Changes in fermentation pattern and quality of Italian ryegrass (Lolium multiflorum Lam.) silage by wilting and inoculant treatments

  • Objective: This study was conducted to investigate the effects of wilting and microbial inoculant treatment on the fermentation pattern and quality of Italian ryegrass silage. Methods: Italian ryegrass was harvested at heading stage and ensiled into vinyl bags (20 cm×30 cm) for 60d. Italian ryegrass was ensiled with 4 treatments (NWNA, no-wilting noadditive; NWA, no-wilting with additive; WNA, wilting no-additive; WA, wilting with additive) in 3 replications, wilting time was 5 hours and additives were treated with 106 cfu/g of Lactobacillus plantarum. The silages samples were collected at 1, 2, 3, 5, 10, 20, 30, 45, and 60 days after ensiling and analyzed for the ensiling quality and characteristics of fermentation patterns. Results: Wilting treatment resulted in lower crude protein and in vitro dry matter digestibility and there were no significant differences in acid detergent fiber (ADF), total digestible nutrient (TDN), water-soluble carbohydrate (WSC), ammonia content, and pH (p>0.05). However, wilting treatment resulted in higher ADF and neutral detergent fiber content of Italian ryegrass silage (p<0.05), and the WNA treatment showed the lowest TDN and in vitro dry matter digestibility. The pH of the silage was higher in the wilting group (WNA and WA) and lower in the additive treatment group. Meanwhile, the decrease in pH occurred sharply between the 3-5th day of storage. The ammonia nitrogen content was significantly lower in the additive treatment (p<0.05), and wilting had no effect. As fermentation progressed, the lactic and acetic acid contents were increased and showed the highest content at 30 days of storage. Conclusion: The wilting treatment did not significantly improve the silage fermentation, but the inoculant treatment improved the fermentation patterns and quality of the silage. So, inoculation before ensiling is recommended when preparing high quality of Italian ryegrass silage, and when wilting, it is recommended to combine inoculation for making high quality silage.

Effect of Selected Inoculant Applications on Chemical Compositions and Fermentation Characteristics of High Moisture Rye Silage

  • The aim of this study was to investigate the effect of isolated lactic acid bacteria (LAB) on the quality of high moisture rye silage. Rye forage (Secale cereale L.) was harvested at the heading stage (27.3% of dry matter (DM)) and cut into approximately 3-5 cm lengths. Then, the forage divided into 4 treatments with different inoculants: 1) No additives (CON); 2) Lactobacillus brevis strain 100D8 at a 1.2 × 105 colony-forming unit (cfu)/g of fresh forage (LBR); 3) Leuconostoc holzapfelii strain 5H4 at a 1.0 × 105 cfu/g of fresh forage (LHO); and 4) Mixture of LBR and LHO (1:1 ratio) applied at a 1.0 × 105 cfu/g of fresh forage (MIX). About 3 kg of forage from each treatment was ensiled into a 20 L mini-bucket silo in quadruplicate for 100 days. After silo opening, silage was collected for analyses of chemical compositions, in vitro nutrient digestibilities, fermentation characteristics, and microbial enumerations. The CON silage had the highest concentrations of neutral detergent fiber and acid detergent fiber (p = 0.006; p = 0.008) and a lowest in vitro DM digestibility (p < 0.001). The pH was highest in CON silage, while lowest in LBR and MIX silages (p < 0.001). The concentrations of ammonia-N, lactate, and acetate were highest in LBR silage (p = 0.008; p < 0.001; p < 0.001). Propionate and butyrate concentrations were highest in CON silage (p = 0.004; p < 0.001). The LAB and yeast counts were higher in CON and LHO silages compare to LBR and MIX silages (p < 0.001). However, the mold did not detect in all treatments. Therefore, this study could conclude that L. brevis 100D8 and Leu. holzapfelii strain 5H4 can improve the digestibility and anti-fungal activity of high moisture rye silage.

