카페인 섭취가 흰쥐의 혈청내 지질과 무기질 함량에 미치는 영향 (The Effects of Caffeine on Lipid and Mineral Content in the Serum of Rats)

  • This study was conducted to investigate the effects of caffeine on lipid and mineral content in the serum of rats given a caffeine free diet(FC), a low caffeine diet(LC), a medium caffeine diet(MC), a high caffeine diet(HC) or a super-high caffeine diet(SHC) for 5 weeks. Thirty male Sprague-Dawley rats(body weight, $110{\pm}0.3$ g) were blocked into 5 groups and fed diets with or without pure caffeine. Caffeine intake models showed a lower mean-weight gain, food intake and food efficiency in the high caffeine diet groups(MC, HC and SHC groups) than the groups receiving a caffeine free or low caffeine diet(FC and LC groups). Serum total lipid, total cholesterol and LDL-cholesterol levels decreased, but the serum HDL-cholesterol level increased according to the increase in caffeine intake. Serum total lipid, HDL-cholesterol and serum triglyceride were significantly lower in the HC and SHC groups than the FC group. All of the serum minerals decreased as caffeine intake increased. Serum iron, calcium, magnesium and phosphorus significantly decreased in the HC and SHC groups compared to the FC group. Caffeine intake was associated with less weight gain and reduced serum total cholesterol, triglyceride and total lipid. The results suggest that rats fed high amounts of caffeine may be susceptible to osteoporosis due to their low levels of calcium, magnesium and phosphorus.

Curcumin-Induced Autophagy Augments Its Antitumor Effect against A172 Human Glioblastoma Cells

  • Glioblastoma is the most aggressive common brain tumor in adults. Curcumin, from Curcuma longa, is an effective antitumor agent. Although the same proteins control both autophagy and cell death, the molecular connections between them are complicated and autophagy may promote or inhibit cell death. We investigated whether curcumin affects autophagy, which regulates curcumin-mediated tumor cell death in A172 human glioblastoma cells. When A172 cells were incubated with $10{\mu}M$ curcumin, autophagy increased in a time-dependent manner. Curcumin-induced cell death was reduced by co-incubation with the autophagy inhibitors 3-methyladenine (3-MA), hydroxychloroquine (HCQ), and LY294002. Curcumin-induced cell death was also inhibited by co-incubation with rapamycin, an autophagy inducer. When cells were incubated under serum-deprived medium, LC3-II amount was increased but the basal level of cell viability was reduced, leading to the inhibition of curcumin-induced cell death. Cell death was decreased by inhibiting curcumin-induced autophagy using small interference RNA (siRNA) of Atg5 or Beclin1. Therefore, curcumin-mediated tumor cell death is promoted by curcumin-induced autophagy, but not by an increase in the basal level of autophagy in rapamycin-treated or serum-deprived conditions. This suggests that the antitumor effects of curcumin are influenced differently by curcumin-induced autophagy and the prerequisite basal level of autophagy in cancer cells.

Whitten 배양액내 인간양수의 첨가가 생쥐 수정란의 체외발달 및 체외신장에 미치는 영향 (Effects of Human Amniotic Fluid Supplemented to Whitten's Medium on Development and Outgrowth of Mouse Embryo)

