Molecular Characterization and Phylogenetic Analysis of Season Influenza Virus Isolated in Busan during the 2006-2008 Seasons (부산지역에서 유행한 계절인플루엔자바이러스의 유전자 특성 및 계통분석('06-'08 절기))

  • To monitor newly emerged influenza virus variants and to investigate the prevalence pattern, our laboratory performed isolation of the viruses from surveillance sentinel hospitals. In the present study, we analysed influenza A/H1N1, A/H3N2, B viruses isolated in Busan during the 2006/07 and 2007/08 seasons by sequence analysis of the hemagglutinin (HA1 subunit) and neuraminidase (NA) genes. The isolates studied here were selected by the stratified random sample method from a total of 277 isolates, in which 15 were A/H1N1, 16 were A/H3N2 and 29 were B. Based on the phylogenetic tree, the HA1 gene showed that A/H1N1 isolates had a 96.7% to 97.7% homology with the A/Brisbane/59/2007, A/H3N2 isolates had a 98.4% to 99.7% homology with the A/Brisbane/10/2007, and B isolates had a 96.5% to 99.7% homology with the B/Florida/4/2006(Yamagata lineage), which are all the vaccine strains for the Northern Hemisphere in 2008~2009 season. In the case of the NA gene, A/H1N1 isolates had 97.8% to 98.5% homologies, A/H3N2 isolates had 98.9% to 99.4% homologies, and B isolates had 98.9% to 100% homologies with each vaccine strain in the 2008~2009 season, respectively. Characterization of the hemagglutinin gene revealed that amino acids at the receptor-binding site and N-linked glycosylation site were highly conserved. These results provide useful information for the control of influenza viruses in Busan and for a better understanding of vaccine strain selection.

Length-Weight Relations and Condition Factor (K) of Zacco platypus Along Trophic Gradients in Reservoir Ecosystems (인공호의 부영양화에 따른 피라미(Zacco platypus) 개체군의 전장-체중 관계 및 비만도 지수)

  • The objective of this study was to determine the weight-length relations and condition factor (K) of Zacco platypus, along the trophic gradients from oligotrophic to eutrophic state in six reservoir ecosystems ($B_aR$, $Y_yR$, $J_yR$, $G_pR$, $Y_dR$, and $M_sR$), during 2008~2010. The species was selected as a sentinel species for the study, due to its wide distribution and wide trophic gradient. The analysis of trophic state index (TSI), based on total phosphorus (TP) and chlorophyll-a (Chl-a), indicated that reservoirs of $Y_yR$ and $B_aR$ were classified as to be in an oligotrophic state (30~40), the $J_yR$ and $G_pR$ as mesotrophic (40~50), and the $Y_dR$ and $M_sR$ as eutrophic state (50~70). Total 47 species and 26,226 individuals were sampled from 6 reservoirs and sensitive species dominated in the oligotrophic reservoirs ($Y_yR$ and $B_aR$). In the mean time, the tolerant speciesdominated the community in the mesotrophic ($J_yR$ and $G_pR$) and eutrophic ($Y_dR$ and $M_sR$) reservoirs. Regression analysis of body weight, against the total length, indicated that the regression coefficient (b value) was lower in the oligotrophic reservoir (2.77~2.79) than the mesotrophic (3.07~3.17) and eutrophic reservoirs (3.15~ 3.21). This result suggests that the population growth rate Zacco platypus reflected the trophic gradients of the reservoirs. The analysis of condition factor (K) against the total length showed positive slopes (b>3.0) in mesotrophic and eutrophic reservoirs, and a negative slope (b<3.0) in oligotrophic reservoir. The variation of the regression slope of "b" in Z. platypus was accounted for 79.7% [$b=0.012{\times}TSI(TP)+2.395$, p=0.017] by the variation of TSI (TP) and 82.2% [$b=0.013{\times}TSI(Chl-a)+2.36$, p=0.013] by the variation of TSI (Chl-a). The proportion of DELT abnormality increased as the trophic state increases in the reservoirs. The overall data suggest that the growth of the fish populations, based on the length-weight relations and condition factor, reflected the trophic state of nutrient and phytoplankton biomass of the reservoir waters. Thus, in spite of the tolerant characteristics of Z. platypus, hypertrophic states might negatively affect the health of the population.

