神朮散이 알레르기 비염 유발 흰쥐 모델에 미치는 영향 (The Effect of Shinchulsan on the Rat Model with Allergic Rhinitis)

  • Objectives : Allergic rhinitis is highly increased in these days. We aimed to investigate therapeutic effect ofShinchulsanby observing changes in blood cells and the protein expression level of IL-4 in spleen of Sprague-Dawley(SD) rats with allergic rhinitis.Methods : Fifteen SD rats were divided into three groups: normal, control, and sample group. Allergic rhinitis was induced in the control and sample group by intraperitional and intranasal sensitization with 0.1% and 0.4% Ovalbumin solution. ThenShinchulsanwas orally administered only to the sample group for 28 days, while the rats in the control group was given normal saline.Results : 1.Shinchulsanshowed significantly decreased eosinophil level on the serum of the rat model.2.Shinchulsanshowed significantly decreased IgE level on the serum of the rat model.3.Shinchulsanshowed significantly decreased the protein expression levels of IL-4 in spleen of the rat model.4.Shinchulsanhad no hepatoxicity, according to AST and ALT on the serum.Conclusion : According to the above results, it is considered thatShinchulsanis helpful in treatment of allergic rhinitis.

  • This studies were to investigate the effects of low power wavelengths Helium-Neon Infra-Red(He-Ne 1R) laser on the changes of the serum immunoblobulin(Ig) concentrations in bum rats, The thirty Spraque-Dawley adult male rats were assigned to the 5 groups: the experimental groups(3). the bum control group(1) and the control group(1). There was made three degree burn by the 290 mW IR on the back oreach rats, from 3 days after being burned. the experimental laser groups were irradiated low power wavelengths(292Hz- 1168Hz, 4672Hz) He-Ne IR laser far 5 minutes every day during the 7days. The results were as follows; 1. The concentrations of immunoglobulin G(Ig G) in serum of all experimental groups on the treated with the low power wavelengths of the He-Ne IR laser during the 5 minutes far 7 days were significantly lower than those of bum groups, but those 1168 Hz group were significantly higher than those of the 292 Hz and 4672 Hz groups. 2. There were significantly decreased the change of the level of immunoglobulin M(Ig M) in serum of all experimental groups on the treated with 5 minutes laser fer 7 days to the burn control group. and were significantly increased on treated with more higher wavelengths groups to those of lower groups. 3. The concentrations immunoglobulin E(Ig E) in serum of the 292Hz wavelengths groups were significantly lower than those in bum control group. As above results. the changes of the immunoglobulin in serum levels on the healing process have meaningful effected with the low power wavelengths role on the treated with He-Ne IR laser.

인삼(人蔘)추출물이 Ovalbumin으로 유도된 천식(喘息)의 Cytokine에 미치는 영향 (Effects of Ginseng Radix Extract on the Cytokine of Experimental Asthma induced by Ovalbumin)

  • This study was performed to investigate the effect of oral administration of GSRE against the asthma. Asthma was induced to Balb/c mouse by i.p. injection and aerosol immunization with ovalbumin. It was observed the change of the eosinophil number in the BALF. Concentrations of IL-4, IL-5 in BALF and splenocyte were assessed by ELISA, IgG and IgE from serum were calculated by same method. Concentration of IL-4 in splenocyte was significantly decreased in GSRE group compared with control group. Concentrations of IL-5 from BALF and splenocyte were significantly decreased in GSRE group compared with control group, respectively. Level of IgE in serum was significantly decreased in GSRE group compared with control group, but not IgG. We found that the effect of GSRE extract in asthma was implicated in reductions of IL-4, IL-5 released from Th2 cell, and decreses of IgE, from plasma cell. These findings suggest that GSRE extract can produce anti-asthmatic effect, which may play a role in allergen-induced asthma therapy.

