바늘구멍 골스캔에 의한 갑상선암 골전이 병소내 중격의 묘출 (Pinhole Bone Scintigraphic Demonstration of Septation in Metastatic Thyroid Carcinoma in Bone)

  • Metastatic thyroid carcinoma in bone shows characteristic expansile, trabeculated lysis on radio-gram. The ordinary scintigraphy manifests with a simple photopenic defect. We were able to portray trabeculation within photopenic defects. We present papillary thyroid carcinoma and follicular thyroid carcinoma, one each, pinhole bone scintigraphic findings of which are cold area with septated increased uptake at the iliae bone. These findings correspond very well with their X-ray pictures.

Improved recovery of active GST-fusion proteins from insoluble aggregates: solubilization and purification conditions using PKM2 and HtrA2 as model proteins

  • The glutathione S-transferase (GST) system is useful for increasing protein solubility and purifying soluble GST fusion proteins. However, purifying half of the GST fusion proteins is still difficult, because they are virtually insoluble under non-denaturing conditions. To optimize a simple and rapid purification condition for GST-pyruvate kinase muscle 2 (GST-PKM2) protein, we used 1% sarkosyl for lysis and a 1 : 200 ratio of sarkosyl to Triton X-100 (S-T) for purification. We purified the GST-PKM2 protein with a high yield, approximately 5 mg/L culture, which was 33 times higher than that prepared using a conventional method. Notably, the GST-high-temperature requirement A2 (GST-HtrA2) protein, used as a model protein for functional activity, fully maintained its proteolytic activity, even when purified under our S-T condition. This method may be useful to apply to other biologically important proteins that become highly insoluble in the prokaryotic expression system.

  • Objective: Ultrasound can be used in the treatment of large intracerebral hematoma. The authors present our experiences with Ultrasound-guided catheter placement for lysis and drainage of ganglionic hematoma, with emphasis on technical aspects. Methods: The authors applied real-time ultrasonography for the aspiration of intracerebral hematoma in 6cases. Ultrasound-guided aspiration via a burrhole was performed under local anesthesia. We selected a temporal entry point instead of the frequently used precoronal approach in ganglionic hematoma. A burrhole was made 4 to 6cm posterior from posterior border of frontal process of the zygomatic bone at the level of 4 to 5cm above the external auditory meatus. Results: In all patients, the catheter was placed accurately into the hematoma target. All patients were irrigated with urokinase once to three times a day. The catheter could be removed within two or three days. The mean hematoma volume was reduced from initially 32mL to 5mL in an average of two days. There were no intraoperative complications related to the use of real-time ultrasonography and no postoperative infections were noted. Conclusion: Ultrasound allows an easy and precise localization of the hematoma and the distance from the surface to the target can be calculated. Ultrasound-guided catheter placement for fibrinolysis and hematoma drainage is a simple and safe procedure.

  • Natural killer (NK) cells play a pivotal role in early surveillance against virus infection and cellular transformation, and are also implicated in the control of inflammatory response through their effector functions of direct lysis of target cells and cytokine secretion. NK cell activation toward target cell is determined by the net balance of signals transmitted from diverse activating and inhibitory receptors. A distinct feature of NK cell activation is that stimulation of resting NK cells with single activating receptor on its own cannot mount natural cytotoxicity. Instead, specific pairs of co-activation receptors are required to unleash NK cell activation via synergy- dependent mechanism. Because each co-activation receptor uses distinct signaling modules, NK cell synergy relies on the integration of such disparate signals. This explains why the study of the mechanism underlying NK cell synergy is important and necessary. Recent studies revealed that NK cell synergy depends on the integration of complementary signals converged at a critical checkpoint element but not on simple amplification of the individual signaling to overcome intrinsic activation threshold. This review focuses on the signaling events during NK cells activation and recent advances in the study of NK cell synergy.

