  • The purpose of this study was to investigate the retention behavior of phenols and to predict their retention in RPLC. The retention data of twenty-five phenols were measured on a $\mu-{Bondapak}\;C_{18}$ and a polymeric $C_{18}$ columns with methanol-water and acetonitrile-water as a mobile phase. From the observation of enthalpy-entropy compensation phenomenon, the following conclusions are drawn with regard to the retention mechanism: 1) the retention mechanism of nitrophenols in different from that of metheyl-and chlorophenols in both mobile phase; 2) in methanol-water mobile phase, the retention mechanism of methyl-and chlorophenols is consistent in the range of methanol-water composition; 3) on the other hand, in the case of acetonitrile-water mobile phase, the retention mechanism depends on the volume fraction of acetonitrile. It means that the retention mechanism can not be explained only by a simple interaction. Based on retention data as compared with two columns, it may be said that the hydrophobic interaction of phenols with polymeric $C_{18}$ column was greater than that with monomeric $C_{18}$ column. The equations for predicting the retention of phenols were derived by using hydrophobic substituent constant $(\pi)$ and the sum of Hammett's constant $(\sigma)$ and Taft's steric constant $(E_s)$.

  • The concentration of selenium in grain samples was determined using isotope dilution method in ORC-ICPMS. The experimental conditions were optimized to $H_2$ mode and the flow rate was $4.0\;mL\;min^{-1}$. ORC in $H_2$ mode proved to eliminate most of polyatomic interferences except $BrH^+$ when Br is present in sample matrix. Chemical removal of Br was very difficult and the mathematical correction was successfully employed. The fraction of $BrH^+$ generated from Br at the current experimental condition was 14.1%. The signal on m/z 82 was corrected and calculated for isotope dilution. The analytical reliability of the propose method was successfully evaluated by analyzing the certified standard reference material NIST SRM 1566 and 1567. The method was applied to real samples and the results are $0.034{\pm}0.001\;{\mu}g\;g^{-1}$ for white rice, $0.059{\pm}0.002_5\;{\mu}g\;g^{-1}$ for brown rice, $0.029{\pm}0.001_4\;{\mu}g\;g^{-1}$ for black rice, and $0.034{\pm}0.002\;{\mu}g \;g^{-1}$ for barley. The detection limits ($3\sigma$) for Se was $0.012\;ng\;g^{-1}$.

  • Immunization of layers against Salmonella gallinarum(S.G.) which causes fowl typhoid resulted in production of anti-S.G. IgY rich eggs. Water soluble fraction was obtained from egg yolk using various gum solutions such as 0.1%(Sigma C-3889) λ-carrageenan; 1% and 2% cold water soluble carrageenan; 1% and 2% hot water soluble carrageenan; and 1% cold water soluble carrageenan with 1% hot water soluble carrageenan. Among them, λ-carrageenan 0.1% treatment showed a high recovery rate, possessing high IgY contents. In the range of pH 5-9, more than 70 percent of IgY was existent. Moreover, Anti-S.G. IgY was relatively heat-stable. This study revealed that immunoglobulin against fowl typhoid could be produced successfully by layers and the IgY was sustainable to further processing due to its pH and heat stability. IgY is promising to be utilized for prevention and treatment of fowl typhoid in industrial scale.

  • In the present study, we investigated the overexpression and characterization of bovine pancreatic (bp)- DNase I in Saccharomyces cerevisiae and Pichia pastoris. The bp-DNase I gene was fused in frame with the GAL10 promoter, $MF{\alpha}$, and GAL7 terminator sequences, resulting in the plasmid, pGAL-$MF{\alpha}$-DNaseI (6.4 kb). Also, the bp-DNase I gene was fused in frame with the AOX1 promoter, $MF{\alpha}$, and AOX1 terminator sequences, resulting in the plasmid, pPEXI (8.8 kb). The recombinant plasmids, pGAL-$MF{\alpha}$-DNaseI and pPEXI were introduced into S. cerevisiae and P. pastoris host cells, respectively. When the transformed yeast cells were cultured at $30^{\circ}C$ for 48 h in galactose or methanol medium, bp-DNase I was overexpressed and the most of activity was found in the extracellular fraction. P. pastoris transformant activity showed 45.5 unit/mL in the culture medium at 48 h cultivation, whereas S. cerevisiae transformant revealed 37.7 unit/mL in the extracellular fraction at 48 h cultivation. The enzymatic characteristics, such as DNA cleavage and half life were investigated. Treatment of the recombinant DNase I from P. pastoris induced degradation of the calf thymus DNA within 1 minute, and this DNA degradation rate was higher than that of commercial bp-DNase I (SIGMA) and the recombinant DNase I from S. cerevisiae.

