길함미생물에 의한 시설재배 딸기 눈마름병의 생물학적 방제 (Biological Control of Strawberry Bud Rot Caused by Rhizoctonia solani AG2-1 with Antagonistic Microorganism)

  • Forth microbial isolates out of 167 isolates from the soil of controlled cultivation areas inhibited mycelial growth of Rhizoctonia solani AG2-1 causing the strawberry bud rot in vitro. Among the isolates, Kr013 and Kr020 showed suppressive effect to R. solani AG2-1 on seedlings of chinese cabbage treated by root immersion, charcoal carrier granule and drenching on 1.0% infested soil in pot. Furthemore, the corresponding effect was also revealed when the charcoal carrier granule of the isolates were treated on the seedling of strawberry that were planted on the planting hole in pot. To examine the effects of biological control in green house, it had been tested the infection rates by using two different treatments. First, the strawberry runner were planted on the nursery soil mixed with 20% charcoal carrier granule of Kr013 and Kr020 isolate respectively, and grown for 20 days before transplanting. Then the young plants form the mother plant were separated and transplanted on the 1.0% infested soil. Another method was that the charcoal carrier of Kr013 and Kr020 isolates applied to planting hole of 1.0% infested soil just before transplanting. Then the young plants were grown for 20 days on the sterilized nursery soil before transplanting. From the results, the effects of biological control was significantly higher on former treatment (e.g. the infection rates were 7.3 and 5.7%, respectively) than on the latter treatment (e.g. the corresponding value were 16.7 and 15.7%, respectively). The antagonistic isolates of Kr013 and Kr020 were respectively identified as Pseudomonas cepacia with the similarity of 55.0% and 60.0% by using the Biolog GN Microplate system.

트랩도어 하강이 일어나는 모형 입자 입상체에서 광탄성 측정 기법을 이용한 흙 아치 구조의 변화 조사 (Investigation of the Change of Soil Arch Structure in Model Particle Assembly Subjected to Displacing Trapdoor via Photoelastic Measurement Technique)

  • 트랩도어 하강이 일어나는 토체 내부에서 발생하는 흙 아치 구조의 변화를 관찰하기 위해서 광탄성 측정 기법을 이용하여 입상체의 다양한 거동을 관찰하였다. 광탄성 반응 재료로 코팅된 원주형 모형 입자를 적층하여 입상체를 구성하였다. 광탄성 기법을 이용하여 입상체 내부에서 발생하는 입자 간 접촉력 전달 구조의 변화를 촬영하여 관찰하였으며 이를 입상체 하부 로드셀에서 측정된 압력 변화 및 내부 활동면의 변화 양상과 비교 분석하였다. 트랩도어 하강 초기에 형성된 흙 아치 구조는 하강이 지속되면 붕괴되지만 다시 확장된 아치 구조가 새롭게 형성되었다. 트랩도어 하강 초기에 일치하였던 흙 아치 구조의 경계면과 활동면은 트랩도어 하강이 진행되면서 활동면이 포함된 영역이 접촉력을 잃은 영역 내부에 포함되었다.

나물용 엉겅퀴의 근권에서 Arbuscular 균근균의 분포 (Distribution of Arbuscular Mycorrhizal Fungi in the Rhizosphere of Cirsium japonicum DC. for Wild Vegetables)

  • of Hangalku per plant sold for wild vegetables were 9.1g and 0.9g, and number of leaves was 10.8. Root fresh and dry weights of Hangalku per plant were 19.2g and 4.1g. Thirty five soil samples were collected from the native soils grown Cirsium japonicum DC., and mycorrhizal spores in soils were separated using wet-sieving methods. Number of mycorrhizal spores per 30g fresh soil sized over 500${\mu}$m, 355~500${\mu}$m, 251~354${\mu}$m, 107~250${\mu}$m and 45~106${\mu}$m were 0.6, 2.1, 6.0, 55.3 and 126, etc. Total number of mycorrhizal spores per 30g fresh soil were 190. Root infection by vesicles, hyphae and arbuscules were 13%, 4% and 3%, respectively. As a result of identification, mass propagated mycorrhizal spores by the host plant of Sudangrass were Glomus sp., Gigaspora sp., and Acaulospora sp., and so on.