Efficient Isolation of Intact RNA from the Soft Coral Scleronephthya gracillimum $(K\ddot{u}kenthal)$ for Gene Expression Analyses

  • A rapid, simple and efficient method to extract RNA from the adult polyps of a soft coral, marine cnidarian, Scleronephthya gracillimum $(K\ddot{u}kenthal)$; was developed in this study. The highest yield and purity of RNA was obtained with the lysis solution containing 35 mM EDTA, 0.7 M LiCl, 7.0% SDS, and 200 mM Tris-Cl (pH 9.0). Approximately $40{\mu}g$ of total RNA was extracted from 200 mg of liquid nitrogen-pulverized polyp tissue. The ratio of absorbance at 260 nm and 280 nm ranged from 1.8 to 2.0. The results of the reverse transcription polymerase chain reaction (RTPCR) with ${\beta}-actin$ gene specific primers and Northern blot analysis using the same gene probe revealed that the RNA extracted by our method had high quality, and was sufficient for subsequent molecular biological analyses. This method was effective for RNA extraction from other soft coral species which belong to the genus Dendronephthya.

DNA ‘Comet Assay’를 이용한 곡류의 방사선 조사 여부 확인 (Detection of Irradiated Grains Using the DNA ‘Comet Assay’)

  • 저선량으로 조사된 곡류의 방사선 조사 여부를 판별 하는데 DNA 'Comet Assay'의 활용이 가능한지를 검토하였다. 참깨, 들깨, 밀, 보리 및 쌀을 0, 0.1, 0.3, 0.5, 0.7, 1.0 kGy로 조사하여 시료로 사용하였다. 시료에 PBS 용액을 가하여 세포 현탁액을 조제하고 agarose gel과 혼합하여 microscope slide 상에서 lysis한 후 전기영동하여 염색한 다음 DNA 절편의 이동에 의해 형성된 ‘comet’의 tail length를 측정하여 분석하였다. 0.3 kGy 이상으로 조사한 모든 시료에서 tail length가 비조사한 시료와 차이를 나타내어 조사 여부를 확실히 판별할 수 있었다. 또한 비조사된 시료에서는 tail이 없거나 아주 짧은 tail을 가진 세포들이 관찰되었으며, 조사된 시료에서는 조사선량의 증가에 따라 DNA의 손상정도가 증가함으로서 tail length가 길어지는 것이 관찰되었다. 특히 참깨, 들깨 및 밀의 경우는 0.1 kGy의 낮은 조사선량에서도 현미경을 통하여 육안으로 비조사한 시료와의 구별이 가능하였다. 따라서 DNA 'Comet Assay'는 간단하고 신속하며 비교적 저렴하게 실시할 수 있는 곡류의 방사선 조사 여부를 판별하는 데 활용할 수 있음을 확인하였다.

감마선 조사된 감자, 마늘, 생강에서 조사선량과 저장기간에 따른 DNA fragmentation의 변화 (Changes of DNA Fragmentation by Irradiation Doses and Storage in Gamma-irradiated Potato, Garlic and Ginger)

  • The changes in DNA damage were investigated during storage after irradiation. Potato, garlic were irradiated at 0.05, 0.07, 0.1 and 0.15 kGy and stored for 3 months. Ginger was irradiated at 0.01, 0.02, 0.03, 0.04 and 0.05 kGy and stored for 1 month. The comet assay was applied to the sample immediately after irradiation and at the end of storage. Samples were isolated, grounded and the suspended cells were embedded in an agarose layer. After lysis of the cells, they were electrophoresed for 1 min. and then stained. DNA fragmentation in seeds caused by irradiation was quantified as tail length and tail moment (tail length ${\times}%$ DNA in tail) by comet image analyzing system. Right after irradiation, the differences in tail length between unirradiated and irradiated samples were significant(p<0.05) in potato, garlic and ginger. With increasing the irradiation doses, statistically significant longer extension of the DNA from the nucleus toward anode was observed. The results represented as tail moment showed similar tendency to those of tail length. Similarly in the stored samples, even 1 or 3 months after irradiation, all the irradiated samples significantly showed longer tail length than the unirradiated controls. These results indicate that the comet assay could be one of the simple methods of detecting irradiated samples. Moreover, the method could detect DNA damage even after 1 or 3 months after irradiation.