BALB/c mice에서 quercetin의 경구투여가 picryl chloride로 유발된 접촉성 피부 알레르기의 예방에 미치는 영향 (Allergy Prevention Effect of Oral Administration of Quercetin on Picryl Chloride-induced Contact Dermatitis in BALB/c Mice)

  • 본 연구를 통해 BALB/c 마우스에서 quercetin의 경구투여가 PCL로 유도된 접촉성 피부 알레르기의 예방에 미치는 효과에 대해 알아보았다. Quercetin을 투여 농도에 따라 대조군(0 mg/kg), 저용량군(50 mg/kg), 고용량군(100 mg/kg)으로 나누고 8일에 걸쳐 총 8번 경구투여를 실시한 후 알레르기 유발물질인 PCL을 마우스의 양쪽 귀에 감작시켜 접촉성 피부 알레르기를 유발 시켰다. Quercetin의 투여 농도별 ear swelling 변화를 확인한 결과 quercetin을 투여하지 않은 대조군에 비해 quercetin을 투여한 군의 ear swelling 증가폭이 낮게 나타났고, 100 mg/kg (고용량군)에서 농도 유의적으로 ear swelling의 증가폭이 현저히 낮게 나타났다. Quercetin의 투여 농도에 따른 혈청 내 염증성 매개 물질의 농도 변화를 알아보기 위한 IgE 및 histamine level 측정 결과에서 quercetin을 투여하지 않은 대조군에 비해 quercetin을 투여한 50 mg/kg (저용량군), 100 mg/kg (고용량군)에서 낮은 수준의 IgE, histamine 수치가 나왔다. H&E염색과 Toluidine blue stain을 통한 조직병리학적 검사결과에서 quercetin을 투여한 고용량군의 귀 두께가 대조군에 비해 얇게 관찰되었고, 비만세포의 유무를 알아보기 위한 Toluidine blue stain의 결과 대조군에 비해 고용량군에서 적은 수의 비만세포들이 관찰되었다. 따라서 ear swelling과 IgE, histamine level, 조직병리학적 결과를 종합해 봤을 때 식물성 flavonoid 성분인 quercetin은 접촉성 피부 알레르기의 예방에 상당한 효과가 있다고 판단되며, 부작용이 발생하는 기존의 치료제를 대체할 수 있는 후보 물질로써 중요한 가치가 있다고 사료된다.

항 아토피 화장품이 아토피 동물모델 Hairless Mice에 미치는 영향 (The Effect of Anti-atopic Cosmetic in Hairless Mice)

  • 이 연구는 아토피 피부염이 유발된 hairless mice에 항 아토피 제품 처리를 통해 아토피 증상 개선 효과에 대해 알아 본 실험이다. 항 아토피 물질인 ceramide와 astaxanthin을 melt-homogenization 방법을 이용해 고형 지질 나노입자를 제조하고 입자 주위에 silk fibroin을 코팅시켰다. 동시에 고형 지질 나노입자를 함유하는 항 아토피 로션(lotion)과 크림(cream)을 만들어 타 사의 항 아토피 제품과 비교하였다. 아토피 피부염이 유발된 마우스의 외형적인 모습 관찰을 통해 ceramide와 silk fibroin을 함유한 항 아토피 제품인 FL과 FC는 아토피 피부염으로 손상된 피부 장벽을 재생 시키는 것을 확인하였고, 아토피 피부염이 유발된 피부 두께 역시 감소됨을 알 수 있었다. 면역학적 평가를 통해 silk fibroin과 astaxanthin 이 Th2 cytokine인 IgE와 IL-4의 발현을 억제하고 $IFN-{\gamma}$의 발현을 증가시켜 Th cell 사이의 균형을 이루게 하는 것이 밝혀졌다. 또한, 비장수치를 통해 T 림파구의 따른 IgE, IL-4 발현 억제능, 그리고 $IFN-{\gamma}$ 발현을 확인하였다.

폐흡충(Paragonimus westermani) 감염이 흰쥐의 IgE 생성 및 비장림프구 아군분포에 미치는 영향 (Changes of IgE production, splenic helper and suppressor T lymphocytes in mice infected with Paragonimus westermani)