Ensiling of Sweet Potato Leaves (Ipomoea batatas (L.) Lam) and the Nutritive Value of Sweet Potato Leaf Silage for Growing Pigs

  • The effect of adding carbohydrate-rich feedstuffs to sweet potato leaves (SPL) on silage quality was studied using a total of 180 laboratory silos. Silage quality was assessed by changes of pH, dry matter (DM), crude protein (CP) and ammonia nitrogen ($NH_{3}$-N). Pre-wilted SPL was mixed with cassava root meal (CRM), sweet potato root meal (SPM) or sugar cane molasses (Mo) at levels of 0, 30, 60 and 90 g $kg^{-1}$ (air-dry weight of additives to pre-wilted weight of SPL). Samples for assessing silage quality were collected after mixing the SPL with the additive and thereafter at 7, 14, 28 and 56 days of ensiling. There was a marked decrease in pH after 7 days and the pH remained low and stable until day 56. Addition of 60 and 90 g $kg^{-1}$ resulted in a lower pH (p<0.05) than the other treatments. The DM content of the silage increased (p<0.05) with increasing levels of additive, while there were no differences in DM with time of ensiling. The CP content of the silage decreased (p<0.05) with increasing levels of additive. The CP content did not change up to 28 days, but was lower (p<0.05) after 56 days in all treatments. The $NH_{3}$-N levels were increasing (p<0.05) with time of ensiling, and were lower (p<0.05) with additive levels of 60 g $kg^{-1}$ or higher. Also, the additive source affected the $NH_{3}$-N values, with the lowest values found for Mo. Castrated male pigs (Large White$\times$Mongcai) were used in 4$\times$4 Latin square design to study the total tract digestibility and nitrogen (N) utilisation of diets with inclusion of ensiled SPL. The diets were based on cassava root meal with inclusion of protein from either fish meal (C) or SPL ensiled with CRM (D1), SPL ensiled with SPM (D2) and SPL ensiled with Mo (D3). The digestibility of DM, organic matter (OM) and CP were higher (p<0.05), and the digestibility of crude fibre (CF) was lower (p<0.05), in diet C than in diets D1, D2 and D3. However, there were no differences (p>0.05) in digestibility of dietary components between diets D1, D2 and D3. Also, the excretion of N in faeces was higher (p<0.05) and the N retention was lower (p<0.05) in diets D1, D2 and D3 than in diet C. It can be concluded from the present experiments, that a good quality silage can be produced from pre-wilted SPL by addition of 60 g $kg^{-1}$ of either CRM, SPM or Mo. Diets with inclusion of 450 g ensiled SPL $kg^{-1}$ DM showed a high digestibility of dietary components and thus ensiled SPL should be considered as a potential feed resource for growing pigs.

옥수수 사일리지용 미생물의 발효능력 평가 (Evaluation of Fermentation Ability of Microbes for Corn Silage Inoculant)

  • 옥수수는 우리나라에서 매우 중요한 사료작물이다. 대부분 사일리지 형태로 이용이 되고 있으며 많은 비율의 곡실을 함유하고 있어 반추가축의 우수한 사료로 이용된다. 본 시험은 옥수수 사일리지용 미생물 첨가제 개발을 위해 수집된 균주의 발효능력을 평가하기 위해 수행되었다. 양질의 옥수수 사일리지에서 선발한 젖산균을 0.02%의 Sodium azide가 들어있는 MRS 배지에 접종을 한 후 균을 분리하여 MRS broth에 접종하여 산 생성 능력을 조사하였다. 총 6종의 젖산균을 선발하였고 동정결과 C3-2, B13-1 및 CC9-1은 Lactobacillus plantarum으로 판명되었고 C11-4는 Lactobacillus fermentum, B14-1은 Lactobacillus paracasei, 그리고 A3-1은 Leuconostoc lactis로 동정되었다. 선발된 5종의 균주와 시판되는 2종 첨가제 그리고 무처리구를 두고 황숙기에 수확된 옥수수에 접종하여 사일리지를 제조하고 2개월 후에 개봉하여 분석한 결과 B13-1 및 CC9-1 처리구의 사일리지가 pH가 낮았고 젖산함량도 높았다. 프리그 점수와 품질등급에 있어서도 B13-1 및 CC9-1 처리구에서 가장 높게 나타났다. 이상의 결과를 종합하여 볼 때 Lactobacillus plantarum B13-1 및 CC9-1는 사일리지 접종제로 사용이 권장되었다.

