Minerals is required small amounts among various nutrients, but it has a significant impact on sow longevity and reproduction performance. This study was carried out to see the beneficial effects of marine-derived Ca-Mg complex on the reproductive performance of sows during four-parity periods. Seventy-two gilts ([Yorkshire × Landrace] × Duroc), with an average body weight of 181 kg, were randomly allocated to three groups; CON (basal diet), 0.3LC (CON - MgO - 0.3% limestone + 0.4% Ca-Mg complex), and 0.7LC (CON - MgO - 0.7% limestone + 0.4% Ca-Mg complex). During parity 3 and 4, the expression level of SCD gene was lower in the umbilical cord of piglets born to 0.3LC and 0.7LC sows compared with the CON sows. During parity 2, 3 and 4, SLC2A2 and FABP4 gene expressions were higher in the umbilical cord of piglets born to 0.7LC sows and the placenta of sows from 0.3LC groups, respectively. Ca-Mg complex increased (p < 0.05) Ca and Mg concentrations in sows and their piglets' serum as well as in colostrum regardless of parities. The serum vitamin D concentration was higher (p < 0.05) in their first parity, whereas serum prolactin and estrogen concentrations were higher (p < 0.05) during the fourth and third parity, respectively. The growth hormone concentrations were higher (p < 0.05) in the piglets born to sows during the first and second parity. The fat and immunoglobulin A (IgA) concentrations in colostrum were higher (p < 0.05) during the third and fourth parity, respectively. A reduction (p < 0.05) in salivary cortisol, epinephrine, and norepinephrine concentrations was observed in 0.3LC and 0.7LC sow groups compared with CON after farrowing regardless of parity, however before farrowing, a reduction in norepinephrine was observed. Before farrowing, the epinephrine and norepinephrine concentrations were higher (p < 0.05) during the first and second parity. After farrowing, the concentration of these hormones was higher during the second parity. Taken together, sows' parity and dietary Ca-Mg complex supplementation influenced serum metabolites, colostrum nutrients, stress hormones as well as the gene expressions related to lipid and glucose metabolism.

This study hypothesized that dietary feed additive containing probiotics alter either immune-related serum substances or serum metabolites in Hanwoo heifers. A probiotic treatment was given at 0.5% top-dressing of concentrate diet for 6 months. The change of immunological indicators in the blood was analyzed under LPS (Lipopolysaccharide) challenge. One day before administration of LPS, all heifers were fitted with an indwelling jugular vein catheter for serial blood collections. Both a serum tube and an EDTA-coated tube were collected at 30-min intervals from - 2 to 8 hours relative to the LPS challenge at time 0 ($1{\mu}g/kg$ of BW). Serum was used for analyzing albumin (ALB), glucose (GLU), total protein (TP), triglycerides (TG), phosphorus (IP), and non-esterified fatty acids (NEFA). Plasma was used for analyzing white blood cell (WBC), red blood cell (RBC), platelet (PLT) and inflammation-related factors (NE, LY, MO, EO, BA, Hb, HCT, MCV, MCH, MCHC, RDW, MPV). There were significant differences in ALB, GLU, TG, IP, and NEFA concentration with the passage of hours post challenge (p < 0.05). The level of ALB, GLU, TG, and IP showed significant difference (p < 0.05) between treatments. However, none of the data showed interaction between time and treatments (p > 0.05). The level of WBC, EO, LY, and MO were reduced after LPS challenge (p > 0.05). In conclusion, LPS challenge after dietary supplementation of probiotics changed the levels of both serum metabolites and inflammation-related factors. The increase of GLU and TG indicated a probiotics-positive response under LPS challenge (p < 0.05).

Zearalenone (ZEN), a mycotoxin produced by Fusarium in food and feed, causes serious damage to the health of humans and livestock. Therefore, we compared the metabolomic profiles in the liver, serum, and urine of piglets fed a ZEN-contaminated diet using proton nuclear magnetic resonance ($^1H-NMR$) spectroscopy. The spectra from the three different samples, treated with ZEN concentrations of 0.8 mg/kg for 4 weeks, were aligned and identified using MATLAB. The aligned data were subjected to discriminating analysis using multivariate statistical analysis and a web server for metabolite set enrichment analysis. The ZEN-exposed groups were almost separated in the three different samples. Metabolic analysis showed that 28, 29, and 20 metabolites were profiled in the liver, serum, and urine, respectively. The discriminating analysis showed that the alanine, arginine, choline, and glucose concentrations were increased in the liver. Phenylalanine and tyrosine metabolites showed high concentrations in serum, whereas valine showed a low concentration. In addition, the formate levels were increased in the ZEN-treated urine. For the integrated analysis, glucose, lactate, taurine, glycine, alanine, glutamate, glutamine, and creatine from orthogonal partial least squares discriminant analysis (OPLS-DA) were potential compounds for the discriminating analysis. In conclusion, our findings suggest that potential biomarker compounds can provide a better understanding on how ZEN contaminated feed in swine affects the liver, serum, and urine.
