Johne's disease is a chronic inflammatory bowel disease of cattle, sheep, goats and other ruminants, and Mycobacterium paratuberculosis is the etiologic agent of this disease. Many studies have been carried out on paratuberculosis from daily cattle and Korean native cattle in multiple areas around nation, but there is no report in Eastern-Gyeongbuk area. The purpose of this study is to investigate the seroprevalence of bovine paratuberculosis in Eastern-Gyeongbuk area. From July to December in 2007, blood samples were collected from 363 dairy cattle of 27 farms and 281 Korean cattle of 114 farms and the ELISA was conducted. 25 (6.9%) dairy cattle of 6 (22%) farms and 19 (6.8%) Korean cattle of 8 (7.0%) farms were positive in ELISA. In regional analysis, 25 (8.3%) out of 300 dairy cattle in Gyeungju were positive and Pohang were negative in this research. 12 (16.4%) out of 73 Korean cattle in Gyeungju and 7 (9.6%) out of 73 Korean cattle in Uljin were positive. Pohang and Youngdeok of Korean cattle were negative in this research. According to raising scale of dairy cattle, 4 (66.7%) farms out of 6 farms were raising 30 below and 2 (33.3%) farms out of were raising 30$\sim$70. And there were negative raising scale more than 70. In Korean cattle, 6 (75%) farms out of 8 were raising below 10 and 2 (25%) farms were raising 10$\sim$30. And there were negative raising scale more than 30. The rate of seropositive of paratuberculosis dairy cattle and Korean cattle were similar and the positive rate of Eastern-Gyeongbuk area is reported lower than that of any other region.

A major hurdle to the development of RNA interference as therapy for HIV infection is the delivery of siRNA to T lymphocytes which are difficult cells to transfect even in vitro. We have employed a single chain antibody to the pan T cell surface antigen CD7 was conjugated to an oligo-9-arginine peptide (scFvCD7-9R) for T cell-specific siRNA delivery in NOD/SCIDIL2${\gamma}$-/- mice reconstituted with human peripheral blood lymphocytes (Hu-PBL). Using a novel delivery, we first show that scFvCD7-9R efficiently delivered CD4 siRNA into human T cells in vitro. In vivo administration to Hu-PBL mice resulted in reduced levels of surface CD4 expression on T cells. Mice infected with HIV-1 and treated on a weekly basis with scFvCD7-9R-siRNA complexes targeting a combination of viral genes and the host coreceptor molecule CCR5 successfully maintained CD4/CD3 T cell ratios up to 4 weeks after infection in contrast to control mice that displayed a marked reduction in CD4 T cell numbers. p24 antigen levels were undetectable in 3 of the 4 protected mice. scFvCD7-9R/antiviral siRNA treatment also helped maintain CD4 T cell numbers with reduced plasma viral loads in Hu-PBL mice reconstituted with PBMC from donors seropositive for HIV, indicating that this method can contain viral replication even in established HIV infections. Our results show that scFvCD7-9R could be further developed as a potential therapeutic for HIV-1 infection.
Mycoplasma gallisepticum (MG) is major cause of chronic respiratory disease in chickens. M. synoviae (MS) most frequently occurs a subclinical upper respiratory infection but may result in airsacculitis and synovitis in chickens and turkeys. Both mycoplasmas induce economic losses by triggering chronic respiratory signs, airsacculitis and decreased egg production. For prevention of the infections, live attenuated andinactivated vaccines are commercially used for prevention of MG but not MS in Korea. Serum plate agglutination (SPA) and enzyme-linked immunosorbent assay (ELISA) have been commonly used for serological diagnosis for MG and MS. Recently, it is believed that MS spread in chickens is very seriously in Korea and respiratory infection with MS causes substantial loss in poultry farms. In this study, we investigated the serological prevalence of MG and MS in unvaccinated chickens between 2008 and 2009. The overall seroprevalence of MG was 24% of 2,094 for individual chickens and 24% of 189 farms. The overall seroprevalence of MS was 36% in 2,095 chickens and 39% in 198 farms. The results show that seropositive ratio of MS is higher than MG. The geographical prevalence of MG has been estimated in following sequence; Gangwon, Jeolla, Gyeonggi, Gyeongsang, and Chungcheong. The geographical prevalence of MS has been estimated as follows; Gangwon, Gyeonggi, Gyeongsang, Chungcheong, and Jeolla. Seasonal seroprevalencewas also examined, and it found that seroprevalence in spring, fall and winter was higher than that in summer in MG, but not in MS. No significant difference was shown in seroprevalence according to breed. Future study about pathogenicity of MS isolates would be needed and economical losses by MS outbreaks should be analyzed. Moreover, we compared sero-positivity obtained with SPA and ELISA. The kappa value of MG between SPA and ELISA was 0.8061 and the kappa value of MS between SPA and ELISA was 0.7649.
