  • An attractive target for anti-herpes chemotherapy is the herpes simplex virus 1 (HSV-1) protease encoded by the UL26 gene. HSV-1 protease is essential for DNA packaging and virus maturation. To perform high throughput for potent inhibitors, the efficient production of larger amounts of highly purified enzyme and protease activity assay method must be established. In this report, expression in E. coli and purification of the protease gene of HSV-1 strain F was investigated. The protease gene was cloned pET28, and the nucleotide sequence of protease catalytic domain of HSV-1 compared strain F with other strains (KOS and CL101). In these results the F strain was different in base sequence. However, the amino acid sequence was identifical. The HSV-1 protease was purified with His-tagged affinity column. The analysis of HSV-1 protease activity was performed by high performance liquid chromatography.

  • Since traditional linear feature extraction methods are unable to handle nonlinear characteristics often exhibited in hyperspectral imagery, nonlinear feature extraction, also known as manifold learning, is receiving increased attention in hyperspectral remote sensing society as well as other community. A most widely used manifold Isomap is generally promising good results in classification and spectral unmixing tasks, but significantly high computational overhead is problematic, especially for large scale remotely sensed data. A small subset of distinguishing points, referred to as landmarks, is proposed as a solution. This study proposes a new robust and controllable landmark selection method based on the maximum volume of the simplex spanned by landmarks. The experiments are conducted to compare classification accuracies with standard deviation according to sampling methods, the number of landmarks, and processing time. The proposed method could employ both classification accuracy and computational efficiency.

  • We investigated effects of several chemical carcinogens, i.e., $benzo({\alpha})pyrene$ (BP),7,12-dimethylbenz(a)anthracene (DMBA), nitrosomethyl urea (NMU), and nicotine on the replication, cytolyticity, DNA synthesis, and protein synthesis of type 1 herpes simplex virus (HSV-1) in viral infected Vero cell monolayers. We observed that the BP and DMBA did not show such activity. All chemical carcinogens did not inhibit the synthesis of viral DNA, but the expression of gamma viral proteins that are expressed from the newly synthesized progeny viral DNA was somewhat notably inhibited by BP and DMBA. However, the synthesis of alpha and beta viral proteins was not altered by the chemical carcinogens. These data indicate that the gamma viral proteins expressed from the newly synthesized DNA in the presence of chemical carcinogens in the culture medium may be defective. This is further supported by the fact that the virus fail to replicate in the presence of these chemical carcinogens, in spite of viral DNA and proteins are somewhat normally synthesized.

  • This paper presents a branch-and-bound algorithm for solving the concave minimization problem with upper bounded variables whose single constraint is linear. The algorithm uses simplex as partition element. Because the convex envelope which most tightly underestimates the concave function on the simplex is uniquely determined by solving the related linear equations. Every branching process generates two subsimplices one lower dimensional than the candidate simplex by adding 0 and upper bound constraints. Subsequently the feasible points are partitioned into two sets. During the bounding process, the linear programming problems defined over subsimplices are minimized to calculate the lower bound and to update the incumbent. Consequently the simplices which do certainly not contain the global minimum are excluded from consideration. The major advantage of the algorithm is that the subproblems are defined on the one less dimensinal space. It means that the amount of work required for the subproblem decreases whenever the branching occurs. Our approach can be applied to solving the concave minimization problems under knapsack type constraints.

  • The present study was performed to estimate the seroprevalence of larval Anisakis simplex infection among the residents health-examined in 3 hospitals in southern parts of Korea. A total of 498 serum samples (1 serum per person) were collected in 3 hospitals in Susan Metropolitan city, Masan city, and Geoje city in Gyeongsangnam-do (Province) and were examined by IgE-ELISA and IgE-western blotting with larval A. simplex crude extract and excretory-secretory products (ESP). The prevalence of antibody positivity was 5.0% and 6.6% with ELISA against crude extracts and ESP, respectively. It was also revealed that infection occurred throughout all age groups and higher in females than in males. A specific protein band of 130 kDa was detected from 10 patients with western blot analysis against crude extract and ESP among those who showed positive results by ELISA. Our study showed for the first time the seroprevalence of anisakiasis in Korea. The allergen of 130 kDa can be a candidate for serologic diagnosis of anisakiasis.

  • In order to explore the potential of ascorbic acid supplementation for the prevention and treatment of herpes simplex viral diseases, plaque reduction assays were performed. Ascorbic acid as well as copper chloride/ferric chloride were added to wells containing Vero cells infected with herpes simplex virus type 1 (HSV-1), and the infectivity of HSV-1 was determined. Since copper and iron are major transition metals in human plasma, near the normal human plasma concentrations of them were used for experiments. When Cu(II) and Fe(III) were applied, there were no significant differences between virus control and Cu(II)/Fe(III)-treated groups. But, when appropriate concentrations of ascorbic acid were added to wells, meaningful differences between control and ascorbate-treated groups were found. In the presence of Cu(II)/Fe(III) at $5.8/3.7\;{\mu}M$, 72-h treatment with ascorbate at $50\;{\mu}M$ reduced HSV-1 infections to $10.77%{\pm}4.25%$ (P < 0.001) and $500\;{\mu}M$ did to $3.06%{\pm}1.62%$ (P < 0.001). Moreover, the cytotoxicities for Vero cells at those concentrations were insignificant (P > 0.05). Current recommended dietary allowance (RDA) of ascorbic acid is 60 mg/day, and the oral intake of 60 mg/day of ascorbic acid yields plasma ascorbic acid at 45 to $58\;{\mu}M$ in a healthy adult man. Therefore, the results of this study suggest that the maintenance of appropriate level (more than $50\;{\mu}M$) of ascorbic acid in human plasma by appropriate amount (more than the RDA) of ascorbic acid supplementation may be helpful for the prevention and treatment of diseases caused by HSV -1 in an adult man. In addition, this study also suggests that ascorbic acid may be useful for the prophylaxis of fatal HSV-1 infections in neonates and the prevention of HSV-1 reactivation in immunocompromised hosts.

  • In a mixture experiment, one may be interested in estimating not only main effects but also some interactions. Main effects and interactions may be estimated through appropriate designs such as simplex-centroid designs. However, the estimability problems, implied by the sum to one functional relationship among the factors, have strong consequences on the confounding and identifiability of models for such designs. To handle these problems, we address homogeneous polynomial model based on the computational commutative algebra (CCA) instead of using $Scheff{\acute{e}}s$ canonical model which is typically used. The problem posed here is to give how to choose estimable main effects and also some low-degree interactions. The theory is tested using a fraction of simplex-centroid designs aided by a modern computational algebra package CoCoA.

  • As a component development of heavy duty industrial gas turbine combustor development program, pressure swirl simplex injector was designed and tested to figure out spray characteristics and performance. Injector flow rate as a function of pressure drop was measured and compared to the design target. Also spray shape was analyzed qualitatively and spray cone angle was measured from spray visualization image using shadowgraph. The flow test result showed that the injector was designed and manufactured correctly according to the design target and spray cone angle was measured from shadowgraph result. As a next step, PDA (Phase Doppler Anemometry) measurement is planned to figure out more specific spray performance and characterization.
