Control of Fusarium Wilt of Watermelon with the Root-Stock Grafting of Sicyos angulatus L. (안동오이 대목을 이용한 수박 덩굴쪼김병 방제)

  • Watermelon plants grafted with the root-stock of wild-cucumber (Sicyos angulatus) were not infected by Fusarium oxysporum f.sp. niveum in pot inoculation and infected fields tests. Controlling efficacy of the root-stock grafting with S. angulatus on Fusarium wilt of watermelon was more excellent than that of the root-stock grafting with Lagenaria siceraria. The isolates of Fusarium oxysprum from cucurbitaceae plants had a certain host-specific pathogenicity, but they did not express the absolute one forma specialis-one host-plant phenomenon by the root dipping inoculation. The pathogenic isolates of Fusarium oxysproum from cucurbitaceae crops did not infect the root-stock plant such as S. angulatus, L. siceraria and Cucurbita ficifolia. The fast-wilting of watermelon caused by uncertain agents was observed in watermelon plant grafted with L. siceraria in the continuously cropping fields, but it was not observed in watermelon plants grafted with S. angulatus in the same fields.

Effects of Plant Growth Regulators on Callus Formation and Organogenesis of Sicyos angulatus L. (야생식물 안동오이의 기내배양시 캘러스형성과 기관분화에 미치는 생장조절제의 영향)

  • This study was carried out to determine the effects of plant growth regulators on cell culture and organogenesis from Sicyos angulatus L. using explants of leaves, stems and cotyledons. Optimal callus induction for S. angulatus was obtained on MS medium with 0.1mg/$\ell$ BA and 2.0mg/$\ell$ 2,4 -D from cotyledons, 0.1mg/$\ell$ BA and 5.0mg/$\ell$ NAA from leaves explants, Optimal media for subculture and growth of S. angulatus callus were 1/2 MS medium with 0.1mg/$\ell$ BA and 1.0mg/$\ell$ 2,4 -D for solid culture, and 0.1mg/$\ell$ 2,4-D for suspension culture. Many adventitious roots with some shoots were formed were formed from leaf and cotyledon explants of S. angulatus during callus induction with optimal combinations of plants growth regulators.

Callus Induction and Somatic Embryogenesis from Sicyos angulatus L. (야생식물 Sicyos angulatus L.로부터 캘러스 유도 및 체세포배 발생)

  • In order to investigate the possibility of in vitro mass propagation via somatic embryogenesis from Sicyos angulatus L., effects of plant growth regulators and carbon sources on callus induction and somatic embryogenesis were evaluated. Optimal combinations of plant growth regulator for callus induction from cotyledon and inflorescence explants were 2,4-D 2.0 mg/L + BA 0.1 mg/L and 2,4-D 1.0 mg/L + BA 0.1 mg/L in MS basal medium supplemented with sucrose 30 g/L,, respectively. Somatic embryogenesis was observed from cultured inflorescence explants, but it could not be achieved from leaf or cotyledon explants. The most effective plant growth regulators for somatic embryogenesis from callus was NAA 1.0 mg/L + kinetin 10 mg/L in the half strength of MS basal medium supplemented with 20 g/L sucrose.

Herbicidal Activity of Naturally Developed d-Limonene against Sicyos angulatus L. under the Greenhouse and Open Field Condition (온실 및 노지조건에서 천연물질 d-Limonene의 가시박에 대한 살초활성)

  • This experiment was conducted to Investigate the herbicidal activity of naturally developed d-Limonene on the control of Sicyos angulatus L. which is destructive to biodiversity and ecosystem under greenhouse and open field condition. Among the tested concentrations, most efficacy of early foliar application(leaf stage with 1.5 and 3) of d-Limonene was found in $70mg\;mL^{-1}$, followed by $35mg\;mL^{-1}$, $17.5mg\;mL^{-1}$ in that order under greenhouse condition. The higher the d-Limonene concentration, the better the effect in greenhouse condition. In leaf stage with 2, 3 and 5 under open field condition, the higher the d-Limonene concentrations, the higher the weeding effect. The remarkable effect was, particularly, found in d-Limonene with $70mg\;mL^{-1}$ while little was found in the plants with d-Limonene with $35mg\;mL^{-1}$. In the case of 15 leaf stage of S. angulatus L. treated with d-Limonene under outside treatment effect was low. The treatment effect of d-Limonene under open field compared with greenhouse condition was more significant. It can be recommended that d-Limonene with $70mg\;mL^{-1}$ could control S. angulatus L. by 5 leaf stage.

