Study on antimicrobial resistance of Escherichia coli isolated from domestic beef on sale (유통되는 쇠고기에서 분리한 대장균의 항생제 내성 조사.연구)

  • In this study, antimicrobial resistance of E coli isolated from domestic beef on sale in Busan and Gyeongnam province was investigated from March to October 2007. A total of 600 beef samples were collected for the monitoring of antimicrobial resistance, and 92 (15.3%) strains of E coli were isolated. Antimicrobial resistance test was carried out by agar disc diffusion method with 17 antimicrobials. In general, E coli isolates showed the highest antimicrobial resistance to doxycycline (73.9%), followed by tetracycline (70.7%) andcefazolin (63.0%). Then they showed higher resistance to several antimicrobials like norfloxacin (48.9%). However, They had low antimicrobial resistance to amikacin (4.3%), colistin (1.1%). Of 92 isolates, 82 (89.1%) were resistant to more than 2 antimicrobials. Among 17 antimicrobials examined, tetracyclines were the most resistant, followed by cephalosporins, quinolone. The resistance was seemed to be correlated to amounts of antimicrobial use. In the result of this study, we suggest that there be need to regulate the abuse of antimicrobial on food-producing animals in Korea because the concern on antimicrobial resistant is gradually increased worldwide.

Co-infection of Aspergillus spp. with Erysipelothrix rhusiopathiae in a red-crowned crane: a case report

  • This case describes outbreaks of acute aspergillosis in a red-crowned crane. A six-month-old, male, crane had showed clinical signs (i.e. anorexia, performance loss, ruffled feathers and drooped wings and open mouth breathing, etc.) before death. In necropsy examination, spherical to oval nodules disseminated from the respiratory tract to other organs. Those nodules were formed predominantly in air sacs, lung, peritoneum, serosa of esophagus and trachea. The nodules varied in size from 1 mm to over 1cm and the color was white to yellow. Microscopically, most of lung architecture were replaced by multiple foci which were characterized by well demarcated eosinophilic and karyorrhetic debris and surrounded by numerous Inflammatory cell. Most within necrotic center of the nodules, large numbers of fungal hyphae were present. Microbiology result indicated fungal growths on sabroud dextrose agar and bacterial growths on blood agar. Bacteria identified as E. rhusiopathiae using MALDI-TOF (microflex, BRUKER, USA) and fungi identified as A. fumigatus, A. terreus by sequencing the ITS1 and ITS4 regions. To confirm the route of infection, we checked the existence of the same pathogens in cohabitant (i.e. mother crane). The young age and weakened immunity (i.e. bacterial infection, etc.) causes fatal aspergillosis in birds.

Evaluation of SERS Nanoparticles to Detect Bacillus cereus and Bacillus thuringiensis

  • Purpose: This research evaluated five types of nanoparticles to develop a surface-enhanced Raman spectroscopy (SERS) method for the rapid detection of two Bacillus species (Bacillus cereus and Bacillus thuringiensis) that are commonly found on fresh produce, which can cause food poisoning. Methods: Bacterial concentrations were adjusted to a constant turbidity, and a total of $30{\mu}L$ of each Bacillus cell suspension was prepared for each nanoparticle. A point-scan Raman system with laser light source of wavelength 785 nm was used to obtain SERS data. Results: There was no qualitative difference in the SERS data of B. cereus and B. thuringiensis for any of the five nanoparticles. Three gold nanoparticles, stabilized in either citrate buffer or ethanol, showed subtle differences in Raman intensities of two Bacillus species at $877.7cm^{-1}$. Conclusions: Among the three types of nanoparticles, the gold nanoparticles stabilized in citrate buffer showed the lowest standard deviation, followed by gold nanoparticles stabilized in ethanol. This result supports the potential application of gold nanoparticles for SERS-based detection of B. cereus and B. thuringiensis.

  • The importance of service industries is growing as the portion of the service sector increases fast in the recent decades. This research deals with service prototyping and testing in a service laboratory. While products are generally tested through prototyping in new product development processes, services are difficult to test because of the characteristic of service, intangibility. A service laboratory, named s-Scape, is the experiment environment which has been developed to test services in virtual space for the purpose of analysis and improvement of real-world services such as hospitals and automobile show rooms. In this research, we present a service prototyping tool and language to support service test in the service laboratory. We first analyze key elements of service prototypes, and then design the service prototype diagram (SPD) and the service prototype description language (SPDL). SPD, which is a variant of the service blueprint, is a graphical tool to be used to generate SPDL. SPDL is an executable language of describing a service prototype of a real-world service in extensible markup language (XML) to experiment the service environment in virtual space. SPD reflects the control and interface of virtual reality devices, as well as key elements of service modeling. SPD represents a service process in which service providers and customers interact with each other in a service scape.

