Porcine epidemic diarrhea virus: an update overview of virus epidemiology, vaccines, and control strategies in South Korea

  • Porcine epidemic diarrhea virus (PEDV) has posed significant financial threats to the domestic pig industry over the last three decades in South Korea. PEDV infection will mostly result in endemic persistence in the affected farrow-to-finish (FTF) herds, leading to endemic porcine epidemic diarrhea (PED) followed by year-round recurrent outbreaks. This review aims to encourage collaboration among swine producers, veterinarians, and researchers to offer answers that strengthen our understanding of PEDV in efforts to prevent and control endemic PED and to prepare for the next epidemics or pandemics. We found that collaboratively implementing a PED risk assessment and customized four-pillar-based control measures is vital to interrupt the chain of endemic PED in affected herds: the former can identify on-farm risk factors while the latter aims to compensate for or improve weaknesses via herd immunity stabilization and virus elimination. Under endemic PED, long-term virus survival in slurry and asymptomatically infected gilts ("Trojan Pigs") that can transmit the virus to farrowing houses are key challenges for PEDV eradication in FTF farms and highlight the necessity for active monitoring and surveillance of the virus in herds and their environments. This paper underlines the current knowledge of molecular epidemiology and commercially available vaccines, as well as the risk assessment and customized strategies to control PEDV. The intervention measures for stabilizing herd immunity and eliminating virus circulation may be the cornerstone of establishing regional or national PED eradication programs.

  • Software compliance is an essential process when Open Source Software is included in software development to avoid such as license violation issue. However, analyzing quite big software which involves many developers requires enormous time and hard difficulty. To resolve these kinds of problem, SPDX formalizes and standardize the metadata about the software package. When the use of SPDX is activated, software package analysis would be simple and could contribute fair Open Source Software distribution. In this paper, we develop blockchain based SPDX distribution platform which fulfills the requirement of SPDX lifecycle to provide SPDX database which does not depend on particular centralized service but serve as distributed ledger and control by user's certification and their purpose. Moreover, to contribute invigoration of blockchain based SPDX distribution platform, we develop SPDX document generation plug-in for integrated development environment such as Visual Studio.

Identification of heat shock protein70-2 and protamine-1 mRNA, proteins, and analyses of their association with fertility using frozen-thawed sperm in Madura bulls

  • Objective: This study aims to identify heat shock protein70-2 (HSP70-2) and protamine-1 (PRM1) mRNA and protein in Madura bull sperm and demonstrate their relation as bull fertility biomarkers. Methods: The Madura bull fertility rates were grouped based on the percentage of first service conception rate (%FSCR) as high fertility (HF) (79.04%; n = 4), and low fertility (LF) (65.84%; n = 4). mRNA of HSP70-2 and PRM1 with peptidylprolyl isomerase A (PPIA) as a housekeeping gene were determined by quantitative real-time polymerase chain reaction, while enzyme-linked immunoassay was used to measure protein abundance. In the post-thawed semen samples, sperm motility, viability, acrosome integrity, and sperm DNA fragmentation index were analyzed. Data analysis was performed on the measured parameters of semen quality, relative mRNA expression, and protein abundance of HSP70-2 and PRM1, among the bulls with various fertility levels (HF and LF) in a one-way analysis of variance analysis. The Pearson correlation was used to analyze the relationship between semen quality, mRNA, proteins, and fertility rate. Results: Relative mRNA expression and protein abundance of HSP70-2 and PRM1 were detected and were found to be highly expressed in bulls with HF (p<0.05) and were associated with several parameters of semen quality. Conclusion: HSP70-2 and PRM1 mRNA and protein molecules have great potential to serve as molecular markers for determining bull fertility.

Visual detection of porcine circovirus 2 by loop-mediated isothermal amplification (LAMP) with hydroxynaphthol blue dye (육안 판독 등온증폭법을 이용한 돼지 써코바이러스 2형 신속 진단법)

  • In this study, we developed a loop-mediated isothermal amplification (LAMP) with hydroxynaphtol blue dye (HNB) for rapid and direct visual detection of porcine circovirus 2 DNA with high sensitivity and specificity. The LAMP was completed in 40 min at $63^{\circ}C$, and the results of the LAMP can be confirmed by naked eye without any detection process. The sensitivity of the LAMP was 10-fold higher than that of the commercial PCR (cPCR) and real time PCR (rPCR) previously reported. In clinical application, the PCV2 detection rate of the LAMP was the same on porcine tissue samples (75.0%, 36/48) between porcine blood samples (75.0%, 39/52). The PCV2 detection rate (75.0%) of LAMP was higher than those of the cPCR and rPCR (67.3%, 35/52) in blood samples. In conclusion, the LAMP developed in the study could be an useful alternative method for the detection of PCV2 in the swine disease diagnostic laboratories.