딸기 시설재배에서 Arbuscular 균근균의 분포 (Distribution of Arbuscular Mycorrhizal Fungi in Greenhouse Strawberry Plants)

  • This study was conducted to investigate into the distribution of arbuscular mycorrhizal fungi (AMF) in the greenhouse soils grown strawberry plants in Damyang and Jangheung districts. Twenty three soil samples were collected from strawberry plants under greenhouse conditions, and mycorrhizal spores in soils were separated using wet-sieving methods. Number of mycorrhizal spores per 30g fresh soil sized over 500${\mu}$m, 355~500${\mu}$m, 251~354${\mu}$m, 107~250${\mu}$m and $45{\sim}106{\mu}m$ were 0.3, 1.0, 4.2, 50.4 and 119, etc. Total number of spores per 30g fresh soil were l73.9. Root infection by vesicles and hyphae were 25% and 4%, respectively. Mycorrhizal root infection by arbuscules was not shown in strawberry roots. Isolated mycorrhizal spores were inoculated into the host plant of sudangrass to identify the genus of arbuscular mycorrhizal fungi, and propagated for 4 months. As a result of identification, mass propagated mycorrhizal spores were Glomus sp., Gigaspora sp., and Acaulospora sp., and so on.

Characterization of Clay Minerals in Ranch Pasture

  • This study deals with the distribution of the clay minerals separated from clay fractions of ranch pastures in Korea and their chemical and mineralogical properties. Crystalline phases of the clay minerals were identified by powder X-ray diffraction (XRD) pattern and FT-IR spectra, and their relative chemical compositions were also analyzed by X-ray flourescence spectrometry (XRF). Primary minerals consisted mainly of quartz and mica and chlorite and kaolinite along with a trace of swelling micas were identified as secondary clay minerals. However, the relative content of these clay minerals was different with the locations, which led to significant effects on physical and chemical properties of soils like inorganic elemental composition. In particular, $SiO_2$ content was higher in Gochang ranch pasture than in other ranch pasture. Infrared (IR) spectra did not indicate any significant differences in organic functional groups among the locations. This study clearly showed that ranch pastures had different relative content of clay minerals and chemical properties depending on the location and consequently that those properties are worthy to be taken into account for soil amendment.

CRS 압밀시험에 의한 점성토의 팽창특성에 관한 연구 (The Swelling Characteristics of Clayey Soil by CRS Consolidation Test)

  • In this study, the swelling characteristics of reconstituted clayey soil were investigated by STD and CRS test. The strain rate during loading was constant i.e. 0.05 %/min, 0.03 %/min and during unloading was varied in proportion to 1/1, 1/5, 1/10 and 1/15 of strain rate during loading. From this study the following conclusions were obtained; (1) There were similar values, especially, during unloading in case of 1/10 or 1/15 of strain rate during loading and the test results between STD and CRS were much to be alike. (2) The cross point of effective stress versus excess pore water pressure ratio curve, was increased during unloading, while the stress level of the cross point was decreased. The stress level can be separated into two zones according to the swelling index named Cs1 and Cs2. From the test results, the values of Cs1 were approximately constant irrespective of strain rate during unloading, but the values of Cs2 were much influenced by strain rate. (3) In CRS consolidation tests, it was found that unloading strain rate did not affect on the existence of zone.

유묘 뿌리썩음병 진전에 따른 이산재배 토양의 유별 (Grouping the Ginseng Field Soil Based on the Development of Root Rot of Ginseng Seedlings)

  • Disease incidence (DI), pre-emergence damping-off (PDO), days until the first symptom appeared (DUS), disease progress curve (DPC), and area under disease progress curve (AUDPC) were investigated in vivo after sowing ginseng seeds in each of 37 ginseng-cultivated soils which were sampled from 4 regions in Korea. Non linear fitting parameters, A, B, K and M, were estimated from the Richards' function, one of the disease progress models, by using the DI at each day from the bioassay. Inter- and intra-relationships between disease variables and stand-missing rate (SMR) in fields were investigated by using the simple correlation analysis. Disease variables of the root rot were divided into two groups: variables related to disease incidence, e.g., DI, AUDPC and A parameter, and variables related to disease progress, e.g., B, K and M parameters. DI, AUDPC, and DUS had significant correlations with SMR in ginseng fields, and then it showed that the disease development in vivo corresponded with that in fields. Soil samples could be separated into 3 and 4 groups, respectively, on the basis of the principal component 1 (PC1) and the principal component 2 (PC2), which were derived from the principal component analysis (PCA) of Richards' parameters, A, B, K and M. PC1 accounted for B, K and M parameters, and PC2 accounted for A parameter.