  • After single fraction of 2, 5, 10 Gy irradiation on submandibular gland of 40 male rats, weighing 150gm, respectively, these animal were sacrificed two hours after 0.1㎎/g bromodeoxyuridine (Sigma) peritoneal injection in 1, 3, 7, 15 hours, 1, 3, 7 days after irradiation. And excised submandibular gland were fixed in Carnoy's and Bouin's solution for 2 hours. Paraffin sections were stained with H&E, and PAS for the observation of the change of salivary gland tissue, and with Feulgen for the study of the DNA distribution, and immunohistochemically stained with anti-bromodeoxyuridine (Sanbyo Co.) for detection of DNA synthetic cells in order to study the distribution of DNA synthetic cells of salivary gland tissue and endothelium after irradiation in 5 different sites of 6 slides on X 200 high power field. The results were as followings. 1. In PAS staining 3 days after 5Gy irradiation, decreased mucine secretion of serous cells were found, and 7 days after l0Gy irradiation, decreased mucine secretion of mucous cells were found. 2. In histopathologic features, degeneration of serous cells were found in 3 days after 2 Gy irradiation and there was little change in mucous cells and excretory duct cells. 3. In Feugen staining, 3 days after 2 Gy, 5 Gy irradiation, more high percentage of DNA synthetic cells were found in intercalated duct cells, striated duct cells and excretory duct cells than in BrdU staining. 4. In immunohistochemical features, DNA synethsis of serous cells and granular convoluted tubular cells abruptly decreased in early period after irradiation and showed no recovery in 7 days after irradiation but there was an increase in DNA synthesis of intercalated duct cells, striated duct cells and excretory duct cells, which have less S-phase cells comparatively, in 7 days after 2 Gy, 5 Gy irradiation. 5. In immunohistochemical features, the DNA synthesis of endothelial cells was continuously decreased after irradiation but showed slight increase in 7 days after 2 Gy and S Gy irradiation.

  • The dynamic softening mechanisms of AISI 316, AISI 304 and AISI 430 stainless steels were studied with torsion test in the temperature range of $900 - 1200^{\circ}C$ and the strain rate range of $5.0x10^{-2}-5.0x10^0/sec$. The austenitic stainless steels, such as AISI 316 and AISI 304 were softened by dynamic recrystallization (DRX) during hot deformation. Also, the evolutions of flow stress and microstructure of AISI 430 ferritic stainless steel show the characteristics of continuous dynamic recrystallization (CDRX). To establish the quantitative equations for DRX of AISI 316 stainless steel, the evolution of flow stress curve with strain was analyzed. The critical strain (${\varepsilon}_c$) and strain for maximum softening rate (${\varepsilon}^{*}$) could be confirmed by the analysis of work hardening rate ($d{\sigma}/d{\varepsilon}={\theta}$). The volume fraction of dynamic recrystallization ($X_{DRX}$) as a function of processing variables, such as strain rate ( $\varepsilon$ ), temperature (T), and strain ( $\varepsilon$ ) were established using the ${\epsilon}_c$ and ${\varepsilon}^{*}$. For the exact prediction the ${\varepsilon}_c,\;{\varepsilon}^{*}$ and Avrami' exponent (m') were quantitatively expressed by dimensionless parameter, Z/A, respectively. It was found that the calculated results were agreed with the experimental data for the steels at my deformation conditions. Also, we can reasonably conclude that the DRX, CDRX and grain refinement of stainless steels can be achieved by large strain deformation at high Z parameter condition.

  • Supernovae type Ia (SNe Ia) cosmology is providing the only direct evidence for the presence of dark energy. This result is based on the assumption that the look-back time evolution of SNe Ia luminosity, after light-curve shape correction, would be negligible. However, the most recent compilation of SNe Ia data shows systematic difference in the Hubble residual (HR) between the E and Sd/Irr galaxies, indicating that the light-curve fitters used by the SNe Ia community cannot quite correct for a large portion of the population age effect. In order to investigate this possibility more directly, we have obtained low-resolution spectra for 30 nearby early-type host galaxies. This data set is used to estimate the luminosity-weighted mean ages and metallicities of host galaxies by employing the population synthesis models. We found an interesting trend between the host galaxy age and HR, in the sense that younger galaxies have positive residuals (i.e., light-curve corrected SNe Ia luminosity is fainter). This result is rather independent of the choice of the population synthesis models employed. Taken at face value, this age (evolution) effect can mimic a large fraction of the HR used in the discovery of the dark energy. This result is significant at 1.4 - 3 sigma levels, depending on the light curve fitters adopted, and further observations and analyses are certainly required to confirm the trend reported here.