Analysis of Influenza Virus Isolates in Seoul during 2003-2004 Season (2003-2004 절기 서울지역의 인플루엔자 바이러스 분리 및 아형 분석)

  • Influenza is an important public health problem which occurs almost every winter in temperate climates and is often associated with increased rates of hospitalization and death. In 1999, our influenza surveillance was initiated with 4 voluntary sentinel physicians and the Public Health Center. During the 2003-2004 influenza season, 124 influenza viruses were isolated from 401 clinical specimens, which were collected from patients with Influenza-like illness(ILI) in Seoul. The case definition of ILI is a case with fever more than $38^{\circ}C$ and systemic symptoms; cough, or sore throat. ILI was the highest at the 20-49 age $group(23\%)$ and the rate of virus isolation was the highest at the 7-19 age $group(50\%)$. Among 124 influenza viruses, isolates 83 were identified as A/H3N2 type and others were subtyped as influenza B viruses in 2003-2004 season. Influenza viruses were collected $39.1\%$ at Nowon-Gu, $13.5\%$ Gangnam-Gu and Seocho-Gu etc. and the isolate rate of virus had the area difference; Yongsan-Gu $66.7\%$, Gangnam-gu $50.0\%$, Nowon-Gu $39.9\%$, Kangbuk-Gu $36.8\%$, Seocho-Gu $27.8\%$, Dongjak-Gu $21.2\%$. Out of 401 individuals, 160 was vaccinated $(40\%)$ and the vaccination rate was the highest at the 20-49 age $group(32\%)$. These findings may contribute to the recommondation of the influenza vaccine formulation and the development of influenza control measure.

Serological evidence of Anaplasma spp., Borrelia burgdorferi and Ehrlichia canis in dogs from the Republic of Korea by rapid diagnostic test kits

  • Background: Emergent and re-emergent canine tick-borne infections are attracting increasing attention worldwide. The rise in pet ownership and the close relationship between dogs and their owners are the most concerning factors because dogs may act as competent reservoirs for human tick-transmitted infectious agents. Objectives: This study contributes to the epidemiological surveillance of canine tick-transmitted infections with zoonotic risk in the Republic of Korea (ROK) by investigating the seroprevalence of the pathogens, Anaplasma spp., Borrelia burgdorferi, and Ehrlichia canis. Methods: Four hundred and thirty whole blood samples from domestic dogs were collected in seven metropolitan cities and nine provinces in the ROK and tested using SensPERT Ab test kits (VetAll Laboratories®) to detect seroreactive animals. Results: The seroprevalence rates identified were 9.8% (42/430) for Anaplasma spp., 2.8% (12/430) for B. burgdorferi, and 1.4% (6/430) for E. canis. The risk factors evaluated in this study that could be associated with the development of a humoral immune response, such as sex, age, and history of tick exposure, were similar. There was only one exception for dogs seroreactive to Anaplasma spp., where the risk factor "tick exposure" was statistically significant (p = 0.047). Conclusions: This serological survey exhibited the widespread presence of Anaplasma spp., B. burgdorferi, and E. canis throughout the ROK. Hence, dogs may play a key role as the sentinel animals of multiple zoonotic infectious agents in the country.

Epidemiological Characteristics of Influenza in Children during the 2017-2018 and 2018-2019 Influenza Seasons in Jeju, Korea (2017-2018, 2018-2019 절기 제주 지역 소아 인플루엔자의 역학적 특성)

  • Purpose: In Korea, seasonal influenza is an important respiratory illness afflicting children every year. We aimed to investigate the childhood epidemiology in Jeju during the 2017-2018 and 2018-2019 seasons. Methods: Children aged <13 years, who were tested for influenza at the Jeju National University Hospital during the 2017-2018 and 2018-2019 influenza seasons, were included. Demographics and the influenza test results were retrospectively reviewed from their medical records. Results: This study included 5,219 cases of influenza-like illness (ILI) (2017-2018: n=2,279; 2018-2019: n=2,940). The mean age of the eligible children was 2.85±2.79 years, and the most common age among ILI patients in each season was 1 year group. There were 902 (17.3%, 902/5,219) confirmed influenza cases during the 2 seasons. The rate of influenza confirmed by rapid influenza diagnostic test or polymerase chain reaction among ILI patients in the 2017-2018 and 2018-2019 seasons was 10.4% (236/2,279) and 10.3% (303/2,940) for influenza A, and 9.1% (208/2,279) and 5.3% (155/2,940) for influenza B, respectively. The mean age of influenza-confirmed cases was 4.09 years and 5.05 years in the 2017-2018 and 2018-2019 seasons, respectively (P<0.05). Weekly distribution of influenza was similar to that of ILI in the clinical sentinel surveillance system in both seasons. Conclusion: The difference in the influenza epidemic trend and age-group distribution between the 2017-2018 and 2018-2019 seasons was distinct in Jeju. Steady epidemiological studies on influenza in Jeju are needed for comparison with other regions of Korea.

