Studies on the Separation and Identification of Acids in Izmir Tobacco Leaves (Izmir 잎담배 중 Acids 성분의 분리 및 확인에 관한 연구)

  • This study was carried out to separate and identify the acidic compounds in tobacco leaves. Izmir tobacco leaves was extracted with isopropyl ether and the extract was concentrated. The concentrate was extracted with 6% NaHCO3 aqueous solution. The aqueous extract was acidified with sulfuric acid, and extracted with diethyl ether. The acidic material was fractionated on silicic acid column using a benzene-methanol mixture with a stepwise increasing methanol concentration. The resulting fractions were esterified with diazomethane, and then identified by GC, GC/MS using SPB -5 fused silica capillary column. Most of acidic compounds in Izmir tobacco leaves were elected from fraction B which was benzene-methanol(98 : 2) mixture on silicic acid column chromatography. The identified acidic compounds of Izmir tobacco leaves were 18 saturated acids, 8 unsaturated acids, 5 dicarboxylic acids, 13 aromatic acids and 7 terpenoid acids. The major acidic compounds of lzmir tobacco leaves were 2- methylbutanoic, 3-methyl butanoic, 3- methylpentanoic, hexanoic, nonanedioic, phenylacetic, benzoic, 4- methoxybenzoic, 3, 5- dimethoxybenzoic, methoxycinnamic and 3, 4- dimethoxycinnamic acid. Antitumor Effect of Mugwort (Artemisia princeps Pampan.) in vivo

  • This study was devised to observe the antitumor activity of mugwort (Artemisia princeps Pampan.) against ICR mice inoculated with sarcoma-180 cells. The antitumor compounds were partially purified from petroleum ether extract of mugwort by silicic acid column chromatography. The active fraction used in in vivo test was obtained under the elution with acetone in silicic acid column chromatography. When the acetone fraction was intraperitoneally injected to the mice which had been subcutaneously inoculated on the left groin with sarcoma-180, the growth rate of tumor (sarcoma-180 mass) was inhibited by 30%. In case the acetone fraction was injected to the mice which had been inoculated intraperitoneally with sarcoma-180, the average life span was prolonged by 20%. After the injection of the active fraction, the spleen index and ${\gamma}-globulin$ ratio (%) were increased significantly (p<0.05). The administration of acetone fraction did not cause any abnormality in the body and the homeostasis of mice. Those observations suggest that the acetone fraction of mugwort extract has an antitumor effect in vivo.

Effect of Curing Temperature on Geopolymeric Polycondensation of Blast Furnace Slag (고로슬래그의 지오폴리머 축중합반응에 미치는 양생온도의 영향)

  • The effect of curing temperature on basic geopolymeric reactivity and hardening behaviour of blast furnace slag were investigated using the mixture of pulverized slag and several alkaline solutions of relatively high concentration. For the pastes prepared at several different temperatures between 20$^{\circ}C$ and 90$^{\circ}C$, setting time and heat of reaction were examined while mineralogical and morphological examinations were performed for the hardened paste after curing period at same temperature. The geopolymeric reaction of slag was revealed to be accelerated strongly according to the curing temperature regardless of the sort and concentration of the alkaline solution. The increase of concentration of the alkaline solution within 9M and the existence of silicic ion in the solution also promoted the reaction severely. The mineral component and their ratio of the hardened paste were revealed to be influenced by the chemical species and silicic ion contained in alkaline solution rather than by the curing temperature. The higher temperature and longer period of curing stage were effective for the sustained formation of geopolymer and succeed improvement of density and uniformity of morphology of the final hardened body.

