Purification and Characterization of Angiotensin I-Converting Enzyme Inhibitors from Sinapis alba L.

  • To separate ACE inhibitors from edible plants, spices, and herbs, 285 extracts of 95 sources were screened for ACE inhibitory activity. The extract of Sinapis alba L. had the most potent ACE inhibitory activity. Mustard seeds were crushed homogeneously and extracted with hexane and water successively. Lyophilized water extract was fractionated with $H_2O$:butanol(1:1). The ACE inhibitor was purified from butanol fraction by methanol precipi-tation, gel filtration, HPLC, and FPLC with Superdex peptide HQ 10/30 column. The active fraction has been purified to homogeneity, which was proven by gel filtration using FPLC system. The yield was 0.02%. The com-pound has a molecular weight of about 640. The compound competitively inhibited ACE activity and the $IC_{50}$ value was 79$\mu\textrm{g}$/ml. The purified compound showed uterus contraction activity in isolated rat uterus.

CD4+ T cells에서 백개자가 IFN-$\gamma$와 IL-4 생성에 미치는 영향 (Effect of Sinapis alba L. on expression of interferon-gamma and interleukin-4 production in anti-CD3/anti-CD28-stimulated CD4(+) T cells)

  • Objective : Sinapis alba L. (SA) is a korean traditional herbal medicine that is usually used to prevent or treat inflammatory diseases, such as respiratory infection and rheumatoid arthritis. However, the effects of SA supplementation in vitro on serum antibody levels, splenocyte and peritoneal macrophage immune responses have not yet been determined. In this study, we examined the effect of SA on the production of Th1/Th2 cytokines. Methods : Splenocytes were isolated from naive C57BL/6 mice. Cells were enriched for CD4+ cell populations by first staining the cells with anti-CD4 (BD PharMingen, Calif, USA). CD4+ T cells were selected on a (CS) column, and the flow-through was collected as CD4+ T cells. Isolated cells were activated by overnight incubation on 24-well plates coated with $1{\mu}g/mL$ anti-CD3, $1{\mu}g/mL$ anti-CD28 and with SA ($100{\mu}g/mL$). Primary macrophages were collected from the peritoneal cavities of mice (8-week-old female C57BL/6). The peritoneal macrophages were washed and plated with RPMI-1640 overnight for the experiments. After 48-hours cultures, samples were centrifuged at 2000 rpm for 10 minutes, and the supernatants were stored at $-80^{\circ}C$. Mouse IL-4, IFN-$\gamma$ and TNF-$\alpha$ were quantified using ELISA kits (BioSource International, Camarillo, Calif, USA) according to the manufacturer's protocols. Results : SA at 100ug/ml decreased the generation of Th1 cytokine (IFN-$\gamma$) by 0.5-fold. However, SA has no effect on Th2 (IL-4) production. Conclusions : These results suggest that SA may play an important role in the control of T-cell-mediated autoimmunity by down-regulation of Th1 cytokine (especially IFN-$\gamma$, TNF-$\alpha$). These data may contribute to the design of new immunomodulating treatments for a group of autoimmune diseases.

클로버씨스트선충에 대한 백겨자 식물자원의 저항성 스크리닝 (Resistance screening of white mustard(Sinapis alba) plant resources against clover cyst nematode, Heterodera trifolii)

  • 클로버시스트선충(Heterodera trifolii, CCN)은 국내에서 중요한 식물기생선충 중 하나로, 2017년부터 강원도 고랭지배추에 심각한 피해를 주고 있다. 훈증성 살선충제는 CCN 관리에 좋은 방제 방법이지만, 강원도 고랭지배추 재배지는 경사지가 많아 비닐 피복이 어렵기 때문에 훈증성 살선충제 처리가 어렵다. 이러한 이유로 CCN 관리를 위한 대체 소재의 개발이 필요한 상황이며, 살선충 피복작물과 저항성 품종이 대안 중 하나이다. 따라서, 우리는 선충방제용 녹비작물 또는 저항성 배추 품종 육성을 위한 육종소재로 활용하기 위해 씨앗은행의 백겨자(Sinapis alba) 82개 자원을 대상으로 CCN에 대한 저항성 검정을 수행하였다. 1차 시험에서는 15개의 백겨자 자원이 CCN에 저항성인 것으로 확인되었으며 나머지 자원은 CCN에 감수성이었다. 선발된 15개 자원 중 13개 자원을 대상으로 CCN 저항성의 재현성 검정을 위해 2차 시험을 수행하였다. 그 결과, 백겨자 5개 자원(IT297309, IT297312, IT302951, IT302953, IT302954)이 CCN에 저항성으로 나타났다. 최종 선발된 백겨자 5개 자원은 선충 방제용 녹비작물로 활용하거나 CCN에 저항성인 고랭지배추 품종 육성을 위한 유용한 유전자원으로 판단된다.

