rapidly increased in spring and fall, while it remained low throughout winter. During summer, the dynamics of zooplankton community seemed to be largely affected by hydrological parameters such as, precipitation and inflow. Total zooplankton abundance and biomass below AVI was much higher than that of pelagic zone (L1) in Lake Paldang. Copepoda and cladocera represented the main bulk of the zooplankton community from summer to fall at the both sites. Copepods were more dominant at AVI area, while cladocera were more dominant at pelagic zone (L1). Water quality, prey and habitat condition, species competition between zooplankton seemed to play important roles in dominance of the copepoda and cladocera in zooplankton community at AVI area. Our results conclude that artificial vegetation island provide the stable habitat and besides phytoplankton, diverse food to zooplankton, and consequently influence the diversity and richness of zooplankton community.
Bentic macroinvertebrate and fish fauna were investigated from September 2008 to October 2009 in the middle reach of the Suyoung river in Busan. During the survey period benthic macroinvertebrates were collected total 35 species at the three sites (Hanmul-gyo, Dongcheon-gyo, Seokdae Jct.) of Suyoung river. At that sites, species diversity index was 1.9, 1.5, 1.6, dominance index was 0.75, 0.81, 0.86 and Korean Saprobic Index (KSI) was 1.8(B), 4.4(D), 4.4(D) on average, respectively. Dominant species was Caenis Kua, Chironomidae sp., at Hanmul-gyo site and Limnodrilus gotoi, Chironomidae sp., at Dongcheon-gyo and Seokdae Jct. sites. Fish fauna was also investigated at the two sites (Hanmul-gyo and Seokdae Jct.) of Suyoung river. Total caught fishes were 10 species 32 individuals in the first survey and 6 species 26 individuals in the second survey. Dominant species was Carassius auratus in the both surveys. Index of Biological Integrity (IBI) value was C grade at the Hanmul-gyo and D grade at the Seokdae Jct.

The Saemangeum project is one of the biggest reclamation efforts in Korea and may cause coastal ecosystem change due to altered environments and habitat loss. Since February 2002, benthic environment and community structure in the Saemangeum studied area were studied to assess the influence of the project on macrofaunal community. The result of seasonal study from February, 2002 to August 2005 is reported here. Overall, changes of species numbers and dominant species of benthic animals in the periods before (1988) and after $(2002{\sim}2005)$ the Saemangeum dike construction were not evident both inside and outside the dike. However, local environmental and community change were noted The partial completion of Saemangeum dike $(4^{th}\;dike)$ in June 2003 altered water circulation and sediment deposition patterns both inside and outside the dike. Fine sediment was accumulated inside and outside the $4^{th}$ dike while coarse sediment dominated near the main channel (Sinsi gate). Benthic community resl)ended to the altered sediment type in these areas. Species number and diversity in both site was low compared to other sites. The dominant species in these areas were composed of the benthos that had not commonly occurred in the Saemangeum subtidal area.

The C-terminus ends of the second putative transmembrane domains of both m1 and m2 muscarinic receptors contain a triplet of amino acid residues consisting of leucine (L), tyrosine (Y) and threonine (T), This triplet is repeated as LYT-LYT in m2 receptors at the interface between the second transmembrane domain and the first extracellular loop. Interestingly, however, it is repeated in a transposed fashion (LYT-TYL) in the sequence of m1 receptors. In this work we employed site-directed mutagenesis to investigate the possible significance of this unique sequence diversity for determining the distinct differential drug-receptor interaction and cellular function at m1 muscarinic receptor. Mutation of the LYTTYL sequence of m1 receptors to the corresponding m2 receptor LYTLYT sequence, however, did not result in a significant change in the binding affinity of the agonist carbachol or in the affinity of the majority of a series of receptor antagonists which are able to discriminate between wild-type m1 and m2 receptors. Surprisingly, the LYTLYT ml receptor mutant demonstrated markedly enhanced coupling to activation of phospholipase C without a change in its coupling to increased cyclic AMP formation. There was also an enhanced receptor sensitivity in transducing elevation of intracellular Ca$\^$2+/. These changes were not due to alterations in the rate of receptor. desensitization or sequestration, On the other hand, the reverse LYTLYT-LYTTYL mutation in the m2 receptor did not alter its coupling to inhibition of adenylate cyclase, but slightly enhanced its coupling to stimulation of PI hydrolysis, Our data suggest that the LYTTYL/LYTLYT sequence difference between ml and n12 muscarinic receptors is not involved in determining receptor pharmacology. On the other hand, while these differences might play a role in the modulation of muscarinic receptor coupling to PI hydrolysis, they are not important for specifying coupling of various subtypes of muscarinic receptors to different cellular signaling pathways.

