Recent Research Trends in Korean Medicine Treatment of Diabetes mellitus (당뇨의 한의학적 치료에 대한 최근 연구동향)

  • Objective The purpose of this study was to analyze the directions of Korean Medicine treatment of diabetes mellitus. Methods We reviewed the 52 studies about diabetes mellitus which had been published from 2000 to 2007. We selected those studies from the search engine of the web site of five journals. Those were the Journal of Korean Oriental Medical Society, Korean Journal of Oriental Physiology & Pathology, the Journal of Korean Acupuncture & Moxibustion Society, Korean Journal of Oriental Internal Medicine and the Journal of Korean Pharmacopuncture Institute. Results 1. The types of diabetes mellitus model in studies were the model used Alloxan(2cases), the model used Streptozotocin(41cases), NOD mice(1case), ob/ob mice(1case), db/db mice(5cases) and rats fed highfat diet(2cases). 2. The types of method in studies were pharmacopuncture(8cases), herbal medcine(47case) and both pharmacopuncture and herbal medcine(3cases). the types of materials in studies were single herb(24cases), multiple herbs(32cases) and both single and multiple herbs(4cases). 3. The types of evaluation criteria in studies were glucose, pancreas, liver, kidney, serum lipid, oxidative stress, nervous system, vascular system and immunity. Conclusions There have been reported many studies of diabetes mellitus in Korean Medicine. It requires to study further types of diabetes mellitus, kinds of herbs and complications of diabetes mellitus for Korean Medicine treatment of diabetes mellitus was covered the ground.

Randomized Clinical Controlled Trials of a Herb Remedies in Korea - Systematic Review (한약을 이용한 한국에서의 무작위 임상시험 개괄적 검토)

  • Objective: The study aimed to review all randomized controlled clinical trials (RCTs) performed in Korea with herbal medicine, to produce further useful information for herbal-drug development. Methods: All papers reporting RCT with herbal drugs were collected via public database of Korea Science and Technology Integration Services, and various Oriental medicine journals. Then, the clinical question, herbal medicine, result, RCT design, and its conductor were analyzed. Results: A total of 20 RCTs were finally selected, and most of them were performed after 2003. The main subjects of RCT were related to heart and blood circulation, dermatitis, respiration disorder, Sasang constitution, or psychiatric disorders. Eight RCTs were done with a single herb while 12 RCTs were with multiple herbal formulae. In particular, three RCTs for atopic dermatitis with three formulae showed positive results. The quality of RCT was considered as better than average. Conclusions: This study revealed the current status of RCT using herbal drugs. This result would provide helpful information for research and drug development using traditional herbal medicine.

  • Medicinal herbs are significant source of synthetic and herbal drugs. In the commercial market, medicinal herbs are used as raw drugs, extracts or tinctures. Isolated active constituents are used for applied research. For the last few decades, phytochemistry (study of plants) has been making rapid progress and herbal products are becoming popular. According to Ayurveda, the ancient healing system of India, the classical texts of Ayurveda, Charaka Samhita and Sushruta Samhita were written around 1000 B.C. The Ayurvedic Materia Medica includes 600 medicinal plants along with therapeutics. Herbs like turmeric, fenugreek, ginger, garlic and holy basil are integral part of Ayurvedic formulations. The formulations incorporate single herb or more than two herbs (poly-herbal formulations). Medicinal herb contains multitude of chemical compounds like alkaloids, glycosides, saponins, resins, oleoresins, sesquiterpene, lactones and oils (essential and fixed). Today there is growing interest in chemical composition of plant based medicines. Several bioactive constituents have been isolated and studied for pharmacological activity. R. cordifolia is an important medicinal plant commonly used in the traditional and Ayurvedic system of medicine for treatment of different ailments. This review illustrates its major constituents, pharmacological actions substantiating the claims made about this plant in the traditional system of medicine and its clinical applications.

Synthesis of Bismuth Vanadate as Visible-light Photocatalyst by Precipitation Reaction (침전 반응에 의한 가시광 광촉매 Bismuth Vanadate 합성)

  • Bismuth vanadate($BiVO_4$) with monoclinic phase as photocatalyst under visible light is synthesized by precipitation reaction in hot water. Properties such as crystal phase, particle morphology and visual light absorbance as well as the effects of thermal treatment for $BiVO_4$ powders are investigated. $BiVO_4$ powders with both single monoclinic phase and 0.2 ${\mu}m$ in particle size are synthesized when precipitate is stirred in water for 5 h at 95$^{\circ}C$. Well-developed monoclinic phase and light absorption property under 535 nm are observed as a result of thermal treatment for 1 h at 300$^{\circ}C$ after precipitation reaction in water for 5 h at 95$^{\circ}C$. Degradation of monoclinic crystal is found in firing above 350$^{\circ}C$, and particle growth is occurred in firing above 550$^{\circ}C$.

