The continuous production of citric acid from dairy wastewater was investigated using calcium-alginate immobilized Asrergillus niger ATCC 9142. The citric acid productivity and yield were strongly affected by the culture conditions. The optimal pH, temperature, and dilution rate were 3.0, 30$^{\circ}C$, and 0.025 h$\^$-1/, respectively. Under optimal culture conditions, the maximum productivity, concentration, and yield of citric acid produced by the calcium-alginate immobilized Aspergillus niger were 160 mg L$\^$-1/ h$\^$-1/, 4.5 g/L, and 70.3%, respectively, The culture was continuously perfored for 20 days without any apparent loss in citric acid productivity. Conversely, under the same conditions with a batch shake-flask culture, the maximum productivity, citric acid concentration, and yield were only 63.3 mg L$\^$-1/h$\^$-1/, 4.7 g/L and 51.4%, respectively, Therefore, the results suggest that the bioreactor used in this study could be potentially used for continuous citric acid production from dairy wastewater by applying calcium-alginate immobilized Aspergillus niger.
The production of itaconic acid by Aspergillus terreus NRRL 1960 was studied. The optimal culture conditional such as pH, inoculum size and medium composition were established. Maximum production of itaconic acid, $19.18g/\ell$, was obtained when the cultivation was carried out at $37^{\circ}C$ and pH 2.5 for 7days, with medium containing 5%(w/v) glucose, 0.5%(w/v) NH4Cl, 0.2%(w/v) yeast extract 0.1%(w/v) CaC12, 0.1%(w/v) MgSO4 and 0.2%(w/v) NaCl. A proper medium for inoculum culture was found to be 2%(w/v) malt extract. The batch production of itaconic acid with free cells in a stirredtank reactor was not efficient compared to the shake-flask culture.
Microbial desulfurization characterlstics of a bituminous coal have been determined by using Thiobacillus ferrooxidans. The effects of process variables (such as coal pulp density, particle size and addition of surfactants) on pyrite removal have been investigated in both shake and airlift-bioreactor culture experiments. In shake experiments, pyrite could be removed over 78% for pulp densifies below 20% (w/v) and removed below 40% for pulp densities over 30% (w/v) in 8 days. Pyrite removal decreased with increasing pulp densities, and it also decreased sharply with increasing particle sizes. In airlift bioreactor experiments, pyrite at 50% (w/v) pulp density could be removed about 50%. Its value is much higher than 15% at the same pulp density in a shake experiment. With addition of surfactants, pyrite removal was enhanced in shake experiments significantly, whereas it was slightly decreased in an airlift bioreactor experiment.
Kojic acid production by Aspergillus flavus strain S44-1 using sucrose as a carbon source was carried out in a 250-mL shake flask and a 2-L stirred tank fermenter. For comparison, production of kojic acid using glucose, fructose and its mixture was also carried out. Kojic acid production in shake flask fermentation was 25.8 g/L using glucose as the sole carbon source, 23.6 g/L with sucrose, and 6.4 g/L from fructose. Reduced kojic acid production (13.5 g/L) was observed when a combination of glucose and fructose was used as a carbon source. The highest production of kojic acid (40.2 g/L) was obtained from 150 g/L sucrose in a 2 L fermenter, while the lowest kojic acid production (10.3 g/L) was seen in fermentation using fructose as the sole carbon source. The experimental data from batch fermentation and resuspended cell system was analysed in order to form the basis for a kinetic model of the process. An unstructured model based on logistic and Luedeking-Piret equations was found suitable to describe the growth, substrate consumption, and efficiency of kojic acid production by A. flavus in batch fermentation using sucrose. From this model, it was found that kojic acid production by A. flavus was not a growth-associated process. Fermentation without pH control (from an initial culture pH of 3.0) showed higher kojic acid production than single-phase pH-controlled fermentation (pH 2.5, 2.75, and 3.0).
A. niger IMI 41873 was shake-cultured in Cassava starch medium, then mycelial dry weight and mycelial protein were measured. The effects of Cassava starch concentrations, various nitrogen sources and concentrations on the levels of mycelial production and mycelial protein were investigated. The results were as follows; In each medium containg 4%, 6%, 8% and 10% of Cassava starch, mycelial production was 13.35 mg/ml, the medium containg 6% of Cassava starch was proved to be most effective. The levels of mycelial protein in shake culture with medium of 6% Cassava starch and $NaNO_3(1.5\;g/l)$ were the highest. Among nitrate, ammonium sulfate, and urea, the nitrate was the most effective on mycelial production.