The purpose of this study is to provide the fundamental material and information for the plant maintenance after rooftop planting through physiochemical characteristics. The characteristics of artificial soils after rooftop planting from 1993 to 1999 was investigated. Fourteen investigation areas were selected from 4 cities(2 areas selected by each year). The analysis of the circumstances of the areas, the physical characteristics, and the chemical characteristics of the soil were conducted. The artificial soil pH ranged 5.26∼7.40 showing that after construction the soil pH tended to decrease. The soil bulk density of the site was lowest in 1999, 0.15g/㎤, and used to increase toward 1993. We found the fact that the soil bulk density increased gradually after rooftop application . The coefficients of permeability of the soils range from 0.016 to 0.052 cm/sec, which seemed to be in good permeability level. The artificial soils had relatively high water moisture capacity of 62.69∼71.36%. The soil organic matter content of the artificial soils ranged from 0.43 to 1.34%. The exchangeable caution concentration in the artificial soil ranged, Na, 2.36∼4.71mg·{TEX}$kg^{-1}${/TEX}, Mg 0.88∼2.84mg·{TEX}$kg^{-1}${/TEX},K 2.97∼9.61 mg·{TEX}$kg^{-1}${/TEX}, and Ca 9.39∼28.23 mg·{TEX}$kg^{-1}${/TEX}. The amount of total N ranged from 0.003 to 0.286% in study sites. Soil chemical properties varied year to year and showed little tend. The research results showed that some characteristics of the artificial soil were changed after rooftop planting, i.e., soil pH and soil bulk density. Soil bulk density had a negative relationship with the coefficient of permeability, showing that the drainage condition might be limited after some period. This study suggests that a diversity of the research in the changes of the plant growth basis on the areas after construction.
Although thermal effluent from coastal power plants affects marine ecosystems, few studies have examined the spatiotemporal distribution of macrobenthic communities in a coastal area of Korea with thermal discharge. This study analyzed the species composition and abundance of a macrobenthic community in relation to environmental variables for a period of 1 year. In total 334 macrobenthic faunal species were collected; the mean density was 3,221 ind/$m^2$. The number of species and the density of macrobenthic fauna increased with distance from the thermal discharge site. Cluster analysis indicated that the macrobenthic community could be divided into two groups: group I in shallow (< 30 m deep) and group II in the outer areas (> 30 m deep). Group I showed the lowest species density and diversity. Four polychaetes, including Magelona japonica, Spiophanes bombyx, Scolotoma longifolia and Chaetozone setosa, all of which have been dominant species since 1987, exhibited higher mean densities in the area distant from the thermal discharge (the outer and north areas). Conversely, the warm-adapted and opportunistic species, such as the amphipods Urothoe convexa and Mandibulophoxus mai, the bivalve Felaniella sowerbyi, and the polychaete Rhynchospio sp., were more abundant in the thermal discharge region. The results of this study indicate that thermal effluent influences macrobenthic communities in the shallow area, while other environmental variables, such as depth, sediment grain size, and TOC, are more important determinants of the macrobenthic communities in deeper regions (> 30 m deep).
Polyhydroxyalkanoate (PHA) is a class of biodegradable plastics that have great potential applications in the near future. In this study, the micro-biodiversity and productivity of PHA-accumulating bacteria in activated sludge from a domestic wastewater treatment plant were investigated. A previously reported primer set and a self-designed primer set (phaCF1BO/phaCR2BO) were both used to amplify the PHA synthase (phaC) gene of isolated colonies. The new primers demonstrated higher sensitivity for phaC, and combining the PCR results of the two primer sets was able to widen the range of detected genera and raise the sensitivity to nearly 90%. Results showed that 85.3% of the identified bacteria were Gram-negative, with Ralstonia as the dominant genus, and 14.7% were Gram-positive. In addition, Zoogloea and Rhizobium contained the highest amounts of intracellular PHA. It is apparent that glucose was a better carbon source than pentone or tryptone for promoting PHA production in Micrococcus. Two different classes, class I and class II, of phaC were detected from alphaproteobacteria, betaproteobacteria, and gammaproteobacteria, indicating the wide diversity of PHA-accumulating bacteria in this particular sampling site. Simultaneous wastewater treatment and PHA production is promising by adopting the high PHA-accumulating bacteria isolated from activated sludge.

In this work, thus, the enhanced stimulation of PI hydrolysis in the LYTLYT $M_1$ receptor mutant was further investigated. The stimulation of PI hydrolysis by carbachol was enhanced in the mutant $M_1$ receptor, and this change was not due to alterations in the rate of receptor desensitization or sequestration. The observed larger response to carbachol at mutant $M_1$ receptors was also not due to an artifact resulting from selection of CHO cells which express higher levels of G-proteins or phospholipase C. Our data suggest that although the LYTTYL sequence in $M_1$ muscarinic receptors is not involved in determining receptor pharmacology, mutation of the sequence enhanced the coupling of $M_1$ receptors to the stimulation of phospholipase C.

Studies about architectural planning for elderly residences have been carried out to examine various factors including the unit plan, site planning, and size planning. However, no studies have been undertaken that focus on the residents' daily life patterns. This study offers the design guidelines to support the daily life patterns of the elderly through the planning of elderly housing. For this study, elderly residents in Tokyo, Yokohama etc. in Japan were interviewed and surveyed. The results indicate that the changes to the daily life patterns of the residents living in elderly housing are the reduction of household, the increase of free time, and the change of social relationships. The elderly want to utilize their free time and focus on their social relationships. However, the elderly residences that we surveyed pay no consideration to social spaces and programs for their residents except for the cafeteria. To support the daily life patterns of the elderly, elderly housing should set a high value on the location, shared spaces, and unit plans of the residences in order to consider the personal diversity of the residents. Accessibility to public transportation systems and facilities should to be considered in the initial location planning. The revitalization of the public spaces should to be considered during the architectural planning stage. Considerations of personal taste during the unit planning stage are also necessary.