Analytic Study on Treatises of the Jounal of Oriental Gynecology (대한한방부인과학회지(大韓韓方婦人科學會誌)에 수재(收載)된 논문(論文)에 대한 분석고찰(分析考察))

  • Objectives : Every year tens of treatises have been published on the journal of oriental gynecology. But until now there were no papers on the analysis of those treatises and no efforts to evaluate them. Method : To analyze the treatises of journal of oriental gynecology, we examined 19 journals published from 1987 to 2001. There were no rules to analyze treatises, we followed the way of Seo, classifying treatises into bibliographic, experimental and clinical categories. Results and conclusions : 1. Until 1994 most of treatises were bibliographic but from 1995 experimental treatises were increased. 2. Many(29.5%) of them were studied about the diseases in pregnancy, Many(25.4%) of experimental treatises were reserched about the diseases on leukorrhea. 3. In the clinical treatises most of them were case reports. 4. In the bibliographic treatises 90% of them were studied about the classic contents categories. 5. In the clinical treatises the way of treatment showed about same percentage between single herb medication and acupuncture-herb medication complex.

Putative multiple reaction monitoring strategy for the comparative pharmacokinetics of postoral administration Renshen-Yuanzhi compatibility through liquid chromatography-tandem mass spectrometry

  • Background: Exploring the pharmacokinetic (PK) changes of various active components of single herbs and their combinations is necessary to elucidate the compatibility mechanism. However, the lack of chemical standards and low concentrations of multiple active ingredients in the biological matrix restrict PK studies. Methods: A putative multiple reaction monitoring strategy based on liquid chromatography coupled with mass spectrometry (LC-MS) was developed to extend the PK scopes of quantification without resorting to the use of chemical standards. First, the compounds studied, including components with available reference standard (ARS) and components lacking reference standard (LRS), were preclassified to several groups according to their chemical structures. Herb decoctions were then subjected to ultrahigh-performance liquid chromatography coupled with quadrupole time-of-flight mass spectrometry analysis with appropriate collision energy (CE) in MS2 mode. Finally, multiple reaction monitoring transitions transformed from MS2 of ultrahigh-performance liquid chromatography coupled with quadrupole time-of-flight mass spectrometry were used for ultrahigh-performance liquid chromatography coupled with triple quadrupole mass spectrometry to obtain the mass responses of LRS components. LRS components quantification was further performed by developing an assistive group-dependent semiquantitative method. Results: The developed method was exemplified by the comparative PK process of single herbs Radix Ginseng (RG), Radix Polygala (RP), and their combinations (RG-RP). Significant changes in PK parameters were observed before and after combination. Conclusion: Results indicated that Traditional Chinese Medicine combinations can produce synergistic effects and diminish possible toxic effects, thereby reflecting the advantages of compatibility. The proposed strategy can solve the quantitative problem of LRS and extend the scopes of PK studies.

Development of High Cordycepin-Producing Cordyceps militaris Strains

  • Cordyceps militaris, known as Dong-Chong-Xia-Cao, produces the most cordycepin among Cordyceps species and can be cultured artificially. For these reasons, C. militaris is widely used as herb or functional food in the East Asia. In this study, we developed a new strain of C. militaris that produces higher cordycepin content than parent strains through mating-based sexual reproduction. Twenty parent strains were collected and identified as C. militaris based on internal trasncrived spacer and rDNA sequences. Seven single spores of MAT 1-1 idiomorph and five single spores of MAT 1-2 idiomorph were isolated from 12 parent strains. When 35 combinations were mated on the brown rice medium with the isolated single spores, eight combinations formed a stroma with a normal perithecia and confirmed mated strains. High pressure liquid chromatography analysis showed that mated strain KSP8 produced the most cordycepin in all the media among all the tested strains. This result showed due to genetic recombination occurring during the sexual reproduction of C. militaris. The development of C. militaris strain with increased cordycepin content by this approach can help not only to generate new C. militaris strains, but also to contribute to the health food or medicine industry.

