Affinity Filtration Chromatography of Proteins by Chitosan and Chitin Membranes: 2. Separation of BSA and Lysozyme (키토산 및 키틴 막에 의한 단백질의 친화 여과 크로마토그래피: 2. BSA 및 Lysozyme의 분리)

  • Porous affinity chitosan and chitin membranes with good mechanical strength and high protein binding capacity were prepared by using silica particles as porogen. The maximum binding capacity of affinity chitosan membrane for BSA protein is 21.8mg/mL, and that of affinity chitin membrane for lysozyme enzyme is 26.1mg/mL. Chromatographic separations of BSA and lysozyme proteins using the porous affinity chitosan and chitin membranes were performed with change of the flow rate, loading amount and concentration of protein loading solutions. Protein eluted amount and binding yield were calculated from the filtration chromatograms consisted of loading/washing/elution sequences. Protein binding amount and yield were increased with decreasing of flow rate, increasing of loading amount and concentration of protein loading solutions. Those results suggest that the porous chitosan and chitin membranes prepared by using silica particles as porogen are suitable in affinity filtration chromatography for large scale separation of proteins.

Development of Adsorptive Permeation Membrane (APM) and Process for Separation of $CO_2$ from gas mixtures (이산화탄소 분리를 위한 흡착투과막 및 공정 개발)

  • Adsorptive permeation hollow fiber membrane (APM) has been developed for effectively separating $CO_2$ from gas mixture. Inside the APM, zeolite 13X particles were uniformly dispersed without covering their surfaces by a symmetric porous structure of polypropylene lattice. In this study, $CO_2/N_2$ mixture was used as a simulated gas mixture. Separation was achieved by adsorbing $CO_2$ on the zeolite particles in the APM and then permeating $N_2$ into permeate side in passing all the feed gas through the APM. Adsorptive permeation tests were carried out with a set of APM modules, and the adsorptive permeation performances of the modules were analyzed from the test results. After saturation of the adsorbent with $CO_2$, the APM was regenerated by desorption of $CO_2$ from it through vacuuming both inside of outside of the APM hollow fiber, and the regeneration process of the APM by vacuuming was discussed in terms of regeneration efficiency and energy consumption.

Change in the Characteristics of Particle Separation and Particle Size Distribution of Weathered Granite Soil from the Yecheon Area (Eastern South Korea) after Water Washing (물 세척한 예천지역 화강풍화토의 입자분리와 입도분포 변화 특성)

  • In this study, sieve analysis testing was performed on weathered granite soil from Yecheon (eastern South Korea) before and after water washing in accordance with the sieve analysis regulations of KS F 2302. The changes in particle separation and particle size distribution after washing with water were analyzed. Image analysis using an optical microscope revealed that soil particles were separated into smaller particles by water washing. The change in the particle size distribution curve was assessed using five index values. The increase in the fine particle fraction (<0.075 mm) was 13.67%, the increase in the 0.075-0.25 mm fraction was 19.44%, and the mean particle diameter (D50) decreased by 0.663 mm. In addition, the maximum passage width (BM) of the particle size distribution curve increased by 21.08% for the #30 sieve, and the moving area (A) of the particle size distribution curve was 69.28%·mm. These results suggest that washing with water is an effective way to prevent underestimation of the fine particle content in soil.

Numerical investigation of ceramic particle movement for injected gas flow rate in cyclone separator system (사이클론 분리기 시스템 내에서의 가스 주입 유속에 따른 세라믹 입자 거동 전산모사)

  • Using computational fluid dynamics (CFD) method, we investigated three-dimensional fluid flow field and particle movement with respect to the injected gas flow rate variation in typical cyclone separator system. The results of numerical investigation were deduced by coupling the analysis of fluid flow field with Wavier-stokes equation and the tracking of the particle trajectory with Langrangian approach. It was shown that the increasing of injected gas flow rate resulted in the increasing of pressure loss in the separator. This change of inner pressure had an effect on an aspect of the fluid flow in the separator. Particle movement was determined by fluid flow in the separator and was fully depended on a diameter of particles under the fixed flow rate. Increasing of injected gas flow rate was led to an increasing of the trace of particle, so the particles moved to the lower part of the separator. For this reason, the minimum diameters of the particles were decreased and increased the separation rate under the fixed particle diameter. In conclusion, the changes of injected gas flow rate have an important factor to the fluctuation of the fluid flow field and particle trajectory in the separator.

Separation characteristics of particles in a self-rotating type centrifugal oil purifier

  • The centrifugal oil purifier is used in an engine for lubrication and to remove impurities. The momentum needed for the rotation of the cylindrical chamber is obtained by jet injections. An impure particle in the oil is separated by the centrifugal forces moving to the inner wall of the rotating cylindrical chamber body. The dust particles are eliminated when the particles are absorbed onto the surface of the inner wall of the chamber body. The flow characteristics and the physical behaviors of particles in this centrifugal oil purifier were investigated numerically and the filtration efficiencies was evaluated. For calculations, a commercial code is used and the SST (Shear Stress Transport) turbulence model has been adopted. The MFR (Multi Frames of Reference) method is introduced to consider the rotating effect of the flows. Under various variables, such as particle size, particle density and rotating speed, the filtration efficiencies are evaluated. It has been verified that the filtration efficiency is increased with the increments in the particle size, the particle density and the rotating speed of the cylindrical chamber.

