A SEM STUDY OF RADIATION EFFECTS ON THE RAT MOLAR ENAMEL FORMATION (방사선조사가 백서 구치 법랑질형성에 미치는 영향에 관한 주사전자현미경적 연구)

  • The purpose of this study was to investigate the effects of radiation on the formation of rat molar enamel at the developmental stage. The experimental animals were divided into five groups and were irradiated single dose of 396cGy ; 1 st group on 14th day of gestation, 2nd group on 19th day of gestation, 3rd group on 3 days after birth, 4th group on 8 days after birth, 5th group on 28 days after birth. The control and 1, 2, 3, and 4th experimental groups were sacrificed on 2, 4, and 6 weeks and the 5th groups were sacrificed on 1 day and 2 weeks after irradiation. Distal 1/2 and occlusal 1/3 enamel surface of lingual side of lingual cusp, and fractured surface of lingual side of lingual cusp in a longitudinal direction of the mandibular first molar were examined using scanning electron microscope. The following results were obtained. 1. The roughness of enamel surface and enamel hypoplasia were increased in a sequence of 4th, 1st, 2nd, and 3rd experimental group, and the enamel cracks were increased in the 1st and 2nd experimental group. 2. The pattern of enamel hypoplasia had a network form on the 1st and 2nd experimental group, and appeared a linear shape on the 3rd experimental group, and then the crator-like enamel defects were observed in all experimental groups (especially 1st and 2nd experimental group) except 5th. 3. Dentinoenamel junction showed the clear-cut and straight appearance except 5th experimental group. 4. There was no significant difference between 5th experimental and control group.

  • Alumina hydrates were prepared by the neutralization of AlCl3.6H2O solution with NH3 gas diluted with N2 gas. The values of pH in reaction solution influenced the formation of alumina hydrates minerals. Amorphous alumina hydrates, for example, were formed at ${\gamma}$-Al2O3longrightarrow$\delta$-Al2O3longrightarrow$\theta$-Al2O3longrightarrow$\alpha$-Al2O3. (2) Bayeritelongrightarrowamorphouslongrightarrow${\gamma}$-Al2O3longrightarrow$\delta$-Al2O3longrightarrowη-Al2O3longrightarrow$\theta$-Al2O3longrightarrow$\alpha$-Al2O3. Shot Change Detection Using Multiple Features and Binary Decision Tree (다수의 특징과 이진 분류 트리를 이용한 장면 전환 검출)

  • Contrary to the previous methods, in this paper, we propose an enhanced shot change detection method using multiple features and binary decision tree. The previous methods usually used single feature and fixed threshold between consecutive frames. However, contents such as color, shape, background, and texture change simultaneously at shot change points in a video sequence. Therefore, in this paper, we detect the shot changes effectively using multiple features, which are supplementary each other, rather than using single feature. In order to classify the shot changes, we use binary classification tree. According to this classification result, we extract important features among the multiple features and obtain threshold value for each feature. We also perform the cross-validation and droop-case to verify the performance of our method. From an experimental result, it was revealed that the EI of our method performed average of 2% better than that of the conventional shot change detection methods.

Morphological and Genetic Stability of Dormant Apple Winter Buds After Cryopreservation

  • Twenty apple germplasm accessions from the Korean Genebank were successfully cryopreserved using two-step freezing to back up genetic resources maintained by field collections. This study examined the morphological and genetic stability of cryopreserved dormant apple buds that were stored in liquid nitrogen, and then rewarmed and regrown. Whole plants were regenerated directly from dormant buds through budding without an intermediary callus phase. The cryopreserved buds produced high levels of shoot formation (76.2-100%), similar to those of noncryopreserved buds (91.3-100%), with no observed differences between cryopreserved and noncryopreserved materials. Three of the twenty cryopreserved apple germplasm accessions were used to assess morphological and genetic stability. No differences in morphological characteristics including shoot length, leaf shape, leaf width/length ratio, and root length were observed between controls (fresh control and noncryopreserved) and cryopreserved plantlets. The genetic stability of regenerants (before and after cryopreservation) was investigated using inter simple sequence repeat (ISSR) markers. The ISSR markers produced 253 bands using four primers, ISSR 810, SSR 835, ISSR 864, and ISSR 899. These markers showed monomorphic banding patterns and revealed no polymorphism between the mother plant and regenerants before and after cryopreservation, suggesting that cryopreservation using two-step freezing does not affect the genetic stability of apple germplasm. These results show that two-step freezing cryopreservation is a practical method for long-term storage of apple germplasms.