Effects of Gelling Agents on the Saikosaponin Content of Bupleurum falcatum Callus (배지응고제가 시호캘러스의 Saikosaponin함량에 미치는 영향)

  • In order to product the saikosaponin which is one of the secondary product from Bupleurum falcatum efficiently through the tissue culture, several levels of agar and gellan gum as the gelling agent, 2,4-D as the growth regulator, and L-phenylalanine as the precursor were used with single or combination treatment on MS basal medium. Gellan gum was more effective than agar as the gelling agent in fresh and dry weight increase of callus induced from Bupleurum falcatum leaf segment. Gellan gum medium supplemented with L-phenylalanine produced 1.6 times of fresh weight more than that of agar. The fresh weight was remarkably high in gellan gum when the calli was treated with the combination of 2,4-D and L-phenylalanine similar to the single treatment of 2,4-D or L-phenylalanine. However, the saikosaponin content in callus was high in gellan gum with the single treatment of L-phenylalanine. Especially, the saikosaponin content in gellan gum supplemented with 1.0mg/L L-phenylalanine was 2 times(2.4 mg/g) higher than that in agar medium supplemented with 0.1 mg/L 2,4-D(0.9 mg/g).

Inhibition of Nitric Oxide Production and Hyaluronidase Activities from the Combined Extracts of Platycodon grandiflorum, Astragalus membranaceus, and Schisandra chinensis (길경, 황기와 오미자 혼합추출물의 NO 억제활성과 Hyaluronidase 억제활성 효과)

  • In this study, the optimal extraction conditions for three medicinal herbs as functional sources against inflammatory and arthritic diseases were developed. Traditional medicinal herbs were screened for their inhibition of hyaluronidase (HAse) activity and nitric oxide (NO) synthesis. For the screening of anti-inflammatory properties, ethanolic extracts of 53 species of traditional medicinal herb were examined. We confirmed that Astragalus membranaceus (A.R.), Schisandra chinensis (S.F.), and Platycodon grandiflorum (P.G.) inhibit NO production. For extraction from all three herbs simultaneously, an ethanol concentration of 95%, a 1:2:1 mixture ratio, and at 50 rpm mixing speed, for over 12 h and at $30^{\circ}C$ was the best condition for optimal extract yield and NO inhibition effects. HAse inhibition from the three herb extraction was three fold higher than single samples. The ethanol extracts were fractionated with various solvents (n-hexane, chloroform, ethyl acetate, n-butanol, and water). The ethyl acetate-soluble fraction of the herb mixture showed the highest extract yield (13%) and NO inhibition effects (73%). In conclusion, this study provides experimental evidence that a mixture of P.G., A.R., and S.F. could be used as a source of antioxidant ingredients in the food industry.

Analysis of Experimental Study Tendency on Oriental Herbal Medicines for Male Infertility - Focusing on Domestic Theses for a Degree - (남성 불임에 관한 실험연구 동향 분석 - 학위논문 중심으로 -)

  • Objectives: The tendency of experimental studies on the male infertility was analyzed through focusing on domestic theses for a degree to indicate the hereafter direction for its study in oriental herbal medicine. Methods: 35 domestic theses for a degree published after 2003 were analyzed. And theses were classified by year, study design, degree and subjects. Results: The followings are the results of this study. 1. 35 theses were reviewed. 11 master dissertations and 24 doctorate dissertations 2. In the annual publishing tendency, the number of theses began to increase from 2003 and the theses have been published actively from 2005. 3. Classified by study design, 19 in vitro & in vivo and 16 in vitro experimental studies were performed. 4. In the medication and prescription, the studies with single medication were 32, and prescription were 3. In the studies with single medication, four studies on Ginseng Radix (人蔘) were the most. 5. In theses related to single medication, BoYang-Yak was most prefered, followed BoYeum-Yak, BoKi-Yak, SabJung-ChukNyo-JiDae-Yak. 6. Analysed 35 theses by subject, concentrational experiments were 23, periodical experiment were 3, concentrational & periodical experiment was 1. 7. In theses related to herb medication, the themes were Body & testicular weight, sperm parameters, testosterone level, CREM mRNA level & CREM protein, seminiferous tubule and antioxidant activity. 8. Related to herb medication's antioxidant activity efficacy were 7 theses. Conclusions: If there are more diverse studies on medication, prescription, external treatment methods and experiment methods in the future, this will be very helpful for the clinical treatment of male infertility.