  • Dielectrophoresis(DEP) is useful in manipulation and separation of micro-sized particles including biological samples such as bacteria, blood cells, and cancer cells in a micro-fluidic device. Especially, those separation and manipulation techniques using DEP in combination of micro fabrication technique have been researched more and more. Recently, it is revealed that a window structure of insulating layer in microfluidic DEP chip is key role in trap of micro-particles around the window structure. However, the trap phenomenon-driven by DEP force gradient did not fully understand and is still illusive. In this study, we characterize the trap mechanism and efficiency with different shapes of window in a microfluidic DEP chip. To do this characterization, we fabricated 4 different windows shapes such as rhombus, circle, squares, and hexagon inside a micro-fluidic chip, and performed micro-sized particles manipulation experiments as varying the frequency and voltage of AC signal. Moreover, the numerical simulation with the same parameters that were used in the experiment was also performed in order to compare the simulation results and the experimental results. Those comparison shows that both results are closely matched. This study may be helpful in design and development of microfluidic DEP chip for trapping micro-scaled biological particle.

Flow Characteristics of Neutrally Buoyant Particles in 2-Dimensional Poiseuille Flow through Circular Capillaries

  • An experimental study has been conducted to quantitatively characterize the motion of neutrally buoyant particles in 2-dimensional Poiseuille flow through the micron-sized circular capillaries in the range of Re (Reynolds number) $\approx0.1\sim100$. $A{\mu}-PTV$ (Particle Tracking Velocimetry) system is adopted, which consists of a double-headed Nd:YAG laser, an epi-fluorescence microscope and a cooled CCD camera. Since high shear rate can be induced due to the scale effect even at low Re, it is shown that in micro scale neutrally buoyant particles in Poiseuille flow drift away from the wall and away from the center of the capillary. Consequently, particles accumulate at the equilibrium position of $0.52\sim0.64R$ with R being the radius of the capillary, which is analogous to that of tube flow in macro scale. There is a plateau in equilibrium position at small Re, while equilibrium position starts increasing at $Re\approx30$. The outermost edge of particle cluster is closer to the center of the capillary than that in previous studies due to low Re effect. The present study quantitatively presents characteristics of particle motion in circular capillaries. Furthermore, it is expected to give optimum factors for designing microfluidic systems that are to be used fur plasma separation from the blood.

Effects of the Counter Ion Valency on the Colloidal Interaction between Two Cylindrical Particles

  • In this study, the effects of counter ion valency of the electrolyte on the colloidal repulsion between two parallel cylindrical particles were investigated. Electrostatic interactions of the cylindrical particles were calculated with the variation of counter ion valency. To calculate the electrical repulsive energy working between these two cylindrical particles, Derjaguin approximation was applied. The electrostatic potential profiles were obtained numerically by solving nonlinear Poission-Boltzmann (P-B) equation and calculating middle point potential and repulsive energy working between interacting surfaces. The electrical potential and repulsive energy were influenced by counter ion valency, Debye length, and surface potential. The potential profile and middle point potential decayed with the counter ion valency due to the promoted shielding of electrical charge. On the while, the repulsive energy increased with the counter ion valency at a short separation distance. These behaviors of electrostatic interaction agreed with previous results on planar or spherical surfaces.

  • Drilling mud was used once in the step of separating the gas and powder they were transported to a surge tank. At that time, the fine powder, such as dust that is not separated from the gas, is included in the gas that was separated from the mud. The fine particles of the powder are collected to increase the density of the powder and prevent air pollution. To remove particles from air or another gas, a cyclone-type separator generally can be used with the principles of vortex separation without using a filter system. In this study, we conducted numerical simulations of air-particle flow consisting of two components in a cyclone separator in a mud handling system to investigate the characteristics of turbulent vortical flow and to evaluate the collection efficiency using the commercial software, STAR-CCM+. First, the single-phase air flow was simulated and validated through the comparison with experiments (Boysan et al., 1983) and other CFD simulation results (Slack et al., 2000). Then, based on one-way coupling simulation for air and powder particles, the multi-phase flow was simulated, and the collection efficiency for various sizes of particles was compared with the experimental and theoretical results.

Experimental study on feasibility test for removing particles in air scouring membrane backwash water with metal membrane (금속막을 이용한 저압 막 공기병용 역세척수 처리 타당성 연구)

  • The main objective of this research is to study feasibility for applying metal membrane to remove particles from air scouring membrane backwash water. Also, the research was conducted to investigate the influence of polyamine coagulation on floc growth in membrane backwash water as pretreatment for removal particles. From the results of experiments for evaluating the influence of polyamine coagulation on floc growth, it was investigated that particles in the rage of $2{\sim}50{\mu}m$ grew up to $30{\sim}5,000{\mu}m$. In addition, all six metal membranes showed lower removal efficiency, which was 0.87~13.89%, in the case of no polyamine coagulant. On the other hand, in the case of injecting polyamine coagulant, those did extremely high efficiency in 56~92%. From the SEM(Scanning Electron Microscope) images of filtered wiremesh and metal foam membrane, sieve effects were predominant for liquid solid separation in wiremesh and adsorption and diffusion capture effects were predominant in metal foam membrane.