  • 본 연구는 생쥐 초기배 및 부화후 체외신장에 미치는 인간 양수의 첨가효과를 조사하고자 실시하였다. 생쥐 수정란의 체외발달에 미치는 인간 양수의 적정농도를 확인하고자 기본배양액 (Whitten's medium)에 16주령의 양수 농도를 각각 달리하여 체외배양을 실시한 결과, 20%를 첨가한 구에서 가장 높은 배반포로의 발달율과 부화율을 나타내었으며, 그 이상의 농도에서는 배발달율이 저하되었다. 포유동물 수정란 체외배양시 가장 많이 사용되는 첨가제인 fetal calf serum (FCS)과 bovine serum albumin(BSA)을 첨가한 구화의 배발달 성적을 비교한 결과, 임신중기 양수를 20% 첨가한 구의 배반포로의 발달율 (92.8%)과 부화율 (75.7%)은 기본배양액에서 배양된 것보다 배반포로의 발달율 (82.8%)과 부화율 (31.3%)은 높았으나 0.3% BSA (90.5%, 70.8%)나 10% FCS 첨가구 (94.3%, 74.3%)와는 유의한 차이가 인정되지 않았다. 임신주령에 따른 양수의 첨가효과를 조사한 결과 20%의 임신말기 양수를 첨가한 구의 배반포로의 발달율 (71.9%)과 부화율 (57.3%)은 20% 임신중기 양수를 첨가한 구보다 낮았으며, 부화후 배의 체외신장은 임신중기 양수와 FCS이 첨가된 구에서는 유기되었으나, 임신말기 양수와 BSA가 첨가된 구에서는 배의 체외신장이 유기되지 않았다. 이상의 본 연구결과를 통해 임신중기 양수 내에는 배발달 촉진인자와 배의 체외신장을 유기시키는 물질이 함유되어 있어 포유동물 배의 체외배양에 상당한 영향을 미치고 있다고 사료된다.

BcI-2 Over-expression Reduced the Serum Dependency and Improved the Nutrient Metabolism in a NS0 Cells Culture

  • The over-expression of Bcl-2 has greatly improved the culture period, specific growth rate, and maximum viable cell density of NS0 cells culture under low serum condition. Further analysis of these data suggests that a saturation model of the Monod type can be used to represent the relationships of specific growth rate and initial serum concentration. The ${\mu}_{max}$ and $K_s$ for the Bcl-2 cell line is $0.927day^{-1}\;and\;0.947\%(v/v)$ respectively, which are $21\%$ greate and $7\%$ lower respectively than its control counterpart. Study on the amino acid supplementation revealed that Bcl-2 cell lines possess greater improvement in the specific growth rate and maximum viable cell density compared to the control cell lines. A further increase in the amino acid supplementation has resulted a $17\%$ decrease in specific growth rate and no improvement in maximum viable cell density in the control culture. However, the Bcl-2 cell line exhibited a better growth characteristic in this culture condition compared to that of control cell lines. The higher specific growth rate and maximum viable cell density of the Bcl-2 cell line in medium fortified with serum and MEM EM suggested a more efficient nutrient metabolism compared to that in the control cell line. The low serum and amino acid utilisation rate and the higher cell yield may prove to be important in the development of serum/protein free culture.

The Effect of Spent Medium Recycle on Cell Proliferation, Metabolism and Baculovirus Production by the Lepidopteran Se301 Cell Line Infected at Very Low MOI

  • The aim of this paper was to study the effect of spent medium recycle on Spodoptera exigua Se301 cell line proliferation, metabolism, and baculovirus production when grown in batch suspension cultures in Ex-Cell 420 serum-free medium. The results showed that the recycle of 20% of spent medium from a culture in mid-exponential growth phase improved growth relative to a control culture grown in fresh medium. Although both glucose and glutamine were still present at the end of the growth phase, glutamate was always completely exhausted. The pattern of the specific glucose and lactate consumption and production rates, as well as the specific glutamine and glutamate consumption rates, suggests a metabolic shift at spent medium recycle values of over 60%, with a decrease in the efficiency of glucose utilization and an increase in glutamate consumption to fuel energy metabolism. Baculovirus infection provoked a change in the metabolic pattern of Se301 cells, although a beneficial effect of spent medium recycle was also observed. Both growth rate and maximum viable cell density decreased relative to uninfected cultures. The efficiency of glucose utilization was dramatically reduced in those cultures containing the lowest percentages of spent medium, whereas glutamine and glutamate consumption was modulated, thereby suggesting that infected cells were devoted to virus replication, retaining their ability to incorporate the nutrients required to support viral replication. Recycle of 20% of spent medium increased baculovirus production by around 90%, thus showing the link between cell growth and baculovirus production.

