삼두해정탕이 사염화탄소(四鹽化炭素) 및 d-Galactosamine에 의(依)하여 유발(誘發)된 실험적(實驗的) 흰쥐의 간손상(肝損傷)에 미치는 영향(影響) (The effects of Samduhaejungtang on liver injury of rats induced by $CCl_4$ and d-galactosamine)

  • In order to investigate the protective effect of Samduhaejungtang on the liver injury of rats induced by $CCl_4$ and d-galactosamine, the serum transaminase(GOT& GPT), alkaline phosphatase(ALP), lactic dehydrogenase(LDH) for enzyme activities and triglyceride, total bilirubin amounts for serum component were measured. All animals were divided into 4 groups, those were normal group(untreated), control group(treated with vehicle 0.9% Saline solution), sample Ⅰ group(1000mg/kg administrated), sample Ⅱ group(2000mg/kg administrated). The results were as follows : 1. The inhibitory effects of the serum GOT activities in rats induced by $CCl_4$ were noted in both sample Ⅰ group(p<0.01) and sample Ⅱ group(p<0.001). In serum GPT activities, sample Ⅱ group(p<0.05) only showed the inhibitory effects. 2. The inhibitory effects of the serum ALP activities in rats induced by $CCl_4$ were noted in both sample Ⅰ group(p<0.01) and sample Ⅱ group(p<0.001). 3. The inhibitory effects of the serum LDH activities in rats induced by $CCl_4$ were noted in sample Ⅱ group(p<0.05). 4. The increases of the serum triglyceride content level in rats induced by $CCl_4$ were inhibited in both sample Ⅰ group(p<0.01) and sample Ⅱ group(p<0.001). 5. The increases of the serum total bilirubin content level in rats induced by $CCl_4$ were inhibited in both sample Ⅰ group(p<0.01) and sample Ⅱ group(p<0.001). 6. The inhibitory effects of the serum GOT& GPT activities in rats induced by d-galactosamine were noted in sample Ⅱ group(p<0.001), but sample Ⅰ group was not recongnized. 7. The significantly inhibitory effects of in the serum LDH activities in rats induced by d-galactosamine were note in sample Ⅱ group(p<0.05). 8. The increases of the serum triglyceride content level in rats induced by d-galactosamine were inhibited in sample Ⅱ group(p<0.01). 9. The increases of the serum total bilirubin content level in rats induced by d-galactosamine were inhibited in sample Ⅱ group(p<0.05). According to the above results, it is considered that Samduhaejungtang has protective effect against liver injury in rats induced by $CCl_4$ and d-galactosamine.

Influence of Long-term Supplementation with Korean Red Ginseng on in vivo Antioxidant Capacities in Rats

  • Effects of ginseng on in vivo antioxidant capacities with age were studied in rats. All rats were reared in the conventional system. Ginseng-treated rats were supplied with ginseng water extracts (25 mg/kg/day) continuously from 6 weeks of age to spontaneous death. None of the rats showed any discernible adverse effects of treatment with ginseng-containing water. There was no significant difference in body weight (BW) gains with age between treated and control groups. However, ginseng extracts did cause a decrease in the level of serum low density lipoprotein (LDL)-cholesterol, glucose, and thiobarbituric acid reactive substances (TBARS) in the treated rats. The activities of superoxide dismutase (SOD), catalase, and glutathione peroxidase in liver cytosol decreased with age in the control group. However, these enzyme activities were well maintained in the ginseng-treated rats and, especially, catalase and glutathione peroxidase activities were consistently higher than in control rats. The levels of total sulfhydryl group (T-SH) and glutathione reductase (GR) were unchanged, and glutathione-s-transferase (GST) activity gradually decreased with age in both groups. There were no differences in T-SH, GR, or GST between the control and treatment groups. These results indicate that long-term administration of ginseng retards age-related deterioration in some biochemical parameters such as cholesterol, glucose, and lactate dehydrogenase in serum and it has an enhancing effect on antioxidant capacity in the liver.