NALYSIS OF THE ECLIPSING BINARY SDSS J1021+1744: A WDMS SYSTEM WITH UNUSUAL DIPS

  • We present our recent observations of SDSS J102102.25+174439.9, a new eclipsing white dwarf - main sequence WDMS binary with an orbital period of 0.14 days. This system belongs to the post common-envelope binary group as shown by the spectrum from the Sloan Digital Sky Survey. We obtained our data using the ULTRASPEC instrument installed on the 2.4-m telescope at the Thai National Observatory (TNO). Our multi-band observations reveal an unusual and persistent drop in brightness after the primary eclipse. These dips, which appear to show variations in amplitude, also have a complex shape that changes within days. Dips in WDMS systems have been observed on only one other occasion, in the light curve of QS Vir prior to the eclipse of the white dwarf. The dips in SDSS J1021+1744 are unique because they are present at different wavelengths and they occur approximately at similar phases. Hosting a DA white dwarf and an M4 companion star, this system is known to be the only WDMS to show these kind of dips in its light curve. It is possible that these dips are caused by ejected materials from an active companion star, such as in QS Vir. The light curve in the g' filter exhibits deep and narrow features, implying that the material which passes in front of the white dwarf in SDSS J1021 must be dense and small in size. Furthermore, we try to constrain the stellar and orbital parameters of SDSS J1021+1744 using the Binary Maker 3 software. We use g' and r' data for our light curve analysis to have a better approximation for the red dwarf star.

  • Transformation of an auxotrophic requirement for uracil in Pleurotus florida P101 has been achieved using chimeric vector containing Aspergillus nidulans ans 1, and Neurospora crassa pyr 4 DNA. Protoplasts of $Ura^-$strains of P. florida were incubated with plasmid pDJB3 containing the cloned pyr 4 gene in the presence of polyethylene glycol and $CaCl_2$. Transformants could grow on MMM showing mitotical stability. Southern hybridization analysis of DNA isolated from transformants showed that the Neurospora pyr 4 gene and vector sequence might be integrated into the P. florida chromosomes. As the transformants were monokaryon, each transformant was mated with the other monokaryon. Fruitbody shape of untransformant was eroded type but those of transformants were eroded type, funnel type, plane type and ungrowing cap type.

Design and Implementation of the Security System for the Moving Object Detection (이동물체 검출을 위한 보안 시스템의 설계 및 구현)

  • In this paper, we propose a segmentation algorithm that can reliably separate moving objects from noisy background in the image sequence received from a camera at the fixed position. Image segmentation is one of the most difficult process in image processing and an adoption in the change of environment must be considered for the increase in the accuracy of the image. The proposed algorithm consists of four process : generation of the difference image between the input image and the reference image, removes the background noise using the background nois modeling to a difference image histogram, then selects the candidate initial region using local maxima to the difference image, and gradually expanding the connected regions, region by region, using the shape information. The test results show that the proposed algorithm can detect moving objects like intruders very effectively in the noisy environment.

