한국산 쏘가리의 기원과 분자계통진화적 위치 (Origin of the Korean Mandarin Fish, Siniperca scherzeri and Its Molecular Phylogenetic Relationships to Other Siniperca Fishes)

  • 이 연구는 cytochrome b 유전자 서열을 이용하여 쏘가리속 어류의 분자계통 진화적 관계와 쏘가리 지역 개체군 간의 유전적 차이를 조사함으로써 한극산 쏘가리의 분자진화적인 위치와 유래를 알기 위하여 실시하였다. 그 결과, 쏘가리속 어류의 진화초기에 S. roulei가 가장 먼저 분화하였으며 그 후 조사대상 어류인 쏘가리속 6개 종 (S. schezeri, S. undu-lata, S. fortis, S. obscura, S. knerii 및 S. chuatsi) 이 분화한 것으로 생각된다. 그러나 이들 어류들의 분화 우선순위는 통계학적으로 강하게 지지되지 못해서 명확하게 밝히기는 어려웠다. 한편 쏘가리 개체군은 크게 세 개의 집단으로 구분되었다. 첫 번째 집단은 한국산 개체군과 중국북부 (Liaoning, Henan) 개체군이다. 두 번째 집단은 Anhui, Fujian 및 Guangxi 개체군이며, 세 번째 집단은 Zhejiang 개체군이다. 첫 번째 집단 내 한국산 쏘가리 개체군과 중국 북부(Liaoning, Henan)개체군 사이의 염기서열 차이는 1~5 base pairs (bp)였으며 첫 번째 집단과 두 번째 집단의 염기서열 차이는 31~43 bp였다. 그리고 두 번째 집단과 세 번째 집단 사이의 염기서열 차이는 37~44 bp를 나타냈으며, 첫 번째 집단과 세 번째 집단 사이의 염기서열 차이는 27~29 bp였다. 따라서 한국산 쏘가리의 유래는 중국의 북부 개체군이 신생대 3기 Pliocene 기간 중에, 즉 초기 빙하기 이전 시기에 중국 중부 또는 남부의 쏘가리 개체군으로부터 최초로 분화된 후 빙하기를 거치면서 한반도로 그 분포범위를 확장함으로써 생겨난 것으로 추정된다.

  • 쏘가리는 국내 주요 경제성 어종이며 방류 및 복원사업의 대상 어종 중 하나이다. 본 연구는 금강의 보 설치구간에서 쏘가리의 이동 특성 및 서식처 범위 등을 평가하기 위해 Radio telemetry를 이용하여 연구를 실시하였다. 연구결과 산란기에 방류한 개체들의 경우 방류 즉시 상류로 10km 이상 소상하였지만, 비 산란기에 방류한 개체들은 2개체를 제외한 모든 개체들이 방류 지점으로부터 400 m 이내에서 정착하였다 (Ss11, 3.2 km; Ss 15, 1.4 km). 산란기에 상류로 소상한 개체들이 정착한 지점의 서식처 특성은 기존에 알려진 쏘가리의 산란장 특성과 유사하게 나타났으며, 현재 퇴적이 진행되고 있는 상태였다. 쏘가리가 감지는 수변부에서 높은 비율로 나타났으며, 이는 수변부에 큰 돌이 많이 분포하는 것, 쏘가리가 주로 돌 틈에 숨어서 서식하는 특성과 관련이 있는 것으로 판단된다. 쏘가리의 개체 특성과 이동거리와의 상관관계 분석 결과 무게와 비대지수가 이동거리와 양의 상관관계를 보였으며 성별과 크기는 이동거리와 관련이 없는 것으로 나타났다. 일주기 이동 범위 분석 결과$214.94m^2{\sim}3,257.19m^2$로 나타났으며 주로 수변부에 국한되어 있는 것으로 확인되었다. 이와 같은 결과는 쏘가리 방류사업, 혹은 복원에 유용한 자료로 활용될 수 있을 것으로 판단된다.

Disc 전기영동법에 의한 수종 담수어 혈장 단백질의 분획 (Fractionation of Plasma Protein on the Several Fresh Water Fishes by Disc Electrophoresis)

  • The plasma proteins of fresh water fishes have been fractionated by disc electrophoresis in acrylamide gels utilized as an electrophoretic supporting medium. The species of fishes examined in this experiment were Anguilla japonica, Misgurnus mizolepis, Parasilurus asotus, Siniperca scherzeri, Pelteobagrus fulvidra, Carassius carassius, Cyprinus carpio, and Hemibarbus labeo, obtained in the Han River. Disc electrophoresis was performed as described by Ornstein and Davis. Gels and buffer solution were prepared by the method developed by W.J.Kim. The separation gels were 7% acrylamide gel. The fractionation of plasma proteins showed 13 bands in Anguilla japonica, 10 in Misgurnus mizolepis, 15 in Parasilurus asotus, 12 in Siniperca scherzeri, 11 in Pelteobagrus fulvidra, 13 in Carassius carassius, 9 in Cyprinus carpio, and 13 in Hemibarbus labeo. The patterns of plasma protein on the each species of fishes were different in the number of bands, ratio of contents, relative mobilities, and forms of fractionation.