Dimethylformamide가 사람 혈청의 과산화지질 농도와 Superoxide dismutase 활성도 변화에 미치는 영향 (Effects of Dimethylformamide on Lipid Peroxide Level and Activity of Superoxide Dismutase in Human Serum)

  • The variation in the enzyme activities of human liver usually represents the particular physiological conditions of each individuals. Thus, we investigated the variation in the activities of SOD, HR and LPO of (1) non-exposed workers (56 subjects), and (2) exposed workers to DMF (43 subjects) in synthetic leather process. Serum levels of enzyme activities of exposed workers (AST:$30.26{\pm}20.041$U/L, ALT:$32.72{\pm}23.393$U/L, GGT:$28.47{\pm}18.635$ U/L, ALP:$81.77{\pm}34.879$ U/L)were slightly higher than those in nonexposed workers (AST:$24.00{\pm}9.441$ U/L, ALT:$23.89{\pm}18.305$U/L, GGT:$21.95{\pm}17.970$U/L, ALP:$70.84{\pm}24.678$U/L), but only the level of ALT was statistically significant (p< 0.05). Serum levels of LDH, TRF and CHOL in non-exposed workers were slightly higher than those of exposed workers. However, they were not statistically significant (p > 0.05). Serum levels of HR and LPO of the exposed workers appeared to be reduced, but not those of the non-exposed workers. The SOD activities of exposed workers were also slightly higher than those of non-exposed workers, but the results were not statistically significant (p > 0.05). The level of HR was increased with age, but the SOD level was not. These results suggest that the intermittent exposure to DMF at time-weighted average (TWA) level (10 ppm/$m^3$) has affected on the activities of enzymes such as AST, ALT, TRF, but not on the generation of HR, activity of SOD. However, if high dose of DMF was used, there would be severe effects for the generation of HR and LPO.

Effects of grape pomace on the antioxidant defense system in diet-induced hypercholesterolemic rabbits

  • The effects of grape seeds extract and grape peels extract prepared from grape pomace on the activity of antioxidant enzymes, degree of lipid peroxidation in serum and liver tissue were investigated in rabbits fed on high cholesterol diet. New Zealand white rabbits were divided as follows ; 1) NOR (normal group); 2) CHOL (cholesterol group); 3) GSH (cholesterol + grape seed extract group); 4) GPE (cholesterol + grape peel extract); 5) GSP (cholesterol + grape seed powder); 6) GPP (cholesterol + grape peel powder); 7) GE (cholesterol + grape seed and peel extract); 8) GP (cholesterol + grape seed and peel powder). Eight groups of rabbits were studied for 8 weeks. At the end of the experimental period, rabbits were sacrificed and the liver tissue were removed. Then, GSH, GPx, GST, CAT and MDA in the liver were measured. In liver tissues, total glutathione contents (GSH), glutathione peroxidase (GPx) and catalase (CAT) activity, which was significantly higher by grape seed extract supplementation. The level of malondialdehyde (MDA) was lower in the serum of rabbits fed grape seed extract or grape peel powder plus cholesterol than in the serum of rabbits fed cholesterol alone. It is therefore likely that grape seed extract prepared from grape pomace functioned as antioxidants in vivo, negating the effects of the oxidative stress induced by 1% cholesterol diet. The grape seed extract was found effective in converting the oxidized glutathione into reduced glutathione, and in removing $H_2O_2$ that is created by oxidative stress. The grape peel powder was found to have small influence on reduced glutathione content, CAT and GPX activity, but it increased GST activity in liver tissues, resulting in promoting the combination of lipid peroxide and glutathione (GSH), and further, lowering the formation of lipid peroxide in the serum. Therefore, grape pomace (grape seed extract and grape peel powder) supplementation is considered to activate the antioxidant enzyme system and prevent damage with hypercholesterolemia.

Phaleria macrocarpa Suppresses Oxidative Stress in Alloxan-induced Diabetic Rats by Enhancing Hepatic Antioxidant Enzyme Activity

  • Oxidative stress is caused by an imbalance between the production of reactive oxygen and an ability of a biological system, to readily detoxify the reactive intermediates or easily repair the resulting damage. It has been suggested that developmental alloxan-induced liver damage is mediated through increases in oxidative stress. The anti-diabetic effect and antioxidant activity of Phaleria macrocarpa (PM) fractions were investigated in alloxan-induced diabetic rats. After two weeks administration of PM, the liver antioxidant enzyme and hyperglycemic state were evaluated. The results showed that oral administration of PM treatments reduced blood glucose levels in diabetic rats by oral administration (P < 0.05). Serum glutamic-oxaloacetic transaminase (sGOT) and serum glutamic-pyruvate-transaminase (sGPT) were also diminished by PM supplementation. The superoxide dismutase (SOD), catalase (CAT) and glutathione-peroxidase (GPx) activities, and glutathione (GSH) level in the alloxan-induced diabetic rats were significantly decreased (P < 0.05) compared to those in the normal rats but were restored by PM treatments. PM fractions also repressed the level of malondialdehyde (MDA) in the liver. Glutathione reductase (GR), glutathione-S-transferase (GST) and $\gamma$-glutamylcysteine synthase (GCS) were also reduced in alloxan-induced diabetic rats. PM fractions could restore the GR and GST activities, but the GCS activity was not affected in rat livers. From the results of the present study, the diabetic effect of the butanol fraction of PM against alloxan-induced diabetic rats was concluded to be mediated either by preventing the decline of hepatic antioxidant status or due to its indirect radical scavenging capacity.

