In order to increase the utilization of defatted sesame meal (DSM), a by-product of sesame oil production, the conditions of extraction of insoluble proteins from DSM by enzyme treatment were investigated. As a result of comparing the treatment results of proteolytic enzymes Alcalase, Flavorzyme, Neutrase, and Protamex with control, Protamex was effective in increasing the total solid and protein content. At the reaction conditions of Protamex (50 ℃, pH 6.0), the dosage of enzymes was appropriate for 1% of DSM and 3 h of enzyme reaction time. To improve the efficiency of enzymatic treatment, the protein content extracted increased as the heat treatment temperature increased, and slightly increased above 110 ℃. As a result of investigating the effect of the combination treatment of cell lytic enzyme (Tunicase) and protease (Protamex) on protein solubilization, it was most effective to treat the cell lytic enzyme after processing the protease. After heat treatment (110 ℃, 10 min), sequential treatment of Protamex and Tunicase increased the protein content by about 3.5 times (9.85→35.58 mg/mL) of the non-heated control and 2.2 times (15.83→35.58 mg/mL) of the heat treated control.

This study analyzed total concentrations and existing forms of heavy metals in soils of Ulsan using a sequential extraction method. Soil samples were collected from 6 categorized areas including green, residential, heavy traffic, petrochemical industrial complex(IC), mechanical and shipbuilding IC, and non-ferrous metal IC areas. which represent different emission characteristics. The highest total concentrations of heavy metals by a sequential extraction analysis were observed in the soils collected from the non-ferrous metal IC area, followed by the mechanical and shipbuilding IC and heavy traffic areas. Dominant(> 50%) existing forms of Cd, Cr and Ni were residual forms followed by Fe and Mn oxides in almost areas. Residual fractions in the non-ferrous metal IC areas were relatively lower than those in other areas. However, the fractions of organic and sulphides in the IC areas were higher. The dominant farms of Cu were much different with the investigated areas. In most areas, the dominant forms of Pb and Zn were Fe and Mn oxides, followed by residual fraction for Pb. The exchangeable and carbonate fractions represent mobility of metallic elements in soils. They are also significantly affected by the environmental renditions, such as pHs of soil and rainfall. In this study the exchangeable and carbonate fractions were lower than other fractions. Because the total concentrations of heavy metals in the soils of the non-ferrous metal IC area were extremely high, however, the mobile fractions of heavy metals in the IC area would be significant. Thus a large amount of heavy metals can be released into plants, water bodies, and soils. Therefore, urgent measures, such as source control for soil remediation of heavy metals, in the non-ferrous metal IC areas are essentially required. Analysis results obtained from the sequential extraction and the aqua regia extraction showed a high correlation, whose determination coefficients(R2) of heavy metals except Cd approximately ranged from 0.7 to 0.9.
This study was carried out to determine the heavy metal contents in sluges of Keumho river and her branches. The heavy metal contents of sludge are the highest in Sincheon, Kongdancheon and Dalseocheon among the branches. The large part of heavy metal exists in $0{\sim}5cm$ from sludge surface. The sequential extraction with various reagents showed that the residual and organically bound fraction were the most abundant pool. Decomposition of organic matter caused sludge to release heavy metals. On the extraction of sludge with various solution having different pH, it was found that the lower the pH, the more heavy metal was extracted. However, considerable amount of heavy metal was not extracted even with pH 3 solution.
This study investigates the dispersion and behavior of Au and heavy metals in the water system (soil, AMD and stream sediment) at the abandoned Bonjeong gold mine, based on XRD, aqua regia, sequential extraction, and physico-chemical analyses. The XRD analyses targeted quartz and kaolinite in the mine waste soil and quartz and goethite in stream sediment. The physico-chemical analyses of AMD with increasing distance from water system showed that pH increased from 3.00 to 3.19 and Eh decreased from 450 to 396 mV. The Au content in AMD ranged from 0.68 to 0.97 mg/L upstream, but was not detected downstream. The Au content of stream sediment was 13.76 to 22.85 mg/kg. Sequential extraction from stream sediment revealed 10.84% exchangeable (STEP I), 11.09% carbonates (STEP II), 25.53% Fe-Mn oxides (STEP III), 26.62% organic matter (STEP IV), and 24.61% residual (STEP V).
