Perfluorooctanoic acid (PFOA), a member of the perfluoroalkyl acids that have wide commercial applications, is persistent organic pollutants widely spread throughout the environment and human population. But little is known about the adverse biological effects of the PFOA. In the present study, the toxicological effects of PFOA were investigated in rats. Sprague-Dawley rats (N = 10 in each group) were orally administered with PFOA in drinking water for 4 weeks (0, 100, 200, or 400 ppm in male, and 0, 200, 400, or 800 ppm in female). Three female rats given 800 ppm died during the study. PFOA treatment decreased the body weight gain and increased the liver weights in both genders. Serum biochemical investigations revealed significant increases in the aspartate aminotransferase, alanine transaminase, alkaline phosphatase, blood urea nitrogen, total cholesterol, and total bilirubin in male but in female. Serum estradiol (E2) levels were increased in all treated rats. Histopathologically, hepatocellular hypertrophy around central vein was noted in the liver of treated rats. No significant histopathological changes were noted in other organs. In conclusion, PFOA induced toxicological changes in the liver and increased serum E2 level which was not related to histopathological changes of endocrine and reproductive system.
This studies were to investigate the effects of low power wavelengths Helium-Neon Infra-Red(He-Ne IR)laser on the changes of the blood biochemical components in burn rats. The thirty Spraque-Dewely adult male rats were assigned to the 5 groups; the experimental groups(3), the burn control group(1) and the control group(1). There was made three degree burn by the 250mW IR on the back of each rats, from 3 days after being, burned, the experimental laser groups were irradiated low power wavelengths (292Hz, 1168Hz, 4672Hz) He-Ne IR laser for 5 minutes every day during the 7days. The results wee as follows: The activities of aspirate aminotransferase(AST), alanine aminotransferase(ALT), and lactate dehydrogenase(LDH), on treated with the wavelengths ((292Hz, 1168Hz, 4672Hz) laser groups during 5 min. for 7 days were significantly increased to the control group respectively, but those of total cholesterol(CHOL) and alkaline phosphatase(ALP) were significantly decreased to the a control group (p<0.05). there were significantly decreased on the activity of alanine aminotransferase and alkaline phosphatase on the all experimental groups to the burn control group and those of lactate dehydronase on treated with 292Hz and 4672Hz wavelength9 groups to the burn control group but, were significantly increased on those of urea nitrogen (292Hz and 1168Hz)(p<0.05). The activity of AST, LDH, on treated with 1168Hz wavelengths were significantly decreased of the 292Hz and 4672Hz wavelengths groups that of ALT on the treated with 1168Hz group was significantly increased to the 4672 Hz wavelength group but those of CHOL and LDH on the 1168Hz and 4672Hz wavelength groups were dignificatly decreased to the MHz wavelengths group. As above results, the changes of the activities of biochemical conponents in the serum levels on the healing process have meaningful effected of the role of low power wavelengths He-Ne IR laser.
Severe protein-calorie malnutrition, common in patients with advanced liver disease, can seriously undermine the capacity for regeneration and functional restoration of liver. Nutritional supplementation for these patients can improve biochemical and hormonal abnormalities. However, these effects were not identified in patient with nonalcoholic liver cirrhosis. To determine effects of nutritional supplementation in patients with nonalcoholic liver cirrhosis, 77 subjects aged 29 to 69 years participated in this study for 12 weeks and were subdivided into three groups; normal diet group (Control group, n = 16), branched-chain amino acid supplementation group (BCAA group, n = 31), nutritional supplementation group (NS group, n = 30). Anthropometric parameters, hemoglobin, hematocrit, blood cell counts, serum levels of lipids, vitamins, minerals and fatty acid composition, and plasma amino acids were examined. The mean values of age and height, and the initial values of weight and body mass index (BMI) were not different among all groups. After 12 weeks, there were no significant changes in these values in Control group. Only NS group showed significant increases in weight, lean body mass, midarm circumference, triceps skinfold thickness. Serum transferrins were increased both in BCAA and NS groups. Plasma levels of branched-chain amino acids, urea amino acids and glutamic acid were also significantly increased in these groups, but plasma levels of ammonia, serum LDL cholesterol and atherogenic index were decreased. However, there were no significant changes in serum levels of vitamin and mineral and composition of fatty acids in phospholipids in these groups. These results showed that the nutritional supplementation for patients with nonalcoholic liver cirrhosis can more improve nutritional status in these people together with increases of weight, body fat and lean body mass, compared to only BCAA supplementation. To ascertain and investigate the appropriate nutritional supplementation for patients with nonalcoholic liver cirrhosis, further studies are necessary.