QuEChERS-based determination of tissue residues and acute toxicity of pyraclofos in rat (QuEChERS 법을 이용한 Rat 조직내 Pyraclofos 잔류 분석 및 급성독성 평가)

  • Environmental pesticides used for insect control can be transferred from plants to animals even to livestock animals through food chain. Human beings also can be exposed to pesticides by consuming polluted dairy products, including meats, eggs and other milk products. Therefore, the Ministry of Food and Drug Safety (MFDS) established Standard for Pesticide Residue Limits in dairy products. The QuEChERS (quick, easy, cheap, effective, rugged and safe) methods for detecting residual pesticides are relatively well established for fruits and vegetables, however, the methods for meat have not been appropriately studied yet. In the present work, pyraclofos was used as an organophosphate pesticide to examine its tissue residue in experimental animals by QuEChERS methods. For this, pyraclofos (150 mg/kg body weight) was orally administered to male rats once a day for 2 days. After 6, 12, and 24 hr of the treatment, the tissue residues in liver and femoral muscle of the rats were determined using QuEChERS methods followed by HPLC analyses. In preliminary studies, the recovery rates of spiking samples of pyraclofos demonstrated approximately 109~110% from the tissues. In previous study, pyraclofos tissue residues were observed with significantly high levels in livers and muscles at 6 hr of oral treatment. Then, they were almost completely disappeared after 24 hr of the administration, indicating the orally exposed pyraclofos is rapidly absorbed and distributed to body organs, then quickly excreted from the body with a negligible level of tissue residue. The alterations in blood chemistry as well as the histopathology of heart, lung, liver, spleen and kidney have also been investigated in the experimental animals for assessing acute toxic effects of pyraclofos. The obtained blood chemistry indexes (ALT and AST) showed maximum peak values at 12 hr after the oral administration and decreased to the normal levels at 24 hr of the treatment. Histopathologic observation exhibited acute hepatic damages at 24 hr of the treatment. In conclusion, we suggest that QuEChERS method can be adequately optimized for the analysis of pyraclofos residues in animal tissues.

Detection of etiologic agents in diarrhea fecal samples from calves in Gyeongnam province, Korea (경남지역에서 송아지 설사병 병원체 검출 조사)

  • The objective of this study was to determine the infection patterns of etiological agents causing calf diarrhea in the Gyeongnam province, Korea. In this study, from January 2011 to December 2011, feces and necropsy specimens from 249 calves diagnosed with diarrhea (<7 months old) were examined by reverse transcriptase-polymerase chain reaction assay and bacteria & coccidium isolation for detection pathogenic organism. The results of this study showed that 78 cases (31.3%) in spring, 71 cases (28.5) in summer, 62 cases (24.9%) in fall and 38 cases (15.3%) in winter were diagnosed with calf diarrhea, respectively. Calf diarrhea-causing pathogens were diagnosed as bacteria 113 (45.4%), viruses 97 (39.0%), coccidium 1 (0.4%), unknown cases 13 (5.2%), and mixed infections 25 (10.0%). We isolated three virus types from fecal samples (97), which were classified as BVD 64 (66.0%), BRV 21 (21.6%), and BCV 12 (12.4%). Moreover, co-infected pathogens were 25 cases, consisting with BVD & BRV 11 (44%), BVD & BCV& BRV 7 (28.0%), E. coli & BCV 3 (12%), and BVD & IBR 1 (4.0%). In summary, we demonstrated that the enteropathogens of bacteria, viruses, and parasite were detected in samples from cattle with diarrhea, principally in young calves less than 7 months of age. Future studies of infectious diarrhea in cattle should include assays for this etiologic agent.

Polymorphism in the intron 20 of porcine O-linked N-acetylglucosamine transferase