The purpose of this study was to investigate the effectiveness of a stabilization treatment for As contaminated soil. A combination of hydrated lime, Portland cement, $FeCl_3{\cdot}6H_2O$, and NaOH were used as stabilizing agents. The effectiveness of stabilization treatment was evaluated by the Korean Standard Test (KST) method (1N HCl extraction). Sequential extractions were performed to investigate the As distribution after treatment. Following the application of the treatment, curing periods of up to 7 and 28days were investigated. The experimental results showed that a combination of hydrated lime/Portland cement was more effective than treatments of hydrated lime or Portland cement at immobilizing As in the contaminated soil. The treatment of 25wt% hydrated lime and 5wt% Portland cement was effective in reducing As leachability less than the Korean warning standard of 20 mg/kg. However, the treatments of hydrated lime and Portland cement failed to meet the Korean warning standard even when up to 30 wt% was used. The treatment utilizing hydrated lime and $FeCl_3{\cdot}6H_2O$ was not effective in properly reducing As leachability. The addition of $FeCl_3{\cdot}6H_2O$ was negative in terms of pH condition. Moreover, the treatment with hydrated lime/NaOH was effective in reducing As leachability but not as much as hydrated lime/Portland cement. The sequential extraction results indicated that the residual phase was greatly increased upon the treatment of hydrated lime/Portland cement. It was concluded that the hydrated lime/Portland cement treatment was the best among the other combinations studied at achieving trace As concentrations.

Rain events are extremely important for phosphorus (P) dynamics in rivers since large portions of annual river P loads can be transported in particulate forms during only a few major events. Despite their importance, a precise estimation of P contribution in river sediments after rainy seasons has rarely been reported. This study estimated the longitudinal variation in the concentrations of different inorganic P fractions in bed sediments of the South Han River over a rainy season, through using the sequential extraction method. Non-apatite P was the dominant form, representing more than 60% of total inorganic P (TIP) content in sediments. Although no significant variation of TIP contents was observed, the proportion of bioavailable P in TIP pools decreased after the rainy season. The concentrations of individual inorganic P fractions ($NH_4Cl-P$, $NH_4F-P$, NaOH-P, and $H_2SO_4-P$) were significantly different across sites and after the rainy season (p < 0.05, two-way ANOVA). $NH_4F-P$ and NaOH-P concentrations in sediments increased in a downstream direction. After the rainy season, $NH_4Cl-P$ concentrations in sediments decreased whereas $NH_4F-P$ and $H_2SO_4-P$ concentrations increased. The redistribution of individual P fractions in sediments observed after rainy seasons were possibly due to the changing contribution of various sources of runoff and the variation in flow related particle size. Current estimation of P in bed sediments of the South Han River suggests a lower potential of internal P loading from sediments after the rainy season.
The metals contamination of farmland soil nearby abandoned metal mine was serious problem in Korea. Stabilization of contaminated soil was reported using various stabilizers. Application of limestone and steel refining slag was reported as effective stabilizers in the stabilization of metals. The batch studies confirmed that the mixture of limestone and steel refining slag was suitable for stabilization of metals in contaminated soil. The limestone and steel refining slag mixture (2 : 1 and 3 : 2) were used in column studies and it was confirmed that the stabilizers effectively stabilized heavy metals in contaminated soil. The pH of the soil was increased with the addition of stabilizers. Total leached concentration of metals from the column study was reduced 44, 17, and 93% in comparison to the control at arsenic, cadmium and copper, respectively. The sequential extraction studies showed that the exchangeable fraction was changed into carbonate bound fraction (Cd and Cu) and Fe-Mn oxide bound fraction (As). Based on the results we confirmed that 2:1 ratio of limestone and steel refining slag effectively stabilizes the heavy metals. The mixed treatment of lime stone with steel refining slag would be an effective and feasible method for controlling metals leaching in contaminated soil.

