비만아의 체성분 및 생화학적 측정치와 혈중 Leptin 농도와의 관계 (Relationship among Body Composition, Biochemical Measurements and Serum Leptin Level in Obese Children)

  • 대구 및 대구 인근 경북지역 초등학교 4∼5학년에 재학 중인 남아 42명, 여아 55명을 대상으로 조사대상자들의 비만도에 따른 신체 계측치와 체성분 측정치, leptin를 비롯한 생화학적 자료들을 비교 분석하고, 이들 인자들간의 상호 관련성을 분석한 결과는 다음과 같다. 1) 여러 가지 방법으로 비만을 판정한 결과 체질량지수 25를 기준으로 하였을 때 비만군의 분포는 총 조사대상자의 30.9%, 상대체중 120%를 기준으로 하였을 때는 27.8%, 체지방을 25% 이상을 기준으로 했을 때는 56.7%로 나타났다. 체질량지수와 상대체중 상호간에는 높은 상관성 (r = 0.87, p <0.01)을 나타내었으나, 체지방율과는 중 정도의 상관성을 나타내었다 (r = 0.54, r = 0.59, p < 0.01). 2) 신체계측 및 체성분 분석 결과, 비만군이 대조군보다 신장, 체중 체지방량 제지방량, 근육량, 체지방을, 복부지방을, 체질량지수, 상완위 근육둘레가 남녀 모두 유의적으로 높게 나타났다 (p < 0.001). 특히 남아의 경우 비만군이 근육형태의 분포에서는 대조군과는 달리 저근육형 과체중의 분포가 매우 높게 나타났고, 신체발달점수도 유의하게 낮게 나타났다. 전체 조사대상자의 반 이상이 상하체 균형이 허약한 것으로 나타났다. 3) 혈액의 생화학적 검사 결과, 남아의 경우 HDL-cholesterol을 제외한 모든 측정치가, 여아의 경우 중성지방과 iron의 농도가 비만군이 대조군보다 유의적으로 높게 나타났다. 혈중 leptin농도는 남녀 모두 비만군이 대조군보다 유의적으로 높게 나타났으나, 성별에 따른 혈중 leptin농도는 유의적인 차이를 나타내지 않았다. 그러나 대조군에서는 여아가 남아보다 비만군에서는 오히려 남아가 여아보다 높은 경향을 보였다. 4) Leptin농도와 신체계측치 및 체성분 측정 치와의 관계에서 혈청 leptin함량은 측정된 모든 신체 계측치 및 체성분 분석치와 유의적인 높은 양의 상관관계를 나타냈다. 다변량분석 결과 전체대상자 중에서는 체질량지수가. 비만여부에 따라 분석하였을 때 대조군의 경우는 총지방량이 비만군의 경우는 체질량지수가 각각 독립적으로 leptin 농도에 실질적으로 예언력을 갖는 것으로 나타나 체지방 분포도에 따른 차이보다도 총체지방량 및 체질량지수와 관련이 더 있음을 보여주었다. 5) Leptin농도와 생화학적 지표와의 관련성을 분석한 결과, leptin 농도는 HDL-cholesterol를 제외한 측정된 모든 생화학적 지표와 유의적인 양의 상관관계를 나타내었다. 다변량분석 결과 전체대상자 중에서는 total-cholesterol, iron, 중성지방, ferritin의 농도가 각각 독립적으로 leptin 농도에 실질적인 예언력을 갖는 것으로 나타났고 비만 여부에 따라 분석하였을 때 비만군에서만 total-cholesterol, iron의 농도가 각각 독립적으로 leptin농도에 실질적인 예언력을 갖는 것으로 나타났다.

곽향정기산가미방(藿香正氣散加味方)이 고지방식이(高脂肪食餌)로 유식(誘導)된 흰쥐의 체지방에 미치는 영향(影響) (Effects of Kwakhyangjungkisangamibang Extract on the Adipose Tissues Induced by a High Fat Diet in Rats)

