  • The usefulness of single, random measurements of serum FSH and LH in the diagnosis of primary amenorrhea by radioimmunoassay was investigated. The 16 patients were divided into 3 groups by the level of serum FSH and LH. The first group with increased level of serum FSH and LH is five patients, all of these are related to the acquired or congenital abnormality of the ovary. Further studies indicated include buccal smear, chromosome analysis, gynecography and laparosocopy. The second group with normal serum FSH and LH is nine patients, four patients of these are related to the developmental anomaly of the Mullerian duct and five patients are undo etermined origin. Further studies indicated include laparoscopy and gynecography. The third group with decreased serum FSH and normal or decreased serum LH is two patients, one of these is related to the pituitary function, isolated FSH deficiency, the other is undetermined origin. Further studies indicated include the pituitary function test, LH-RH stimulation test, skull radiography. Determination of serum FSH and LH levels does not permit a specific etiologic diagnosis of primary amenorrhea. However the serum levels of FSH and LH can be used to differentiate the principal area of the investigation and can be of assistance in choosing more specific testing procedures.

  • This study was done to compare serum levels of lipids, trace minerals, ceruloplasmin and ferroxidase activity between female college smokers and nonsmokers. Serum levels of lipids, Cu, Zn, ceruloplasmin and ferroxidase activity were determined in 33 smokers and 42 nonsmokers residing in the Seoul area. Serum levels of TG, total cholesterol and HDl-cholesterol were measured using test kits. Serum Cu Zn were determined by the AAS. Serum ceruloplasimin level and ferroxidase activity were determined by spectrophotometric methods. All data were statistically analyzed by the SAS PC package program. Mean and standard errors were calculated for each item, and the significance of the difference between two groups was evaluated by Student's t-test at p〈0.05. There was no significant difference in levels of total cholesterol, TG, VLDL-cholesterol, LDL-cholesterol, LDL/HDL and atherogenic index, but HDL-cholesterol level was significantly lower(p$\ll$0.05) in female college smokers than in nonsmokers. Serum zinc level showed no significant difference, whereas copper level was significantly higher in smokers, consequently yielding higher Cu/Zn ratio in smoking college women. Ceruloplasmin level was 32.6mg/dl in smoking college women, which was significantly higher(p$\ll$0.05) than that of 28.6mg/dl in nonsmokers. However, cerulopasmin ferroxidase activity, 0.34U/mg in nonsmokers was higher(p$\ll$0.05) than that of 0.29U/mg in smokers. From these results, it is confirmed that cigarette smoking itself has a very detrimental effect on health and mineral nutritional status although the smoking history is relatively short among college women(pack-years=1.7). • To establish the protocol of a standardized exercise test for evaluating exercise intolerance and degree of fitness in Thoroughbred racehorses, we examined serum lactate concentrations related to exercise intensities using the high speed treadmill. Twelve clinically healthy Thoroughbred racehorses with or without previous training or racing history were assigned to two gorups, fit and unfit group, respectively. The protocol used for the standardized exercise test was consisted of two stages : stage of warm-up and that of acceleration. During the warm-up, the horses exercised 5 min at 1.8m/s and 3 min 3.4m/s without inclination. At the acceleration stage, exercise test was performed at 10% slope and the speed was increased from the initial 5m/s to the maximal speed which each tested horse could keep up with. The speed was increased with incremental steps of 1 m/s every minute. During the last 15 sec of each step, blood samples were collected for serum lactate determination. $V_{max}$(maximal treadmill speed which tested horses could keep up with) of the fit group ($10.93{\pm}0.33m/s$, mean${\pm}$SE, n = 6) was higher than that of the unfit group ($9.52{\pm}0.23m/s$, mean${\pm}$SE, n = 6). Serum lactate concentrations increased exponentially according to exercise intensities. $V_{La4}$(speed producing a serum lactate concentration of 4mmol/l) of the fit group, $6.45{\pm}0.26m/s$, was higher than that of the unfit group, $5.45{\pm}0.23m/s$. $La_{peak}$(peak plasma lactate concentration during the exercise test) was lower in the fit group ($20.34{\pm}1.62mmol/l$ at 1 min after maximal intensity exercise) than in the unfit group ($24.78{\pm}1.09mmol/l$ at 2 min after maximal exercise step). $t_{50%}$(time required for the recovery of lactate concentration to be one-half of $La_{peak}$ after maximal exercise) of the unfit group and the fit group were 40.0 and 18.0 min, respectively. Therefore, the protocol of the incremental standardized exercise test utilized in this study seems to be reliable for the assessment of fitness and exercise intolerance for the Thoroughbred racehorses.