The chemical forms of Cd, Cu, Pb, and Zn were analysed by sequential extraction technique to evaluate the effects of drying and heating of dredged sediments from Lake Chungcho. The most abundant fraction of Cd, Cu, and Zn in the wet and untreated sediment was organic/sulfidic fraction that is state in reducing environment such as the bottom condition of Lake Chungcho, while Pb dominated in residual fraction. This means that the source of Cd, Cu, and Zn in the Chungcho lake sediment is related to the organic degradation and Pb to the erosion from surrounding rocks. With drying and oxidation by dredging, heating treatment, and disposal of the lake sediment, the chemical forms of studied metals changed greatly from organic/sulfidic fraction to adsorbed and reducible fractions which are more labile in oxygenated environment. Organic/sulfidic fraction of Cd, Cu and Pb in the wet sediment was transformed with drying and heating treatments to the labile ones like adsorbed and reducible fraction, but Zn to carbonate and reducible fraction. Heating of the sediment at $320^{\circ}C$ greatly increased the labile fraction of Cd and Cu, while that at $105^{\circ}C$ for Pb and Zn. It is believed that the increase in labile forms of heavy metals in the sediments by drying and heating is caused by the contact with oxygen during drying and heating and by the increase of pH of the pore water at the expense of organic/sulfidic fraction. It is concluded that the drying and oxidation currently used in the treatment of dredged sediment can increase labile forms of heavy metals in the sediment, and the potential of the metal availability from the sediment.
Stream sediment samples were collected in the vicinity of the Sambo Pb-Zn-barite mine in order to investigate dispersion patterns and pollution levels of heavy metals, and to find out their mode of occurrences. Those samples were analyzed for Cd, Cu, Pb, Zn, Fe, and Mn by using partial extraction and sequential extraction methods. Stream sediments of this studied area has been contaminated severely by Pb, Zn, and Cd. Dispersion patterns of heavy metals in those sediments are different according to their pollution source and sampling distance from the mine. The stream sediment pH is generally neutral in control area, and is slightly acidic in the lower part of tailing dam. The main pollution source of stream sediments was confirmed as tailing and mine drainage. Their pollution level depends mainly on inflowed water and distance from the mine. The tolerance index ranges from -0.9 to 0.7. The mode of occurrances of heavy metals in polluted stream sediment are identified as Fe-Mn oxides, organic bounded and carbonate-bounded compound which show high potential of bioavailablity.

To determine Pb species in soils following the immobilization process, sequential extraction has been used despite the possibility of overestimating Pb species from unintended reactions during chemical extraction. Meanwhile, the application of extended X-ray absorption fine structure (EXAFS) has been shown to provide a more precise result than chemical extraction. In this study, the immobilization of Pb in contaminated soils treated with liming materials such as oyster shell (OS) or eggshell (ES) was evaluated with thermodynamic modelling and EXAFS analysis. Thermodynamic modelling by visual MINTEQ predicted the precipitation of $Pb(OH)_2$ in OS and ES treated soils. In particular, the values of saturation index (SI) for $Pb(OH)_2$ in OS (SI=0.286) and ES (SI=0.453) treated soils were greater than in the control soil (SI=0.281). Linear combination fitting (LCF) analysis confirmed the presence of $C_{12}H_{10}O_{14}Pb_3$ (lead citrate, 44.7%) by citric acid from plant root, Pb-gibbsite (Pb adsorbed gibbsite, 26.4%), and Pb-kaolinite (Pb adsorbed kaolinite, 20.3%) in the control soil. On the other hand, $Pb(OH)_2$ (16.8%), Pb-gibbsite (39.3%), and Pb-kaolinite (25.6%) were observed in the OS treated soil and $Pb(OH)_2$ (55.2%) and Pb-gibbsite (33.8%) were also confirmed in the ES treated soil. Our results indicate that the treatment with OS and ES immobilizes Pb by adsorption of Pb onto the soil minerals as a result of the increase in soil negative charge and the formation of stable $Pb(OH)_2$ under high pH condition of soils.