  • Objetives This experimental study is designed to investigate the effects of kwakhyangjungkisangamibang extract on the changes of adipose tissues, serum insulin and leptin levels in rats induced by a high fat diet. The leptin has been proposed to be involved in the role of food intake and energy expenditure. Methods During the 8 weeks of experimental period, male Sprague-Dawley rats were fed on the 2 different diets: the normal diet AIN-76A and high fat(beef tallow) diet ad libitum, of which 40% of the calories intake was fat. 9 rats in the control and high fat diet group were killed for the baseline experiment at 4 weeks of age. The high fat diet group was divided into three groups: control group, experimental group I(high fat diet plus kwakhyangjungkisangamibang extract), and experimental group II(high fat diet plus mahuang). The body weight, hypodermal fat tissue, serum lipid profile, insulin and leptin levels were measured after the administration of the high fat diet and extract. Results 1. The body weight was decreased in experimental group II, but there was not a significance difference compared to the control group. 2. The peritoneal, visceral, and hypodermal adipose tissue was decreased in experimental group I, but there was not a significance difference compared to the control group. 3. The lipid profile was decreased in experimental group I, but not in group II. 4. The insulin concentration was increased in experimental group I and II, but there was not a significance difference compared to the control group. 5. The leptin level was increased significantly in experimental group I(p<0.05) and II(p<0.01) compared to the control group. Conclusion The results of this study indicate that kwakhyangjungkisangamibang has an effect on the regulation of obesity.

성인여성의 체지방률에 따른 영양소섭취, 렙틴, 아디포넥틴, 코티졸 및 인슐린농도 (Nutrient Intake, the Concentrations of Leptin, Adiponectin, Cotisol & Insulin by the Body Fat Content of Women)

  • The aim of this study was to compare nutrient intakes, serum hormones (leptin, adiponectin, insulin), salivary cortisol and ${\alpha}$-amylase of middle-aged women by the percentage of body fat (% fat). Subjects were assigned to 3 groups by body fat (%) group I (27.5%), group II (32.5%), group III (37.7%). WHR of group II (0.97) was significantly higher than of group I, III (0.95) (p < 0.05). Nutrient intakes were not different among 3 groups. Serum leptin levels of group III ($16.53{\mu}g/ml$) were higher than in group I ($10.07{\mu}g/ml$), group II ($12.24{\mu}g/ml$) (p < 0.05). Salivary cortisol levels of group II ($0.39{\mu}g/dl$) were higher than in group I ($0.17{\mu}g/dl$) and group III ($0.15{\mu}g/dl$) (p < 0.05). Adiponectin concentrations were negatively correlated with TAS (r = -0.29) and positively correlated with HDL cholesterol (r = 0.27). Insulin levels were negatively correlated with total cholesterol (r = -0.33), Zn intake (r = -0.31) and positively correlated with WHR (r = 0.31). The overall anthropometric indices showed positive relations with leptin levels. Salivary cortisol levels were positively corelated with WHR (r = 0.28), total cholesterol (r = 0.31), MDA (r = 0.29) and intakes of SFA (r = 0.35) and MUFA (r = 0.3). Salivary amylase levels were positively correlated with overall nutrient intakes (energy, CHO, fat, cholesterol. Fe, SFA, MUFA, Zn, Na, vitamin $B_2$, r = 0.24-0.5) and was negatively correlated with HDL cholesterol (r = -0.34). These results suggested that 1) WHR would be a helpful index in the assessment of metabolic risk diseases. 2) Understanding of individual stress exposure should be considered in developing strategies for prevention and treatment of obesity.

남성 근로자의 비만이 adiponectin과 leptin의 생리적 농도와 대사증후군 진단지표에 미치는 영향 (Effects of Obesity on the Physiological Levels of Adiponectin, Leptin and Diagnostic Indices of Metabolic Syndrome in Male Workers)

  • Purpose: The aim of this study was to examine the effects of obesity on the physiological levels of adiponectin, leptin and components of metabolic syndrome (MS) in male workers, aged 30-40 years. Methods: Body mass index (BMI) was measured with Anthropometric equipment. Blood pressure and serum parameters were measured with an automatic digital sphygmomanometer and autochemical analyzer, respectively. Adiponectin and leptin were analysed by ELISA kits and MS was defined based on the NCEP-ATP III. Results: Body fat mass of waist and hip, systolic and diastolic blood pressure were significantly higher, as expected, in the BMI>25kg/$m^2$ in comparison with the $BMI{\leq}25kg/m^2$. While fasting glucose, insulin, HOMA-IR and leptin in the BMI>25kg/$m^2$ were also significantly higher compared with $BMI{\leq}25kg/m^2$, HDL-cholesterol and adiponectin were significantly higher in $BMI{\leq}25kg/m^2$. On multiple logistic regression analysis for the components of MS, exercise, adiponectin and leptin were an only independent factor for MS in non-obese male workers($BMI{\leq}25kg/m^2$) after adjustment for age, cigarette smoking and drinking habits. Conclusion: These results suggested that the obesity in men was associated with physiological levels of adiponectin and leptin contributing to feedback control of MS and that dysfunction and/or declination in feedback control system associated with changes in physiological levels of neurptrophics: adiponectin and leptin might ultimately induce MS.

