The Parasite of Microsporidia in Insects (야외곤충에 기생하는 미포자충류에 대하여)

  • Silkworms have been found cross infected with other microsporidia of insects in mulberry trees, forest and fruit gardens. Even the unidentified microsporidian species were not seriously pathogenic to silkworms, the silkworm egg producers lose their profit because of the elimination of eggs laid from moths which are infected with any kind of microsporidian species. Recently, the microsporidian cross infection to silkworm is in tendency of increase and the authors have investigated the field insects to examine the microsporidia. The number of species of insects infected with microsporidia was 10 and they were Boettcherisca peregrina (Robineau-Dewvoidy), Apis melifera linnaeus, Artogenia rapae Linnaeus, Tipula aino Alexander, Altica cacrulescens (Baly), Anomela daimiana Harold, Eilema griseola (Jubner), Rbalbistylun speciosum Uller, Anisodactylus signatus illiger, Oulema oryzae (Kuwayama). From the Boettcherisca peregrina (Rogineau-Desvoidy), three different species of micrsporidia were isolated and the microsporidia isolated from Boettcherisca peregrina (Robineau-Desvoidy), Apis melifera Linnaeus, Artogenia rapae Linnaeus donot have infectivity to silkworm larvae, Bombyx mori L.

Effect of Potassium Nitrate Supplementation on Economic Parameters of the Silkworm Bombyx mori L.

  • The effect of supplementation with potassium nitrate on economic parameters was analysed following treatment of last larval stadium. The treated larvae showed a significant increase in larval weight at 500$\mu\textrm{g}$/ml and silk gland weight in all the treated groups along with other enhanced larval, cocoon and adult parameters. The larval period was significantly decreased in all the treated groups with increased female and male cocoon weights its shell weights filament lengths at 500$\mu\textrm{g}$/ml. The denier was significantly decreased in all the groups. The filament weight moth emergence percentaged length of the ovariole were unaffected. However, eggs per ovariole, fecundity and hatching percentage significantly decreased compared with the corresponding parameters of the carrier control.

Effect of Hydrocortisone on the Economic Parameters of the Domestic Silkworm, Bombyx mori L.

  • The effect of topical application with hydrocortisone on economic parameters was analysed following treatment of last larval stadium. The treated larvae showed a significant increase in larval weight at higher concentrations along with other enhanced larval, cocoon and adult parameters. This suggests that hydrocortisone can be used effectively for commercial silkworm rearing. The larvas period was significantly increased in all the treated groups with increased female cocoon weights, its shell weights and male cocoon shell weight at 20$\mu\textrm{g}$/ml treated group. Filament length, weight and denier were increased significantly in all the treated groups. Moth emergence percentage, length of the ovariole, eggs per ovariole, fecundity and rind hatching percentage were unaffected when compared with that of the carrier control group. This suggests that hydrocortisone in addition to affecting larval growths also affect silk crop.

Production of the Eggs with Abnormal Shape from the Domestic Silkworm, Bombyx mori, Infected with Autographa californica Nuclear Polyhedrosis Virus

  • The female pupae of the silkworms Bombyx mori, were injected with recombinant Autographa californica nuclear polyhedrosis virus (AcNPV) expressing green fluorescent protein (GFP) by percutaneous inoculation. When the 4 day-old female pupae were injected with 1x10$^{7}$ or 2${\times}$10$^{7}$ plaque forming units (pfu) of the recombinant AcNPV, oviposited number and egg weight were significantly decreased. Furthermore, the shape of the eggs was obviously divides into normal and abnormal shapes. The percentage of the eggs with an abnormal shape was 7.8% and 57.1% at 1${\times}$10$^{7}$ and 2${\times}$10$^{7}$ pfu inoculation, respectively. PCR analysis of the genomic DNA extracted from the eggs revealed that gfp and AcNPV ecdysteroid UDP-glucosyltransferase genes were amplified from both types of eggs with normal and abnormal shapes. The results demonstrate that AcNPV DNA, and gfp gene cloned into the AcNPV genome, injected in pupal stage were transmitted to eggs and remained stable through at least next generation.