  • Silicatein-${\alpha}$, the enzyme extracted from silica spicules in glass sponges, has been studied extensively in the way of chemistry from 1999, in which the pioneering work by Morse, D. E. - the discovery of the enzymatic hydrolysis in Silicatein-${\alpha}$ - was published. Since its reaction conditions are physiologically favored, synthesis of various materials, such as gallium oxide, zirconium oxide, and silicon oxide, was achieved without any hazardous wastes. Although some groups synthesized oxide films and particles, they have not achieved yet controlled morphogenesis in the reaction conditions mentioned above. With the knowledge of catalytic triad involved in hydrolysis of silicone alkoxide and oligomerization of silicic acid, we designed the novel peptide amphiphiles to not only form self-assembled structure, but also display similar activities to silicatein-${\alpha}$. Designed templates were able to self-assemble into left-handed helices for the peptide amphiphiles with L-form amino acid, catalyzing polycondensation of silicic acids onto the surface of them. It led to the formation of silica helices with 30-50 nm diameters. These results were characterized by various techniques, including SEM, TEM, and STEM. Given the situation that nano-bio-technology, the bio-applicable technology in nanometer scale, has been attracting considerable attention; this result could be applied to the latest applications in biotechnology, such as biosensors, lab-on-a-chip, biocompatible nanodevices.

Quantitative Fractionation of Total Lipids and Their Fatty Acid Composition in Korean Yam Tubers. (한국산 마지질의 분획 정량과 지방산조성)

  • To investigate the lipid components of Korean yams(Dioscorea batatas and Dioscorea aimadoimo), silicic acid column, thin layer and gas chromatographies were applied. Total crude lipid contents in D. batatas and D. aimadoimo were 1.70% and 1.31%, and total purified lipid contents were 1.35% and 0.99% on a dry weight basis. Lipids were fractionated and quantitated by silicic acid column chromatography. The contents of neutral lipids, glycolipids and phospholipids were 43.33, 39.60 and 17.07% in D. batatas and 43.94, 34.74 and 21.32% in D. aimadoimo. Neutral lipids were the most abundant, in which triglycerides and sterol ester were found as the major components. Glycolipids were mainly composed of monogalactosyl and trigalactosyl diglycerides. Phosphatidyl choline and phosphatidyl ethanolamine were the major components of phospholipid. Fatty acids of the total lipids were mainly linoleic and palmitic acids. The fatty acid compositions of neutral lipids, glycolipids and phospholipids were the same tendency as that of total lipids. However, the content of linolenic acid in glycolipids was higher than in neutral lipids and phospholipids when fractionated.

Analytical Studies on the Lipids in Carotenoprotein purified form Saimo Salar Eggs (연어알에서 분리 정제한 carotenoprotein 지질 성분에 관한 연구)

  • 이온교환 크로마토그라피와 젤 여과법으로 연어알에서 carotenoprotein을 충분히 정제하였다. CHCls/MeOH (211)용매로 지질 성분을 추출한 후, Sephadex G-25로 정제하고 silicic acid 컬럼으로 분획하여 중성지질, 당지질, 인지질 그리고 콜레스테롤의 량을 각각 정량하였다. T.L.C.와 HPLC로 개별 성분을 분리, 정량한 결과 중성지질은 tripalmitate와 cholesteryl oleate로 구성되었고, 당지질은 esterified steryglycoside와 소량의 cerebroside, 인지질은 p.c, co, lp.c 및 p.e로 구성 되었음을 밝혔다. 결합당 성분으로서는 galactose만이 검출되었다. 지방산은 palmitate와 oleate가 대부분이었으며, 6종류의 sterol성분을 분리 확인하였다. 지질과 결합된 단백질의 아미노산도 분석하여 이들로부터 carotenoprotein의 본성을 추론하였다.

Effects of Contaning Pigments of Dried Grasshopper on the Lipid Deterioration (벼메뚜기 건제품중의 지질열화에 함유색소성분이 미치는 영향)

  • Changes in pigments of freeze dried grasshopper in refrigerator were determined to asses the storage stability. In storage at $5^{\circ}C$, chlorophyll contents were drastically decreased but pheopytins were increased. The degradation of chlorophylls get under controlled with vacuum package. The pigments extracted from 98 days stored samples were fractionated by silicic acid column and obtained 3 portions (3a, 3b and 3d). These portions were compared Rf values (0.75, 0.56, 0.50, respectively) and absorption spectra (408, 666nm; 419, 651nm; 433, 655nm, respectively) with that of standard and were identifed pheophytin a and b and 'changed' chlorophyll a. Especially, pheophytins a and b were great strength accelerated to lipid oxidation.

