Novel splice isoforms of pig myoneurin and their diverse mRNA expression patterns

  • Objective: The aim of this study was to clone alternative splicing isoforms of pig myoneurin (MYNN), predict the structure and function of coding protein, and study temporal and spatial expression characteristics of each transcript. Methods: Alternative splice isoforms of MYNN were identified using RNA sequencing (RNA-seq) and cloning techniques. Quantitative real-time polymerase chain reaction (qPCR) was employed to detect expression patterns in 11 tissues of Large White (LW) and Mashen (MS) pigs, and to study developmental expression patterns in cerebellum (CE), stomach (ST), and longissimus dorsi (LD). Results: The results showed that MYNN had two alternatively spliced isoforms, MYNN-1 (GenBank accession number: KY470829) and MYNN-2 (GenBank accession number: KY670835). MYNN-1 coding sequence (CDS) is composed of 1,830 bp encoding 609 AA, whereas MYNN-2 CDS is composed of 1,746 bp encoding 581 AA. MYNN-2 was 84 bp less than MYNN-1 and lacked the sixth exon. MYNN-2 was found to have one $C_2H_2$ type zinc finger protein domain less than MYNN-1. Two variants were ubiquitously expressed in all pig tissues, and there were significant differences in expression of different tissues (p<0.05; p<0.01). The expression of MYNN-1 was significantly higher than that of MYNN-2 in almost tissues (p<0.05; p<0.01), which testified that MYNN-1 is the main variant. The expression of two isoforms decreased gradually with increase of age in ST and CE of MS pig, whereas increased gradually in LW pig. In LD, the expression of two isoforms increased first and then decreased with increase of age in MS pig, and decreased gradually in LW pig. Conclusion: Two transcripts of pig MYNN were successfully cloned and MYNN-1 was main variant. MYNN was highly expressed in ST, CE, and LD, and their expression was regular. We speculated that MYNN plays important roles in digestion/absorption and skeletal muscle growth, whereas the specific mechanisms require further elucidation.

Some Motifs Were Important for Myostatin Transcriptional Regulation in Sheep (Ovis aries)

  • Many motifs along the 1.2 kb myostatin promoter (MSTNpro) in sheep have been found by the MatInspecter program in our recent study. To further verify the role of the motifs and better understand the transcriptional regulation mechanism of the myostatin gene in sheep, the reporter gene EGFP (enhanced green fluorescent protein) was selected and the wild-type (W) vector MSTNPro$^W$-EGFP or motif-mutational (M) vector MSTNPro$^M$-EGFP were constructed. The transcriptional regulation activities were analyzed by detecting the fluorescence strength of EGFP in C2C12 myoblasts transfected with the vectors. The results showed that E-box (E) 3, E4, E5 and E7, particularly E3, E5 and E7, had important effects on the activity of the 1.2 kb sheep myostatin promoter. In addition, we also detected several other important motifs such as MTBF (muscle-specific Mt binding factor), MEF2 (myocyte enhancer factor 2), GRE (glucocorticoid response elements) and PRE (progesterone response elements) along the sheep myostatin promoter by the mutational analysis.

The Commencement Period of the Korean Type Bronze Dagger Culture, Seen from the Condition of the Section Polishing Technique - Through the Chronology of Chinese Data - (구분마연 기술로 본 한국식동검문화의 개시 연대 - 중국 자료의 편년을 통하여 -)

  • The purpose of this study is to grasp the commencement date of Korean-type bronze dagger through the chronology of Chinese data. It focuses on the fact that the same section polishing technique appears both in Korean type bronze dagger and Dongzhou type bronze dagger. Dongzhou type bronze dagger in Anqiu Shandong, in which A1 type section polishing technique is observed, was said to have been collected remains in 1958, but the clear excavation cannot be identified. Therefore, this study presents Tomb No.1 Zuojiawa Jinan, Dongzhou type bronze dagger, and associated products. As associated products, bronze weapon and bronze ware were excavated, whose periods are estimated to be in the Spring and Autumn period, the transition period of Warring States, and the former part of the China's Warring States. Accordingly, the Korean bronze dagger, excavated in the remains of the Han Peninsula appears to have run parallel with the Dongzhou type bronze dagger of the A1 type section polishing technique, excavated in China for a fixed period. In addition, the chronology of Tomb No. 61MI grave in Wanrongmiaoqian, Shanxi is estimated to range from the former part to the middle part of the China's Warring States, which is identified to be connected to the A1 type section polishing technique. Examining the data of the relative date, we can find out that the Commencement Period of the Korean type bronze dagger Culture is seen to be the transition period and the former part of the China's Warring States, which is estimated to be the 5th and 4th centuries BC. This chronology is followed by Tomb No.6512 Zhengjiawazi Shenyang, recorded as the 6th century B.C. which reveals that Liaoning type bronze dagger culture and Korean-typed Bronze Dagger Culture are naturally connected. Furthermore, the A1 type section polishing technique was distributed in the southwestern area of the Korean peninsula and Shandong, China, while the A2 type section polishing technique was distributed in the southern area of the Korean peninsula, Shanxi-Province in China, and Northern Kyushu region in Japan. Seen from the weapon-shaped bronze ware of the section polishing technique, Shanxi area(Central Plains area), China. the southwestern area of the Korean peninsula (northwest area), and Northern Kyushu region in Japan are set up as one traffic road(spreading route). This demonstrates that the section polishing technique emerged around the Han Peninsular, spreading the technique regionally.