  • The south and southwestern parts of Jeonra-namdo has been known as an alunite province in Korea. The alunite deposits investigated for the present study are Okmaisan, Seongsam, Bugog, Gasado south, Gasado north, Jangsando, Dogcheon and Jungyongri deposits. The main purpose of this study is to depict the genetical origin of the alunite deposits. The rocks distributed in the areas mentioned above consist chiefly of rhyolitic tuff, breccia tuff and andesitic tuff of Cretaceous age which represent different episodes of volcanic activities during Cretaceous epoch. The attitude of bedding of the tuffaceous rocks varies from place to place but generally dips very gently. The alunite deposits are embedded mostly in the rhyolitic tuff so that they appear as layered deposits, this occurrence may be the result of stratigraphic and lithologic controls. The result of this study can be summarized as below. The mineral sequence studied by the mineral paragenesis and the result of the spectrograph anlyses is such that (1) alunite was formed at first and pyrophyllite was nearly contemporaneous with alunite but pyrophyllite formation can be recognized as a secondary mineralization products, (2) kaoline was succeeded to form later and hematite finally deposited, and (3) pyrite was deposited from the begining to the end of the above mineralization period. The compositional change of host rocks is such that CaO, $SiO_2$ and $Na_2O$ were largely removed from the parent rocks and some $Al_2O_3$ and $SO_3$ were transported by the solution so as to enrich the rocks. The sequencial process of such mineralization has resulted in forming those distinguish mineral zones; alunite, kaoline, pyrophyllite, silicifide and sulphide zone which manifest irregular shape. These deposits were formed by hydrothermal solution which was possibly low temperature and contained sulphuric acid originated from $H_2S$ and $SO_2$ gases.

First Report of Green Mold Disease Caused by Trichoderma hengshanicum on Ganoderma lingzhi

  • Ganoderma lingzhi is a well-known source of natural fungal medicines which has been given for the treatment of several diseases. China is one of the major commercial producers of Ganoderma mushroom worldwide. However, with the expansion of the commercial cultivation, the occurrence of the fungal diseases on G. lingzhi has also been increased. The green mold disease symptoms were observed in the cultivation base of G. lingzhi in Zuojia Town, Jilin City, Jilin Province, China, causing the basidiomes to be rotten and withered, and the green mycelium layer generated gradually. The pathogenicity tests showed the same symptoms as appeared naturally in Zuojia mushroom base. Morphology characters revealed conidia green, ellipsoid, globose, 2.56-4.83 × 2.09-4.22 ㎛, length-width ratio was 1.1-1.2 (n = 10). Conidiophores trichoderma-like, often asymmetry, branches solitary, paired or in whorls of 3 phialides formed solitary, paired or in whorl, variable in shape, lageniform, sometimes ampulliform or subulate. While using molecular methodology, comparing with the sequences of Trichoderma hengshanicum from GenBank, the analyzed sequence showed 97.32% homology with the RPB2 sequences, 100% with the TEF1-a sequences. A fungus isolated from the diseased tissues was identified based on morphology and molecular studies as T. hengshanicum. This is the first report of T. hengshanicum causing the green mold disease of G. lingzhi in China.

Sclerotium Rot of Sponge Gourd Caused by Sclerotium rolfsii (Sclerotium rolfsii에 의한 수세미오이 흰비단병)

  • Sclerotium rot of sponge gourd occurred at the experimental field of Gyeongsangnam-do Agricultural Research and Extension Services in August 2010. The infected fruits showed water-soaked and rot symptoms. White mycelial mats spread over lesions, and then sclerotia were formed on fruit and near soil line. The sclerotia were globoid in shape, 1-3 mm in size and white to brown in color. The optimum temperature for mycelial growth and sclerotia formation on PDA was $30^{\circ}C$ and the hyphal width was 4-8 ${\mu}m$. The typical clamp connections were observed in the hyphae of the fungus grown on PDA. On the basis of mycological characteristics, ITS rDNA sequence analysis, and pathogenicity to host plants, this fungus was identified as Sclerotium rolfsii Saccardo. This is the first report of sclerotium rot on sponge gourd caused by S. rolfsii in Korea.