Fractionation and Quantitative Analysis of Lipid Components in Korean Yam (Dioscorea) Tubers (한국산 마의 지질 성분의 분리 및 분석)

  • Using the lipids extracted from Korean yam(Dioscorea) tubers, D. batatas, D. aimadoimo and D. japonica, fractionation and identification of lipid components and their fatty acid compositions were analysed. Lipid contents determined by Folch's method in D. batatas, D. aimadoimo and D. japonica were 11.0 mg/g, 11.4 mg/g and 6.6 mg/g, respectively. Lipids extracted were fractionated into neutral lipid, glycolipid and phospholipid by silicic acid column chromatography. The content of neutral lipid was over about 60% in lipid. Neutral lipid was composed of sterol ester, triglyceride, 1,3-diglyceride, 1,2-diglyceride and monoglyceride. Main constituents of glycolipid were acylsterylglycoside, monogalactosyldiglyceride, sterylglycoside, digalactosyldiglyceride and sulfolipid, and phospholipid contained phosphatidylethanolamine, phosphatidylcholine and phosphatidylinositol. The fatty acids of the total lipid and its three lipid fractions were analyzed by GC. The major fatty acids were palmitic and linoleic acids. Content of the saturated fatty acids was less than that of the unsaturated fatty acids.

Volcanisms and igneous processes of the Samrangjin caldera, Korea (삼랑진 칼데라의 화산작용과 화성과정)

  • The Samrangjin Caldera, a trapdoor-type, formed by the voluminous eruption of the silicic ash-flows of the Samrangjin Tuff which is above 630m thick at the northern inside of the caldera and thinnerly 80m at the southern inside. The caldera volcanism eviscerated the magma chamber by a series of explosive eruptions during which silicic magma was ejected to form the Samrangjin Tuff. The explosive eruptions began with phreatoplinian eruption, progressed through small plinian eruption and transmitted with ash-flow eruption. During the ash-flow eruption, contemporaneous collapse of the roof of the chamber resulted in the formation of the Samrangjin caldera, a subcircular depression subsiding above 550m deep. During postcaldera volcanism after the collapse, flow-banded rhyolite was emplaced as cental plug along the central vent and ring dikes along the caldera margins. Subsequently rhyodacite porphyry and dacite porphyry were emplaced along the inner side of the ring dike. After their emplacement, residual magma was emplaced as a hornblende biotite granite stock into the southwestern caldera margin. In the northeastern part, the eastern dikes were cut final intrusions of granodioritic to granitic composition along the fault zone of $^{\circ}$W trend.

  • Lipids extracted and purified from Herbs Capsellae leaf ana root were studied, To investigate those, silicic acid column chromatography, TLC, GLC was used for analysis of lipids and Fatty acids composition. Following results were obtained. 1. From result of total lipid classes in Herba Capsellae leaf and root, NL were obtained 62.6%, 58.5% respectly. GL 20.8%, 17,8%. PL 16.6%, 23.7%, respectively. 2. Among the NL in Herba capsellae leaf and root, TG were predominant 77.5%, 80.3%, respectly. SE 9.2%, 8.4%. FFA 4.8%, 3.4%. 1.2-DG 4.1%, 3.6%. FS 2.2%, 2.2%. and 1, 3-DG 1.3%, 1.0%, in turn. 3. The major fatty acids in the lipid classes were linoic acid, palmitic acid, oleid acid, in all. And total unsaturated fatty acid was NL 78.0%, GL 66.6%, and PL 75.2% in Herbs capsellae leaf, and NL 66.1% GL 63.6% and PL 61.2% in Herbs capsellae root.
