Synthesis of Silver Nanoplates with Fibronectin Nanofibril Template and Their SERS Applications

  • In this work, a novel strategy is provided to prepare silver nanoplates by a fibronectin (Fn) nanofibril template. First, Fn molecules were controlled to assemble into amyloid-like nanofibrils in highly concentrated ethanol aqueous solution. The resultant nanofibrils could serve as a soft template to direct the formation of silver nanoplates. It is worth noting that the silver nanoplates are excellent surface-enhanced Raman scattering (SERS) substrate with 4-aminothiophenol (4-ATP) molecule as a test probe. This high active SERS substrate can also be used to detect drug molecule, 2-thiouracil with high sensitivity.

  • Russell-Silver syndrome is a genetic disorder characterized by intrauterine and/or postnatal growth restriction and typical facies. The clinical feature is various due to heterogeneous genetic characters. Their common findings are short stature without catch-up growth, normal head size for age, a distinctive triangular face with prominent forehead and a pointed chin, low set ears and clinodactyly of the fifth fingers. Intraoral features of the syndrome are microdontia, delayed tooth eruption, hypodontia, and crowding. More than 400 case have been reported in the literature, and estimated incidence is from 1 in 3000 to 1 in 100,000. In this case we performed caries treatment under the general anesthesia for the patient with Russell-Silver syndrome. Dentist have to consider microstomia for the management of patients with Russell-Silver syndrome.

THE THERAPEUTIC EFFECT OF SILVER NANOCRYSTALLINE OINTMENT ON TMJ CAPSULITIS (은 나노 연고가 측두하악관절낭염의 환자의 치료에 미치는 영향에 관한 연구)

  • The silver nanocrystalline is widely used for biological field because of its biocompatibility and anti-microbial effect. The objective of this study was to evaluate the therapeutic efficacy of the silver nanocrystalline ointment on the temporomandibular joint (TMJ) capsulitis. Total 39 patients were included in this study and all patients were received single topical application of the silver nanocrystalline ointment (group A, n=30) or placebo ointment (group B, n=19). Measured variables were maximum mouth opening (MMO), visual analog scale (VAS) for pain, and VAS for function. In results, we could not assess any therapeutic efficacy of single application in the chronic TMJ capsulitis (p>0.05). However, the single application of silver nanocrystalline ointment showed significant improvement in MMO and VAS for pain compared to placebo effect in the acute TMJ capsulitis (p<0.05). We could not find any complications related to ointment application in both groups. In conclusion, the single application of silver nanocrystalline ointment was effective in improving patient's symptom in acute TMJ capsulitis without any noticing complications.

  • White rot, which is caused by Sclerotium cepivorum, is a lethal disease affecting green onions. Three different types of nanosilver liquid (WA-CV-WA13B, WA-AT-WB13R, and WA-PR-WB13R) were tested in several different concentrations on three types of media to assess their antifungal activities. Results from in vitro experiments showed that all three of the nano-silver liquids had more than 90% inhibition rates at a concentration of 7 ppm. Greenhouse experiments revealed that all of the nano-silver liquids increased biomass and dry weights, and there were minimal changes in the population of various bacteria and fungi from the soil of greenhouse-cultivated green onions. In addition, a soil chemical analysis showed that there were minimal changes in soil composition.

  • In this study, the experiments for surface enhancement of silver mirror substrates were done, where we checked the characteristics of silver surface made by Tollen's method. The surface enhancement of Methyl orange was analyzed by silver surfaces. We observed the Surface Enhanced Raman Spectra of Methyl orange. The assignments of the vibrational bands shown in SERS spectra are given based on both literature and the semi-empirical calculations at the PM3 methods. We deduced that the adsorption orientation of Methyl orange was little tilted parallel to the silver surfaces by using of the surface selection rules.

  • 2-Mercaptothiazoline is very sensitive reagent for silver, which have been used for highly selective amperometric determination of silver. A method is described for the determination of micro-quantity of silver by amperometric titration with 2-mercaptothiazoline in ammonical solution. Direct titration of milligram amount of silver (0.05∼1.00mg) is possible in the presence of a number of foreign ions in ammonical solution containing ethylenediaminetetraacetic acid as masking agent under atmosphere. The interfering elements are gold and platinum. The milligram amounts of silver can be determined by the proposed method within an error ${\pm}$5%.

Effects of Nanometer Sized Silver Materials on Biological Toxicity During Zebrafish Embryogenesis

  • Commercial nanometer sized silver is widely used for its antibacterial effect; however, nanoparticles may also have ecotoxicological effects after being discharged into water. Nanometer sized silver can flow into aquatic environments, where it can exert a variety of physiologically effects in living organisms, including fish. The present study aimed to investigate the effect of nanometer sized silver on the development of zebrafish embryos, analyze the properties of commercial nanometer sized silver and define the toxicity relationship between embryogenesis and hatched flies. The commercial nanometer sized silver was analyzed in the $Ag^+$ ion form. The hatch rate decreased in the nano-silver exposed groups (10 and 20 ppt); furthermore, the hatched flies had an abnormal notochord, weak heart beat, damaged eyes and curved tail. The expression of the Sel N1 gene decreased in the nano-silver exposed groups, and the catalase activities of the exposed groups increased relative to those in the control group. Therefore, the $Ag^+$ ions in commercial nanometer sized silver could accumulate in aquatic environments and seriously damage the development of zebrafish embryos.

  • Silver nanoparticles has been prepared by the $\gamma$-irradiation and in situ reduction methods. Based on the Raman spectra, TEM images, X-ray Diffraction (XRD) patterns and UV-vis spectra, the in situ reduction method is more stable and the average size of the silver nanoparticles is also smaller than by the $\gamma$-irradiation reduction method. It is identified that the silver ions interacting with nonbonding electrons of oxygen atom in the carbonyl group of polyvinylpyrrolidone (PVP) by the in situ reduction method. It is also found advantages of the in situ reduction method including no additional reducing agents, without $\gamma$-irradiations treatment and the room temperature treatment suitability.

  • The translation elongation factor 1A, eEF1A, catalyzes the binding of aminoacyl-tRNA to the A-site of the ribosome by a GTP-dependent mechanism. By subtractive suppression hybridization technique, we have isolated a soybean low-temperature inducible gene, SLTI100 encoding translation elongation factor 1A. Multiple sequence alignments and phylogenic analysis showed that SLTI100 and other eEF1As originated from diverse organisms are highly conserved. RNA expression of SLTI100 was specifically induced by low temperature, high salt, ABA, or drought stress. Based on the subcellular localization of the corresponding gene product fused to GFP, we were able to confirm that SLTI100-GFP was restricted to the nucleus and cytoplasm. We propose that soybean eEF1A may play an important role in translational regulation during abiotic stress responses in plants.
