Rapid Propagation through Tissue Culture of Cudrania tricuspidata, Medicinal Plant

  • An effective rapid propagation method was established through in vitro cultures of the medicinal plant, Cudrania tricuspidata. In vitro plantlets were obtained from in vitro germinated seeds. The various levels of cytokinins (BAP, Kinetin and TDZ) were tested on multiple shoot formation from plantlets. BAP (1.0 mg/l) treatment induced highest number of multiple shoots. Single shoot cultures gave higher initial shoot numbers than 5 shoots per culture. Among the various culture media, the shoot elongation was optimal on 2 MS basal medium without growth regulators. The IAA (2.0 mg/l) treatment induced highest number of roots. IBA (2.0 mg/l) treatment more promoted in vitro root growth than other concentrations. Rooted shoots were transferred directly to small pots with an artificial soil and successfully acclimatized.

In Vitro Propagation of Cindium officinale Makino Through Shoot Tip Culture (천궁의 경정배양을 통한 기내번식)

  • This experiment was conducted to identify the optimal in vitro propagation condition of Cnidii rhizoma (Cnidium officinale Makino). It was effective to reduce contamination and improve regeneration of shoot when shoot tips taken in July were cultured in 1/2 strength Murashige and Skoog medium supplemented with 500 mg/L carbenicillin disodium 1.0 mg/L BA and 1.0mg/L $GA_3$followed by surface sterilization of explant source in solution of 1% sodium hypochlorite for 20 minutes. When shoot tips were 쳐cultured in 1/2 strength MS medium with 0.5 mg/L BA and 60 g/L sucrose, shoot elogation and subsequent multiplication of the formed shoot were favorable than in other media. Regenerants were well rooted in 1/2 strength MS medium containing 3.0 mg/L NAA.

  • This study was conducted to examine the effects of explant sources and plant growth regulators on mass propagation of Cyclamen persicum. Tuber, cotyledon, and petiole tissues were cultured on MS medium supplemented with different concentrations and combinations of auxins and cytokinins. Shoots were not induced from calli on cotyledon and petiole explants cultured on MS medium containing various concentrations of 2,4-D or NAA. However, multiple shoots were formed directly from tuber explants cultured on the medium containing 0.5 and 1.0 mg/L 2,4-D or NAA. In MS medium with cytokinin alone, TDZ was more effective in shoot formation than BA or kinetin in all explants. The combinations of NAA and BA was found to be most effective in shoot formation from tuber, cotyledon and petiole explants. Especially, shoots were formed from all the tuber explants on the medium containing 0.5 mg/L of NAA and BA. Hormonal effects on root formation were examined by subculturing single shoots on MS medium containing NAA or IBA. The medium with 0.5 mg/L IBA was most effective in root induction and subsequent plantlet regeneration.

Effect of Medium Composition and Growth Regulators on Mass Propagation of Virus-Free Plant from the Meristem Cultures of 'Jarang' Grape ('자랑' 포도 생장점 배양으로부터 무병주 식물 대량번식에 미치는 배지 구성물질과 생장 조절제의 영향)

  • This study was performed to clarify the effect of medium compositions and plant growth regulators on the shoot, root formation and growth of 'Jarang' grape for mass propagation of virus-free plant. The formation and growth of shoot were considerably favorable in half-concentration of MS medium. However, the formation of adventitious root per explants (avg. 2.1) was effective in higher concentration (two times) of MS medium. For sucrose concentration, 1% for the shoot formation, 3% for the adventitious root formation and 1% for the growth were observed as yield significant results. With the addition of 0.05% of activated carbon, the shoot growth was improved, and it was effective for the adventitious root formation and growth as well. A pH of 6.8 in the medium was the most suitable for mass propagation; the results showed significant enhancement in the number of nodes and the length of the shoot, 3.9 and 1.3 cm, respectively. The shoot growth was the most vigorous in BA 1.0 mg/L due to the impact of the growth regulator on the mass propagation in it. Consequently, 16.9 shoots per explant were formed in NAA 1.0 mg/L so good results were obtained.