Impact of Fungus on Egg Shell of Tropical Tasar Silk Worm, Antheraea mylitta: An Ultra-structural Approach

  • The egg shell of the tropical tasar silkworm, Antheraea mylitta is formed from the substances secreted by the follicular epithelium during the late vitellogenic stage. TEM study reveals the inner travecular and outer lamellar layer of chorion. The travecular layer is composed of the innermost wax layer, inner and outer chorionic layer. The inner and outer chorionic layers are connected to each other by vertical pillers forming of cavities. The lamellar layer is perforated by the aeropyles. SEM study reveals the differentiation of an anterior surface of the egg shell into four zones-micropylar, edge, aeropyles crown and disc zone. In the mycosis infected eggs the aeropyles and egg-shell surface are fully packed with the hyphae of the fungus, Aspergillus sydowi blocking of plastron respiration and causing the death of developing embryo so that mycosis infected eggs become sterile.

Relationship between Egg Size and Quantitative Characteristics in Silkworm, Bombyx mori (교잡원종에 있어서 잠란의 크기가 계량형질에 미치는 영향)

  • This study was investigated the relationship between egg size and manifestation of quantitative characteristics such as larval growth, cocoon weight and cocoon shell weight etc. There was a high positive correlation between egg size and body weight of newly hatched larva. Until 2nd instar, larval growth was affected by egg size. The body weight of grown larva from big eggs were heavier on all strains except for strain El6 and the larval duration from big egg were shorter on the Japanese and the Chinese skins. These facts show that the characteristics of larval body weight and larval duration were affected more by the strain than by the egg size. The cocoon characteristics of big eggs were better in comparison than those of regular eggs.

Study on the effect of different temperatures on the main productive characters of the silkworm larvae, Bombyx mori under Egyptian agro-climatic conditions (에집트 기후조건 특히 온도의 변화가 원원잠품종의 주요 유전형질에 미치는 영향)

  • Eleven inbred lines of silkworm, Bombyx mori L. were exposed to two main different temperature (23C and 25C) during the larval period in order to elucidate the effect of the temperature differences on their main characters under the, prevailing in Egyptial agro-climactic conditions. The results show that the temperature differences did not affect significantly the pupation ratio and cocoon shell ratio. However, they affected their larval duration to become shorter with the high temperature treatment ($27^{\circ}C$) and their cocoon shell weight and cocoon yield per box of silkworm eggs to be higher in the lower temperature($23^{\circ}C$). It was recommended that some inbred lines should be bred at the 23C throughout the whole larval period, particularly in being reared with high nutritional leaves in order to maximize the hybridization of cocoon yields.

Production of the BmCecB1 antimicrobial peptide in transgenic silkworm

  • This peptide has antibacterial activity against several Gram-positive and Gram-negative bacteria. Bombyx mori cecropinB1(BmCecB1) is antimicrobial peptides from Bombyx mori and belongs to cecropin family. Antimicrobial peptides are important components of the innate immune systems in all living organism. To produce the BmCecB1 antimicrobial peptide, we constructed transgenic silkworm that expressed BmCecB1 gene under the control BmA3 promoter using piggyBac vector. The use of the 3xP3-driven EGFP cDNA as a marker allowed us to rapidly distinguish transgenic silkworm. Mixtures of the donor vector and helper vector were micro-injected into 600 eggs of bivoltin silkworms, Baegokjam. In total, 49 larvae (G0) were hatched and allowed to develop into moths. The resulting G1 generation consisted of 22 broods, and we selected 2 broods containing at least 1 EGFP-positive embryo. The rate of successful transgenesis for the G1 broods was 9%. We identified 9 EGFP-positive G1 moths and these were backcrossed with wild-type moths. With the aim of identifying a BmCecB1 as antimicrobial peptide, we investigated the Radical diffusion Assay (RDA) and then demonstrated that BmCecB1 possesses high antibacterial activities against Gram-negative bacteria.
