Determination of Silybin B in the Different Parts of Silybum marianum using HPLC-UV

  • Silymarin is the standardized extract from Silybum marianum which consists mainly of flavonoids and polyphenols. It is highly regarded for its hepatoprotective ability. Silybin B is a flavonolignan and one of the active components of silymarin. The content of silybin B in various parts of S. marianum was analyzed by HPLC-UV. Results show that the extract of seeds contain the highest amount of silybin B (7.434 mg/g DW). The petioles of S. marianum showed a low content of silybin B. This study revealed that seeds of S. marianum contain high amount of silybin B and could be a good source of the compound.

Silybum marianum seed extract supplementation positively affects the body weight of weaned piglets by improving voluntary feed intake

  • This study was conducted to evaluate the effects of dietary supplementation of Silybum marianum seed (SMS) extract on the growth performance, nutrient digestibility, fecal noxious gas emission, and hematology parameters in weaned piglets. A total of 120, 21-day-old weaned piglets ([Yorkshire × Landrace] × Duroc) were randomly assigned to 3 groups based on the average initial body weight (6.46 ± 0.45 kg). There were 8 replicate pens per treatment and 5 pigs (mixed sex) per pen. The experimental period was 42 days. Dietary groups included a basal diet, and a basal diet supplemented with 0.05% or 0.10% SMS extract. Feeding weaned piglets with SMS extract containing diet significantly increased average daily gain and average daily feed intake. Additionally, the supplementation of SMS extract had no significant effects on nutrient digestibility, serum hematology, and fecal noxious gas emission parameters. We considered that the supplementation of SMS extract had positive effects on the voluntary feed intake in weaned piglets, thus improving growth performance.

The Pharmacological Properties of Silymarin and Its Constituents

  • Silymarin is a standardized extract obtained from the seeds and fruits of Silybum marianum L., or commonly called milk thistle, a member of Carduus marianum family that contains mix of flavonolignans. Some epidemiological and preclinical studies revealed that S. marianum L. has been used for herbal remedies for centuries for its pharmacological activity. In this review, pharmacological studies in vitro and in vivo of silymarin are discussed thoroughly stressing on antioxidant, antimicrobial, antiviral, and anti-carcinogenic aspects of silymarin. In addition, the protective influences of silymarin on some organs such as heart, liver, bone, and neuron tissue are reviewed as well. This review would be useful for further study regarding the potential of natural plant, notably silymarin, and its therapeutic potential in the prevention and treatment of diseases.

흰무늬엉겅퀴 열매 추출물의 자외선에 대한 피부 보호 효과 (Photoprotective Effects of Silybum marianum Extract)

  • 자외선은 피부 광노화를 일으키며 홍반, 일광 화상 등 피부 광손상의 원인이 된다. 실리마린은 밀크 씨슬로도 알려져 있는 흰무늬엉겅퀴(Silybum marianum; S. m) 추출물에 존재하는 폴리페놀 화합물로 항산화 효과 및 자외선에 대한 피부 보호 효과가 알려져 있다. 본 연구에서는 피부의 표피층에서 실리마린을 함유하는 S. m 추출물의 광보호 효과를 이해하고자 하였다. 표피층을 구성하는 세포들이 자외선에 노출되면 DNA 손상이 발생하는데 S. m 추출물이 DNA repair를 촉진할 뿐만 아니라 UV filter로 작용하여 DNA 손상을 방지하는 것도 확인할 수 있었다. 특히, 홍반이 발생하지 않는 suberythemal dose (SED)의 자외선 노출에도 각질 산화와 DNA 손상이 일어날 수 있으며, S. m 추출물이 이런 미세 손상이 발생하는 것을 억제하는 것도 확인하였다. 또한 자외선에 의하여 각질이 산화되어 카르보닐화 단백질(protein carbonylation)이 증가하는 현상도 S. m 추출물을 도포한 경우는 억제됨을 확인하였다. S. m 추출물은 흡광의 성질이 있지만 광독성 유발 가능성은 낮은 것으로 평가되었다. 따라서 S. m 추출물은 광노화를 방지하고 피부를 보호하는데 활용 될 수 있을 것이다.