Ninety pigs under the age of 120-day-old requested at the diagnostic laboratory of animal diseases in Cheju National University were evaluated for the prevalence of tissue antigen and serum antibody to swine influenza virus (SIV). For histopathologic examination there was sampled at the consolidated area in cranioventral or dorsocaudal lobes of lungs. Lung tissues from all pigs were tested for the antigen of SIV type A by immunohistochemistry (IHC). Sera from 56 pigs were used for the antibody detection to SIV type A (subtype H1N1 and H3N2) by haemagglutinin inhibition test. Pneumonic lesions were observed in 72 cases (80%) of 90 pigs. Broncho-interstitial or interstitial pneumonia were more prevalent than suppurative or fibrinous bronchopneumonia. According to HI test, 46.4% of the tested sera showed seropositive. Positive sera were consisted with 5.3% for SIV H1N1, 28.6% for SIV H3N2, and 12.5% for both subtype to be tested, respectively. SIV antigens were detected in 51 cases(56.6%) of 90 pigs. Most SIV antigens were presented in the epithelium of the bronchi and bronchiole. Necrotizing bronchitis or bronchiolitis were observed in 28(31.1%) cases of all inspected pigs. These results suggested that SIV might be an important role to induce swine pneumonia in Jeju. Also IHC was very useful for the detection of SIV in the lung.
This study was conducted to investigate the characteristics of brucellosis in Korean native cattle in a farm where bovine brucellosis was confirmed 3 times from September 2006 to March 2007. Of 74 bulls serum samples examined, 21 (28.4%) were positive by Rose-Bengal test (RBT) and Standard tube agglutination test (STAT). In the isolation test from seropositive bulls, B abortus was isolated and identified from 2 specimens (testis, intestinal lymph node) among 6 kinds of specimens including blood, urine, feces and soil. Isolation rate of intestinal lymph node and testis was 25% (3/12 cases) and 16.7% (2/12), respectively. B abortus was also isolated from calves below 12 months old, i.e., 1 isolate (25.0%) was confirmed from testis, 4 (40.0%) from supra-mammary lymph nodes and 1 (25.0%) from intestinal lymph node. All isolates had Brucella specific 16s r-RNA with 905-bp band detected by PCR assay. For the more effective control of bovine brucellosis in korea, this paper would like to suggest that all of bulls and calves should be included in the screening tests.
This study was conducted from May to December 2016 to investigate the prevalence of antibody titers for Newcastle disease (ND), Infectious Bronchitis (IB), Egg drop syndrome (EDS), Avian pneumovirus (APV), Fowl Typhoid (FT), Mycoplasma gallisepticum (MG), Mycoplasma synoviae (MS) in laying hens in Incheon area. In ND, 418 (99.5%) of the 420 outbreaks were positive. IB showed 330 (78.5%) positive antibody out of 420 cases. EDS showed 419 (99.7%) positive seropositive rates among 420 cases. In the case of APV, 357 cases (85%) were positive in 420 cases. In the case of PT, the positive rate of antibody was 128 (30.4%) out of 420 cases. Among the 420 cases of MG, 222 cases (52.8%) were positive for antibodies. In the case of MS, 395 out of 420 cases (94.0%) were positive. This result suggests that the positive rate of antibody for PT in Incheon area was low. And it was also clear that MS without vaccine is now rampant in Incheon.
Total of 1085 swine sera (1996-1997) from nation-wide were tested for the presence of antibodies to influenza A virus. Fifty nine percent of the tested sera showed seropositive by HI test. Positive sera consisted of 24--- of H3, 15--- of H1, and 20--- of the sample had both antibodies, respectively. Sera collected from various region represented 7~27--- seropositivity to H1N1, 15~25--- to H3N2, respectively. Swine influenza field isolate from nasal swab was characterized antigenically and genetically to elucidate its relatedness with other known strains of influenza A virus. The study was focused on the HA gene which is related to pathogenecity and antigenic variability of the influenza virus. By RT-PCR using influenza A/H1N1 specific primers, influenza virus H1N1 specific DNA fragment was amplified from A/Swine/Iowa/15/30(H1N1), US field isolate but not in H3N2 strain. PCR products were sequenced by dideoxy chain termination method to determine nucleotide homology with other strains of influenza A virus. The US field isolate and A/Swine/Indiana/1726/88 strain had 97--- of nucleotide homology and 98--- of amino acid homology. Based on the results obtained from this experiment, the field isolate was genetically related to A/Swine/Indiana/1726/88 and had higher homology with A/Swine/Indiana/1726/88 than with classical swine influenza virus, A/Swine/Iowa/15/30. The field isolate had no amino acid changes at the antigenic site compare to that of the A/Swine/Indiana/1726/88. The proteolytic enzyme cleavage site between HA1 and HA2 had no alteration and the amino acid arginine was intact. There is no evidence has been found that the field isolate has genetic shift or genetic drift which might altered antigenic determinant.