  • Objective : These experimental studies were designed to investigate the effects of Sargassum extract and Polygoni multiflori radix extract on the weight and the lipid metabolism of the obese rats induced by 1% cholesterol diet. They were designed to detect changes in the serum and leptin levels in the infant rats. Methods : The serum and leptin levels were measured for a period of 8 weeks for obese SD rats induced by 1% cholesterol diet. Result : 1. The body weight showed a tendency to decrease in samples I & II(P>0.05). 2. The average size of the epididymal fat cell significantly decreased in sample II(p<0.05), and showed a tendency to decrease in sample I(P>0.05). 3. The leptin levels significantly decreased in samples I & II(P<0.05). 4. The total cholesterol levels significantly decreased in samples I & II(P<0.05). 5. The triglyceride levels significantly decreased in sample I & II(P<0.05). 6. The glucose levels significantly decreased in sample I & II(P<0.05).

In Vitro Antioxidant Properties of Equisetum arvense and Its Effects on Serum Lipid Levels in Mice Fed a High-Fat Diet

  • This study analyzed the antioxidant properties of Equisetum arvense and its effects on serum factor levels in mice fed a high-fat diet. The aim was to establish a new effective resource for biologically active materials. E. arvense stem and root extracts were obtained using deionized water at $95^{\circ}C$, and 70.5% ethanol at $85^{\circ}C$. These extracts were used to analyze the total phenolic compounds and antioxidant (ABTS, DDPH, and FRAP) activities. The effects of prepared ground samples were evaluated by feeding them to mice. E. arvense extracts showed strong antioxidant effects. The caffeic acid content was highest in the 70.5% ethanol extract of the vegetative stem, as determined by high-performance liquid chromatography. The blood concentrations of insulin and leptin were significantly lower in mice fed a high-fat diet supplemented with extracts of the root, reproductive stem, and vegetative stem of E. arvense than in mice fed only a high-fat diet. These results suggest that the polyphenolic compounds in E. arvense extracts exert various antioxidant effects. The stems and root of E. arvense can lower the blood levels of insulin and leptin, even after consumption of a high-fat diet.

녹용이 흰쥐의 비만억제에 미치는 영향 (Effect of Deer Antler on the Inhibitory Action of Obese Rats)

  • Objective: We investigated the effects of deer antler extract (DAE) on the expression of the weight, leptin, and serum levels (lipid metabolism) of the obese SD rats induced by a $2\%$ cholesterol diet in vivo. Methods: In vivo, the weight, leptin and serum levels were measured after a period of 7 weeks for obese SD rats induced by a $2\%$ cholesterol diet and treated with DAE of three concentrations (0.938 g/kg/5ml, 1.875 g/kg/5ml, 3.75 g/kg/5ml). Results: 1. Body weight didn't change significantly. 2. Leptin levels significantly decreased (p<0.05). 3. Total cholesterol, triglyceride, LDH, sGOT & sGPT levels significantly decreased in the sample (p<0.05). 4. The HDL cholesterol levels significantly increased in the sample (p<0.05). 5. The average size of the epididymal fat cells significantly decreased as the concentration of deer antler extract increased (p<0.05). Conclusion: Deer antler effects inhibition of obese rats.

They were designed to change values in the serum and leptin levels in the rats. Materials and Methods : The serum and leptin levels were measured for a period of 8 weeks for obese SD rats induced by 2% cholesterol diet. Results : The body weight showed a tendency to decreased significantly in sample (p<0.05). The Total cholesterol and Triglyceride level significantly decreased in sample (p<0.05). The HDL Cholesterol levels significantly increased in sample (p<0.05). The LDH levels did not show a significant change in sample. The ALP levels significantly decreased in sample (p<0.05). The SGOT and SGPT level did not show a significant change in sample. The Leptin levels significantly decreased in sample (p<0.05). The average size of the epididymal fat cell significantly decreased in sample (p<0.05).