노인에게 적용한 이압요법이 야뇨와 불면증에 미치는 효과 (Effects of Auricular Acupressure Therapy on Nocturia and Insomnia in the Elderly)

  • Purpose: The purpose of this study was to confirm the effects of auricular acupressure therapy on insomnia and nocturia in the elderly. Methods: This study was a nonequivalent control group pre-post test design. Data collection was conducted from July 1 to August 31, 2013. Subjects were set for totally 36 people with 18 people in the experimental group and 18 people in the control group among the elderly over the age of 60 in silver town, welfare center, and senior citizen center where are located in Seoul area. For the experimental group, auricular acupressure therapy was applied two times during 5 days by fixing it to auricular reaction site with tape using Sinapis alba L. seeds. With SPSS/WIN 19.0 program, the collected data were analyzed. Results: Compared to the control group, the sleeping score and sleeping satisfaction were improved and nocturia frequency was lowered in the experimental group who applied auricular acupressure therapy. Conclusion: Auricular acupressure therapy was considered to be likely useful therapeutic method for the effects of relieving nocturia and insomnia in elderly.

Determination of the Effect of Different Ground Mustard Seeds on Quality Characteristics of Meatballs

  • This study investigated the effect of yellow, black, and brown mustard seeds on color, lipid oxidation (thiobarbituric acid-reactive substances [TBARS]), and microbiological and sensory qualities of meatballs during storage. Heat treatment of mustard seeds affected the TBARS value of meatball samples (p<0.0001). The addition of mustard seeds decreased TBARS value of meatball samples (p<0.0001). Heat treatment of mustard seeds decreased the $L^*$, $a^*$ and $b^*$ values of meatball samples (p<0.0001). The meatball samples with mustard seeds increased $b^*$ value of meatball samples however it decreased $a^*$ value of meatball samples (p<0.0001). The addition of mustard seeds decreased aerobic mesophilic bacteria count (p<0.0001), Enterobacteriaceae count (p<0.0001), psychrophilic bacteria count (p<0.0001) and yeast and mold count of meatball samples (p<0.0001). On a given storage day, the yellow mustard added meatballs sample was given higher color, appearance, flavor, acceptability ratings than those added black and brown mustard. Regarding sensory and microbiological properties, mustard seed contributed to microbiological quality and sensorial properties of meatball samples.

Thermal Inactivation of Myrosinase from White Mustard Seeds

  • Myrosinases (thioglucosidases) catalyze the hydrolysis of a class of compounds called glucosinolates, of which the aglycones show various biological functions. It is often necessary to minimize the loss of myrosinase activity during thermal processing of cruciferous vegetables. Myrosinase was isolated from a popular spice, white mustard (Sinapis alba), and its thermal inactivation kinetics was investigated. The enzyme was extracted from white mustard seeds and purified by a sequential processes of ammonium sulfate fractionation, Concanavalin A-Sepharose column chromatography, and gel permeation chromatography. At least three isozymes were revealed by Concanavalin A-Sepharose column chromatography. The purity of the major myrosinase was examined by native polyacrylamide gel electrophoresis and on-gel activity staining with methyl red. The molecular weight of the major enzyme was estimated to be 171 kDa. When the consecutive step model was used for the thermal inactivation of the major myrosinase, its inactivation energy was 44.388 kJ/mol for the early stage of destruction and 32.019 kJ/mol for the late stage of destruction. When the distinct two enzymes model was used, the inactivation energy was 77.772 kJ/mol for the labile enzyme and 95.145 kJ/mol for the stable enzyme. The thermal inactivation energies lie within energy range causing nutrient destruction on heating.