총체 벼 사일리지용 미생물의 발효능력 평가 (Evaluation of Fermentation Ability of Microbes for Whole Crop Rice Silage Inoculant)

  • 본 시험은 사료용 총체 벼 사일리지 전용 미생물 첨가제 개발을 위하여 2003년부터 2005년까지 축산연구소에서 수행되었다. 새로운 총체 벼 전용 첨가제 개발을 위해 수집된 28점의 총체 벼 사일리지로부터 미생물을 선발하여 전체 5개 균주(R4-1, R7-1, R7-2, R10-1 및 R12-1)를 분리하였다. 분리된 균주중 4종(R4-1, R7-1, R7-2 및 R10-1)은 Lactobacillus plantarum으로 동정되었고 R12-1은 L. pentosus로 동정되었다. 황숙기에 수확된 총체 벼는 사일리지를 제조할때 선발된 5개 균주 이외에 시중에서 판매되는 첨가제 3종을 접종을 하여 60일간 실온에서 저장한 후 평가한 결과는 다음과 같다. 저장된 사일리지의 pH와 초산 함량은 첨가제를 처리한 구에서 낮았고 젖산 함량은 높았다(p<0.05). 특히 R7-1은 초산 함량이 가장 낮았고 젖산 함량이 가장 높았다. 조단백질 함량은 R7-2 첨가제 처리구에서 가장 높았으며 ADF 및 NDF 함량은 R7-1에서 가장 낮았다(p<0.05). 선발된 균주들은 대부분 사일리지 품질개선효과를 나타내었으나 R7-1이 그 중에서 가장 효과적이었다. 본 미생물은 한국농용미생물 협회에 NLRI 401로 등록되었다.

Silage preparation and fermentation quality of natural grasses treated with lactic acid bacteria and cellulase in meadow steppe and typical steppe

  • Objective: In order to improve fermentation quality of natural grasses, their silage preparation and fermentation quality in meadow steppe (MS) and typical steppe (TS) were studied. Methods: The small-scale silages and round bale silages of mixed natural grasses in both steppes were prepared using the commercial lactic acid bacteria (LAB) inoculants Chikuso-1 (CH, Lactobacillus plantarum) and cellulase enzyme (AC, Acremonium cellulase) as additives. Results: MS and TS contained 33 and 9 species of natural grasses, respectively. Stipa baicalensis in MS and Stipa grandi in TS were the dominant grasses with the highest dry matter (DM) yield. The crude protein (CP), neutral detergent fiber and water-soluble carbohydrate of the mixed natural grasses in both steppes were 8.02% to 9.03%, 66.75% to 69.47%, and 2.02% to 2.20% on a DM basis, respectively. All silages treated with LAB and cellulase were well preserved with lower pH, butyric acid and ammonia-N content, and higher lactic acid and CP content than those of control in four kinds of silages. Compared with CH- or AC-treated silages, the CH+ AC-treated silages had higher lactic acid content. Conclusion: The results confirmed that combination with LAB and cellulase may result in beneficial effects by improving the natural grass silage fermentation in both grasslands.

Effect of Glucose and Formic Acid on the Quality of Napiergrass Silage After Treatment With Urea

  • Urea as a silage additive increases crude protein but reduces fermentation quality of silage by increasing pH and enhancing clostridial bacteria growth, especially in low sugar forages. Glucose and formic acid might be expected to compensate these defects caused by urea addition to grass silage. Thus, in this experiment urea formic acid or urea with glucose was applied to improve N content and the quality of napiergrass (Pennisetum purpureum Schumach.) silage. The first growth of napiergrass was harvested at 85 days of age and about 700 g of the grass was ensiled in laboratory silos (1.0 liter polyethylene containers) for 2, 7, 14, and 30 days at room temperature ($28^{\circ}C$). The treatments were no additives (control), urea, urea+glucose or urea+formic acid. Urea was added before ensiling at 0.5% of fresh weight of napiergrass and glucose and formic acid were added at 1% of fresh weight, respectively. After opening the silo, pH, dry matter content (DM), contents on DM basis of total N (TN), volatile basic nitrogen (VBN), lactic acid (LA), acetic acid (AA) and butyric acid (BA) were determined. The control at 30 days of fermentation showed 5.89 for pH with 13.8% for VBN/TN and 1.51% for AA. The addition of urea increased TN by about 1.5% units but decreased the fermentation quality by increasing pH from 5.89 to 6.86, increasing VBN/TN from 13.8% to 24.63%, increasing BA from 0.02% to 0.56%, and decreasing LA from 1.03% to 0.02%. Glucose addition with urea significantly decreased VBN/TN from 13.8% to 4.44% by reducing pH from 6.86 to 4.83 because of higher production of LA (2.62%). Adding urea and formic acid resulted in a more pronounced depression of VBN/TN and fermentation than the addition of urea and glucose. This study suggested that the combination of 1% glucose or 1% formic acid with 0.5% urea will improve nutritive value and fermentation quality of napiergrass silage.

