Identification of Highly Transcribed Genes in Japanese Oak Silkworm, Antheraea yamamai, Using PCR-Based cDNA Library

  • Determined sequences of 384 randomly selected clones in a PCR-based cDNA library of Antheraea yamamai could identify expressed sequence tags (ESTs) of highly expressed gene. One EST (fibroin) appeared 15 times, one EST (40S ribosomal protein S18) twelve times, one EST (ribosomal protein S24a) eleven times, ten times (ribosomal protein S8), nine times (60S ribosomal protein L10A), seven times (60S ribosomal protein S15A, S17, S17 and seroin), six times (ribosomal protein S8), five times (ribosomal protein S24, mariner transposase and P8 protein), four times (serpin 2), three times (heat shock protein 70 and poly A binding protein), and the remaining 6 ESTs twice (amylase, KIAA1006, elongation factor-1, transposon mag, translation initiation factor 4C, QM protein, transposase). Therefore, the 94 EST make it possible to identify 24 redundant clones that are candidates for highly expressed genes in posterior silk gland of this insect. The 24 redundant EST clones were identified in GenBank, but none of them was related to A. yamamai, suggesting that there are many unidentified genes which are highly expressed in the A. yamamai genome.

Changes in Properties of Silk Monofilament Caused by Drawing and Hydrolysis (견 Monofilament의 연신과 가수분해에 의한 특성변화)

  • The middle silk gland, that is a liquid silk thread gland consisting of silk protein, was taken out and a silk monofilament was made by drawing rapidly to approximately 3 times. In order to deteriorate the inter molecular hydrogen bonding force and to stretch in, the drawn silk filament was swoolen in boiling water. The results obtained are as follows ; The silk gland sample that just dried silk gland was occupied in crystalline region of silk-I type and random amorphous region. According to the examination of X-ray diffraction and thermal analysis, silk-II type crystal begins to appear partially in monofilament sample and spread to almost complet silk-II type crystal in 65.2% drawn sample. And, orientation of silk fibroin mlecule increased suddenly in early stage with a rise of drawing ratiofrom birefringence and density, and it was found that orientation of fibroin molecule was completed. As drawing ratio increases relation with time of hydrolysis, birefringence appeared almost fixed a tendency. Crystallization collapse by hydrolysis was not found in X-ray diffraction and thermal analysis. But, amorphous region began to flow by treated hydrolysis, that orientation of crystallization part was disturbed was supposed.

Effect of Silk Fibroin Hydrolysate on the Apoptosis of MCF-7 human Breast Cancer Cells

  • Breast cancer is one of the most common cancers among women worldwide. Recently anticancer agents have been developed using natural substances. To evaluate the anticancer effect of hydrolysates of silk fibroin (HSF), we investigated the effect of HSF on cell viability and apoptosis of a breast cancer cell line, MCF-7, induced through the mitochondrial pathway. The result showed that HSF decreased cell viability in MCF-7 cells in a dose- and time-dependent manner, resulting in an increase in the sub-G1 phase cell population. HSF increased the level of the pro-apoptotic Bax protein and decreased the levels of the anti-apoptotic Bcl-2 protein. In addition, HSF induced apoptosis in MCF-7 cells through a mitochondria-dependent pathway by increasing levels of cytochtome c, and cleavage of PARP. Taken together, these findings suggest that HSF inhibits the proliferation of MCF-7 breast cancer cells through a mitochondria and caspase dependent apoptotic pathway.

Effect of Opuntia Ficus-indicts variety Saboten Makino and Satsuma Mandarin Dyeing on Physical Characteristics and Colors of Protein Fabrics (선인전 및 감귤염색에 의한 단백질 직물의 물성 및 색상에 관한 연구)

  • The physical characteristics of silk and wool fabrics dyed with OFS-FR and SM-FR were examined and color difference and colorfastness of these fabrics were analyzed. The results are as follows; 1. The thickness, density and weight of both dyed fabrics were increased than those of undyed fabrics independently of dyeing materials and mordants. 2. Air permeability of all the silk samples increased than that of undyed silk, on the other hand, air permeability of most of wool samples decreased than that of undyed wool. 3. Thermal insulation values of silk samples were not consistent with dyeing conditions, but those of wool samples increased than those of undyed fabrics regardless of dyeing materials and mordants. 4. Color difference was clear between OFS-FR and SM-FR dyed fabrics. The colors dyed with OFS-FR-Al and OFS-FR-chitos an showed violet, those dyed with SM-FR displayed yellowish, in addition to in case of wool, dyed with OFS-FR-Cu the color was greenish yellow, in case of silk dyed with SM-FR-Cu that was yellowish red. 5. Sense of silk fabrics by sensory-test was soft and smooth. Feeling of colors in silk fabrics was wick, warm and slightly brilliant. In case wool, sense of dyed fabrics was slightly soft and slightly rough. Feeling of colors in wool fabrics was warm but not elegant. Therefore the result of sensory-test in silk might be better than that of in wool. 6. Colorfastness to dry cleaning and perspirations was good, but that to washing and sunlight was not fine.

Effect of ethanol concentration on the infrared spectroscopic characteristics of silk beads (알코올의 농도에 따른 실크 비드의 적외선 분광 특성)

  • The structural transition of silk protein has been induced by various method including alcohol treatment. To know the effect of alcohol concentration on silk beads conformation, silk beads were prepared in different alcohol concentration (100%, 70%, and 50%) and then examined the infrared spectra of silk beads. Silk beads prepared in 100% alcohol showed at $1265cm^{-1}$ attributed ${\beta}-sheet$ conformation and did not showed a characteristic absorption peak from model drug. However, silk beads in 70% and 50% alcohol showed some peaks originated from model drug including $2933cm^{-1}$, $1069cm^{-1}$, and $973cm^{-1}$. These results means that the micro-environment of silk beads was affected by alcohol concentration.

