Objectives : This study was performed to investigate the effects of Rhizoma Arisaematis water extract on lipid and glucose metabolism and histochemical change of obese rats. Methods and materials : 10 rats were divided into normal, control and RA (Rhizoma Arisaematis) groups. We fed the control group a high-fat diet and administered normal saline for 8 weeks. We fed the experimental group of rats a high-fat diet and administered an extract of Rhizoma Arisaematis for 8 weeks. At the end of the experiment, the rats were sacrificed to determine their chemical composition. The groups were examined for effects on blood serum lipids, blood sugar, blood insulin concentration and epididymal fat cells. Results : 1. Serum total cholesterol, triglyceride, HDL-cholesterol and glucose of the RA group decreased compared with those of the control group. These decreased rates were significant(P<0.05). 2. Serum LDL-cholesterol, total lipid, free fatty acid and the average size of epididymal fat cells of the RA group decreased compared with those of the control group. These decreased rates were significant(p<0.01). Conclusions : These results suggest that RA may be used to prevent or cure the obesity iniduced by a high-fat diet.
This study was performed to investigate the effect of various levels of Zn(0, 30, 3000 ppms) and different kinds of lipid(corn oil, sesame oil or butter.) in the diet upon lipid metabolism and Zn & Cu concentration in various organs and tissues in weanling rats. The results obtained were summarized as following : 1) Food consumptions, body weight gains and F.E.R.s showed no significant differences among groups. 2) The contents of total lipids and total cholesterols in serum tended to increase with increase in lipid saturation and Zn levels. Those in per g liver were higher in butter groups and tended to be higher in low Zn groups. Total lipids contents in per g muscle tended to be higher in butter and high Zn groups, but total cholesterols contents in per g muscle tended to be higher in corn oil and low Zn groups. 3) Liver, serum and fecal Zn concentrations tended to be higher in high Zn groups than other Zn groups. The Cu concentrations in liver and serum tended to be lower in high Zn groups. On the contrary, the Cu concentrations in urine tended to be higher in high Zn groups.
Adiponectin is secreted specifically by adipose tissue and regulates lipid and glucose metabolism. In addition, adiponectin has been found to inhibit inflammatory process and possibly atherogenesis. This study was done to compare hematological parameters, serum lipids with serum adiponectin level according to body mass index (BMI) and gender in eighty eight Korean adults aged 40 to 68. In this study, the association between serum adiponectin and other variables including RBC counts, hemoglobin, hematocrit and albumin were investigated. RBC, hemoglobin status, was inversely associated with serum adiponectin levels in normal adult women. Adiponectin levels were significantly lower in men than women (p<0.01). In men, serum adiponectin levels were negatively correlated with body mass index (BMI) (p<0.05). However, no correlations were found in women. These results might imply that the regulation of key adipokines such as adiponectin might be a strategy for the prevention or treatment of obesity-associated diseases.
This study is to investigate the effect of dietary L-carnitine supplementation on lipid metabolism in rats fed with isolated soy protein and casein for their source of protein. Four experimental groups were organized and each group had eight Sprague-Dawley male rats with the initial weight of around 180g. The four groups were CO (casein only supplemented group); CC (casein and 3% L-carnitine supplemented group); ISO (isolated soy protein only supplemented group); ISC (isolated soy protein and 3% L-carnitine supplemented group). All groups were supplemented with the experimental diet for four weeks and carnitine comprised 3% of. their diet. The results were as follows; 1. There was no significant difference in food intake among the groups. 2. Final weight gain was significantly lower in the groups supplemented with isolated soy protein than in the groups supplemented with casein (P<0.05). The groups with supplemented casein and carnitine showed the effect of weight reduction (p<0.05). 3. Food efficiency ratio was lower in the groups supplemented with isolated soy protein than in the groups supplemented with casein (p<0.01). The groups supplemented with casein and carnitine showed low food efficiency ratio. 4. The serum total lipid was higher in the groups supplemented with casein than in the groups supplemented with isolated soy protein (p<0.05). 5. Serum total cholesterol was higher in the groups supplemented with casein than in the groups supplemented with isolated soy protein. 6. There was no significant difference in triglyceride, HDL-cholesterol, and LDL-cholesterol in serum among the groups. 7. Out of the groups supplemented with isolated soy protein the total cholesterol level in liver was low in the groups to which carnitine was supplemented (p< 0.05). However, there was no significant difference of liver total lipid and triglyceride among the groups. 8. There was no difference in TBARS levels and GSH-Px activities in liver among the groups.