  • Porcine circovius type 2 (PCV 2) is a novel virus of Circoviridae familiy which is considering the cause of postweaning multisystemic wasting syndrome (PMWS). This study was carried out to investigate the prevalence of PCV 2 infection of swine in eastern areas of Gangwon province from February to June in 2005. Polymerase chain reaction (PCR) were conducted to identify the PCV 2 genome against 80 pigs. The number of infection and its rate of 4 areas, 8 farms and 80 pigs were 3 (75.0%), 7 (88.0%) and 44 (55.0%), respectively.

  • Fowl typhoid (FT) is a septicemic disease caused by Salmonella gallinarum. The purpose of this study was to investigate the antimicrobial resistance and pulsed -field gel electrophoresis (PFGE) patterns of S gallinarum isolated from broiler. During 1999 to 2004, there was isolated a total of 26 strains in liver and spleen. The biochemical characteristics of S gallinarum isolates was nonmotile, no production of $H_2S$, glucose gas, non-fermented rhamnose, indole-negative, fermentation of dulcitol, mannitol, maltose, and ornithine decarboxylase. At antimicrobial susceptibility, all of isolates were susceptible to amoxicillin/clavulanic acid, amikacin, neomycin, kanamycin, and cephalothin. Twenty-six isolates were divided into 19 resistant patterns and 6 strains was 8-multi-drug resistance. PFGE of Xba I restriction fragments of S gallinarum isolates was 22 patterns.

Examination of seroprevalence and detection of Avian pneumovirus from layer hens in Gyeongbuk province (경북지역 산란계에서 Avian pneumovirus에 대한 항체가 및 바이러스 검출 조사)

  • Avian pneumovirus(APV), also known as avian rhinotracheitis(ARTV), affects both turkeys and chickens and is known to be the primary causative agent of turkey rhinotracheitis(TRT). The aim of this study was to establish the presence or absence of antibodies to APV by an enzyme-linked immunosorbent assay(ELISA) and confirm APV by a reverse transcriptase-polymerase chain reaction(RT-PCR). The tested serum and feces were collected from laying hens in Gyeongbuk province. The positive farms with antibody against APV by ELISA were 90(96.7%) of 93 and positive serum samples were 433(93.1%) of 465 different sera. By regional group, sera from Uiseong, Cheongsong and Bonghwa were noted as 100% positive and positive rates of samples from Yeongju, Andong and Yeongyang were 93.3%, 85.7% and 50%, respectively. However, APV was not detected in feces samples by RT-PCR.

  • BSE (bovine spongiform encephalopathy)-like symptoms salivation, gait disorder, and the downer syndrome occurred to 8 heads of cattle at six cattle farms in Gyeonju, east eastern Gyeongsang province in 2013. Eight brain tissue samples using a vibration saw (Multimaster$^{(R)}$ FEIN, Germany) were collected referring to "The brain removal technique of transmisible spongiform encephalopathy sampling DVD" by Animal And Plant Quarantine Agency. Brain removal techinque using a vibration saw on the thick, solid frontal bone of cattle is more convenient for incision and removal than an ax for BSE testing.

  • This survey was carried out to determine 5 residual sulfonamides(sulfamerazine : SMR, sulfamethazine : SMT, sulfamonomethoxine : SMM:, sulfadimethoxine : SDM, sulfaquinoxaline : SQX) in muscle of mouse. For this investigation, pertaining detection wavelength, residual levels and residual times in muscle of mouse were summarized as follows ; 1. Pertaining detection wavelength of 5 residual sulfonamides(SMR, SMT, SMM, SDM, SQX) was 270nm by HPLC/UV detection. 2. After 1 day put a stop to sulfonamides administration, residual levels was 1∼l.5ppm, but were not detected at day 7 3. Withdrawal time of 5 sulfonamides were about 7 days suspectly.

A case of rape poisoning in a Hanwoo (한우 유채중독 증례)

  • This is a case report about rape poisoning in a Hanwoo. Non-protein nitrogen (NPN) compound in rape may cause poisoning in domestic animals. Rape poisoned cattle may show big and rapid breath with opening its mouth, edema and erosion of skin with itching, drooling, dyspnea and indigestion. Besides there can be shown neurological (blindness) and urological (red water) symptoms in some cases. There is no effective therapy for rape poisoning, therefore stopping the rape supplying is basically important. Clinical symptoms and history taking are very important factors in diagnosis of rape poisoning, due to the absence of diagnostic standard of judgment for rape poisoning. A Hanwoo 38 months old cow, in a farm located in Gyeoungbuk province showed typical rape poisoning symptoms such as breath with opening its mouth, drooling, dyspnea and hyperphotosensitivity, and dramatical recovery were occurred after stopping supplying of rape. The cow were diagnosed as the rape poisoning based on observation of clinical symptoms and history taking.