Phytochemical analysis of the antioxidant properties of Silybum marianum L

  • A growing body of evidence supports lipid peroxidation as having a role in the pathogenesis of liver disease. Although the probable cause of damage to human hepatocytes may be multifactorial, free radicals have been implicated in a variety of liver diseases, particularly in the presence of iron overload and toxic substances such as ethanol. Consequently, antioxidants, particularly those of plant origin such as flavonoids, may help to reduce the risk of developing these diseases. Silybum (S.) marianum, a medicinal plant widely used in traditional European medicine for the treatment of liver disorders, was evaluated for antioxidant activity. Thin layer chromatography and High Performance Liquid Chromatography analyses of crude extract of the plant confirmed the presence of a number of flavonoids reported in the literature. The antioxidant activity of these flavonoids was measured through inhibition of lipid peroxidation and 1, 1-diphenyl-2- picrylhydrazyl radical scavenging. The crude plant extract showed marked antioxidant activity in both assays. These results suggest that S. marianum contains flavonoids with antioxidant activity, capable of inhibiting or scavenging free radicals, thus supporting its traditional use as a hepatoprotective agent.

HPLC, NMR Based Characterization, Antioxidant and Anticancer Activities of Chemical Constituents from Therapeutically Active Fungal Endophytes

  • Fungi generate different metabolites some of which are intrinsically bioactive and could therefore serve as templates for drug development. In the current study, six endophytic fungi namely Aspergillus flavus, Aspergillus tubigenesis, Aspergillus oryzae, Penicillium oxalicum, Aspergillus niger, and Aspergillus brasiliensis were isolated and identified from the medicinal plant, Silybum marianum. These endophytic fungi were identified through intra transcribed sequence (ITS) gene sequencing. The bioactive potentials of fungal extracts were investigated using several bioassays such as antibacterial activity by well-diffusion, MIC, MBC, anti-biofilm, antioxidant, and haemolysis. The Pseudomonas aeruginosa PAO1 was used to determine the antibiofilm activity. The ethyl acetate extract of Aspergillus flavus showed strong to moderate efficacy against Staphylococcus aureus, Escherichia coli, P. aeruginosa, and Bacillus spizizenii. Aspergillus flavus and Aspergillus brasiliensis exhibited significant antibiofilm activity with IC50 at 4.02 and 3.63 mg/ml, while A. flavus exhibited maximum antioxidant activity of 50.8%. Based on HPLC, LC-MS, and NMR experiments kojic acid (1) and carbamic acid (methylene-4, 1-phenylene) bis-dimethyl ester (2) were identified from A. flavus. Kojic acid exhibited DPPH free radical scavenging activity with an IC50 value of 99.3 ㎍/ml and moderate activity against ovarian teratocarcinoma (CH1), colon carcinoma (SW480), and non-small cell lung cancer (A549) cell lines. These findings suggest that endophytic fungi are able to produce promising bioactive compounds which deserve further investigation.

생약 조성물의 알코올 분해 및 항염증 효과 (Effect of Herbal Composition on Alcohol Degradation and Anti-inflammatory Activity in Mice)

  • These studies were conducted to investigate the effects an herbal extract composition(MHE) containing $Hovenia$ $dulcis$ T., $Artemisa$ $capillaris$ T., $Pueraria$ $thunbergiana$ B., $Polygonatum$ $falcatum$ A., $Agastache$ $rugosa$ O., $Silybum$ $marianum$ L. and $Glycyrrhiza$ $uralensis$ F. in alcohol administered mice. Prophylactic administration of different doses of MHE(20~100 mg/kg) had beneficial actions toward alcohol degradation in acute alcohol treated mice. In addition, intraperitoneal administration of the MHE showed anti-inflammatory effects in inhibition tests of vascular permeability produced by acetic acid. MHE also reduced the concentrations of nitric oxide(NO) and tumor necrosis factor(TNF)-${\alpha}$ in macrophages that were activated by LPS. These results demonstrate that MHE possesses the potential to stimulate alcohol degradation and inhibit inflammatory effects in mice.