Ginsenoside Rd alleviates mouse acute renal ischemia/reperfusion injury by modulating macrophage phenotype

  • Background: Ginsenoside Rd (GSRd), a main component of the root of Panax ginseng, exhibits anti-inflammation functions and decreases infarct size in many injuries and ischemia diseases such as focal cerebral ischemia. M1 Macrophages are regarded as one of the key inflammatory cells having functions for disease progression. Methods: To investigate the effect of GSRd on renal ischemia/reperfusion injury (IRI) and macrophage functional status, and their regulatory role on mouse polarized macrophages in vitro, GSRd (10-100 mg/kg) and vehicle were applied to mice 30 min before renal IRI modeling. Renal functions were reflected by blood serum creatinine and blood urea nitrogen level and histopathological examination. M1 polarized macrophages infiltration was identified by flow cytometry analysis and immunofluorescence staining with $CD11b^+$, $iNOS^+$/interleukin-12/tumor necrosis factor-${\alpha}$ labeling. For the in vitro study, GSRd ($10-100{\mu}g/mL$) and vehicle were added in the culture medium of M1 macrophages to assess their regulatory function on polarization phenotype. Results: In vivo data showed a protective role of GSRd at 50 mg/kg on Day 3. Serum level of serum creatinine and blood urea nitrogen significantly dropped compared with other groups. Reduced renal tissue damage and M1 macrophage infiltration showed on hematoxylin-eosin staining and flow cytometry and immunofluorescence staining confirmed this improvement. With GSRd administration, in vitro cultured M1 macrophages secreted less inflammatory cytokines such as interleukin-12 and tumor necrosis factor-${\alpha}$. Furthermore, macrophage polarization-related pancake-like morphology gradually changed along with increasing concentration of GSRd in the medium. Conclusion: These findings demonstrate that GSRd possess a protective function against renal ischemia/reperfusion injury via downregulating M1 macrophage polarization.

Growth, Clonability, and Radiation Resistance of Esophageal Carcinoma-derived Stem-like Cells

  • Objective: To separate/enrich tumor stem-like cells from the human esophageal carcinoma cell line OE-19 by using serum-free suspension culture and to identify their biological characteristics and radiation resistance. Methods: OE-19 cells were cultivated using adherent and suspension culture methods. The tumor stem-like phenotype of CD44 expression was detected using flow cytometry. We examined growth characteristics, cloning capacity in soft agar, and radiation resistance of 2 groups of cells. Results: Suspended cells in serum-free medium formed spheres that were enriched for CD44 expression. CD44 was expressed in 62.5% of suspended cells, but only in 11.7% of adherent cells. The suspended cells had greater capacity for proliferation and colony formation in soft agar than the adherent cells. When the suspended and adherent cells were irradiated at 5 Gy, 10 Gy, or 15 Gy, the proportion of CD44+ suspended cells strongly and weakly positive for CD44 was 77.8%, 66.5%, 57.5%; and 21.7%, 31.6%, 41.4%, respectively. In contrast, the proportion of CD44+ adherent cells strongly positive for CD44 was 18.9%, 14.%, and 9.95%, respectively. When the irradiation dose was increased to 30 Gy, the survival of the suspended and adherent cells was significantly reduced, and viable CD44+ cells were not detected. Conclusion: Suspended cell spheres generated from OE-19 esophageal carcinoma cells in serum-free stem medium are enriched in tumor stem-like cells. CD44 may be a marker for these cells.

Apoptosis and Apoptosis Related Gene Expression of Preimplantation Porcine Diploid Parthenotes Cultured in Different Protein Supplements