Antioxidant Activity and Anti-inflammatory effects of Sicyos angulatus L. extract (가시박 추출물의 항산화 및 항염증 효과)

  • This study was carried out to find out the physiological activity effect of Sicyos angulatus L. extract which is known as an ecosystem disturbance plant and confirm the availability as a functional cosmetic material. Total polyphenol and flavonoid contents were measured, and DPPH radical scavenging, intracellular ROS, and its inhibitory effect on the expression Nitric oxide of and COX-2 were evaluated. The content of polyphenol and flavonoid was found to be 3.079 mg(CA)/100g and 72 mg(Q)/100g of Sicyos angulatus L. extract and antioxidant activity through high radical scavenging activity was confirmed. ~Significant cytotoxicity was not observed up to a concentration of $100{\mu}g/mL$ in RAW 264.7 cells and HDF cells and concentration-dependent inhibition of ROS production in HDF cells, inhibition of high NO production and inhibition of COX-2 protein expression in RAW 264.7 cells were confirmed. Through these results, we found the possibility of use as a functional cosmetic material with excellent antioxidant effect and antioxidant and anti-inflammatory effects through inhibition of intracellular ROS generation, inhibition of NO generation, and inhibition of COX-2 protein expression.

FISH Karyotype Analysis of Four Wild Cucurbitaceae Species Using 5S and 45S rDNA Probes and the Emergence of New Polyploids in Trichosanthes kirilowii Maxim

  • Wild relative species of domesticated crops are useful genetic resources for improving agronomic traits. Cytogenetic investigations based on chromosome composition provide insight into basic genetic and genomic characteristics of a species that can be exploited in a breeding program. Here, we used FISH analysis to characterize the ploidy level, chromosome constitution, and genomic distribution o f 5S and 4 5S r ibosomal DNA (rDNA) in four wild Cucurbitaceae species, namely, Citrullus lanatus (Thunb.) Mansf. var. citroides L. H. Bailey (2n = 22), Melothria japonica Maxim. (2n = 22), Sicyos angulatus L. (2n = 24), and Trichosanthes kirilowii Maxim. (2n = 66, 88, 110 cytotypes), collected in different areas of Korea. All species were diploids, except for T. kirilowii, which included hexa-, octa-, and decaploid cytotypes (2n = 6x = 66, 8x = 88, and 10x = 110). All species have small metaphase chromosomes in the range of $2-5{\mu}m$. The 45S rDNA signals were localized distally compared to the 5S rDNA. C. lanatus var. citroides and M. japonica showed one and two loci of 45S and 5S rDNA, respectively, with co-localization of rDNA signals in one M. japonica chromosome. S. angulatus showed two co-localized signals of 5S and 45S rDNA loci. The hexaploid T. kirilowii cytotype showed five signals each for 45S and 5S rDNA, with three being co-localized. This is the first report of hexaploid and decaploid cytotypes in T. kirilowii. These results will be useful in future Cucurbitaceae breeding programs.

Enhancement of Seed Germination by Aging, Cold-stratification, and Light Quality during Desiccation in Burcucumber (Sicyos Angulatus L.)

  • Seeds of burcucumber were treated with accelerated aging, cold-stratification, and light quality illuminated during desiccation to enhance their germination and seedling emergence. The germination was increased by aging and cold-stratification although the latter treatment showed greater effectiveness than the former one. In the combined treatment of aging 6 days at $45^{\circ}C$ and cold-stratification, the germination was promoted under longer period of cold-stratification to reach nearly 100% in 3 week cold-stratification on the ninth day from sowing. In the sequentially combined treatment of aging, cold-stratification, and light quality during 24 hour desiccation at $35^{\circ}C$, no-stratified seeds showed the highest rate in red light treatment but the lowest in far-red light. This implies that the phytochrome action run during the desiccation of imbibed seeds. The red light exposure during drying for the cold-stratified seeds after aging accelerated the germination even more than the dark treatment and germinated 100% on the next day of sowing. It is concluded that the sequential treatment of aging, cold-stratification, and red light illumination during desiccation can highly promote percentage and speed of burcucumber seed germination.

Effect of Shading Using Different Colored Fabrics on Burcucumber(Sicyos angulatus L.) Seedling Growth and Cotyledon Chlorophyll Content (차광막의 색상이 안동대목 유묘의 생장과 자엽의 엽록소 함량에 미치는 영향)

  • Nowadays burcucumber(Sicyos angulatus L.) has been widely used as a rootstock of Cucurbitaceae. The study was done to examine the effects of shading with different colored fabrics(white, red and aluminum-coated) on growth and cotyledon chlorophyll content of its seedling. Shading with the white colored fabric lengthened seedling height and hypocotyl length, but shading with the red colored fabric enlarged true leaf area. Total dry weight of each seedling was decreased in the order of no shading, white, red and aluminum, in which resulted from reductions of cotyledon dry weight in white colored fabric, cotyledon and true leaf dry weight in red colored fabric, and dry weight of cotyledon, true leaf and root in aluminum colored fabric. Cotyledon chlorophyll content was reduced by shading treatments although the 3 shading treatments showed nearly same content.
