Structural and Physical Properties of Antheraea pernyi Silk Fibroin Fiber Treated with $I_2-KI$ Aqueous Solution

  • Silk fibroin (SF) fiber from the Antheraea pernyi silkworm was treated with a 1.23 N iodine-potassium iodide ($I_2-KI$) aqueous solution, and the structure and physical properties were investigated to clarify the effects of the iodine treatment. The noticeably high weight gain value of SF fiber, about 25 wt% was attributed to the absorption of polyiodide ions in the form of $I_3{^-}\;and\;I_5{^-}$. Fourier transform infrared spectroscopy and X-ray diffraction measurements suggested that polyiodide ions mainly entered the amorphous region. In addition, a new sharp reflection on the meridional direction, corresponding to a period of $7.0{\AA}$, was observed and indicated the possibility of the formation of mesophase structure of ${\beta}$-conformation chains. Dynamic viscoelastic measurements showed that the damping tan ${\delta}$ peak at $270^{\circ}C$ gradually shifted to lower temperature in the iodinated SF fibers, suggesting an enhancement of the molecular motion of the fibroin chains induced by the presence of polyiodide ions. With heating above $254^{\circ}C$, the iodine component introduced intermolecular cross-linking of SF, and the melt flow of the sample was inhibited. The thermal decomposition stability of fibroin molecules was greatly enhanced by iodine treatment.

Gelation of silk fibroin solution via β-sheet formation promoted by riboflavin-mediated photo-crosslinking

  • Di-tyrosine photo-crosslinking of silk fibroin (SF) is recently highlighted as a biocompatible hydrogel fabrication process, because this method does not need potentially harmful chemical species. However, the resulting crosslinking density is often insufficient to obtain a mechanically stiff hydrogel unless additional oxygen is provided during the reaction. In this study, we proposed a combinational crosslinking method to form an SF hydrogel via the di-tyrosine photo-crosslinking with riboflavin (photoinitiator) and physical interaction of SF chains. In the UV light-irradiated SF solution, small particles formed and these particles promoted β-sheet formation of SF molecules, resulting in quick gelation. The di-tyrosine photo-crosslinking produced nuclei that might trigger regular assembly of SF molecules in high temperature condition. Conclusively, this process would contribute to a development of biocompatible hydrogel fabrication for biomedical uses of SF hydrogels.

  • 작잠은 가잠과는 종속이 다른 섬유 곤충으로 우수한 섬유용 재료를 만든다. 최근 비섬유용 소재로 작잠 견단백질을 활용하기 위한 기초 연구의 일환으로 작잠 견단백질을 용해 조건에 대하여 연구 결과 작잠견사의 연감율은 16.8%로 나타났으며, 작잠견사는 염화칼슘 에탄올 혼합 용매에는 용해되지 않으나 질산칼슘수화물의 용융액에는 용해되었다. 질산칼슘과 에탄올의 조성비를 조정하여 낮은 농도의 질산칼슘 농도에서도 작잠견사가 용해되었다. 작잠 실크단백질의 주요 아미노산 조성은 알라닌, 글리신, 세리신으로 작잠 정련사와 유사한 조성을 보였다. 따라서, 금후 작잠 견단백질의 적절한 용해시스템 확보 및 소재개발을 위한 기초 자료로 활용될 수 있을 것으로 사료된다.

  • 염화아연에 의한 작잠 견사의 용해 조건을 살펴본 후 투석법에 의하여 얻은 작잠 견피브로인 분말의 형태와 적외선 분광 특성 및 X-선 회절 특성을 살펴본 결과 다음과 같은 결론을 얻었다. 1. 작잠 견사는 고농도의 염화아연 수용액에 용해되었으나 용해 조건을 조절하여 완전하게 작잠 견사를 용해할 수 있었다. 2. 재생 작잠 분말의 구조 특성을 살펴보기 위하여 적외선 분광분석과 X-선 회절분석을 행한 결과 작잠분말은 $\beta$-sheet 구조와 $\alpha$-helix 구조로 이루어져 있었다. 3. 염화아연을 사용하여 제조한 작잠 견피브로인 용액으로부터 순수한 작잠 견피브로인을 얻기 위한 새로운 방법이 개발되어야 함을 알 수 있었다.

견 피브로인의 Rat 콜라겐 성장 효과 (Collagen Growth Effects in Rats using B. mori Fibroin)

  • The collagen growth effects in rats using B. mori fibroin sponge sheet, pure fibroin solution and chitosan sponge sheet were investigated. Histopathological inspection of the wound 12 days later showed the increse of a vascular ingrowth and the absence of inflammatory cells. Wound healing effects were accelerated in the order of Fibroin sponge sheet$\geq$Fibroin dressing solution〉Chitosan sponge sheet〉Control. The mean percentage of collagen production in rats after 12 days was 46.12$\pm$4.58% and 42.11$\pm$5.67% in fibroin sponge sheet and dressing solution-treated rats, respectively. These values were higher than the relative content in the chitosan sponge sheet and control groups, which was 23.87$\pm$3.24 and 2.42$\pm$0.32%, respectively.