  • Objective: O-linked N-acetylglucosamine (O-GlcNAc) transferase (OGT) catalyzes the addition of O-GlcNAc and GlcNAcylation has extensive crosstalk with phosphorylation to regulate signaling and transcription. Pig OGT is located near the region of chromosome X that affects follicle stimulating hormone level and testes size. The objective of this study was to find the variations of OGT between European and Chinese pigs. Methods: Pigs were tested initially for polymorphism in OGT among European and Chinese pigs by polymerase chain reaction and sequencing at the U.S. Meat Animal Research Center (USMARC). The polymorphism was also determined in an independent population of pigs including European and Chinese Meishan (ME) breeds at the National Institute of Animal Science (NIAS, RDA, Korea). Results: The intron 20 of OGT from European and Chinese pigs was 514 and 233 bp, respectively, in the pigs tested initially. They included 1 White composite (WC) boar and 7 sows ($2Minzu{\times}WC$, $2Duroc\;[DU]{\times}WC$, $2ME{\times}WC$, $1Fengzing{\times}WC$) at USMARC. The 281-bp difference was due to an inserted 276-bp element and GACTT in European pigs. When additional WC and ME boars, the grandparents that were used to generate the $1/2ME{\times}1/2WC$ parents, and the 84 boars of 16 litters from mating of $1/2ME{\times}1/2WC$ parents were analyzed, the breeds of origin of X chromosome quantitative trait locus (QTL) were confirmed. The polymorphism was determined in an independent population of pigs including DU, Landrace, Yorkshire, and ME breeds at NIAS. OGT was placed at position 67 cM on the chromosome X of the USMARC swine linkage map. Conclusion: There was complete concordance with the insertion in European pigs at USMARC and NIAS. This polymorphism could be a useful marker to identify the breed of origin of X chromosome QTL in pigs produced by crossbreeding Chinese and European pigs.

The Study on the Economic Appraisal of Fishing Port Investments (어항투자사업의 경제성 평가에 관한 연구)

  • From the economic point of view the fishing port is the complex of installations on land, organized to serve the fishing fleet and its cargo, and is the main link in the production chain of all components of the fishing industry, with the aim of achieving the planned targets with the minimum cost. Fishing port investment decisions have had significant impact on the development aims of Korean fisheries. Fishing port investments in Korea are made mostly by public or semipublic port authorities. Such investments should be judged not purely on the basis of financial profitability but rather on the extent to which they serve the development aims of the fishing industry. This makes the economic appraisal process more complex and presents certain problems in correctly quantifying the economic costs and benefits of the fishing port projects. This study concentrates more on the theoretical economic appraisal models than on the purely financial aspects of fishing port investments and points out the difference between the two approaches. In the result, there is clearly an element of judgment as to whether or not a shadow price needs to be used in estimating economic benefits and costs. From this viewpoint, some attempts are made to provide definitions of the possible economic benefits and costs, and methods for estimating and evaluating them in Part III and IV. Especially queueing theory is applied in the calculation of economic benefits. When a project is contemplated and analysis shows it to Lave a positive NPV, one question that arises is whether it should be implemented now or delayed. In this paper, the first year rate of return method is regarded as a more concise way of solving the timing of investment, At the end of Part IV, risk analysis of fishing port investments is considered. It can be handled in a number of ways, ranging from informal judgment to complex statistical analyses involving large-scale computer models, This paper recommends that evaluators of fishing port investments use the sensitivity analysis indicating exactly how much NPV will change in response to a given change in an input variable, other things held constant. Decisions regarding the amount of capacity to provide must be made in fishing port investments. Providing too much service would involve excessive capital costs. On the other hand, not providing enough service capacity would cause the waiting line of fishing vessels to become excessively long at times. Therefore, in Part V, the optimal number of berths and berth productivity in fishing port are defined as follows: Minimize E(TC) = E(WC)+E(SC) The minimum of this function is the solution and that is the optimal number of berth and berth productivity in fishing port.

  • External costs emerge as a consequence of negative external effects. The EU commission and national government in Europe made themselves the internalization of external costs and began to make various measures. The traffic sector plays an important role. At the same time, it affects as financial loads to logistics service providers and carriers in Europe. This study examines current development in european logistic market conditions. It describes the reaction of different market players and stakeholders in the logistic value chain in EU, Switzerland and Germany. A portfolio of different strategic answers to the changing market environment is being developed. Therefore, different options for the internalization of external costs are proposed. This study finally derives propositions for future research.

  • Last-Mile delivery optimization plays a key role in the urban supply chain operation, which is the most expensive and time-consuming and most complicated part of the whole delivery process. The urban consolidation center (UCC) is regarded as a significant asset for supporting customer demand in the last-mile delivery service. It is the key benefit of UCC to improve the load balance of vehicles and to reduce the total traveling distance by finding the better route with the well-organized multi-leg vehicle journey in the urban area. This paper presents the model using multiple scenario analysis integrated with mathematical optimization techniques using Geographic Information System (GIS). The model aims to find the best solution for the distribution network consisted of DC and UCC, which is applied to the case of Ulaanbaatar Mongolia. The proposed methodology integrates two sub-models, location-allocation model and vehicle routing problem. The multiple scenarios devised by selecting locations of UCC are compared considering the general performance and delivery patterns together. It has been adopted to make better decisions the quantitative metrics such as the economic value of capital cost, operating cost, and balance of using available resources. The result of this research may help the manager or public authorities who should design the distribution network for the last mile delivery service optimization using UCC within the urban area.