  • Enzymatic hydrolysis of different forms of silk fibroin (soluble, gel and insoluble forms) by industrial and commercial enzyme preparations to obtain aqueous and powdered silk fibroin in relatively mild conditions was investigated. A mono-enzymatic hydrolysate systems were tested for hydrolysis of water-soluble form of fibroin as most productive form of protein substrate. Insoluble forms of substrate usually were hydrolyzed less effective. In some cases from soluble fibroin substrate gel was formed during hydrolysis process. This hindered intermixing and decreased rates of hydrolysis. Insoluble sediments were formed in enzymatic hydrolysates in other cases. These sediments and also sediment after chemical hydrolysis were purified and tested on amino acids content for comparison. Sediments formation in these conditions are considered as pure tyrosine isolation method. Obtained hydrolysates were characterized by gel-chromatography analysis and other standard biochemical methods. Possibility of application of enzymatic hydrolysis for preparation of silk fibroin hydrolysates is discussed.

  • Using gel filtration chromatography, high molecular silk fibroin with high purity was obtained and silk flbroin microsphere particles (SFMP) could be simply made by spray dryer method. Also, some of the physicochemical properties of SFMP and morphology were investigated. The average molecular weight of pure silk fibroin protein dissolved in calcium chloride is about 61,500g/㏖ as measured by gel permeation chromatography. SFMP was spherical in shape, and particles, sized average of 2 ${\pm}$ 10 ${\mu}$, were observed by SEM and particle analyzer, respectively. Obtaining microspheres particles by spray dryer method accelerated the transition from the random coil to the $\beta$-sheet structure during spray dryer treatment. It was identified by the basic fourier transform infrared spectroscopy of SFMP. The swelling ratio of SFMP is majorly dependent on the pH of the solution, not on the occurred gelation. The characteristic structure, which might be applicable to immobilization of drugs is superior to other matrix materials for the use of biomaterials with skin affinity.

Anti-adipogenic Effect of Hydrolysate Silk Fibroin in 3T3-L1 Cells

  • Hydrolysate silk fibroin (HSF) is a fibrous protein composed of parallel $\beta$-structures and is made from pure silk elements including 18 amino acids, with glycine, alanine, and serine comprising of over 80% of the amino acids. Numerous studies have documented a range of effects of HSF, including moisturizing, antioxidant activity, nervous system disorders, and many more. We investigated whether HSF has anti-obesity effects in vitro. The effects of HSF inhibition on lipid accumulation and acceleration of lipid degradation in 3T3-L1 cells were studied. Treatment of 3T3-L1 cells with HSF caused significant inhibition of cell viability, an increase in glycerol release, and a decreased in adipocyte differentiation. Moreover HSF stimulated downregulated of adipogenic enzyme expressions (PPAR${\gamma}$ and C/EBP${\alpha}$) and up-regulated of fatty oxidation enzyme expressions (CPT-1 and UCP-2). Based on these results, hydrolysate silk fibroin can be suggested as a potential therapeutic substance as part of a prevention or treatment strategy for obesity.

Antioxidation and anti-inflammatory effects of gamma-irradiated silk sericin and fibroin in H2O2-induced HaCaT Cell

  • Oxidative stress in skin cells can induce the formation of reactive oxygen species (ROS), which are critical for pathogenic processes such as immunosuppression, inflammation, and skin aging. In this study, we confirmed improvements from gamma-irradiated silk sericin (I-sericin) and gamma-irradiated silk fibroin (I-fibroin) to skin cells damaged by oxidative stress. We found that I-sericin and I-fibroin effectively attenuated oxidative stress-induced ROS generation and decreased oxidative stress-induced inflammatory factors COX-2, iNOS, tumor necrosis factor-α, and interleukin-1β compared to the use of non-irradiated sericin or fibroin. I-sericin and Ifibroin effects were balanced by competition with skin regenerative protein factors reacting to oxidative stress. Taken together, our results indicated that, compared to non-irradiated sericin or fibroin, I-sericin, and I-fibroin had anti-oxidation and antiinflammation activity and protective effects against skin cell damage from oxidative stress. Therefore, gamma-irradiation may be useful in the development of cosmetics to maintain skin health.

Dyeability and Functionality of Bamboo Extracts (Part II) -Dyeing Properties of Protein Fiber- (대나무 추출물의 염색성과 기능성 (제2보) -단백질섬유에 대한 염색성-)

  • Dyeing properties of protein fiber with bamboo colorants were studied by investigating the effect of dyeing conditions such as dye concentration, dyeing temperatures, times on dye uptakes, effect of mordants, and color change. The various colorfastness of dyed fabrics were evaluated for practical use. In addition, the antimicrobial ability, ultraviolet-cut ability, and deodorant ability were estimated. The dye uptake increased as the dyeing concentration increased. Bamboo colorants showed relatively good affinity to protein fiber and produced a yellow color. Dye uptake increased as the dyeing time and temperature increased. Post-mordanting was more effective than pre-mordanting. Mordants, Fe and N.Fe, were effective for increasing dye uptake. The color of fabric mordanted with Cu and N.Cu changed to GY. Colorfastness of dyed fabrics showed a relatively good rating, and mordanting had no significant effect on colorfastness. Dyed silk fabric showed very good antimicrobial abilities of 99.9%. Also, ultraviolet-cut ability and deodorant ability were improved in silk fabric dyed with bamboo extracts.