Fermentation Quality of Ensiled Water Hyacinth (Eichhornia crassipes) as Affected by Additives

  • A lab-scale ensiling study was carried out to investigate the fermentation quality of water hyacinth (WH) supplemented with molasses, rice bran, as an absorbent, and an inoculant in the form of fermented vegetable juice and their combinations. After wilting the water hyacinths for 7 h to a dry matter (DM) content of 240 to 250 g/kg, the following treatments were applied: i) Control (C), WH only; ii) WH with sugarcane molasses at 40 g/kg WH (CM); iii) WH inoculated with fermented vegetable juice at 10 ml/kg WH (CI); iv) CM and CI (CMI) combined; v) WH with 150 g rice bran/kg WH (CA); vi) CA and CI combined (CAI); vii) CA and CM combined (CAM); and viii) CA, CM and CI combined (CAMI). After application of additives, the differently treated forages were mixed and ensiled in triplicates in 1,500-ml polyethylene jars. After ensiling for 3 d, pH values in all treatments, except C and CI, had decreased to approximately 4.0 and remained low till 14 d. After 56 d, pH had increased between 0.4 to 0.9 pH-units compared to those at 14 d. The ammonia nitrogen ($NH_3$-N) concentration ranged from an acceptable level in treatment CM (8 g/kg N) to a high $NH_3$-N value in treatment CMI (16 g/kg N). Lactic acid formation was higher in CI than in all other treatments. Butyric acid contents, which indicate badly fermented silages, were low in all silages (<2 g/kg DM). There were two-way interactions (p-values from <0.001 to 0.045) for almost all fermentation end-products and pH, except for the molasses${\times}$inoculant interaction on $NH_3$-N (p = 0.26). Significant 3-way interactions were found on all observed variables except for weight losses of silages. It is concluded that conserving wilted WH as silage for ruminants may be improved by the addition of molasses or rice bran.

Improving sugarcane bagasse quality as ruminant feed with Lactobacillus, cellulase, and molasses

  • The objective of the study was to evaluate the effects of Lactobacillus, cellulase, and molasses on chemical composition, fermentation qualities, and microorganism count of sugarcane bagasse silage after 30-days fermentation. The treatments were arranged according to a factorial arrangement (2 × 2 × 2) + 1, in a complete randomized design. The first factor consisted of two levels of Lactobacillus casei TH14 (TH14, 0 and 0.05 g/kg fresh matter; the second factor consisted of two levels of cellulase enzyme (C, 0 and 104 U/kg fresh matter); and the third factor consisted of two levels of molasses (M, 0 and 5 g/ 100 mL distilled water). A treatment (+1) referred to the use of rice straw without any treatments. The result showed that dry matter increased by 4% and neutral detergent fiber decreased by 2% of sugarcane bagasse when ensiled as a combination of additives as compared to untreated sugarcane bagasse. The pH and ammonia nitrogen were significantly dropped to 3.5 and 2.3 g/kg dry matter. Furthermore, lactic acid was increased by 64% when compared to untreated sugarcane bagasse, respectively. Lactic acid bacteria count was increased by 28% as compared to untreated sugarcane bagasse. Based on this experiment, fermenting with L. casei TH14, cellulase, and molasses in combination resulted in the promotion of the best qualities of sugarcane bagasse silage.