Physico-chemical Characteristics and In situ Fish Enclosure Bioassays on Wastewater Outflow in Abandoned Mine Watershed (폐광산 지역의 유출수에 대한 이.화학적 수질특성 및 Enclosure 어류 노출시험 평가)

  • The objectives of this study were to evaluate the physico-chemical water quality, trophic and tolerance guilds in the control ($C_o$) and impacted streams of the abandoned mine, along with the ecological health, using a multimetric health model and physical habitat conditions of Qualitative Habitat Evaluation Index (QHEI), during the period of three years, 2005~2007. Also, eco-toxicity ($EE_t$) enclosure tests were conducted to examine the toxic effects on the outflows from the mine wastewater, using the sentinel species of Rhynchocypris oxycephalus, and we compared the biological responses of the control ($C_o$) and treatment (T) to the effluents through a Necropybased Health Assessment Index ($N_b$-HAI). Tissue impact analysis of the spleen, kidney, gill, liver, eyes, and fins were conducted in the controlled enclosure experiments (10 individuals). According to the comparisons of the control ($C_o$) vs. the treatment (T) in physicochemical water quality, outflows from the abandoned mine resulted in low pH of 3.2, strong acid wastewater, high ionic concentrations, based on an electrical conductivity, and high total dissolved solid (TDS). Physical habitat assessments, based on Qualitative Habitat Evaluation Index (QHEI) did not show any statistical differences (p>0.05) in the sampling sites, whereas, the $M_m$-EH model values in a multimetric ecological health ($M_m$-EH) model of the Index of Biological Integrity (IBI), using fish assemblages, were 16~20 (fair condition) in the control and all zero (0, poor condition) in the impacted sites of mine wastewater. In addition, in enclosure eco-toxicity ($EE_t$) tests, the model values of $N_b$-HAI ranged between 0 and 3 in the controls during the three years, indicating an excellent~good condition (Ex~G), and were >100 (range: 100~137) in the impacted sites, which indicates a poor condition (P). Under the circumstances, organ tissues, such as the liver, kidney, and gills were largely impaired, so that efficient water quality managements are required in the outflow area of the abandoned mine watershed.

Role of PKR and EGR-1 in Induction of Interleukin-S by Type B Trichothecene Mycotoxin Deoxynivalenol in the Human Intestinal Epithelial Cells (B형 트리코테센 곰팡이 독소 데옥시니발레놀에 의한 인체 장관 상피세포 염증성 인터루킨 8유도에서의 PKR과 EGR-1의 상호 역할 규명)

  • Mucosal epithelia sense external stress signals and transmit them to the intracellular cascade responses. Ribotoxic stress-producing chemicals such as deoxynivalenol (DON) or other trichothecene mycotoxins have been linked with gastrointestinal inflammatory diseases by Fusarium-contamination. The purpose of this study was to test the hypothesis that DON evokes the epithelial sentinel signals of RNA-dependent protein kinase (PKR) and early growth response gene 1 (EGR-1), which together contribute to the pro-inflammatory cytokine interleukin 8 (IL-8) in human intestinal epithelial cells. PKR suppression by the dominant negative PKR expression attenuated DON-stimulated interleukin-8 production. Moreover, 1L-8 transcriptional activation by DON was also reduced by PKR inhibition in the human intestinal epithelial cells. Treatment with the PKR inhibitor also suppressed EGR-1 promoter activity, mRNA and protein induction, although mitogen-activated protein (MAP) kinases such as extracellular signal-regulated protein kinases (ERK) 1/2, p38, c-Jun N-terminal Kinase (INK) were little affected or even enhanced in presence of a PKR inhibitor. These patterns were also compared in the EGR-1-suppressed cells, which showed much more suppressed production of 1L-8. All things taken into consideration, DON-activated sentinel signals of EGR-1 via PKR mediated interleukin-8 production in human intestinal epithelial cells, which provide insight into the possible general mechanism associated with mucosal inflammation as an intestinal toxic insult by ribotoxic trichothecene mycotoxins.

Feasibility Assessment of Spectral Band Adjustment Factor of KOMPSAT-3 for Agriculture Remote Sensing (농업관측을 위한 KOMPSAT-3 위성의 Spectral Band Adjustment Factor 적용성 평가)

  • As the number of multispectral satellites increases, it is expected that it will be possible to acquire and use images for periodically. However, there is a problem of data discrepancy due to different overpass time, period and spatial resolution. In particular, the difference in band bandwidths became different reflectance even for images taken at the same time and affect uncertainty in the analysis of vegetation activity such as vegetation index. The purpose of this study is to estimate the band adjustment factor according to the difference of bandwidth with other multispectral satellites for the application of KOMPSAT-3 satellite in agriculture field. The Spectral band adjustment factor (SBAF) were calculated using the hyperspectral satellite images acquired in the desert area. As a result of applying SBAF to the main crop area, the vegetation index showed a high agreement rate of relative percentage difference within 3% except for the Hapcheon area where the zenith angle was 25. For the estimation of SBAF, this study used only one set of images, which did not consider season and solar zenith angle of SBAF variation. Therefore, long-term analysis is necessary to solve SBAF uncertainty in the future.