미소유체시스템을 위한 실용적인 패키징 기술 (Practical Packaging Technology for Microfluidic Systems)

  • 본 논문은 다기능 미소유체시스템의 일체형 패키징을 위한 MSI (microfluidic system interface) 기술을 제안하고, 이를 설계, 제작, 시험 평가하였다. MSI 기술을 통해 플러그 방식의 유체 인터커넥터, 유체제어를 위한 미소밸브, 광학 인터페이스를 위한 광학창을 유체시스템에 일체형으로 쉽게 구현할 수 있었다. MSI 기술의 유용성을 보이기 위해 미소 유전자시료전처리시스템에 적용되었으며, 미소 유전자시료전처리시스템은 세포정제, 세포분리, 세포용해, DNA 고체상추출, 중합효소연쇄반응, 그리고 모세관전기영동 기능으로 구성되었다. 나아가 MSI 기술이 적용된 미소 유전자시료전처리시스템의 DNA 고체상추출 및 중합효소연쇄반응의 실험결과로부터 MSI가 미소유체시스템을 위한 실용적 패키징 기술임이 검증되었다.

감마선 조사 육류, 가금류에서 저장전과 후의 조사선량에 따른 DNA fragmentation의 변화 (Changes of DNA fragmentation by Irradiation Doses and Storage in Gamma-irradiated Meats and Poultry)

  • The changes in DNA damage were investigated during storage after irradiation. Beef, pork and chicken were irradiated at 1.0, 3.0 and 5.0 kGy and stored for 6 months at $-20^{\circ}C$. The comet assay was applied to the sample muscles at the beginning of irradiation and at the end of storage. Muscles were isolated, sliced, and the suspended cells were embedded in an agarose layer. After lysis of the cells, they were electrophoresed for 2 min. and then stained. DNA fragmentation in tissues caused by irradiation was quantified as tail length and tail moment (tail length ${\times}$ % DNA in tail) by comet image analyzing system. Right after irradiation, the differences in tail length between unirradiated and irradiated muscles were significant(p<0.05) in beef, pork and chicken. With increasing the increasing doses, statistically significant longer extension of the DNA from the nucleus toward anode was observed. Similarly even 6 months after irradiation, all the irradiated muscles significantly showed longer tail length than the unirradiated controls. The results represented as tail moment showed similar tendency to those of tail length, but the latter parameter was more sensitive than the former. These results indicate that the comet assay could be one of the simple methods of detecting irradiated muscles. Moreover, this method suggest that using comet assay, we were able to detect DNA damage differences even after 6 months after irradiation.

낭종성 섬유성 이형성증 (Cystic Fibrous Dysplasia in the Kong Bone)

  • 목적: 장골에서는 드물게 보고되어 있는 낭종성 섬유성 이형성증(cystic fibrous dysplasia)의 임상적, 방사선학적, 병리학적 소견과 더불어 단순 소파술과 골이식술의 결과를 기술하고자 한다. 대상 및 방법: 1996년 1월부터 2003년 3월까지 가톨릭 의과대학 3개 부속병원에서 섬유성 이형성증으로 치료받은 98명의 환자 중 커다란 낭종성 변화를 동반한 11명을 대상으로 하였다. 전례에서 단순 골소파술, 동종골 이식술 및 내고정술을 시행하였다. 6명은 남자였고, 5명은 여자였으며, 환자의 나이는 20세에서 46세로, 평균 36.7세였다. 결과: 6명은 수개월 이상 통증을 호소했으며, 4명은 사소한 외상 후에 발생한 통증을 호소하였다. 1명에서는 병적 골절이 발생했다. 10명의 환자는 단골성(monostotic) 이었고, 1명의 환자는 다골성(polyostotic) 이었다. 병변의 발생부위는 7예에서 대퇴골, 3예에서 상완골 그리고 1예에서는 요골이었다. 단순방사선 사진 상 간유리 음영과 더불어 보여지는 현저한 용해성 병변을 보였고, 자기공명영상에서는 섬유성 이형성증을 시사하는 신호강도와 낭종을 의미하는 신호강도가 동시에 보였다. 조직학적으로는 전형적인 섬유성 이형성증 소견과 더불어 비 특이적인 낭종성 변성 변화를 보였다. 최종 추시 관찰시 국소재발 없이 모두 만족스러운 결과를 보였다. 결론: 장골의 낭종성 섬유성 이형성증은 지금까지 보고된 것처럼 드물지 않은 것으로 사료된다. 자기공명영상으로 양성 낭종성 변화와 악성 변화를 쉽게 구분할 수 있으나, 비 특이적 낭종성 변성변화와 동맥류성 골낭종을 구분하기 위해 조직검사가 필요하다. 낭종성 섬유성 이형성증은 수술적 치료의 대상이며, 단순 골소파술 및 동종골 이식술은 효과적인 치료로 사료된다.