This study aimed to assess the effects of neutral detergent fiber (NDF) from rice straw on blood metabolites and productivity of dairy cows in the tropics. Eight Holstein ${\times}$ indigenous (75-87.5% Holstein) were randomly allocated to dietary treatments according to a double $4{\times}4$ Latin square design. Contents of roughage NDF from rice straw were 12.8, 14.8, 16.8 and 18.8% dry matter (DM) and concentrate NDF content was 10.2% DM, so that levels of dietary NDF were 23, 25, 27 and 29% DM. Dietary treatments were isonitrogenous and isocaloric diets. The average maximum and minimum temperature humidity index during the experimental period were 85.6 and 76.9, respectively. Plasma glucose (p<0.05), dry matter intake and 4% fat corrected milk (p<0.01) decreased with increasing contents of roughage NDF from rice straw, while rectal temperature, serum urea, cortisol, low density lipoprotein-cholesterol (p<0.05), serum total protein and ${\beta}$-hydroxybutyrate (p<0.01) increased. Serum triglyceride, total cholesterol and high density lipoprotein-cholesterol were not affected (p>0.05). Decreased rectal temperature and concentrations of serum cortisol were associated with decreasing levels of ${\beta}$-hydroxybutyrate and serum urea, but with increased concentrations of plasma glucose, DMI and 4% fat corrected milk indicating that lactating cows fed lower fiber diets were in lower oxidative stress. Therefore higher intake and metabolic efficiency in dairy cows fed diets lower in roughage NDF from rice straw would be of an advantage in productive systems under tropical conditions.
The objective of the current study was to determine seasonal changes in glucose, cholesterol, non-esterified fatty acids (NEFA), serum inorganic phosphorous (SIP), calcium, magnesium and iron concentrations in Nguni and crossbred cattle on the sweet and sour rangelands of the Eastern Cape Province of South Africa. Body weights, body condition scores (BCS) and serum concentrations of energy-related metabolites and mineral profiles were determined in late cool-dry, hot-dry, hot-wet, post-rainy and early cool-dry seasons in 100 cattle raised on communal rangelands from August 2007 to May 2008. Nguni cattle had lower (p<0.05) and higher (p<0.05) serum concentrations of glucose in the hot-wet and post-rainy seasons, respectively, compared to crossbreds in the same seasons. Serum cholesterol and NEFA concentrations in Nguni were lower (p<0.05) than in the crossbreds. Nguni and crossbred cattle had higher (p<0.05) serum NEFA concentrations on the sweet rangeland during the late cool-dry season than on sour rangeland. Nguni cattle had higher (p<0.05) SIP concentration in the hot-wet season than the crossbreds. Generally, both breeds had lowest SIP concentration during the hot-wet season on the sour rangeland. The lowest magnesium and highest iron concentrations were observed in the hot-wet and post-rainy seasons, respectively, compared to other seasons. Cattle on the sour rangeland had lower (p<0.05) iron concentrations than those on the sweet rangeland. It was concluded that Nguni cattle had lower cholesterol and NEFA, and higher SIP concentrations in the hot-wet season than crossbreds and energy deficits mostly occurred during the late cool-dry season on the sweet rangeland.
This study was designed to observe the change of body composition and nutrient metabolites and the excretion of minerals during complete fasting for 10 days in thirty women. Average loss of body weight was 7.98kg and body fat was gradually reduced after 9 days fasting, but the loss of lean body mass was reduced after 7 days fasting. Serum glucose level was sustained at constant level, but serum levels of blood urea nitrogen, free acid and $\beta$-hydroxybutyrate were significantly increased during fasting but decreased after re-feeding. Urinary excretions of 3-methylhistidine, total creatinine, and urea were high in the beginning of fasting but gradually decreased. Serum level of parathyroid hormone was significantly reduced by fasting but regained after re-feeding. Serum level of minerals was at the constant level throughout the experimental period. The urinary excretion of minerals(Ca, K, Mg, P) was significantly increased in the early stage of fasting and then reduced from 7 days, but the excretion of Zn was continuously increased until the late stage of fasting. These results showed that amino acid fatty acid released from the breakdown of muscle protein and body fat, respectively, were utilized for energy during fasting. Body weight and BMI were reduced because of the increased muscle protein breakdown and body water excretion during early stage of fasting, but the significant body fat loss was after 9 days fasting. Therefore, it could be suggested to fast for longer than 10 days if the reduction of body fat was planned by fasting, and recommed to exercise and ingest more vitamins and minerals to replace the excreted minerals during fasting. Chemical toxicants are metabolically converted to numerous metabolites in the body. Toxicokinetic characteristics of metabolites could be therefore used as biomarker of exposure for human risk assessment. Biologically based dose response (BBDR) model was proposed for future direction of risk assessment. However, this area has not been developed well enough for human application. Benzo(a)pyrene (BP), for example, is a well-known environmental carcinogen and may produce more than 100 metabolites and BPDE-DNA adduct, a covalently bound form of DNA with benzo(a)pyrene diolepoxides (BPDES), has been applied to qualitatively or quantitaively estimate human exposure to BP. In addition, di(2-ethylhexyl) phthalate (DEHP), a widely used plasticize. in the polymer industry, is one of endocrine-disrupting chemicals (EDCs) and has been monitored in humans using urinary or serum concentrations of DEHP or its monomer MEHP for exposure and risk assessment. However, it is difficult to estimate the actual level of toxicants using these biomarkers in humans using. This presentation will discuss a methodology of exposure and risk assessment by application of metabolic profiling kinetics.