다이옥신 저해능에 대한 어성초의 항산화 활성 효과 (Effects of Houttuynia Cordata thunb on Antioxidative Activity against TCDD Damage)

  • This study investigated the effects of Houttuynia Cordata thunb(HCT) administration on the biochemical parameters of function in liver of 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) treated rats. Houttuynia Cordata thunb (200mg/kg) was administeres into rats intraperitoneally for four weeks , seven days after the injection of TCDD(1$\mu\textrm{g}$/kg). We examined the antioxidative enzymatic activity by measuring the level of AST, ALT , SOD and Catalase in serum and liver tissue of rats. HCT group showed 49% of inhibitive effect in AST activity compared to TANO group. ALT level of HCT group was decreased to the level of NO group. SOD and Catalase in TANO group were lower than in NO group, but SOD and Catalase in HCT group were increased by 46% and by 50% respectively compared to TANO group.

  • The purpous of the study was to find an effect of oral administered medium chain triglyceride(MCT) diet on the serum and enzyme activities of Spraque-Dawly rats when they were oral fed with MCT, soybean oil and palm oil. The result was as follow. 1) The body weight gains in MCT diet group was lower than that of the all experimental group. 2) Total cholesterol levels in serum and liver of MCT diet group were very lower than that of the all experimental groups. 3) HDL-cholesterol level of MCT diet group was higher than that of the all experimental group except normal group and VLDL, LDL-cholesterol levels was very higher. 4) TG and PL levels of MCT diet group higher than that of the control group. 5) The activities of GOT with soybean oil diet group and of GPT with MCT diet group were lower than that of the all experimental group. 6) HDL-levels in the serum lipoprotein of MCT diet group was increased and LDL levels was decreased.

한냉에 노출된 흰쥐에서 갑상선 호르몬이 체열 생산인 미치는 영향 (Effect of Cold Exposure on Thyroid Thermogenesis in Rats)

  • It has been well documented that animals exposed to cold show increased activity of thyroid gland. The calorigenic action of thyroid hormone has been demonstrated by a variety of in vivo and in vitro studies. According to Edelman et al., the thyroid thermogenesis is due to activation of energy consuming processes, especially the active sodium transport by the hormone in target tissues. If so, the increase in thyroid activity during cold exposure should induce increased capacity of sodium transport in target tissue and the change in tissue metabolism should be precisely correlated with the change in Na+_K+_ATPase activity of the tissue. This possibility was tested in the present study: in one series, changes in oxygen consumption and Na+_K+_-ATPase activity of liver preparations were measured in rats as a function of thyroid status, in order to establish the effect of thyroid hormone on the tissue respiration and enzyme system in another series, the effect of cold stimulus on the serum thyroid hormone level, hepatic tissue oxygen consumption and Na+_K+_ATPase activity in rats. The results obtained are as follows: 1. The Na+_dependent oxygen consumption of liver slices, the oxygen consumption of liver mitochondria and the Na+_K+_ATPase activity of liver preparations were significantly inhibited in hypothyroidism and activated in hyperthyroidism. Kinetic analysis indicated that the Vmax. of Na+_K+_ATPase was decreased in hypothyroidism and increased in hyperth)'roidism. 2. In cold exposed rats, the serum triiodothyronine (T₃) level increased rapidly during the initial one day of cold exposure, then declined slowly to the control level after two weeks. The serum thyroxine (T₄) level decreased gradually throughout the cold exposure. Accordingly the T₃/T₄ratio increased. The mitochondrial oxygen consumption and the Na+_dependent oxygen consumption of liver slices increased during the first two days and then remained unchanged thereafter The activity of the Na+_K+_ATPase in liver preparations increased during cold exposure with a time course similar to that of oxygen consumption. Kinetic analysis indicated that the Vmax. of Na+_K+_ATPase increased. 3. Once the animal was adapted to cold, induction of hypothyroidism did not significantly alter the hepatic oxygen consumption and Na+_K+_ATPase activity. These results indicate that: 1) thyroid hormone increases capacities of mitochondrial respiration and active sodium transport in target tissues such as liver; 2) the increased T₃level during the initial period of cold exposure facilitates biosynthesis of Na+_K+_ATPase and mitochondrial enzymes for oxidative phosphorylation, leading to enhanced production and utilization of ATP, hence heat production.