유산소 운동이 폐경 후 비만여성의 혈청지질, 렙틴, 그렐린 및 인슐린저항성지수에 미치는 영향 (Effects of Aerobic Exercise on Serum Blood Lipids, Leptin, Ghrelin, and HOMA-IR Factors in Postmenopausal Obese Women)

  • 본 연구는 체지방률이 32% 이상인 폐경 후 비만여성 36명을 대상으로 유산소운동이 혈청지질, 렙틴, 그렐린 및 인슐린저항성지수에 미치는 영향을 구명하기 위하여 운동집단(18명)과 통제집단(18명)으로 나누어 유산소운동인 라인댄스를 16주간 실시한 후 체조성, 혈청지질, 렙틴, 그렐린 및 인슐린저항성지수를 운동전과 후로 측정하여 비교분석하였다. 본 연구의 결과 운동 후에 운동집단에서 체중, 체지방률(%BF), 체질량지수(BMI), 내장지방면적(VFA), 렙틴(Leptin), 인슐린(Insulin), 글루코스(Glucose), 인슐린저항성지수(HOMA-IR), 총콜레스테롤(TC), 중성지방(TG), 아포리포프로틴(Apolipoprotein) B, 저밀도지단백 콜레스테롤(LDL-C) 및 수축기혈압(SBP)이 유의하게 감소하였다. 또한 아포리포프로틴(Apolipoprotein) A-1, 그렐린(Ghrelin) 및 고밀도지단백 콜레스테롤(HDL-C)이 유의하게 증가하였다. 유산소운동집단의 내장지방면적에 영향을 미치는 에너지대사조절인자의 회귀분석결과에서 주요요인으로 그렐린, 인슐린, 글루코스 및 HOMA-IR 나타났다. 따라서 규칙적이고 지속적인 유산소운동이 폐경 후 비만여성의 체조성, 내장지방, 혈청지질, 렙틴, 그렐린 및 HOMA-IR을 개선시키는 데 효과가 있을 것으로 생각된다.

이진탕가미방(二陳湯加味方)이 비만(肥滿)에 미치는 실험적(實驗的) 연구(硏究) - 지질대사, 혈압, 뇌혈류량을 중심으로 - (The Experimental Study of EjinTang-Gamybang (Erchentang-jiaweifang) on the Obesity in Rats - Focusing on Lipid-metabolism, Blood pressure, Cerebral Blood Flow -)

  • Objectives : This experimental study was designed to investigate the effects of EjinTang-Gamybang(Erchentang-jiaweifang ; ETG) on the change of some values related to obesity and observe the complications coming from obesity in obese rat induced by high fat diet. Methods : Experimental group were as follows ; normal group were fed normal diet and administered DDW $1.0m{\ell}$ to rat during 7 weeks, control group were fed high fat diet and administered DDW $1.0m{\ell}$ during 7 weeks, sample A were fed high fat diet and administered ETG 500 mg/kg $1.0m{\ell}$ during 7 weeks, sample B were fed high fat diet and administered ETG 700 mg/kg $1.0m{\ell}$ during 7 weeks. Results & Conclusions : 1. Sample A and Sample B significantly decreased body weight, serum LDL-cholesterol level, serum free fatty acid level, serum total lipid level, serum phospholipid level and serum leptin level in comparison with control group. 2. Sample B significantly decreased serum total cholesterol level and serum triglyceride level in comparison with control group. 3. Sample B significantly increased serum HDL-cholesterol level in comparison with control group. According to above results, the author suggested that ETG was able to be used for the herbal medication of obesity. 4. ETG significantly increased rCBF, and increased CMF in a dose-dependent. 5. ETG significantly decreased MABP in a dose-dependent. 6. rCBF was significantly and stably increased by ETG(10 mg/kg, i.p.) during the period of cerebral reperfusion, which contrasted with the findings of rapid and marked increase in control group. This results were suggested that ETG significantly increased rCBF by dilating arterial diameter and activating serum leptin level. So that, the present author thought that ETG had an effects of obesity and complication coming from obesity(ischemic cerebral and cardiac disease).