충주호에서의 쏘가리와 메기의 이동범위 및 행동 (Movement range and behavior of mandarin fish (Siniperca scherzeri) and catfish (Parasilurus asotus) in Chungju Lake)

  • This study was conducted to collect the information on the behavioral characteristics and the habitat environment of mandarin fish (Siniperca schezeri) and catfish (Parasilurus asitus) with acoustic telemetry method in Chungju Lake, Korea. Mandarin fish tended to stay within 1km from the release points in downstream, and had a strong diurnal behavior. They approached to the lakefront at night. They also preferred to stay at deep water off the lakefront. The average swimming speed was faster at night (0.4BL/s) than during a day (0.2BL/s). They swam the shallow water area at night. Catfish frequently moved between upstream and downstream. Catfish tended to act during a day. The average swimming speed was faster during a day (0.3BL/s) than at night (0.2 BL/s). The average swimming depth was 14.3m, and they tended to float about 4m during a day.

쏘가리(Siniperca scherzeri)의 시기에 따른 영양성분 변화 (Seasonal Variation in the Nutritional Composition of the Muscle of Mandarin Fish Siniperca scherzeri)

  • Seasonal variation in the composition and fatty acid, amino acid, cholesterol, and vitamin contents in the muscles of the mandarin fish Siniperca scherzeri, caught in Soyangho Lake, Korea, was studied. The moisture content was 74.5-78.7 g/100 g, lipids ranged between 1.06 and 4.23 g/100 g, protein between 18.5-20.7 g/100 g, and ash between 0.74 and 2.04 g/100 g. The moisture content was lowest in May and tended to increase from June to October. The highest protein levels occurred in May, which coincided with the timing of oocyte maturation, and then decreased at the beginning of spawning. The highest lipid levels occurred in May and November coinciding with the periods before spawning and hibernation. The ratio of EAA/NEAA was higher than the value recommended by FAO for an 'ideal' food protein. There were significant differences in Mg and Fe contents with spawning; they were highest in May. Contents of vitamin A and E were from not detected (ND) to $17.1{\mu}g/100g$ and ND to 0.69 mg/100 g, respectively and were highest in November. The content of niacin tended to decrease in June and increased until November and hibernation. The cholesterol content was 14.9-78.9 mg/100 g; it decreased from March to July and then increased until November. In conclusion, there is potential for expanding the commercial utilization of the mandarin fish as a food resource.

Effect of dietary protein and lipid level on growth, feed utilization, and muscle composition in golden mandarin fish Siniperca scherzeri

  • A feeding trial was designed to assess the effects of dietary protein and lipid content on growth, feed utilization efficiency, and muscle proximate composition of juvenile mandarin fish, Siniperca scherzeri. Six experimental diets were formulated with a combination of three protein (35, 45, and 55%) and two dietary lipid levels (7 and 14%). Each diet was fed to triplicate groups of fish ($8.3{\pm}0.1g$) to apparent satiation for 8 weeks. The results showed that growth performance in terms of weight gain (WG) and specific growth rate (SGR) increased with increasing dietary protein level from 35 to 55% at the same dietary lipid level. At the same dietary lipid content, WG and SGR obtained with diets containing 55% protein was significantly higher than those obtained with diets containing 45 and 35% protein. No significant effect on growth rate was found when the dietary level of lipid was increased from 7 to 14%. While the levels of protein and lipid in the diets had no significant effect on feed intake, other nutrient utilization efficiency parameters including daily protein intake (DPI), feed efficiency (FE), and protein efficiency ratio (PER) showed a similar trend to that of growth rates, with the highest values obtained with diets containing 55% protein. Muscle chemical composition was not significantly affected by the different dietary treatments for each dietary lipid or protein level tested. These findings may suggest that a practical diet containing 55% protein and 7% lipid provides sufficient nutrient and energy to support the acceptable growth rates and nutrient utilization of mandarin fish juveniles.

Feeding ratio affects growth, body composition, and blood chemistry of mandarin fish (Siniperca scherzeri) in recirculating aquaculture system

  • The effects of various feeding ratios on the growth, body composition, and blood chemistry of the juvenile mandarin fish Siniperca scherzeri (initial body weight 9.6 g) were examined in recirculating freshwater system equipped with 21, 300 L tanks at 20 fish per tank. The triplicate groups of seven feeding ratios treatments were prepared: 100% (control), 95%, 90%, 85%, 80%, 75%, and 70% of satiation. The feed amount of control group was determined by supplying with apparent satiation and then the feed amounts of the other six feeding groups were determined based on the feed amount of the control group. Fish were hand-fed with test diet (55.4% crude protein) for 10 weeks. Weight gain (WG) and specific growth rate of fish fed to 100% satiation were not significantly (p > 0.05) different from those of fish fed to ≥ 80% satiation but were significantly higher than those of fish fed to 75% and 70% satiation. Feed efficiency, protein efficiency ratio, and protein retention of 100% satiation were not significantly different from those of 95% and 90% satiation but were significantly (p <0.05) lower than ≤ 85% satiation. Condition factor, hepatosomatic index, and coefficient variation were not significantly (p > 0.05) affected by feeding ratio. Whole body composition and contents of hematocrit, glutamic oxaloacetic transaminase, glutamic pyruvic transaminase, glucose, total protein, and high-density lipoprotein cholesterol in blood serum were not significantly (p > 0.05) affected by the feeding ratio; however, content of total cholesterol tended to decrease as the feeding ratio decreased. Using broken-line analysis of WG, it was suggested that the optimum feeding ratio of juvenile mandarin fish, ranging from 9.0 g to 37.0 g, appeared to be 87.7% of satiation without growth inhibition.