  • We explore the relative importance of the role of small-scale environment and large-scale environment in triggering nuclear activity of the local galaxies using a volume-limited sample with $M_r$ < -19.5 and 0.02 < z < 0.0685 selected from the Sloan Digital Sky Survey Data Release 7. The active galactic nuclei (AGN) host sample is composed of Type II AGNs identified with flux ratios of narrow emission lines with S/N > 6 and the central velocity dispersion of the sample galaxies is limited to have a narrow range between 130 < ${\sigma}$ < 200($km\;s^{-1}$), corresponding to 7.4 < $log(M_{BH}/M_{\odot})$ < 8.1 in order to fix the mass of the supermassive black hole at the center of its host galaxy. In this study, we find that the AGN fraction ($f_{AGN}$) of late-type galaxies are larger than of early-type galaxies and that for target galaxy with late-type nearest neighbor, $f_{AGN}$ starts to increase as the target galaxy approaches the virial radius of the nearest neighbor (about a few hundred kpc scale). The latter result may support the idea that the hydrodynamic interaction with the nearest neighbor as well as tidal interaction and merger also plays an important role in triggering the nuclear activity of galaxy. We also find that early-type cluster galaxies show decline of AGN activity compared to ones in lower density regions, whereas the direction of dependence of AGN activity for late-type galaxies is opposite.

  • The paper attempts to estimate the incubation time of a cavity in the interface between a power law creep particle and an elastic matrix subjected to a uniaxial stress. Since the power law creep particle is time dependent, the stresses in the interface relax. The volume free energy associated with Helmholtz free energy includes strain energies caused by applied stress and dislocations piled up in interface(DPI). The energy due to DPI is found by modifying the result of Dundurs and Mura[4]. The volume free energies caused by both applied stress and DPI are a function of the cavity size(r) and elapsed time(t) and arise from stress relaxation in the interface. Critical radius $r^*$ and incubation time $t^*$ to maximise Helmholtz free energy is found in present analysis. Also, kinetics of cavity formation are investigated using the results obtained by Riede [7]. The incubation time is defined in the analysis as the time required to satisfy both the thermodynamic and kinetic conditions. Through the analysis it is found that 1) strain energy caused by the applied stress does not contribute significantly to the thermodynamic and kinetic conditions of a cavity formation, 2) in order to satisfy both thermodynamic and kinetic conditions, critical radius $r^*$ decreases or holds constant with increase of the time until the kinetic condition(eq. 2.3) is satisfied. there for the cavity may not grow right after it is formed, as postulated by Harris [15], and Ishida and Mclean [16], 3) the effects of strain rate exponent (m), material constant $\sigma$0, volume fraction of the particle to matrix(f)and particle size on the incubation time are estimated using material constants of the copper as matrix.

  • The high-temperature deformation mechanisms of a ${\alpha}+{\beta}$ titanium alloy (Ti-6Al-4V), near-a titanium alloy (Ti-6.85Al-1.6V) and a single-phase a titanium alloy (Ti-7.0Al-1.5V) were deduced within the framework of inelastic-deformation theory. For this purpose, load relaxation tests were conducted on three alloys at temperatures ranging from 750 to $950^{\circ}C$. The stress-versus-strain rate curves of both alloys were well fitted with inelastic-deformation equations based on grain matrix deformation and grain-boundary sliding. The constitutive analysis revealed that the grain-boundary sliding resistance is higher in the near-${\alpha}$ alloy than in the two-phase ${\alpha}+{\beta}$ alloy due to the difficulties in relaxing stress concentrations at the triple-junction region in the near-${\alpha}$ alloy. In addition, the internal-strength parameter (${\sigma}^*$) of the near-${\alpha}$ alloy was much higher than that of the ${\alpha}+{\beta}$ alloy, thus implying that dislocation emission/ slip transfer at ${\alpha}/{\alpha}$ boundaries is more difficult than at ${\alpha}/{\beta}$ boundaries.