Ornithobacterium rhinotracheale (OR) is a recently described gram-negative rod-shaped bacterium associated with respiratory tract infection in poultry. In order to investigate current occurrence of OR infection and to evaluate antibiotic susceptibility, the prevalence of OR antibody in domestic chickens were examined and the minimal inhibitory concentrations(MICs) of 8 antibiotics for 11 OR isolates was determined. All isolates tested were mostly susceptible to three antibiotics, ampicillin (MICs ranging from 0.38 ${\mu}g$/ml to 2 ${\mu}g$/ml), tetracycline (MICs 0.094~3 ${\mu}g$/ml) and doxycycline (MICs 0.047~4 ${\mu}g$/ml) but resistant to genatmicin. Ciprofloxacin, norfloxacin, enrofloxacin, and ofloxacin gave most isolates inhibition only in case of a higher concentration (MICs ranged in most cases from 3 ${\mu}g$/ml to 48 ${\mu}g$/ml). Out of 188 chicken flocks including broilers, broiler breeders, and layers, seropositive flock to OR were detected in 5 broilers (4%), 17 broiler breeders (50%), and 16 layers (55.2%), using commercial OR enzyme-linked immunosorbent assay(ELISA) kits. It suggested that OR infection was widespreaded in poultry farms in Korea.
Background: The seropositivity rate of hepatitis B surface antigen (anti-HBs) antibodies is known to be ≥95% after hepatitis B virus vaccination during infancy. However, a low level or absence of anti-HBs in healthy children is discovered in many cases. Recent studies in adults reported that a reduced anti-HBs production rate is related to obesity. Purpose: To investigate whether body mass index (BMI) affects anti-HBs levels in healthy children following 3 serial dose vaccinations in infancy. Methods: We recruited 1,200 healthy volunteers aged 3, 5, 7, or 10 years from 4-day care centers and 4 elementary schools. All subjects completed a questionnaire including body weight, height, and vaccine type received. Levels of serum hepatitis B surface antigen (HBsAg) and anti-HBs in all subjects were analyzed using electrochemiluminescence immunoassay. The standardized scores (z score) for each sex and age were obtained using the lambda-mu-sigma method in the 2017 Korean National Growth Charts for children and adolescents. Results: Our subjects (n=1,200) comprised 750 males (62.5%) and 450 females (37.5%). The overall anti-HBs seropositivity rate was 57.9% (695 of 1,200). We identified significant differences in mean BMI values between seronegative and seropositive groups (17.45 vs. 16.62, respectively; P<0.001). The anti-HBs titer was significantly decreased as the BMI z score increased adjusting for age and sex (B=-15.725; standard error=5.494; P=0.004). The probability of anti-HBs seropositivity based on BMI z score was decreased to an OR of 0.820 after the control for confounding variables (95% confidence interval, 0.728-0.923; P=0.001). Conclusion: There was a significant association between anti-HBs titer and BMI z score after adjustment for age and sex. Our results indicate that BMI is a potential factor affecting anti-HBs titer in healthy children.
Porcine reproductive and respiratory syndrome (PRRS) causes a significant economic loss in the swine industry not only in Korea but also all over the world. Andong and Hapcheon region were selected for Area Regional Control (ARC) programme to reduce the shedding of PRRS virus (PRRSV) and decrease PRRS outbreaks. Before conducting the PRRS ARC, sera of pigs were tested for both antibody using ELISA and antigen using RT-PCR, then phylogenetic classifications was analysed. Pigs of 138/275 (50.2%) in Andong and 352/425 (82.8%) in Hapcheon were seropositive. Also, the RT-PCR results revealed that 27 heads (8.2%) in Andong, 112 heads (22.0%) in Hapcheon were positive for PRRSV antigen. PRRSVs were mainly detected between the ages of 40 to 60 days. PRRSV ORF5 regions were used to determine genetic clusters based on previous report. All PRRSV type I detected in both Andong and Hapcheon were classified as Cluster I. The PRRSV type II isolates in Andong were assorted to Cluster II, whereas the PRRSV type II isolates in Hapcheon were the viruses were unassembled into any cluster except one identified to Cluster III. Phylogenetic analysis indicated that new clusters of PRRSVs type II were prevalent in Hapcheon.