Peroxyl Radical Scavenging Capacity of the Flavonolignan Silybin, Ginkgo Biloba Extract EGb 761, American Green Tea and a Series of Germacranolides

  • We report on the applicability oj a method recently developed in our laboratory for measuring the antioxidant potential of isolated chemicals and extracts derived from natural products. Peroxyl radicals generated by thermal homolysis of 2,2'-azobis-amidinopropane (ABAP) oxidize $\alpha$-keto-${\gamma}$-methiolbutyric acid (KMBA) to ethylene, which is monitored by gas chromatography. Inhibition of ethylene formation in the presence of antioxidants that compete with KMBA for peroxyl radicals is the basis of the Total Oxyradical Scavenging Capacity Assay (TOSCA; Winston et al., 1998). Antioxidative activities of water-soluble extracts of American green tea, the anti-hepatotoxic flavonolignan from milk thistle (Silybum marianum) silybin, Ginkgo biloba extract EGb 761, and a series of naturally occuring sesquiterpene lactones (all ger-macranolides found in in fungi, liverworts, and plants) were studied. The specific TOSC value per $\mu$M silybin was 5.2, which is essentially comparable to that of Trolo $x^{ⓡ}$, a water-soluble vitamine E analog. Tea and Ginkgo extracts exhibited potent peroxyl radical scavenging capacity with values, respectively of =1700 and 1000 $\mu$mols Trolo $x^{ⓡ}$ equivalent per gram dry matter. The known anti-inflammatory activity of some germacranolides prompted study of their antioxidant capacity. None of the lactones exhibited antioxidant capacity toward peroxyl radicals comparable to Trolo $x^{ⓡ}$; costunilide, the most lipophilic, had a TOSC value = to glutathione. The potential role of peroxyl radicals in lipidperoxidation, other cellular damage, and var-ious disease states suggest a possible preventive role for silybin, green tea and Ginkgo biloba in oxidative stress caused by these free radical species.ecies.

엉겅퀴 추출물 실리마린의 피부 미백효과 (Hypopigmentary Effect of Milk Thistle Extract Silymarin)

  • 천연물로부터 새로운 미백 소재를 개발하기 위하여 식물 추출물들의 미백활성을 조사한 결과, 엉겅퀴의 열매로부터 추출한 silymarin이 우수한 미백효과를 나타내는 것을 발견하였다. Silymarin으로부터 유효 성분을 분리하기 위하여 각종 컬럼 크로마토그래피 및 HPLC 등의 기법을 실시하여 silybin과 isosilybin을 분리한 후 이성질체인 silybin A와B, 그리고 isosilybin A와 B를 각각 순수 분리하였다. Silymarin은 Mel-Ab melanocyte에서 세포독성에 영향을 주지 않는 동시에 멜라닌의 생성을 억제하였고 $IC_{50}$ 값은 28.2 ${\mu}g/mL$이었다. 또한 Silymarin은 cell-based tyrosinase의 활성을 저해하고, western blot 분석 결과 tyrosinase 단백질의 발현을 감소시키는 것을 확인하였다. Silymarin으로부터 분리한 활성 화합물인 silybin 및 isosilybin의 미백 효과를 측정한 결과, 각각 42.25 ${\mu}M$ 및 16.32 ${\mu}M$$IC_{50}$ 값을 가지며 멜라닌의 생성을 억제하였으며 tyrosinase 단백질의 발현을 감소시켰다. Diastereoisomer 형태로 존재하는 silybin A 및 B, 그리고 isosilybin A 및 B의 멜라닌 저해활성을 측정한 결과, 네 가지 화합물 모두 농도 의존적으로 멜라닌의 생성을 억제하는 것으로 나타났다. 또한, 피부 미백 임상연구를 실시한 결과, silymarin 2 % 함유 크림을 사용할 경우 피부 미백 효과가 유효하게 나타남을 확인하였다. 이상의 결과로부터 본 활성물질 silymarin은 피부 미백 효과가 우수한 안전한 화장품 원료로서 사용할 수 있을 것으로 사료된다.