A 42-d study with 384 Hy-line brown laying hens was conducted to assess the effects of dietary octacosanol supplementation on laying performance, egg quality and blood metabolites of laying hens. Hens were randomly allocated into 4 dietary groups of 8 cages each, which were fed basal diet supplemented with 0 (Control), 9 (OCT9), 18 (OCT18), and 27 (OCT27) mg/kg diet of octacosanol isolated from rice bran, respectively. The experiment was conducted in an environmental controlled house and hens were fed twice daily for ad libitum intake. Laying performance was determined over the 42-d period, and egg quality as well as blood metabolites were estimated on d 21 and d 42. Diets in OCT18 and OCT27 increased (p<0.05) laying rate, egg weight, egg mass, egg albumen height, Haugh unit and eggshell strength on d 42, but decreased (p<0.05) feed conversion rate and levels of total cholesterol, triglyceride and low density lipoprotein cholesterol in the serum as compared to those of Control. Feed intake, yolk color, yolk diameter, eggshell thickness and high density lipoprotein cholesterol were similar (p>0.05) among treatments. Results demonstrate that supplementing 18 to 27 mg/kg diet of rice bran octacosanol can improve laying rate and egg quality and reduce blood lipid of laying hens.
This study characterized the effect of liver injury produced by hepatotoxicants on the biliary and urinary excretion of acetaminophen(AA) metabolites. Liver damage was produced in male S.-D. rats, 24 hr after dosing with carbon tetrachloride(4CCl_4,$ 0.75 mι/kg, ip) or thioacetamide(TA, 200 mg/kg, ip), or 16 hr after administration of cadmium chloride(4CdCl_2,$ 3.9 mg/kg, iv). Liver damage without renal injury was confirmed by measuring serum enzymes, creatinine and BUN levels as well as by histopathological examination. AA and its metabolites were measured for 3 hr by HPLC in rats injected iv with 1 mmo1/kg of AA. The excreted amounts of AA-glucuronide into bile were reduced to 60~70% of control rats by hepatotoxicants, but did not change urinary excretion of AA-glucuronide and AA-sulfate. Treatments with $CCl_4,\; CdCl_2$ and TA decreased the total (biliary plus urinary) excretion of thioethers of AA(30~50% of control), suggesting that these toxicants decrease cytochrome P-450-mediated toxification of AA. However, treatments of $CdCl_2$and TA markedly enhanced the excretion of AA-mercapturate into urine. Thus, 4CdCl_2$ and TA not only influence the formation of AA-glutathione, but may also alter the excretory routes (i.e. bile and urine) for the elimination of AA-metabolite.

The aim of this experiment was to determine the effect of a high-energy diet on the level of serum metabolites and on carcass characteristics of Hanwoo steers. High energy diets have been used for enhancing intramuscular adipose tissue in high quality beef cattle. However, there is not much information about the physiological reactions to this diet. We hypothesized that a high energy diet would increase blood metabolites and the meat quality of Hanwoo steers during the early and final fattening periods. A $2{\times}2$ factorial arrangement (High, Control, and Early, Final) in a completely randomized design was used to feed 24 Hanwoo steers. Two steers were kept in the same pen and 12 pens were used for the experiment. Blood was drawn from each steer on the first week of every other month from 11 to 28 months. Overall Average Daily Gain (ADG) and feed efficiency were not different between high energy and control diets (p > 0.05). However, Dry Matter Intake (DMI greater with the control diet than DMI with the high energy diet during the final fattening period (p < 0.05). Serum non-esterified fatty acid (NEFA) concentrations were greater in the control diet group than in the high energy diet group during the final fattening period (p < 0.05). However, serum albumin, glucose, total protein, triglyceride, and phosphorus were greater in the high energy group than those of the control group (p < 0.05). Carcass traits or physico-chemical characteristics were not different between high energy diet treatment and the control. These data indicated that a high energy diet (+ 3% TDN) increased serum triglyceride during early fattening periods and decreased non-esterified fatty acids during final fattening periods in Hanwoo steers.