  • 폐흡충 피낭유충을 BALB/c 마우스에 경구 감염시킨 후 마우스를 시기별(3일, 1주, 2주, 3주, 4주, 5주, 7주, 9주, 13주 및 23주)로 희생시켜 말초혈액내 호산구수와 혈청내 IgE 항체가를 측정하는 한편 비장 림프구를 분리하여 림프구 아군(helper T, cytotoxic / suppressor T, pan T)에 대한 단세포군 항체를 결합시켜 EPICS C fluorocytometer로 분석하였다. 폐흡충에 대한 혈청내 총 IgE 항체가는 감염 3주부터 증가되어 감염 4주에 높게 증가되었으며 13주 이후 23주까지 높은 항체가가 유지되었다. 말초혈액내 호산구수는 감염 2주 부터 증가되어 감염 9주에 $664.1/\textrm{mm}^3$으로 가장 증가 되었으며 그 이후에는 감소하여 23주에는 정상보다 약간 증가되었다. 폐흡충을 감염시키고 감염 3일, 1주, 2주, 3주, 4주, 7주 및 23주에 T 림프구 아군을 분석한 결과 L3T4 helper T 림프구와 Lyt-2 cytotoxic / suppressor T 림프구의 비율은 감염 군에서 감염 초기에 대조군에 비해 약간 감소된 경향을 보였으나 대조군과 유의한 차이를 보이지 않았다. Thy 1.2 pan T 림프구의 비율은 감염 2주와 4주에 대조군에 비해 유의하게 감소되었고(P<0.05) 7주 이후에는 대조군과 비슷하였다. 이러한 결과들로 보아 마우스의 폐흡충감염에서 특이 항원이나 마이토젠의 자극이 없이는 비장킬프구의 아군분포에 큰 변화가 없는 것으로 생각된다. 생각된다.

개의 아토피성 피부염에 대한 집먼지 진드기 항원 특이적 IgE와 림프구 증식 반응의 증가 (Increased Allergen-specific IgE Values and Lymphocyte Proliferation Response to House Dust Mites in Dogs with Atopic Dermatitis)

  • 혈중 항원 특이적 IgE 검사와 피내시험으로 집먼지 진드기 (house dust mites, HDM)에 양성 반응을 보인 20두의 아토피성 피부염으로 진단된 개를 대상으로, 말초혈단핵구 (peripheral blood mononuclear cells, PBMC)를 채취하여 HDM항원에 대한 반응을 검토하였다. PBMC를 분리하여 HDM항원으로 자극한 결과 20두 중 13두 (65%)에서 항원 특이적이 림프구의 증식반응을 확인할 수 있었다. HDM 항원에 증식반응은 아토피성 피부염군에서 대조군에 비해 유의적으로 높은 반응이 확인되었다 (P=0.007). 또한, HDM에 대한 반응은 혈중의 IgE 농도와 유의적으로 상관관계를 나타내었으며 (P=0.035), 이는 체내에서 항원 특이적인 IgE의 생산을 촉진하는 작용을 반영하는 지표가 될 수 있다고 생각되었다. 이러한 결과로, 말초혈액중에 존재하는 HDM 항원 특이적인 림프구는 개의 아토피성 피부염의 병태생리에 관여하고 있는 것으로 시사되었다.

생맥산(生脈散)과 생맥산가미방(生脈散加味方)이 NC/Nga 마우스의 아토피 피부염에 미치는 영향에 대한 비교 연구 (A Comparative Study on the Effects of Saengmaeksan and Saengmaeksan-gamibang on Atopic Dermatitis in NC/Nga mouse)

  • Objective : This study was carried out to compare Saengmaeksan-gamibang(SG) to Saengmaeksan(SM) on the efficacy of moisturization, skin whitening, anti-inflammation, and anti-allergy in atopic dematitis using NC/Nga mice model. Methods : We assessed the effects of SG and SM on tyrosinase, filaggrin, serine-palmitoyl transferase(SPT), COX-2, AP-1 in vitro, on IgE, IL-4, IL-5, IL-6, IgM, IgG1, IFN-${\gamma}$ in vivo with NC/Nga mice. Results : 1. SG increased the tyrosinase inhibition activity and the levels of filaggrin and SPT, decreasing the level of COX-2. 2. On the other hand, SM decreased the tyrosinase inhibition activity and the levels of filaggrin and SPT, increasing the level of COX-2. 3. Serum IgE, IL-4, 5, 6, IgM, and IgG1 were decreased in both SG group and SM group compared to the control group. The decrease degree in these factors was higher in SG than in SM. 4. Serum IFN-${\gamma}$ was increased in both SG group and SM group compared to the control group. The increase degree in IFN-${\gamma}$ was higher in SG than in SM. Conclusion : As the result of the above experiments, it was proved that SG has the moisturization, skin whitening, anti-inflammation, and anti-allergy effects, which are greater than those of SM, and provides the significant efficacy on atopic dermatitis treatment.