  • This study was undertaken to know the possibility of in vitro propagation of Stevia through axillary bud culture and the results indicated that: (1) Addition of NAA (0.01-0.05 mg/l) alone on Murashige-Skoog basal medium promoted shoot differentiation and growth rate. And also additional of kinetin of 0.5-1.0 mg/1 alone showed the same trend as that of NAA: (2) Addition of both NAA (0.01-0.05 mg/l) and kinetin (0.5-1.0mg/l) to MS medium promoted better shoot formation. (3) Shoot differentiation and growth were better on the full salt strength of MS medium (1X MS) than that of half strength ( $\frac{1}{2}$MS), while their effects were reversed for root differentiation

  • An efficient method for the rapid propagation of Smilax china from axillary buds was established. Plants with thick leafage were selected from Korea native S. china population. Axillary buds of S. china collected from selected plant and were cultured in various culture media (2MS, MS, 1/2MS, WPM, B5 and SH medium). Shoot was induced from axillary bud on MS basal medium after 4 weeks of culture. 1/2MS medium showed a higher growth rate than those of the others, while the lowest shoot growth was obtained in 2MS medium. Among the sucrose concentrations, 5% sucrose was the optimum level for shoots growth from axillay buds. Among cytokinins, $0.5mgL^{-1}$ 6-benzylaminopurine (BAP) treatment showed the best performance on shoot multiplication, yielding average shoot multiplication forming about 2.4. Rooting was induced directly near the base of the shoot on 1/2MS medium containing with three-auxins ${\alpha}-napthalene$ acetic acid (NAA), indole acetic acid (IAA) and ${\beta}-indolebutyric$ acid (IBA) (0.5 and $1.0mgL^{-1}$). The $1.0mgL^{-1}$ IBA treatments induced earliest rooting with maximum of root number and root growth. These rooted plantlets were successfully transferred to pots for 4 weeks hardening process, and were transferred to soil with above 90% survival rate.

Silver nitrate and silver-thiosulphate mitigates callus and leaf abscission during Shisham clonal micro-propagation

  • Basal callus formation and leaf abscission is a problem in clonal micropropagation. We have described an in vitro clonal propagation protocol of Dalbergia sissoo Roxb (shisham) 'FRI-14' in which AgNO3 played important role not only in mitigating problem of leaf abscission and basal callus, but also improved shoot induction and multiplication. Best induction and shoot multiplication was obtained on MS media with 1.5 mg/l 6-BAP and 10 mg/l AgNO3 and half-strength MS media with 0.5 mg/l 6-BAP, 2 mg/l AgNO3 and 50 mg/l Adenine sulphate whereas best ex vitro rooting was obtained with 200 mg/l IBA in pulse treatment.

Seed Germination and Softwood Cutting Technique of Kalopanax pictus Nakai (음나무(Kalopanax pictus Nakai)의 종자번식 및 녹지삽목 번식기술)

  • This study was carried out to investigate mass propagation technique by using seed and vegetative propagation of Kalopanax pictus Nakai. For developing seed propagation, seed stratification and 1$0^{\circ}C$ after-ripening treatment induced embryo growth within 1 weeks, resulted in increasing germination rate of seeds up to more than 65% when planted. The softwood cutting using one year old shoot increased rooting rate to 69% whereas more than 1 year old shoot looked like inappropriate for cutting propagation. In the cutting timing, the rooting rate on June, 13 cutting of the first growth shoot was the highest, followed by June 20 and July 4. The most efficient cutting timing seemed to be the middle of June. When cutted shoots were soaked for 30 minute with IBA and NAA 1000mg.$L^{-1}$, rooting rate was increased above 70%. As the concentrations of plant hormone were increased above 2000mg.$L^{-1}$, the rooting rate was slowly decreased.

In Vitro Shoot Tip Culture of Haemaria discolor (Haemaria discolor 경정의 기내배양)

  • The experiments were conducted to obtain culture conditions on optimal cultural conditions for propagation of a topical orchid, Haemaria discolor, through shoot tip cultrue in vitro. Survival percentage of shoot tip explant, in initial culture media was higher in H$_3$P$_4$ medium than in Murashige and Skoog's medium. Explants, cultured in H$_3$P$_4$ medium containing 1.0 mg/L kinetin, showed more shoot elongation when compared with those on other media. When the distal part of stem (pseudobulb) was longitudinally sectioned into half and cultured in H3P4 medium containing 0.1 mg/L 2ip, 0.1, or 1.0 mg/L kinetin, shoots from axillary bud were much more elongated under light condition.