지골피와 생강 정제 추출물이 고지방식이 흰쥐의 체내 지질과 혈청 Cytokine 수준에 미치는 영향 (Effects of the purified extracts from Lycii Cortex Radicis and ginger on lipid statusand serum cytokine levels in rats fed high fat diet)

  • The present study was to investigate the effects of Lycii Cortex Radicis (LCR), the root bark of lycium (Lycium chenese Miller) and ginger (Gin) on body lipid status and serum levels of cytokines. Sprague-Dawley (SD) male rats weighing $193.6{\pm}16.8g$ were divided into five groups, including one low fat (LF) and four high fat groups, i.e. HF-Control, HF-LCR, HF-Gin and HF-LCR + Gin groups. Diets for HF-LCR, HF-Gin and HF-LCR + Gin groups contained purified extracts having 0.2 g LCR tyramine, ginerol and 0.1 g tyramine plus 0.02 g gingerol per kg, respectively. Compared with those of the HF-Control total serum cholesterol level decreased, and HDL-cholesterol level increased in the HF-LCR group and serum triglyceride levels decreased in the three experimental groups fed the purified extracts. Liver cholesterol level was lower in the HF-LCR group than the HF-Control group, but triglyceride levels, which were increased by high fat diets were not changed by significantly by LCR or ginger extracts. Fecal lipid excretion was higher in the HF-LCR and HF-Gin groups, but cholesterol excretion was lower in the HF-Gin group than in the HF-Control group. The activities of liver cytosolic glucose-6-phosphate dehydrogenase and malic enzyme were lower in the HF-LCR + Gin group than in the HF-Control group. Serum adiponectin levels did not differ among the five groups, while leptin level was lower in the HF-Gin group and C-reactive protein levels were lower in the HF-Gin and the HF-LCR + Gin groups than in the HF-Control group. It is concluded that LCR can be utilized as an ingredient for lipid-lowering functional foods in the form of purified extract and addition of small amount of ginger extract would be useful for reducing one of the inflammatory cytokines to help prevent atherosclerosis.

Thioacetamide로 유발한 간손상 모델에서 계혈등(鷄血藤)의 간보호 효과 (Protective Effect of Spatholobi Caulis in Thioacetamide induced Acute Liver Injury of Rat)

  • Objectives : This study was undertaken to investigate the hepatoprotective effect of Spatholobi Caulis water extract (SC) to thioacetamide (TAA)-induced acute liver injury (ALI) in rats. Methods : The rats were injected intraperitoneally with TAA (200 mg/kg body weight) and orally administered SC (100 or 200 mg/kg b.w.) daily for 3 days. Liver biomarkers were assessed by serum glutamic oxaloacetic transaminase (GOT), glutamic pyruvic transaminase (GPT), and ammonia levels. Malondialdehyde (MDA) was measured both serum and liver tissue. In addition, nicotinamide adenine dinucleotide phosphate (NADPH) oxidase, anti-oxidant, and inflammation-related proteins were investigated by western blot analysis. Histological examination further confirmed though hematoxylin and eosin stain. Results : The SC treatment reduced liver function markers like GOT and GPT and also remarkably decreased ammonia level. Moreover, the elevated MDA level in TAA-induced group was significantly reduced by SC treatment. NADPH oxidase expression associated with oxidative stress including NOX2, NOX4, and p47phox markedly inhibited by SC administration. SC treatment exerted anti-oxidant effect through the increase of anti-oxidant enzyme including superoxide dismutase (SOD), Catalase, and heme oxygenase-1 (HO-1). The protein expressions of inflammatory cytokines such as tumor necrosis factor-�� (TNF-��), IL-6, and IL-1�� induced by nuclear factor-kappa B (NF-��B) activation were modulated through blocking the phosphorylation of inhibitor of nuclear factor ��B�� (I��B)��. SC treatment also improved histological alterations. Conclusion : These findings suggested that SC administration may be a potential candidate for the prevention or treatment of ALI.