This study investigated the effects of Gymnema sylvestre (GS) extract on body weight, adiposity, and lipid metabolism in leptin-deficient ob/ob mice. The experimental mice were divided into the following 4 groups: basal diet (AIN-93G diet) and 0.5%, 1%, and 1.5% GS supplemented groups. Each group was fed the experimental diet for 9 weeks. The final body weights, adipocyte sizes, and epididymal fat weights of the GS groups were significantly lower than those of the control group. There were no significant differences in food intake and food efficiency ratios among the treated groups. Serum triglyceride levels and the atherogenic index were significantly lower in the GS groups compared to the control group. Serum HDL cholesterol levels were significantly higher in the 1% and 1.5% GS groups compared to the control group. Serum glutamate oxaloacetate transaminase and glutamate pyruvate transaminase activities did not differ among the four experimental groups. The total hepatic lipid content was significantly lower in the GS groups, and hepatic cholesterol and triglyceride contents tended to be reduced in the GS groups compared to the control group. These results suggest that GS extract may be useful for ameliorating dyslipidemia and fatty liver.
BACKGROUNG/OBJECTIVES: Corn silk (CS) extract contains large amounts of maysin, which is a major flavonoid in CS. However, studies regarding the effect of CS extract on cholesterol metabolism is limited. Therefore, the purpose of this study was to determine the effect of CS extract on cholesterol metabolism in C57BL/6J mouse fed high-fat diets. MATERIALS/METHODS: Normal-fat group fed 7% fat diet, high-fat (HF) group fed 25% fat diet, and high-fat with corn silk (HFCS) group were orally administered CS extract (100 mg/kg body weight) daily. Serum and hepatic levels of total lipids, triglycerides, and total cholesterol as well as serum free fatty acid, glucose, and insulin levels were determined. The mRNA expression levels of acyl-CoA: cholesterol acyltransferase (ACAT), cholesterol 7-alpha hydroxylase (CYP7A1), farnesoid X receptor (FXR), lecithin cholesterol acyltransferase (LCAT), low-density lipoprotein receptor, 3-hyroxy-3-methylglutaryl-coenzyme A reductase (HMG-CoA reductase), adiponectin, leptin, and tumor necrosis factor ${\alpha}$ were determined. RESULTS: Oral administration of CS extract with HF improved serum glucose and insulin levels as well as attenuated HF-induced fatty liver. CS extracts significantly elevated mRNA expression levels of adipocytokines and reduced mRNA expression levels of HMG-CoA reductase, ACAT, and FXR. The mRNA expression levels of CYP7A1 and LCAT between the HF group and HFCS group were not statistically different. CONCLUSIONS: CS extract supplementation with a high-fat diet improves levels of adipocytokine secretion and glucose homeostasis. CS extract is also effective in decreasing the regulatory pool of hepatic cholesterol, in line with decreased blood and hepatic levels of cholesterol though modulation of mRNA expression levels of HMG-CoA reductase, ACAT, and FXR.
The present study was designed to investigate the effect Hypericum Perforatum (HP), on behavioral changes, corticosterone, TNF-${\alpha}$ levels and tryptophan metabolism and disposition in bilateral ovariectomized rats compared to $17{\alpha}$-ethinylestradiol. Behavioral analysis by measuring immobility time in forced swimming test and open field test, serum and hippocampal corticosterone and TNF-${\alpha}$ along with hippocampal kynurenine/tryptophan ratio were determined in mature ovariectomized rats treated orally either by HP at three different doses 125, 250, and 500 mg/kg/day or by $17{\alpha}$-ethinylestradiol $30{\mu}g/kg/day$ for 30 days. Ovariectomized rats showed significant increase in immobility time in the forced swimming test. Along with elevation in serum and hippocampal TNF-${\alpha}$ and corticosterone levels associated with significant increase in hippocampal kynurenine/tryptophan ratio. Immobility time in the forced swimming test was decreased in rats treated by different doses of HP in a dose dependent manner and $17{\alpha}$-ethinylestradiol with no concomitant changes in the open field test. Only Rats treated with HP exhibited significant decrease in the elevated serum and hippocampal TNF-${\alpha}$ and corticosterone, which couldn't explain the associated insignificant effect on hippocampaus kynurenine/tryptophan ratio in comparison to ovariectomized untreated rats. It is concluded that increased tryptophan metabolism toward kynurenine secondary to elevated corticosterone and TNF-${\alpha}$ might be one of the pathohphysiological mechanisms that could explain depression like state observed in this rat model. Further, the observed attenuating effect of HP on TNF-${\alpha}$ and corticosterone could contribute in its antidepressant effect in this animal model by other ways than their effects on tryptophan-kynurenine metabolism pathway.