  • The indoor environment is an important source of exposure to various aeroallergens and pathogenic microorganism. It has been shown that exposure to aeroallergens enhances the risk of indoor inhabitants developing asthma. Since the skin prick test, a typical clinical method for identification of subjects positive to allergens, can rarely cause fatal or non-fatal reactions in susceptible persons, an in vitro assay such as ELISA using serum has been considered for testing positivity against various allergens. We evaluated the validity of a serum ELISA kit for screening positive subjects to major aeroallergens including Dermatophagoides farinae, Dermatophagoides pteronyssinus, cockroach, Alternaria, Cladosporium, Aspergillus, Penicillium, dog hair, cat fur, mugwort, and ragweed. The ELISA results were compared with the skin prick test results, and sensitivity, specificity, and overall accuracy were calculated to each allergen. Higher sensitivities were obtained from D. farinae, (77.8%) and D. pteronyssinus (69.2%), but sensitivities to Aspergillus, Penicillium, dog hair, cat fur, and ragweed were very low down to 0%. Specificity ranged from 88.7% (cat fur) to 100% (mugwort and ragweed). Overall the accuracy of the serum ELISA kit was relatively high, in that the lowest was 85.1% for cat fur and the highest was 98.6% for Alternaria, Cladosporium, and ragweed. Considering specificity and overall accuracy for the serum ELISA kit, it may be considered reliable. However, when the kit is used for screening purpose, positivity to aeroallergens should be carefully determined since sensitivity for the kit is low.

  • The purpose of this study is to determine the effect of oral vitamin C supplements on blood sugar and serum lipid level(total cholesterol, triglyceride, high density lipoprotein, low density lipoprotein) in non-insulin independent diabetes mellitus. The study design was a non equivalent control group pre-test post-test design. Data for the study were collected from June 24 to August 31, 2001. The ninty-five research subject were assigned to experimental group(51) and control group(44). Vitamin C(3g/day) was given to 51 subjects for 4 weeks. Following a 12h overnight fasting, blood sample was obstaind at baseline and at the end of 4week - supplementation. Blood samples were taken for plasma vitamin C concentration, fasting blood sugar, HbA1c and serum lipid level. The pre-equivalent test was used by Chi-squre, t-test and two group's pre and post experimental differences were analyzed by t-tset to compare with each other. The results of this study were as follows ; 1. The difference between the two groups in plasma vitamin C concentration was significant(t=-12.950, p=.000). 2. The difference between the two groups in fasting blood sugar was significant(t=5.293, p=.000). 3. The difference between the two groups in HbA1c was not significant(t=1.758, p=.082). 4. The difference between the two groups in total cholesterol, HDL, LDL was significant(t=3.786, p=.000 ; t=-5.515, p=.000 ; t= 4.169, p = .000). These results suggest that megadose vitamin C supplementation be effective in lowering fasting blood sugar, serum lipids and increasing plasma vitamin C. Thus dietary measures to increase plasma vitamin C may be on important health strategy for reducing the compliance of diabetic patients.

  • In this study, the anti-oxidative effects and components of sprout serum were investigated. In the buds, high levels of zinc, iron and manganese were analyzed in addition to copper, magnesium, and potassium. The free radical (1,1-diphenyl-2-picrylhydrazyl, DPPH) scavenging activity of sprout serum was evaluated with $IC_{50}$. $IC_{50}$ of sprout serum was $58.0\;mgmL^{-1}$, and that of vitamin C was $3.7\;mgmL^{-1}$. In the test of superoxide scavenging activity of sprout serum, the activity was dependent on the concentration of serum. In this case, the $IC_{50}$ was 2.0 wt%. Cell viability was detected by using the MTT method. Cultured human fibroblast was treated with 15 mM $H_2O_2$ and cell viability was 70% in case of control. However, the effect of treating 0.5% of sprout serum was similar to that of 0.0001% of vitamin C.