Background: Tumor markers, designated as a broad group of substances produced by malignancies, could be in the form of biochemical substances, immunological substances, cell surface changes and genetic alterations. Cancer, a disorder of cellular behavior is characterized by alteration of serum glycoproteins. L-fucose, a hexose, which is the terminal sugar in most of the plasma glycoproteins, may be useful as a tumor marker for the detection, monitoring and prognostic assessment of malignancies. The aim of the study was to ascertain the role of serum fucose as a biomarker for early detection of oral cancer and to compare serum fucose levels in healthy controls, leukoplakia and oral cancer patients. Materials and Methods: The study included 60 (100.0%) subjects, who were grouped as 20 (33.3%) control subjects, 20 (33.3%) squamous cell carcinoma patients and 20 (33.3%) leukoplakia patients. Fucose estimation was done using UV-visible spectrophotometry based on the method as adopted by Winzler using cysteine reagent. The results were analyzed statistically using ANOVA with Bonferroni post hoc tests. Results: Results showed a high significance in serum fucose in oral squamous cell carcinoma (OSCC) and leukoplakia subjects compared to normal controls. There was a gradual increase in the values noted from control to leukoplakia and to squamous cell carcinoma. Conclusions: Estimation of serum fucose may be a reliable marker and can be used as an effective diagnostic biomarker in oral squamous cell carcinoma patients.
Objectives : Chekamuiyiin-tang has effects of promoting diuresis, promoting blood circulation, elimating phlegm and strengthening the body's resistance to disease. We experimented with Chekamuiyiin-tang in order to find effects which might possibly cure obesity. Methods : We fed a control group of rats a high fat diet and administered normal saline for 8 weeks. And we fed an experimental group of rats a high fat diet and administered an extract of Chekamuiyiin-tang for 8weeks. Results : The serum total cholesterol level, triglyceride level, free fatty acid level, total lipid level and phospholipid level in the Chekamuiyiin-tang group showed a significant decrease in comparison with the control group. The serum HDL-cholesterol level in the Chekamuiyiin-tang group showed no change in comparison with the control group. The serum LDL-cholesterol level in the Chekamuiyiin-tang group showed a decrease in comparison with the control group, but this result showed no efficacy. The size of the epididymal fat cell and the fat drops area in the hepatic lobule of rats in the Chekamuiyiin-tang group showed a significant decrease in comparison with the control group. Conclusios : According to the above results, Chekamuiyiin-tang has shown to be capable of curing and preventing obesity.
The object of this study is to investigate the toxicity of S-biol (d-allethron-d-transchrysanthemate) and the mode of action between other synthetic pyrethroid insecticides and S-biol in rats. Rats were treated daily with S-biol (25 mg/kg, 50 mg/kg, 100mg/kg) by oral administration for 5 weeks. The experimental results were summerized as follows: Biochemical parameters such as LDH and Glucose in serum were much more increased in control groups. No significant hematological change excepts for MCHC in rats treated with S-biol 100 mg/kg were observed in all groups compared to control groups. In animals treated with S-biol for 4∼5 weeks, the levels of cytochrome P-450 in the liver were significantly increased. In renal microsomal fractions, however, no significant changes of cytochrome P-450 contents were observed. The activitis of ATPase in groups treated with S-biol (50 mg/kg, 100 mg/kg) were decreased compared to those in other groups. TBA values and activities of glucose-6-phosphatase in the liver were increased a little in the groups treated with S-biol (100 mg/kg) for 5 weeks. The activities of cholinesterase in hepatic and serum were not significantly changed in all groups but slightly decreased in animals treated with high dose of S-biol (100 mg/kg). The activities of carboxylesterase in serum and in the liver were slightly increased but not significantly changed.
Critically ill patients admitted in ICU and NCU were studied with respect to their nutritional status and support to them at initial period, and the effect of nutritional support after 3 weeks. The nutritional support was supplied to these patients with the enteral and parenteral nutrition. The results of biochemical test and status of nutritional support on 52(29 males, 23 females) critically ill patients were based on medical records and the anthropometry was measured on 28 patients. The subjects were in mild malnutrition. Their initial calorie intakes were 85% of BEE, 57% of total energy requirements and 49% of protein requirement were provided. Two groups, one group of serum albumin level more than 3.5g/dl and the other group of serum albumin level less than 3.5g/dl, were significant different in total calorie and protein intakes. After 3 weeks, inappropriate nutritional support in the group of normal nutrition results in significant decreasing of serum albumin level but, there were no changes in the group of malnutrition. Nutritional support is one of the mainstays in the management of these critically ill patients and is aimed at preventing malnutrition. Therefore, timely nutritional support is heavily required in cases of critically ill patients whether their initial nutritional status is normal or not.