This study was performed to investigate the effect of chongkukjang intake on lipid metabolism and liver function in ethanol consumed rats. Twenty one Sprague-Dawley male rats aging 4 weeks old were used as experimental animals, which were divided into three dietary groups; casein diet(CA), soybean diet(SB) and chongkukjang diet(CJ). Alcohol was consumed with water as 25%(v/v) ethanol solution. After 4 weeks of experimental period, rats were sacrificed to get blood and liver samples for analysis of lipids, lipid peroxides, antioxidative enzymes and biochemical indices of liver function. The mean body weight, food intake and liver index were not significantly different among three groups. Serum level of total lipid, triglyceride and total cholesterol of chongkukjang diet group was the lowest among three groups although the difference was not significant. HDL-cholesterol level was significantly(p<0.05) higher in chongkukjang diet group than that of casein diet group. LDL-cholesterol level of chongkukjang and soybean diet group was significantly(p<0.05) lower than that of casein diet group respectively. Liver TBARS of chongkukjang and soybean diet group was significantly(p<0.05) lower than that of casein diet group respectively. The superoxide dismutase activity of chongkukjang diet group was significantly(p<0.05) higher than that of casein diet group. Catalase activity was not significantly different among three groups. As indices of liver function, glutamic oxaloacetic transminase(GOT), glutamic pyruvic transminase(GPT), $\gamma$-glutamyl transpeptidase($\gamma$-GTP) and lactate dehydrogenase(LDH) were not significantly different among three groups. Serum alcohol concentration and activities of alcohol dehydrogenase(ADH) and aldehyde dehydrogenase(ALDH) were not significantly different among three groups. The chongkukjang diet seems to give a beneficial effect for improving lipid metabolism by increasing HDL-cholesterol level and SOD activity while reducing liver TBARS level. However, effect on liver function has to be investigated further.
This study was conducted to investigate the supplementary effects of Suwon 464 developed by Rural Development Administration, which has over two times of dietary fiber content compared with normal rice (Ilpum), on lipid metabolism in diabetic mice. We supplied 5 kinds of experimental diets (com starch diet as a control (CO), Ilpum polished rice diet (IP), Ilpum brown rice diet (IB), polished rice diet (SP) and brown rice diet (SB) of Suwon 464) to diabetic mice for 8 weeks, after analyzing dietary fiber contents of 5 experimental diets. Diet intake, body weight, organ weights, and lipids levels of serum, liver and feces were measured. The dietary fiber contents in CO, IP, IB, SP, and SB diets were 1.0, 1.2, l.4, 1.4, and 2.0% respectively. Body weight and liver and epididymal fat pad weights were lower in SB group than the other groups though there was no significant difference in diet intake among experimental groups. The concentrations of serum triglyceride was lower in SP and SB groups than CO and IP groups. The levels of hepatic total lipid and total cholesterol were significantly lower in SP and SB groups than CO group, and the level of hepatic triglyceride was lower in IB, SP and SB groups than CO group. The levels of total lipid and triglyceride excreted in feces were higher in IB, SP and SB, and the level of total cholesterol in feces was higher in SP and SB groups than CO group. These results suggested that the high dietary fiber rice (Suwon 464) decrease the triglyceride or total cholesterol concentrations of serum and liver by increasing of fecal lipid excretion in diabetic mice. The purpose of this study was to investigate the effect of enzyme-treated radish (Raphanus sativus L.) leaves on lipid metabolism in rats fed a high fat diet. Rats were divided into four experimental groups which were composed of a high fat diet group (HF group), a high fat diet with 10% radish leaf powder-supplemented group (MA group), a high fat diet with 5% enzyme-treated radish leaf powder-supplemented group (MB group) and a high fat diet with 10% enzyme-treated radish leaf powder-supplemented group (MC group). Total dietary fiber content of enzyme-treated radish leaves were greater than untreated radish leaves. Body weight gain and food efficiency ratio (FER) of the HF group increased compared to the MA, MB and MC groups. The serum total cholesterol, LDL-cholesterol and atherogenic index contents in the radish leaf powder-supplemented groups were lower than that of the HF group, while those values for the MB and MC groups were significantly lower than that of the HF group. The serum HDL-cholesterol contents of the MB and MC groups increased compared to the HF group. The hepatic triglyceride contents of the MA, MB and MC groups decreased compared to the HF group. In fact, the hepatic triglyceride contents of the MB and MC groups were significantly lower than the MA group. The hepatic total cholesterol contents of the MB and MC groups significantly decreased compared to those of the HF group. The fecal total cholesterol contents of the MA, MB and MC groups significantly increased compared to those of the HF group. These results indicate that supplementation with enzyme-treated radish leaves increase the useful fiber contents. Furthermore, it may have a pronounced impact on lipid metabolism of serum and liver in rats fed a high fat diet.