보중익기탕가미방(補中益氣湯加味方)과 마황신궁산(麻黃辛芎散) 병용이 알레르기 비염 유발 흰쥐 모델에 미치는 영향 (The Effects of Bojungikgitang-gamibang Administration along with Mahwangshingungsan on the Rat Model with Allergic Rhinitis)

  • Objectives : We aimed to investigate therapeutic effect of Bojungikgitang-gamibang(BI) and Mahwangshingungsan(MS) by observing changes in blood cells and the nasal mucosa of Sprague-Dawley(SD) rats with allergic rhinitis. Methods : Twenty-four SD rats were divided into three groups: normal, control, and sample group. Allergic rhinitis was induced in the control and sample group by intraperitoneal and intranasal sensitization with 0.1% and 0.4% Ovalbumin solution. Then BI was orally administered only to the sample group along with MS for 28days, while the rats in the control group was given normal saline. Results : BI and MS showed significantly decreased IgE level on the serum of the rat model, Bl and MS showed significantly decreased eosinophil level on the blood of the rat model. BI and MS inhibited the inflammatory reaction on the nasal mucosal tissue, according to nasal mucosal biopsy. Bl and MS had anti-allergic according to level, eosinophil level, nasal mucosal biopsy. BI and MS had no hepatoxicity, according to AST and ALT on the serum. Conclusion : According to the above results, it is considered that Bl and MS is helpful in treatment of allergic rhinitis.

활혈막성방(活血膜性方)이 Cationized Bovine Serum Albumin투여로 유발된 Mouse의 Membranous Nephropathy에 미치는 영향 (The Effects of Hwalhyulmaksung-bang (Huoxiemoxing-fang) on Membranous Nephropathy Induced by Cationic Bovine Serum Albumin in Mouse Model)

  • Objective: The purpose of our study was to show the effects of Hwalhyulmaksung-bang (Huoxiemoxing-fang, HHMSB) treatment on cBSA-induced in MN Mouse Model. Methods: We divided the 20 mice into 4 groups. One group, named NR, was not treated. The second group, named CT, was treated with cBSA (7mg/kg i.p) only. The third group, named HH-250, was treated with cBSA (7mg/kg i.p) and HHMSB extract (250mg/kg, p.o). The fourth group, named HH-500, was treated with cBSA (7mg/kg i.p) and HHMSB extract (500mg/kg, p.o). 4 weeks after cBSA, proteinuria, serum albumin, total cholesterol, serum creatinine, BUN, total cell number of spleen and kidney of all groups were measured. CD3e+/CD19+ and CD4+/CD8 cells ratio of peripheral blood, kidney and spleen of all groups were analyzed. $IL-1{\beta}$ and TNF-$\alpha$, IL-6, IgG, IgM, and IFN-$\gamma$ levels of all groups were gauged. Histological analysis of kidney tissue and immunohistochemical staining (CD4, CD8) of kidney were observed. Results: The level of proteinuria significantly decreased and serum albumin increased in the group treated with cBSA and HHMSB extract compared with the control. Total cholesterol decreased but not significantly. CD3e+/CD19cells ratio of peripheral blood is decreased, but CD4+/CD8cells ratio has no significancy. CD3e+/CD19+ and CD4+/CD8 cells percentage of kidney and spleen has no significancy. Level of $IL-1{\beta}$, IL-6 is significantly decreased, and IFN-$\gamma$ is significantly increased on HHMSB compared with control. Total IgG level significantly decreased on HHMSB compared with the control. Thickness of GBM decreased on histological analysis of kidney. Deposition of CD4 and CD8 decreased on immunohistochemical staining of kidney. Conclusions: We conclude that Hwalhyulmaksung-bang treatment may could be a useful remedy agents for treating Membranous Neuropathy(MN) induced by cationized bovine serum albumin.