  • This study was conducted to determine effects of polyvinyl alcohol (PVA), fetal bovine serum (FBS) and bovine serum albumin (BSA) on blastocoel formation, cell number, apoptosis and apoptosis-related gene expression of porcine diploid parthenotes developing in vitro. Embryos were collected from 2-cell or late 4-cell diploid parthenotes that activated with electro pulse, and in vitro cultured in the NCSU 23 medium supplemented without or with 0.1% PVA, 10% FBS or 0.4% BSA for day 7. The morphological analysis of apoptosis in embryos was carried out using propidium iodide staining and terminal deoxynucleotidyl transferase mediated dUTP nick end labeling. The expressions of Bcl-xL, Bak and P53 in blastocyst stage parthenotes and in vivo-derived blastocysts were determined using semiquantitative RT-PCR. The addition of 0.4% BSA to the culture medium enhanced the development of 2- or late 4-cell stage parthenotes to the blastocysts stage (P < 0.01) while FBS decreased the incidence of blastocoel formation. FBS also reduced cell numbers of blastocysts developed from both 2- (P < 0.001) and late 4-cell (P < 0.05) embryos and increased percentage of apoptosis in the blastocysts (P < 0.001). The relative abundance of Bcl-xL mRNA in diploid parthenotes cultured from 2-cell stage in the presence of BSA is similar with that in in vivo derived embryos, but is significantly higher than in parthenotes cultured with FBS, PVA or none protein supplement control. Bak mRNA showed a significant increase at the blastocyst stage in FBS supplement medium. This result suggests that apoptosis related gene expression is significantly affected by protein supplements, which may result in alteration of apoptosis and embryo viability of porcine embryos developing in vitro.

인삼사포닌의 고 cholesterol 혈증 강하작용에 관한 연구 (Study on The Preventive Effect of Ginsenosides Against Hypercholesterolemia and Its Mechasnism)

  • The Preventive effect of the saponin fraction of Panax ginseng C.A. Meyer against hyperchole- sterolemia was demonstrated by assaying the cholesterol and triacylglyceride level of the blood serum and liver of rats fed high-cholesterol diet with and/or without ginsenoside. To understand the mechanism of the preventive action of ginsenoside, ginsenoside effect on LDL receptor binding ability, cholesterol level, and cAMP level of Chinese hamster ovary (CHO) cells cultured under various conditions were examined. When LDL (20 $\mu$g/ml) was added to the culture medium for CHO cell culture, LDL receptor binding activity of the cell was suppressed down to 58% of that of normal group. Ginsenosides at 10--2% and 10-3% level (w/v) were shown to exert an appreciable stimulatory effect on CHO cell LDL receptor activity, which partially overcame the suppression due to the presence of LDL (20 $\mu\textrm{g}$/ml) in the medium. Ginsenosides also reduced the increased cholesterol level of test group almost to that of normal group, and it increased cAMP level, which was usually reduced to 55% of that of the normal group due to the presence of LDL in the medium. Comparison of Kd and Bmax value of CHO cells cultured under different conditions revealed that this stimulation was due not to the receptor's binding affinity but to its number. Addition of ginsenoside (10-2%) decreased the uptake of taurocholic acid as much as 20% at the actively transporting everted ileal sacs, but it failed to form a large mixed micelles with taurocholic acid, which was one of the proposed mechanisms by which ginsenoside inhibits bile acid reabsorption. From the above results, it seemed likely that ginsenoside prevented hypercholestrolemia by decreasing cholesterol level in cells thereby relieving the inhibition of LDL receptor synthesis by cholesterol and also by inhibiting bile acid reabsorption from the small intestine.

Effect of Biphenyl Dimethyl Dicarboxylate on the Humoral Immunosuppression by Ketoconazole in Mice

  • The present study was undertaken to investigate the effect of biphenyl dimethyl idcarboxylate (PMC) on the humoral immunosuppression by ketoconazole (KCZ) in ICR mice. PMC at a dose of 6 mg/kg was administered orally to mice daily for 14 consecutive days. KCZ was suspended in RPMI 1640 medium and orally administered at 160 mg/kg/day 2 hrs after the administration of PMC. Mice were immunized an challenged with challenged red blood cells (SRBC). The results of the present study are summarized as follows; a gain of body weight and relative weights of spleen and liver were significantly increased by combination of PMC and KCZ, as compared with those in mice treated with KCZ alone. Splenic plaque forming cells (PFC) and hemagglutination (HA) titers to SRBC were greatly enhanced by the combination of PMC and KCZ, compared with treatment of KCZ alone. The elevation of serum glutamicpyruvic transminase (S-GPT) and total protein levels caused by KCZ were reduced to normal level by the combination of PMC and KCZ. In addition, lower serum albumin and A/G ratio were also increased to normal level. These findings indicate that PMC has a protective effect against KCZ-induced humoral immunosuppression.