The Effect of Sample Handling on the Rheological Measurement of Regenerated Silk Fibroin Formic Acid Solution using Parallel Plate Geometry

  • The effect of sample handling condition on the rheological measurement of regenerated silk fibroin formic acid solution using parallel plate geometry was investigated. In case of loading method, the loading by pouring sample solution resulted in the best reproducibility of rheological measurement. Loading with spoon showed a high variance of viscosity value at low shear rate region ($0.01{\sim}1sec^{-1}$) while loading with syringe exhibited a low reproducibility of viscosity at high shear region ($1{\sim}100sec^{-1}$) with a disappearance of shear thinning phenomenon. It was revealed that the sample loading with small extra amount lead to the most reproducible result. The sample loading with the exact amount for the measuring plate resulted in a lack of reproducibility of high shear viscosity, while the loading with large extra volume produced a limited consistency of low shear viscosity. It was turned out that 3 min. of waiting time before measurement was the optimum condition for reliable result. When the waiting time was less than 1 min., the low shear viscosity was obtained with a lack of consistency. On the other hand, the sample solution started drying when the waiting time increased up to 5 min.

  • Using gel filtration chromatography, high molecular silk fibroin with high purity was obtained and silk flbroin microsphere particles (SFMP) could be simply made by spray dryer method. Also, some of the physicochemical properties of SFMP and morphology were investigated. The average molecular weight of pure silk fibroin protein dissolved in calcium chloride is about 61,500g/㏖ as measured by gel permeation chromatography. SFMP was spherical in shape, and particles, sized average of 2 ${\pm}$ 10 ${\mu}$, were observed by SEM and particle analyzer, respectively. Obtaining microspheres particles by spray dryer method accelerated the transition from the random coil to the $\beta$-sheet structure during spray dryer treatment. It was identified by the basic fourier transform infrared spectroscopy of SFMP. The swelling ratio of SFMP is majorly dependent on the pH of the solution, not on the occurred gelation. The characteristic structure, which might be applicable to immobilization of drugs is superior to other matrix materials for the use of biomaterials with skin affinity.

  • 생체재료로 이용하기 위하여 키토산을 기질로 하여 Bembyx mori로부터 추출한 견 피브로인을 블렌드한 필름에 대한 구조와 특성을 X선 회절분석, 열분석, 적외선 분광분석, 전자현미경 관찰, 접촉각 측정, 흡수율 측정으로 조사하였다. $-\~30wt\%$ 피브로인을 포함하는 블렌드 필름은 리튬이온을 포함하는 초산수용액으로 제조되었다. 키토산/피브로인 블렌드 필름은 키토산의 아미노기와 피브로인의 아미드 카보닐기 사이의 수소결합과 같은 분자간 상호작용의 형성에 의해 결정성이 감소되었다. 피브로인의 비율이 증가할수록 키토산의 무수혈 견정상은 소실되었고, 수화형 결정상은 감소되었으며, 퍼브로인의 P형 결정이 형성되어 혼재된 결정구조를 이루었다. 표면 친수성과 수분 흡수성은 피브로인의 비율이 클수록 증가되었으며, 20 $wt\%$, 이상에서 수화젤 상의 필름을 형성하였다. 전자현미경에 의한 블렌드 필름의 표면과 단면은 균일한 상을 나타내었다.

  • 가잠 실크 피브로인의 아미노산 조성 중 가장 많은 비율을 차지하고 있는 Glycine에 동위체 라벨링([1-13C]-Gly)을 실시하여 5령 1일째부터 고치를 지을 때까지의 생합성의 상태를 NMR법을 이용하여 추적하였다. 그 결과 5령 5일을 기준으로 가잠 실크의 결정부분을 형성하는 아미노산이 형성은 되나 실크의 결정부분을 형성하는 아미노산이 형성은 되나 실크가 되었을 때 결정성의 고배향 구조를 이루지 못하고 결정성의 저배향 구조를 형성하고마는 형태로 된다고 생각되어 졌다. 이 사실은 배향시료의 고체CP-NMR 스펙트럼의 피크 분리로부터 확인할 수 있었다. 그 결과 수소결합에 관여하는 N-H의 방향이 거의 수직이라고 할 수 있는 83˚의 분자축 부분이 18˚의 고배향 분포를 이루면서 82% 존재하고, 또 나는 N-H의 결합방향이 60˚인 분자축 부분이 18% 존재하면서 62˚의 배향분포를 이룬다고 하는 시뮤레이션의 결과로 설명되어졌다.

Preparation of cross-linked silk fibroin film by γ-irradiation and their application as supports for human cell culture

  • This study described about preparation of the cross-linked silk fibroin (SF) film by ${\gamma}$-irradiation of the casted SF film, which is fabricated from aqueous solution regenerated via fibers of cocoons and their application as supports for human cell culture. The properties of cross-linked SF film were evaluated by FT-IR spectroscopy, contact angle, solubility to water, thermal analysis, surface area analyzer, and morphology via scanning electron microscopy (SEM). The cross-linked SF films were not dissolved in water and exhibited the rough surface morphology, large surface area, and good thermal properties. The human fibroblast cell (CCD-986sk) and embryo kidney-ft cell were well growed on the surface of cross-linked SF film supports prepared by ${\gamma}$-irradiation. The cross-linked SF film prepared by ${\gamma}$-irradiation can be used as biomaterials for human cell culture.