1, 2, 4-Trichlorobenzene (1, 2, 4-TCB) is used as a dye carrier, as an intermediate in the synthesis of herbicides, as a flame retardant, and for other purpose. After a single oral administration of 1, 2, 4-TCB (200 mg/kg, 400 mg/kg) in rats, toxic effects were studied by means of serum biochemical and heatological analysis, and liver calcium concentration. Administration of 1, 2, 4-TCB resulted in dose-dependent liver and kidney damage as estimated by increased serum alanine aminotransferase (ALT) activities, liver calcium concentration and blood urea nitrogen (BUN). Pretreatment with DL-buthionine sulfoximine (BSO, 2 mmol/kg, i.p. ) considerably decreased liver glutathione concentration, which was accompanied by markedly elevated serum ALT activites. It is well-known that toxicity of halogenated benzene such as bromobenzene, 1, 4-dichlorobenzene is increased by pretreatment of henobarbital (PB), and protected by pretreatment of cytochrome P450 inhibitor including metyrapone (MP). However, there was no obvious alterations in toxicity of 1, 2, 4-TCB by pretreatment of phenobarbital or metyrapone. In comparison with control group, treatment groups exhibited significant changes in some parameters of hematological analysis but all hematological values remined within normal ranges.
1,2,4-trichlorobenzene (1,2,4-TCB) is used as a dye carrier, an intermediate in the syn[hesis of herbicides, aflame retardant, and for other purpose. After a single oral administration of 1,2,4-TCB (200 mg/kg, 400 mg/kg) in rats, toxic effects were studied by means of serum biochemical and hematological analysis, and liver calcium concentration. Administration of 1,2,4-TCB resulted in dose-dependent manner liver and kidney damage being suggested by increased serum alanine aminbtransferase (ALT) activities, liver calcium concentration and blood urea nitrogen (BUN). Pretreatment with DL-buthionine sulfoximine (BSO, 2 mmol/kg, i.p.) considerably decreased liver glatathione concentration, which was accompanied by markedly elevated serum ALT activites. It is well-known that toxicity of halogenated benzene such as bromobenzene, 1,4-dichlorobenzene is increased by pretreatment of phenobarbital, and protected by pretreatment of cytochrorn P450 inhibitor including metyrapone. However, there were no obvious alterations in toxicity of 1,2,4-TCB by pretreatment of phenobarbital or metyrapone. In comparison with control group, treatment groups exhibited significant changes in some parameters of hematological analysis but all hematological values remained within normal ranges.
This study was conducted to examine whether bone mineral density changes in 55 young Korean college women aged 19 to 26 years over 2 years and nutritional and biochemical factors are related. Bone mineral density (BMD) was measured in the spine (LS), femoral neck (FN), ward's triangle (WT), and femoral trochanter (FT) by dual energy X-ray absorptiometry three times at one-year intervals. Serum osteocalcin (OC), parathyroid hormone (PTH), and urinary cross-linked N-teleopeptides of type collagen (NTx) were measured. Dietary intake was assessed 8 times with 24-hour recall method. Physical activity (PA) was obtained by questionnaire and body fat content was measured by bioelectrical impedance analysis at baseline and after 2 years. Analyses were performed on 34 subjects with all three BMD measurements. The BMDs at the lumbar spine gradually increased over 2 years, while the BMDs of three sites at the femur were sustained or increased. The mean OC, PTH had a similar pattern with the change of BMD at the femur. The mean NTx decreased over 2 years but was still higher than those in other studies. BMI, body fat, vitamin A and zinc intake had a significant correlation with LS-BMD. Femur, PTH, body fat, vitamin A, vitamin B$_2$and calcium intake had a significant correlation with WT-BMD and was mostly influenced by diet. By multiple regression analysis, it was shown that the significant factors affecting the LS-BMD were BMI and vitamin A intake and those affecting FN and WT were age, BMI, PTH and calcium intake. These results indicate that some Korean women still experience increases in BMD and that this was associated with PTH and vitamin A and calcium intake. Therefore, proper diet and diet management is needed to increase changes in BMD among college women.