Effects of habitat differences on the genetic diversity of Persicaria thunbergii

  • To understand the effects of habitat characteristics on the genetic diversity of Persicaria thunbergii, three sites of different environmental conditions in a water system were surveyed. Site A was the closest to the source of the water system, and there was a dam between sites A and B. Site C is located on the lowest downstream in the water system. Vegetation survey of four quadrats at each site was performed, and soil samples were collected for physicochemical analysis. Random amplification of polymorphic DNA (RAPD) analysis of ten P. thunbergii individuals at each site was conducted to calculate population genetic diversity and genetic distance among populations. Soil was sterile sand at site A, whereas loamy soil at sites B and C. A pure stand of P. thunbergii appeared at site A, while other species occurred together (such as Humulus japonicus and Phragmites australis) at sites B (Shannon-Wiener index; $H_B=0.309$) and C ($H_C=0.299$). Similar to the species diversity, genetic diversity (Nei's gene diversity; h) within population of site A ($h_A=0.2381$) was relatively lower than sites B ($h_B=0.2761$) and C ($h_C=0.2618$). However, site C was separated from sites A and B in genetic distance rather than the geographical distance (Nei's genetic distance; A~B, 0.0338; B~C, 0.0685; A~C, 0.0833).

Selection of Plant Growth-Promoting Pseudomonas spp. That Enhanced Productivity of Soybean-Wheat Cropping System in Central India

  • The aim of this investigation was to select effective Pseudomonas sp. strains that can enhance the productivity of soybean-wheat cropping systems in Vertisols of Central India. Out of 13 strains of Pseudomonas species tested in vitro, only five strains displayed plant growth-promoting (PGP) properties. All the strains significantly increased soil enzyme activities, except acid phosphatase, total system productivity, and nutrient uptake in field evaluation; soil nutrient status was not significantly influenced. Available data indicated that six strains were better than the others. Principal component analysis (PCA) coupled cluster analysis of yield and nutrient data separated these strains into five distinct clusters with only two effective strains, GRP3 and HHRE81 in cluster IV. In spite of single cluster formation by strains GRP3 and HHRE81, they were diverse owing to greater intracluster distance (4.42) between each other. These results suggest that the GRP3 and HHRE81 strains may be used to increase the productivity efficiency of soybean-wheat cropping systems in Vertisols of Central India. Moreover, the PCA coupled cluster analysis tool may help in the selection of other such strains.

Effects of transgenic watermelon with CGMMV resistance on the diversity of soil microbial communities using PLFA

  • We compared the composition of phospholipid fatty acids (PLFA) to assess the microbial community structure in the soil and rhizosphere community of non-transgenic watermelons and transgenic watermelons in Miryang farmlands in Korea during the spring and summer of 2005. The PLFA data were seasonally examined for the number of PLFA to determine whether there is any difference in the microbial community in soils from two types of watermelons, non-transgenic and transgenic. We identified 78 PLFAs from the rhizosphere samples of the two types of watermelons. We found eight different PLFAs for the type of plants and sixteen PLFAs for the interaction of plant type and season. The PLFA data were analyzed by analysis of variance separated by plant type (P<0.0085), season (P<0.0154), and the plant type${\times}$season interaction (P<0.1595). Non-parametric multidimensional scaling (NMS showed a small apparent difference but multi-response permutation procedures (MRPP) confirmed that there was no difference in microbial community structure for soils of both plant types. Conclusively, there was no significant adverse effect of transgenic watermelon on bacterial and fungal relative abundance as measured by PLFA. We could reject our hypothesis that there might be an adverse effect from transgenic watermelon with our statistical results. Therefore, we can suggest the use of this PLFA methodology to examine the adverse effects of transgenic plants on the soil microbial community.