Immobilized cells of Aspergillus niger was employed to produce citric acid by fermentation of milk factory waste water. A. niger ATCC 9142 as a citric acid production strain was cultured for 3 days and was entrapped with Ca-alginate bead about 2.5∼3.5 mm. The optimal pH and temperature were estimated to be 3.0 and $30^{\circ}C$, respectively. Dilution rate for fermentation was calculated to be $0.025 h^{-1}$ . Maximum amount of citric acid was obtained at 4.5 g/$\ell$ with the optimized fermentation condition. The yield of citric acid produced by immobilized A. niger ATCC 9143 was 70.3%. The yield was increased by 20% with immobilized cell, compared to that of the shake flask culture. Hence, the milk factory waste water is worthy to be used for the substrate of citric acid fermentation.
This study was concerned with the optimization of liquid culture conditions for mycelial growth and polysaccharide production and its physicochemical properties in Cordyceps militaris. The one factor at a time method was adopted to investigate the effects of medium composition, environmental factors and C/N ratio. Among the these varialbles, glucose 80g/L, yeast extract 10g/L, $MgSO_4{\cdot}7H_{2}O\;0.5g/L$, $KH_{2}PO_4\;0.5g/L$ were proved to be the most suitable carbon, nitrogen, and mineral sources, respectively. The optimal temperature, initial pH, working volume were identified to be $24^{\circ}C$, 7.0 and 100ml, respectively. Under the optimal conditions, the strategies in shake flask culture and 5L jar fermentor led to mycelial growth of 29.43 g/L, 28.88g/L and polysaccharide production of 2.53g/L, 6.38 g/L, respectively. Among the phisicochemical properties, relative concentrations(w/v) of total sugar, uronic acid, protein and hexoseamine were identified to be 74.07%, 1.13%, 0.91%, and 0.46%, respectively. The fraction of neutral and acidic polysaccharide were identified to be 81.9% and 18.1%, respectively.
The Pseudomonas sp. strain NGK1 (NCIM 5120) was immobilized in calcium alginate, agar, and polyacrylamide gel matrices. The salicylic acid-producing capacity of freely suspended cells was compared with immobilized cells in batches with a shake culture and continuous culture system in a packed bed reactor. Freely suspended cells ($4\times10^{10}cfu/ml$) produced 12 mM of salicylic acid, whereas cells immobilized in calcium alginate ($1.8\times10^{11}$cfu/g beads), agar ($1.8\times10^{11}$cfu/g beads), and polyacrylamide ($1.6\times10^{11}$cfu/g beads) produced 15, 11, and 16mM of salicylic acid, respectively, from naphthalene at an initial concentration of 25 mM. The continuous production of salicylic acid from naphthalene was investigated in a continuous packed bed reactor with two different cell populations. The longevity of the salicylic acid-producing activity of the immobilized cells from naphthalene was also studied in semi continuous fermentations. The immobilized cells could be reused 18, 13, and more than 20 times without losing salicylic acid-producing activity in calcium alginate-,agar-, and polyacrylamide-entrapped cells, respectively. The study reveals a more efficient utilization of naphthalene and salicylic acid production by the immobilized Pseudomonas sp. strain NGK1 as compared to the free cells.
A fungal strain capable of producing extracellular cellulase was isolated from farmland. It was identified as Aspergillus niger, and named Aspergillus niger KKS. Production of cellulase and xylanase by the A. niger KKS was studied through a shake-flask culture. The effects of culture conditions such as inoculum size, temperature, pH, and medium composition on the cellulase and xylanase production were examined. The optimum temperature and pH for the enzyme production were $30^{\circ}C$ and pH 7.0, respectively. The optimized medium was composed of 2.0% (w/v) rice straw, 0.5% (w/v) proteose peptone, 0.5% (w/v) $KH_2 PO_4$, 0.05% (w/v) yeast extract, 0.01% (w/v) $CoSO_4 \cdot 7H_2O$, and 0.05% (w/v) $CuSO$_4$\cdot 5H_2O$. When the strain was incubated with the optimized medium, it gave the activities of endoglucanase, $\beta$-glucosidase, $\beta$-xylosidase, xylanase were 3.80, 4.20, 4.00, 80.0 (IU/mL), respectively. Filter paper and cotton activities were 0.68 and 0.045 (IU/mL), respectively. The results of this study show that A. niger KKS is a potential organism with a wide spectrum of enzyme activities, such as those of $\beta$-glucosidase, $\beta$-xylosidase, and xylanase.

The effect of on the cell growth, the elaboration of pullulan, the morphology and were the effect of on the molecular weight of pullulan were investigated. A. pullulans showed maximum pullulan production when initial pH 6.5 was 11.98 g/l in shake-flask culture. In batch culture, the maximum pullulan production of 15.16 g/l was obtained at an aeration rate of 0.5 vvm. The mixture of yeast-like form and mycelial form of cells was found at the constant pH 4.5, at which condition, the elaboration of pullulan was high, about 13.31 g/l. However, pullulan with its higher molecular weight (>1,000,000) was produced at the constant pH 6.5.
